A pilot study of nonmyeloablative allogeneic hematopoietic stem cell transplant for advanced systemic mastocytosis.

Systemic mastocytosis (SM) is a disease characterized by tissue infiltration of neoplastic mast cells originating from hematopoietic stem cells. Patients with advanced SM have a poor prognosis, and there is no mast cell ablative therapy available for most patients who carry an activating point mutation in the c-kit gene. We report results of a prospective study evaluating the safety, engraftment, and possibility of inducing a graft-versus-mast cell (GvMC) effect after allogeneic nonmyeloablative hematopoietic cell transplantation (HCT) from an HLA-identical sibling. Three patients with advance

Histogenesis of symmetrical 1,2-dimethylhydrazine-induced neoplasms of the colon in the mouse.

CF-1 female adult mice were given weekly sc injections of 20 mg symmetrical 1,2-dimethylhydrazine (DMH)-2HCl/kg body weight and killed at various intervals after commencement of the injection. [3H]thymidine (TdR) was given before the animals were killed. The histogenesis of colon neoplasms was investigated by means of autoradiographs prepared from sections of Epon-embedded descending colon, which were stained with periodic acid-Schiff reaction and iron hematoxylin. By 9 weeks after initiation of DMH treatment, the distal 5 cm of the colon became enlarged, the mucosa thickened, and the crypts w

lncRNA EBLN3P Promotes Proliferation, Metastasis and Stemness of Gastric Cancer Cells via miR-141-3p/HMGCS1.

Long non-coding RNAs (lncRNAs) have become an essential factor in gastric cancer (GC) initiation and metastasis. This study is aimed to uncover the functional significance of EBLN3P in GC. In GC, EBLN3P and HMGCS1 was significantly increased. In contrast, miR-141-3p was decreased. Localization assay revealed that EBLN3P primarily localized in the cytoplasm rather than the nucleus. In MKN28 and SGC7901 cells, EBLN3P knockdown suppressed cell proliferation, metastasis and stemness. On the contrary, overexpression of EBLN3P facilitated these cellular processes. The EBLN3P/miR-141-3p/HMGCS1 axis w

Subependymal Giant Cell Astrocytoma Non-Associated With Tuberous Sclerosis Complex and Expression of OCT-4 and INI-1: A Case Report.

Subependymal giant cell astrocytoma (SEGA) is a rare, slow-growing tumor with a dual (neuroglial) component that is typically associated with tuberous sclerosis complex (TSC). We present the case of a healthy 19-year-old man with mild occipital trauma followed by two weeks of intense headache, with no response to analgesics. Imaging studies revealed a well-defined tumor in the left paraventricular zone. A biopsy showed a SEGA (GFAP+, NF+, nestin+, CK-EA3/EA4+, and TTF1+). TSC was ruled out. An immunohistochemistry (IHC) panel showed aberrant cytoplasmic expression of octamer-binding transcript

Mastocytosis and fibrosis: role of cytokines.

Mastocytosis is a rare stem cell disorder characterized by abnormal growth and accumulation of mast cells in one or more organ systems. Clinical heterogeneity is a hallmark of mastocytosis. Recent observations of activating mutations in c-kit may help to understand the abnormal growth of mast cells in mastocytosis. However, this mutation alone does not explain the entire clinical heterogeneity of the disease. Reticulin fibrosis is also commonly associated with systemic mastocytosis. Mast cells are known to be the source of fibrogenic cytokines, including platelet-derived growth factor, transfo

Stem cell transplantation for ischemic stroke.

BACKGROUND: Studies in animal models of ischemic stroke have shown that stem cells transplanted into the brain can lead to functional improvement. However, to date, evidence for the benefits of stem cell transplantation in ischemic stroke patients is lacking. OBJECTIVES: To assess the efficacy and safety of stem cell transplantation compared with conventional treatments in patients with ischemic stroke. SEARCH STRATEGY: We searched the Cochrane Stroke Group Trials Register (last searched February 2010), the Cochrane Central Register of Controlled Trials (CENTRAL) (The Cochrane Library 2009,

Thymoquinone decreases cell proliferation and immune evasion of breast cancer cells by reducing CD55 and CD114 levels.

