Tooth bioengineering leads the next generation of dentistry.

BACKGROUND: As a result of numerous rapid and exciting developments in tissue engineering technology, scientists are able to regenerate a fully functional tooth in animal models, from a bioengineered tooth germ. Advances in technology, together with our understanding of the mechanisms of tooth development and studies dealing with dentally derived stem cells, have led to significant progress in the field of tooth regeneration. AIM AND DESIGN: This review focuses on some of the recent advances in tooth bioengineering technology, the signalling pathways in tooth development, and in dental stem c

Epithelial histogenesis during tooth development.

This paper reviews the current understanding of the progressive changes mediating dental epithelial histogenesis as a basis for future collaborative studies. Tooth development involves morphogenesis, epithelial histogenesis and cell differentiation. The consecutive morphological stages of lamina, bud, cap and bell are also characterized by changes in epithelial histogenesis. Differential cell proliferation rates, apoptosis, and alterations in adhesion and shape lead to the positioning of groups of cells with different functions. During tooth histo-morphogenesis changes occur in basement membra

Phylogenetic memory of developing mammalian dentition.

Structures suppressed during evolution can be retraced due to atavisms and vestiges. Atavism is an exceptional emergence of an ancestral form in a living individual. In contrast, ancestral vestige regularly occurs in all members of an actual species. We surveyed data about the vestigial and atavistic teeth in mammals, updated them by recent findings in mouse and human embryos, and discussed their ontogenetic and evolutionary implications. In the mouse incisor and diastema regions, dental placodes are transiently distinct being morphologically similar to the early tooth primordia in reptiles. T

Effect of Down syndrome on the dimensions of dental crowns and tissues.

Abnormal growth in Down syndrome (DS) is reflected by variable reduction in size and simplification in form of many physical traits. This study aimed to compare the thickness of enamel and dentine in deciduous and permanent mandibular incisor teeth between DS and non-DS individuals and to clarify how these tissues contribute to altered tooth size in DS. Sample groups comprised 61 mandibular incisors (29 permanent and 32 deciduous) from DS individuals and 55 mandibular incisors (29 permanent and 26 deciduous) from non-DS individuals. Maximum mesiodistal and labiolingual crown dimensions were me

Epithelial-mesenchymal interface in the rat mandibular process observed by electron microscopy.

Epithelial-mesenchymal interfaces in the stomodeal side of the rat mandibular processes were observed at Theiler's stages 14 and 15, when no epithelial organogenesis could be detected by electron microscopy. At stage 14, the basal lamina was discontinuous. Disintegrated basal lamina-like materials were seen in the epithelial-mesenchymal interface. A number of coated vesicles and smooth vesicles were seen in epithelial cell processes adjacent to the disintegrated basal lamina-like materials. Some epithelial cell processes penetrated into the mesenchymal space through the basal lamina and touche

The biosynthesis of dentin phosphophoryns by rat incisor odontoblasts in organ culture.

An in vitro system consisting of rat incisor fragments was used to study the process of dentinogenesis. In order to establish the usefulness of the organ culture, the biosynthesis and deposition of the major noncollagenous components of dentin, the phosphophoryns, were followed for specific lengths of time in culture. Three criteria were satisfied: (1) the synthesis of proteins which appeared to be chemically identical to the native proteins of dentin, (2) the accumulation of the phosphophoryns within the matrix or time, and (3) the association of the secreted proteins with the mineral phase o

Sexual dimorphism in the development, emergence, and agenesis of the mandibular third molar.

Bilateral agenesis was encountered in about 9% of the cases, with no significant sexual difference. The right and left mandibular third molars had the same pattern of development and emergence. The slight advance of girls over boys at the crown-completion stage was similar to previous observations on other mandibular teeth, particularly the second molar. The root development course of the third molar was faster in males than in females; this sexual dimorphism was much greater for retarded cases than for advanced cases. At the apex closure, the difference between median ages of males and female

Hepatocyte Growth Factor Promotes Differentiation Potential and Stress Response of Human Stem Cells from Apical Papilla.

Harsh local microenvironment, such as hypoxia and lack of instructive clues for transplanted stem cells, presents the serious obstacle for stem cell therapies' efficacy. Therefore, continued efforts have been taken to improve stem cells' viability and plasticity. Hepatocyte growth factor (HGF) has previously been reported to mitigate the complications of various human diseases in animal model studies and in some clinical trials. Besides, human stem cells from the root apical papilla (SCAP) are deemed a better resource of mesenchymal stem cells due to derived stem cells holding greater amplific

Description of Tooth Ontogeny and Replacement Patterns in a Juvenile Tarbosaurus bataar (Dinosauria: Theropoda) Using CT-Scan Data.

Teeth are continually replaced in most of non-mammalian gnathostomes to maintain their functional dentitions. To clarify the tooth replacement patterns in tyrannosaurid theropod dinosaurs, we examined well-preserved dentitions (both premaxillae, left maxilla, partial right maxilla, and both dentaries) of a juvenile Tarbosaurus bataar (MPC-D 107/7) using X-ray computed tomographic (CT) imaging. Three-dimensional (3D) rendering of the dentitions and staging of replacement teeth allowed quantitative analyses of the tooth ontogeny and replacement patterns in this specimen. These strategies were va

Establishment of a congenital tooth agenesis related gene MSX1 knockout human embryonic stem cell lines by CRISPR-Cas9 technology.

Human MSX1 gene is mapped to chromosome 4 and encodes a 303aa homeobox protein MSX1. MSX1 expression appears during early tooth development of vertebrate embryogenesis. Mutations in this protein are related to human tooth anomalie, cleft lip and palate and congenital ectodermal dysplasia syndrome. Most of the confirmed pathogenic mutations are located in exon2 encoded homeobox domain. Here, we report the establishment of MSX1 gene knockout human embryonic stem (hES) cell lines by CRISPR-Cas9 technology. These cell lines provide good materials for further studies of the roles MSX1 plays in huma

Cell-to-cell communication--periodontal regeneration.

BACKGROUND: Although regenerative treatment options are available, periodontal regeneration is still regarded as insufficient and unpredictable. AIM: This review article provides scientific background information on the animated 3D film Cell-to-Cell Communication - Periodontal Regeneration. RESULTS: Periodontal regeneration is understood as a recapitulation of embryonic mechanisms. Therefore, a thorough understanding of cellular and molecular mechanisms regulating normal tooth root development is imperative to improve existing and develop new periodontal regenerative therapies. However, comp

Dental maturation in children with the syndrome of crouzon and apert.

Purpose : Developing teeth are used to assess maturity and estimate age in a number of disciplines. The purpose of this investigation was to study the dental maturation in children with Crouzon or Apert syndrome compared with nonsyndromic controls. Patients and Methods : Records of 40 children with Crouzon syndrome (18 boys and 22 girls, aged 4.0 to 17.9 years) and 28 children with Apert syndrome (10 boys and 18 girls, aged 3.9 to 15.1 years) were referred to the Department of Orthodontics, Cleft Palate Team and Craniofacial Team, Erasmus MC-Sophia. Data from syndromic children were compared w
