Metal-Mediated Nitrogen Doping of Carbon Supports Boosts Hydrogen Production from Ammonia.

Ammonia is an attractive hydrogen carrier, yet its practical use is limited by the need for efficient catalytic decomposition. We demonstrate that in-situ N-doping of Ru nanoparticles and graphitized carbon nanofiber supports during reaction produces a sharp increase in hydrogen production during the first 40 h, followed by stable activity. Spectroscopic and microscopic analyses, together with density functional theory simulations, reveal that Ru nitridation is rapid and support-independent, resulting in a mechanistic shift from the traditional Langmuir-Hinshelwood to a Mars-van Krevelen pathw

Denitrification Capacity of a Natural and a Restored Marsh in the Northern Gulf of Mexico.

Anthropogenic pressures, such as diking, construction of dams, and oil spills negatively impact coastal marshes creating growing pressure to preserve and to restore salt marshes due to their critical role in permanently removing nitrate runoff through denitrification as well as other ecosystem services they provide. This study determined denitrification rates across a typical northern Gulf of Mexico salt marsh landscape that included a natural marsh, a tidal creek, and a 21-year-old restored salt marsh. Denitrification capacity, measured with the isotope pairing technique on a membrane inlet m

Cysteine-SILAC Mass Spectrometry Enabling the Identification and Quantitation of Scrambled Interchain Disulfide Bonds: Preservation of Native Heavy-Light Chain Pairing in Bispecific IgGs Generated by Controlled Fab-arm Exchange.

Bispecific antibodies (bsAbs) are one of the most versatile and promising pharmaceutical innovations for countering heterogeneous and refractory disease by virtue of their ability to bind two distinct antigens. One critical quality attribute of bsAb formation requiring investigation is the potential randomization of cognate heavy (H) chain/light (L) chain pairing, which could occur to a varying extent dependent on bsAb format and the production platform. To assess the content of such HL-chain swapped reaction products with high sensitivity, we developed cysteine-stable isotope labeling using a

SP3-Enabled Rapid and High Coverage Chemoproteomic Identification of Cell-State-Dependent Redox-Sensitive Cysteines.

Proteinaceous cysteine residues act as privileged sensors of oxidative stress. As reactive oxygen and nitrogen species have been implicated in numerous pathophysiological processes, deciphering which cysteines are sensitive to oxidative modification and the specific nature of these modifications is essential to understanding protein and cellular function in health and disease. While established mass spectrometry-based proteomic platforms have improved our understanding of the redox proteome, the widespread adoption of these methods is often hindered by complex sample preparation workflows, pro

Trophodynamics and health risk assessment of toxic trace metals in the food web of a plateau freshwater lake.

The trophodynamics of toxic trace metals is significant for assessing the threat of toxic trace metals to the aquatic ecosystem and human safety. However, due to the difficulty of accurately calculating the trophic positions of freshwater aquatic organisms in the food web, the comprehensive process of trophodynamics of toxic trace metals in freshwater ecosystems was still rarely known. By integrating the compound-specific nitrogen stable isotopic analysis of amino acids (CSIA-AAs) and the Bayesian stable isotope mixing model (SIMM) as a novel approach, the present study investigated the tropho

An ecologically framed mercury survey of finfish of the lower Chesapeake Bay.

Total mercury (THg) and methylmercury (MHg) concentrations and determents of mercury (Hg) accumulation were examined for muscle tissues of 10 finfish from the lower Chesapeake Bay (LCB) and its tributaries. There was no suggestion of potential human harm from Hg due to LCB fish consumption: None of the sampled fish had THg concentrations approaching the United States Environmental Protection Agency human health screening value. Hg concentrations in different fish species generally increased with the increasing stable isotope of nitrogen 15 (δ(15)N) but not the stable isotope of carbon 13 (δ(13

Legacy and emerging contaminants in common guillemot Uria aalge eggs in Ireland and Wales.

Guillemot eggs from multiple Irish colonies and one Welsh colony were analysed for legacy pollutants such as polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers (PBDEs) and other organochlorine compounds (OCs), as well as metals. Stable isotope ratios of carbon (δ13C) and nitrogen (δ15N) were measured in eggs to understand the influence of diet on contaminant levels detected. Wide-scope target and suspect screening techniques were used on a single guillemot egg, providing novel information on contaminants of emerging concern. Stable isotope ratio analysis showed that guillemots fr

Prenatal effects of maternal nutritional stress and mental health on the fetal movement profile.