Controlling the proliferation and immune evasion of cancer cells can lead to effective strategies for cancer treatment. Thymoquinone, a bioactive compound derived from Nigella sativa exhibits a potent anti-tumor activity. It can be used as a therapeutic approach because it stabilizes the G-quadruplex structure in the promoter regions of oncogenes. This study aims to investigate the effects of thymoquinone on CD55, an inhibitor of the complement system, and CD114, which is involved in cancer cell proliferation. Real-time PCR and Western blot were conducted to verify the expression of CD55 and C

Decreased CD117 expression in hypopigmented mycosis fungoides correlates with hypomelanosis: lessons learned from vitiligo.

Hypopigmented mycosis fungoides is an uncommon clinical variant of cutaneous T-cell lymphoma. We hypothesized that hypomelanosis in hypopigmented mycosis fungoides may have a similar mechanism as in vitiligo, a condition in which it is believed that alterations in expression of CD117 (stem cell factor receptor/KIT protein) on epidermal melanocytes and abnormal interactions between melanocytes and surrounding keratinocytes may play a pathogenic role. To test the hypothesis that similar mechanisms might also explain hypopigmentation in hypopigmented mycosis fungoides, skin specimens from five ca

Differential phthalocyanine photosensitization of acute myeloblastic leukaemia progenitor cells: a potential purging technique for autologous bone marrow transplantation.

The potential value of sulphonated aluminium phthalocyanine (AISPc) as a purging agent for bone marrow autografts in acute myeloblastic leukaemia (AML) has been studied using in vitro clonogenic assays for normal (GM-CFC) and leukaemic (AML-CFC) progenitor cells. In nine out of 13 cases, the leukaemic blasts were found to be highly sensitive to AISPc. In six of the sensitive cases clonogenic assays revealed that only 2 +/- 1% of AML progenitor cells survived AISPc treatment under conditions which permitted a GM-CFC recovery of 60 +/- 11%. AISPc photosensitization was also shown to selectively

JAK2 Negative Polycythemia Vera.

Polycythemia vera (PV) is a stem cell disorder, characterized as a panhyperplastic, malignant, and neoplastic marrow disorder. Several reasons suggest that a mutation on the Janus kinase-2 gene (JAK2) is the most probable candidate gene involved in PV pathogenesis, as JAK2 is directly involved in intracellular signaling, following its exposure to cytokines, to which PV progenitor cells display hypersensitivity. A recurrent unique acquired clonal mutation in JAK2 was found in most patients with PV and other myeloproliferative diseases (MPDs). A female patient of age 50 years, presented with hem

The contribution of endothelial activation and injury to end-organ toxicity following allogeneic hematopoietic stem cell transplantation.

Over the last 25 years, allogeneic hematopoietic stem cell transplantation (HSCT) has been used increasingly as a curative treatment option for patients with hematologic and neoplastic diseases. Despite major advances in transplant immunology and improvements in supportive and critical care medicine, HSCT is still plagued by several life-threatening complications. As such, the establishment of effective therapeutic options for these complications will be crucial as increasing numbers of high-risk transplants are performed each year. This brief review will discuss the contribution of vascular e

Erythrophagocytic acute lymphocytic leukemia with B-cell markers and with a 20q- chromosome abnormality.

An erythrophagocytic neoplastic infiltration of the bone marrow which resembled malignant histiocytosis was found in an elderly man who, when initially examined, had fever and cytopenias. Results of cytochemical and immunologic studies were consistent with an acute lymphocytic leukemia in which the lymphoblasts showed the simultaneous expression of lymphoid stem cell and B-cell markers. Chromosome analysis revealed an abnormal clone with a deletion of part of the long arm of chromosome 20. This case illustrates (1) the occurrence of striking erythrophagocytosis by lymphoblasts at the time of i

Melanoma cancer stem-like cells: Optimization method for culture, enrichment and maintenance.

