Neuro-osteology.

Neuro-osteology stresses the biological connection during development between nerve and hard tissues. It is a perspective that has developed since associations were first described between pre-natal peripheral nerve tissue and initial osseous bone formation in the craniofacial skeleton (Kjaer, 1990a). In this review, the normal connection between the central nervous system and the axial skeleton and between the peripheral nervous system and jaw formation are first discussed. The early central nervous system (the neural tube) and the axial skeleton from the lumbosacral region to the sella turci

Application of the three-dimensional maximum mode in prenatal diagnosis of Apert syndrome.

Apert syndrome is a rare disorder characterized by coronal craniosynostosis, syndactyly, brachycephaly, midfacial hypoplasia, and central nervous system anomalies, among other malformations. We present a case of Apert syndrome examined at 22 + 0 weeks' gestation. Three-dimensional maximum mode was decisive for the correct prenatal diagnosis by demonstrating the cranial deformities.

[Congenital malformations in live-born neonates].

A retrospective study on the incidence and prevalence of congenital malformations in living newborns registered during a period of two years (1987-1988) at the University Hospital "Dr. José Eleuterio González" in Monterrey, Nuevo León was conducted. The data was tabulated based on the classification codes established by the World Health Organization (WHO), grouped according to organs and systems and by sex. Of a total of 9,675 living newborns registered, 224 (2.31%) had a congenital malformation: 102 males, 121 females and one of undetermined sex. The greatest incidence corresponded to the cen

Multiple pregnancies achieved with IVF/ICSI and risk of specific congenital malformations: a meta-analysis of cohort studies.

Studies comparing risk of specific congenital malformations (CM) between multiple pregnancies resulting from IVF/intracytoplasmic sperm injection (ICSI) and those conceived naturally report conflicting results; furthermore, there is a lack of a complete overview. This meta-analysis aimed to address which types of CM are increased in IVF/ICSI multiple pregnancies compared with those conceived naturally. All studies testing the association between IVF/ICSI multiple pregnancies and specific CM identified in various databases were considered. The literature search yielded 856 records, of which 21

[Citalopram taken during pregnancy and the child born with Hirschsprung's disease].

A woman treated with citalopram during the entirety of her pregnancy bore a child with Hirschsprung's disease. Theories on the development of the enteric nervous system support a possible negative effect of SSRI usage. Epidemiological studies confirm a correlation between pregnant women's use of SSRIs and congenital malformations of their children's digestive system, but not specifically Hirschsprung's disease. Certain limitations in these studies might explain this lack of specificity.

A Neonate with CLOVES Syndrome.

Congenital lipomatous overgrowth, vascular malformations, and epidermal nevi (CLOVE) syndrome is a recently delineated disorder that comprises vascular malformations (typically truncal), dysregulated adipose tissue, scoliosis, enlarged bony structures (typically of the legs) without progression, or distorting bony overgrowth. The name CLOVE was subsequently extended to CLOVES to emphasize the association with scoliosis/skeletal and spinal anomalies and seizures/central nervous system malformations. We herein report a very rare case of CLOVES syndrome with the findings of lipomatous overgrowth …

A unifying hypothesis for hydrocephalus, Chiari malformation, syringomyelia, anencephaly and spina bifida.

This work is a modified version of the Casey Holter Memorial prize essay presented to the Society for Research into Hydrocephalus and Spina Bifida, June 29th 2007, Heidelberg, Germany. It describes the origin and consequences of the Chiari malformation, and proposes that hydrocephalus is caused by inadequate central nervous system (CNS) venous drainage. A new hypothesis regarding the pathogenesis, anencephaly and spina bifida is described.Any volume increase in the central nervous system can increase venous pressure. This occurs because veins are compressible and a CNS volume increase may resu

The DNase TREX1 is a substrate of the intramembrane protease SPP with implications for disease pathogenesis.

Signal peptide peptidase (SPP) is an ER-resident aspartyl intramembrane protease cleaving proteins within type II-oriented transmembrane segments. Here, we identified the tail-anchored protein Three prime repair exonuclease 1 (TREX1) as a novel substrate of SPP. Based on its DNase activity, TREX1 removes cytosolic DNA acting as a negative regulator of the DNA-sensing cGAS/STING pathway. TREX1 loss-of-function variants cause Aicardi-Goutières syndrome (AGS), a type I interferonopathy. Cleavage of ER-bound TREX1 by SPP releases a cleavage product into the cytosol. Proteolysis depends on sequence

Syndromology: an updated conceptual overview. VIII. Deformations and disruptions.

A deformation is an abnormal form or position of the body caused by nondisruptive mechanical forces. Clubfoot, congenital hip dislocation, and infantile mandibular asymmetry (not based on mandibular malformation) serve as examples. The musculoskeletal system is involved in almost all deformations. Various disturbances of intrauterine movement are the usual causes and commonly occur during the fetal period. Causes may be mechanical or malformational. The former includes first pregnancies, small maternal size, uterine malformations and tumors, and amnionic rupture leading to oligohydramnios. Mal

Hypervitaminosis A induced teratogenesis.

In the past decade, the toxicology of reproduction has become increasingly important. This branch of toxicology focuses on mutagenic and embryotoxic effects. The study of embryotoxicity requires an extensive knowledge of the interaction of drugs and embryonic tissues, normal and abnormal developmental processes, and the susceptible stages during prenatal development. Hypervitaminosis A is one of the most extensively studied teratogens. It produces defects in almost all organ systems. Therefore, this article will first of all review the vitamin A-induced malformations in several organ systems.

[Megacolon in adults--the spectrum of underlying intestinal innervation disorders].

The association of megacolon in adults and Hirschsprung's disease was reevaluated by the morphological assessment of the enteric nervous system. Whole-mount preparations of the resected colonic segments and an immunohistochemical treatment with the pan-neuronal marker protein gene product 9.5 allowed an optimal visualization of the entire intramural nervous plexus layers. The findings included different forms of intestinal neuronal malformations (hypoganglionosis, neuronal intestinal dysplasia, and heterotopic ganglia) apart from classic aganglionosis, thus indicating their etiologic relevance

Causes and consequences of 'delays' in the diagnosis of spinal dural arterio-venous fistula; An illustrative case series, review and commentary.

BACKGROUND: Delays in management and misdiagnosis are common in patients with spinal dural arteriovenous fistula (sdAVF). In this study we review the incidence of delays, where they occur in the diagnostic process, who is responsible and what are the consequences. METHODS: We report three recent cases that demonstrate the diagnostic challenges of sdAVF. We utilise, and extend, recent systematic and scoping reviews on sdAVF to obtain recent data on time to diagnosis and treatment. We propose a hierarchical flow-chart to document the diagnostic pathway. Using t his template where diagnostic err