PURPOSE: Prenatal sub-optimal nutrition and exposure to maternal stress, anxiety and depression in pregnancy have been linked to increased postnatal morbidity and mortality. Fetal growth is most vulnerable to maternal dietary deficiencies, such as those evident in hyperemesis gravidarum (HG), early in pregnancy. The purpose of this pilot study was to examine the effects of HG on fetal movement profiles as a measure of fetal healthy development in the 3rd trimester of pregnancy, and to assess whether nutritional stress on the mother can be evaluated using isotopic analysis of hair. METHOD: We

Quantification of 3-nitrotyrosine levels using a benchtop ion trap mass spectrometry method.

Oxidative damage by reactive nitrogen species is linked to the pathogenesis of numerous inflammatory disorders, including atherosclerosis. 3-Nitrotyrosine (NO2Tyr), a posttranslational modification of proteins generated by reactive nitrogen species, serves as a "molecular fingerprint" for protein modification by nitric oxide (NO)-derived oxidants. Studies demonstrate that systemic levels of protein-bound NO2Tyr serve as an independent predictor of cardiovascular risks and are modulated by statin therapy. Measurement of NO2Tyr in biological matrices may thus serve both as a quantitative index o

Nitrogen use efficiency in crops under salt stress: from molecular networks to intelligent breeding.

Soil salinization threatens global arable land and agricultural sustainability, severely reducing crop nitrogen use efficiency (NUE) by disrupting root ammonium and nitrate fluxes, impairing nitrogen-assimilation enzymes, and disrupting carbon-nitrogen (C-N) balance. This review synthesizes recent advances in the coordination of salt-stress signaling and nitrogen homeostasis in plants. Two mechanistically distinct regulatory axes have recently been proposed. In one, a nitrate transporter acts as a dual sensor for nitrate and abscisic acid (ABA); in the other, SOS kinase-mediated phosphorylatio

Anthropogenic nutrient discharge and microbial community shifts associated with typhoon-driven runoff in an artificial coastal lake.

This study investigated changes in nutrient sources and microbial community composition across a typhoon-associated runoff period in Lake Sihwa, an anthropogenically influenced artificial coastal lake. Stable isotope analyses (δ13CPOC, δ15NPN, δ15NNO₃, and δ18ONO₃) and water-column environmental DNA (eDNA) metabarcoding were combined to characterize nutrient sources and compare ecological conditions between the pre- and post-typhoon surveys. Before the typhoon-associated rainfall period, nitrate inputs were primarily associated with urban runoff, whereas particulate organic matter (POM) reflec

Influence of food sources and trace elements in the acquisition of antimicrobial resistance in Escherichia coli isolated from white stork nestlings (Ciconia ciconia).

Foraging in landfills enhances the probability of acquiring antimicrobial resistance (AMR) in wildlife and increases exposure to pollutants like metal(loid)s, pharmaceuticals and caffeine. Exposure to metal(loid)s in the environment may cause selective pressure on bacteria, inducing metal resistance genes that drive antimicrobial resistance genes (ARGs) through co-resistance and cross-resistance mechanisms. Hence, white storks fed in landfills could increase AMR acquisition through the combined effect of urban-waste and pollutants. Using a novel approach combining stable isotopes, trace elemen

Modeling Life History Parameters in Marine Mammals: Bayesian Hierarchical Analysis of Ontogenetic Dietary Shifts in Indo-Pacific Humpback Dolphins.

AbstractWeaning represents a pivotal ontogenetic process for mammals, marking the transition from parental provisioning to independent foraging. In monophyodont species, distinct growth layer groups that are deposited in their teeth as the animals age represent a permanent chronological archive of physiological events across the animals' lifetimes. Thus, biochemical analysis of annual dentine increments provides a means to explore animal physiological history. We examined the age-specific pattern, individual-level variations, and sex-related differences in dentine nitrogen isotopic values in t

[Genome-wide identification and stress response analysis of sugarcane glutamine synthetase gene family].