Malignant Melanoma is known for being one of the most aggressive cancers with an incidence that increases every year. Cancer stem cells (CSCs) are involved in the resistance to therapeutic treatments, the metastatic process, and the patient's relapses. Thus, it is of vital importance for researchers to find the methodology that allows us to obtain enriched subpopulations that maintain their stem-like properties without differentiating over time. In the present manuscript, our objective was to compare the ability of conditioned medium obtained from human mesenchymal stem cells (MSCs) isolated b

Overexpression of the truncated form of High Mobility Group A proteins (HMGA2) in human myometrial cells induces leiomyoma-like tissue formation.

The pathogenesis of uterine leiomyomas, the most common benign tumor in women, is still unknown. This lack of basic knowledge limits the development of novel non-invasive therapies. Our group has previously demonstrated that leiomyoma side population (SP) cells are present in tumor lesions and act like putative tumor-initiating stem cells in human leiomyoma. Moreover, accumulated evidence demonstrates that these benign tumors of mesenchymal origin are characterized by rearrangements of the High Mobility Group A proteins (HMGA). In this work, we tested the hypothesis that leiomyoma development

Hormonal signaling in glioblastoma: emerging roles in stemness and tumor plasticity.

Glioblastoma (GBM) is characterized by pronounced cell-state plasticity and functional heterogeneity, which drive therapeutic resistance, tumor recurrence, and disease evolution. Although these processes have traditionally been viewed through a predominantly genomic perspective, emerging evidence identifies endocrine signaling as a critical regulator of glioblastoma cell states. In this review, we propose that hormones function not as independent signaling pathways or merely as promoters of tumor growth, but as systemic regulators that integrate physiological and microenvironmental signals to

Effects of Combined Exposure to Carbon Ions and Protons on the Pool of MCF-7 Breast Cancer Stem Cells In Vitro.

We studied the effects of single and combined action of protons and carbon ions 12C6+ on the pool of MCF-7 human breast cancer stem cells. Single irradiation with a beam of protons or carbon ions had no significant effects on the relative number of cancer stem cells (CSC). The effects of combined irradiation in a total equieffective dose of 4 Gy depended on the sequence of exposure to ionizing radiations: the relative number of CSC did not change after irradiation with carbon ions and then with protons, but increased in the case of the reverse sequence. The most favorable result, i.e. a decrea

Metabolic Heterogeneity of Brain Tumor Cells of Proneural and Mesenchymal Origin.

Brain-tumor-initiating cells (BTICs) of proneural and mesenchymal origin contribute to the highly malignant phenotype of glioblastoma (GB) and resistance to current therapies. BTICs of different subtypes were challenged with oxidative phosphorylation (OXPHOS) inhibition with metformin to assess the differential effects of metabolic intervention on key resistance features. Whereas mesenchymal BTICs varied according to their invasiveness, they were in general more glycolytic and less responsive to metformin. Proneural BTICs were less invasive, catabolized glucose more via the pentose phosphate p

Phenazine derivatives attenuate the stemness of breast cancer cells through triggering ferroptosis.

Breast cancer stem cells (BCSCs) are positively correlated with the metastasis, chemoresistance, and recurrence of breast cancer. However, there are still no drugs targeting BCSCs in clinical using for breast cancer treatment. Here, we tried to screen out small-molecule compounds targeting BCSCs from the phenazine library established by us before. We focused on the compounds without affecting cell viability and screened out three potential compounds (CPUL119, CPUL129, CPUL149) that can significantly attenuate the stemness of breast cancer cells, as evident by the decrease of stemness marker ex

Identification of natural products and FDA-approved drugs for targeting cancer stem cell (CSC) propagation.

Here, we report the identification of key compounds that effectively inhibit the anchorage-independent growth and propagation of cancer stem cells (CSCs), as determined via screening using MCF7 cells, a human breast adenocarcinoma cell line. More specifically, we employed the mammosphere assay as an experimental format, which involves the generation of 3D spheroid cultures, using low-attachment plates. These positive hit compounds can be divided into 5 categories: 1) dietary supplements (quercetin and glucosamine); 2) FDA-approved drugs (carvedilol and ciprofloxacin); 3) natural products (aloe