Glutamine synthetases (GSs) are central enzymes in plant nitrogen assimilation, governing nitrogen uptake, transport, and remobilization. This study systematically identified the GS gene family members in sugarcane (Saccharum spontaneum), analyze the sequence features and spatiotemporal expression profiles of SsGSs, and mined candidate genes functionally linked to high nitrogen use efficiency (NUE), aiming to furnish gene resources for enhancing NUE of sugarcane through molecular breeding. We employed S. spontaneum and its elite hybrid progeny as model materials. First, nitrogen source prefere

Root-Derived Endophytic Diazotrophic Bacteria Pantoea cypripedii AF1 and Kosakonia arachidis EF1 Promote Nitrogen Assimilation and Growth in Sugarcane.

Excessive, long-term application of chemical fertilizers in sugarcane crops disrupts soil microbial flora and causes environmental pollution and yield decline. The role of endophytic bacteria in improving crop production is now well-documented. In this study, we have isolated and identified several endophytic bacterial strains from the root tissues of five sugarcane species. Among them, eleven Gram-negative isolates were selected and screened for plant growth-promoting characteristics, i.e., production of siderophores, indole-3-acetic acid (IAA), ammonia, hydrogen cyanide (HCN), and hydrolytic

Fish eye lens chronologies reveal reservoir-driven changes in carbon biogeochemistry and mercury bioaccumulation.

Reservoir impoundments alter river carbon (C) biogeochemistry as submerged terrestrial material is decomposed, which enhances mercury (Hg) methylation. Methylmercury (MeHg) is a neurotoxin that bioaccumulates in aquatic organisms and biomagnifies along food webs, exposing wildlife and human consumers. Reservoir characteristics (ie., water residence time and land flooded area) regulate C and Hg dynamics, yet their temporal dynamics remain poorly resolved. Fish eye lenses grow incrementally and preserve fine-scale temporal records of Hg bioaccumulation and C biogeochemistry. Lenses from the Roma

A guide for an anatomically sensitive dentine microsampling and age-alignment approach for human teeth isotopic sequences.

OBJECTIVES: Stable isotope analysis of sequential dentine samples is a potentially powerful method to reveal insights into early life-histories of individuals in the past. Dentine incremental growth structures are complex, however, and current approaches that apply horizontal sectioning of demineralized tooth halves or quarters risk combining multiple growth layers and may include unwanted cementum or secondary dentine. They also require destruction of large parts of a tooth. Here, we present a less destructive and relatively straightforward protocol that reduces damage, increases temporal res

15N, 13C and 1H resonance assignments of FKBP12 proteins from the pathogenic fungi Mucor circinelloides and Aspergillus fumigatus.

Invasive fungal infections are a leading cause of death in immunocompromised patients and remain difficult to treat since fungal pathogens, like mammals, are eukaryotes and share many orthologous proteins. As a result, current antifungal drugs have limited clinical value, are sometimes toxic, can adversely affect human reaction pathways and are increasingly ineffective due to emerging resistance. One potential antifungal drug, FK506, establishes a ternary complex between the phosphatase, calcineurin, and the 12-kDa peptidyl-prolyl isomerase FK506-binding protein, FKBP12. It has been well estab

A novel strategy for the targeted analysis of protein and peptide metabolites.

In many biological applications such as epitope discovery or drug metabolism studies, the detection of naturally processed exogenous proteins (e.g. vaccines or peptide therapeutics) and their metabolites is frequently complicated by the presence of a complex endogenous mixture of closely related or even identical compounds. We describe a method that incorporates stable isotope labelling of the protein of interest, allowing the selective screening of the intact molecule and all metabolites using a modified precursor ion scan. This method involves monitoring the low-molecular-weight fragment ion

An objective means of diagnosing anorexia nervosa and bulimia nervosa using 15N/14N and 13C/12C ratios in hair.

An objective means based on the carbon and nitrogen stable isotope analysis of five hairs per individual is presented for distinguishing between individuals with anorexia nervosa and/or bulimia nervosa from non-clinical individuals (i.e. clinically normal controls). Using discriminant analysis, an algorithm has been developed that provides both sensitivity and specificity of 80% in making diagnoses of individuals with these eating disorders. With further refinements, the results suggest that it may be also possible to distinguish between individuals with anorexia or bulimia. Finally, the study
