Regional distribution of protein kinases in normal and odor-deprived mouse olfactory bulbs.

Unilateral naris closure produced dramatic down-regulation of tyrosine hydroxylase (TH) gene expression in periglomerular dopaminergic neurons in the olfactory bulb. To explore molecular mechanisms of TH gene regulation, the present study investigated the regional distribution of protein kinase A (PKAalpha), protein kinase C (PKCalpha), and CaM kinases II (CaMKIIalpha, beta) and IV (CaMKIV) in the normal olfactory bulb and in response to odor deprivation. Strong PKAalpha immunostaining was found in the glomerular, granule cell, external plexiform and olfactory nerve layers. PKCalpha staining w

Activity-dependent regulation of dopamine content in the olfactory bulbs of naris-occluded rats.

Several lines of evidence strongly suggest that reduced olfactory nerve activity results in decreased bulb dopamine content. In the present study, high performance liquid chromatography with electrochemical detection was used to assess catecholamine levels in bulbs from postnatal day 60 rats that had undergone either unilateral naris cautery or a sham surgery on day 30. Thirty days of odor deprivation dramatically reduced dopamine and dihydroxyphenylacetic acid levels in functionally-deprived bulbs (ipsilateral to occluded nares) as compared to contralateral controls, while norepinephrine and

Nitric oxide synthase, cGMP, and NO-mediated cGMP production in the olfactory bulb of the rat.

High levels of nitric oxide synthase and cyclic 3',5'-guanosine monophosphate (cGMP) in the olfactory bulb suggest that nitric oxide, acting as a diffusible intercellular messenger molecule inducing increased synthesis of cGMP, plays an important role in olfaction. The localization of cGMP after sodium nitroprusside stimulation of in vitro slices of rat olfactory bulb was compared with the distribution of nicotinamide adenine dinucleotide phosphatediaphorase, nitric oxide synthase, and glial fibrillary acidic protein. cGMP was detected immunohistochemically in cryostat sections. In the presenc

The olfactory bulb: a critical site of action for oxytocin in the induction of maternal behaviour in the rat.

Expanding on research showing that oxytocin originating in the hypothalamic paraventricular nucleus acts to decrease olfactory processing at the level of the olfactory bulb, we explored the importance of oxytocin acting on the olfactory bulb for the onset of maternal behaviour in Wistar rats. Experiment I was designed to test whether spontaneous maternal behaviour following natural delivery is blocked by bilateral infusions of a low dose (5 fmol) of the oxytocin antagonist d(CH2)5[Tyr(Me)2,Thr4,Tyr-NH2(9)]ornithine-vasotocin into the olfactory bulb immediately after the delivery of the first p

Presynaptic metabotropic glutamate receptors in adult and developing neurons: autoexcitation in the olfactory bulb.

The mitral cell of the olfactory bulb is the primary relay neuron that transmits information from the olfactory receptors to the rest of the brain. This excitatory neuron releases glutamate from presynaptic dendrites and axon terminals. All rat mitral cells studied showed strong, selective, and widespread metabotropic glutamate receptor mGluR1 alpha immunoreactivity on the presynaptic membrane of dendrites, often at the synaptic vesicle release site, when examined with light and electron microscopy. The finding of glutamate receptors on mitral cell secondary dendrites supports the conclusion t

Effects of parturition and maternal experience on noradrenaline and acetylcholine release in the olfactory bulb of sheep.

Acetylcholine (ACh) and noradrenaline (NA) release in the olfactory bulb (OB) of ewes was monitored using microdialysis. Both ACh and NA release increased at parturition in multiparous but not in primiparous ewes. However, vaginocervical stimulation performed 6 hr postpartum induced an increase of ACh and NA release in both primiparous and multiparous ewes, indicating that a maturation process had occurred. Finally, pharmacological challenges to the ACh and NA inputs revealed differential responsiveness between nulliparous and multiparous nongestant ewes. These results suggest that the first p

Developmental expression of neural cell adhesion molecules in the mouse neocortex and olfactory bulb.

Polyclonal antibodies to N-CAM and L1 and monoclonal antibodies to epitopes of N-CAM (designated 12F11, 8A2, and 12F8) were used to investigate the spatial and temporal distribution of these neural cell adhesion molecules during the development of mouse cortex and olfactory bulb. The aim of the study was to correlate developmental events such as cell migration, dendritic and axonal outgrowth, and synaptogenesis with the appearance and disappearance of specific molecules involved in cell-cell interactions. Western transfer studies indicated that 12F8 antibody recognized polysialic acid found on

GABAergic mechanisms are involved in the control of tuberoinfundibular arcuate neurons by the accessory olfactory bulb.

In this study we examined electrophysiologically the involvement of the intrinsic GABAergic system of the accessory olfactory bulb (AOB) in controlling the activity of tuberoinfundibular (TI) arcuate neurons in anaesthetized female mice. Local infusions of the gamma-aminobutyric acid-A (GABAA) receptor antagonist, bicuculline into the AOB enhanced the spontaneous firing activity of TI arcuate neurons with excitatory inputs from the AOB. This finding reveals a neural mechanism responsible for the pregnancy blocking effect of this drug in freely behaving female mice and, taken together with the

Projections of two subclasses of vomeronasal nerve fibers to the accessory olfactory bulb in the rabbit.

The organization of the projections of subclasses of vomeronasal nerve fibers to the accessory olfactory bulb was analysed using monoclonal antibodies generated against a homogenate of the rabbit olfactory bulb. Monoclonal antibody R2D5 labels all the somata of vomeronasal receptor cells in the vomeronasal organ as well as all their axons (vomeronasal nerve fibers). Another monoclonal antibody (R4B12), which has been shown to selectively bind and thus identify a subclass of olfactory nerve fibers, also labels a subclass of vomeronasal nerve fibers. The R4B12-positive subclass of vomeronasal ne

[Spatio-temporal study of the electrical activities of the olfactory bulb of the trout Salmo gairdneri].

The electrical stimulation of the olfactory nerve allows to record steady field potentials in the olfactory bulb of the trout Salmo gairdneri. 200 microns separate recording points which are localised on a tridimensional network. At 18, 24, 30 and 38 ms the amplitudes are measured and plotted in a calculator which determines the isopotentials in any vertical, horizontal and sagittal planes. With this method, we have an instantaneous and global vision of bulbar activity and we observe striking anatomo-functional correlations in the olfactory bulb of Salmo gairdneri.

Immunohistochemical demonstration of serotonin nerve fibers in the olfactory bulb of the rat, cat and monkey.

The distribution of serotonin (5-HT) positive fibers in the olfactory bulb of the rat, cat and monkey was studied using the peroxidase-anti-peroxidase (PAP) immunohistochemical method and highly specific antibodies to 5-HT. In general, 5-HT fibers were present throughout all layers in the olfactory bulb of these species except for the olfactory nerve layer and different as well as restricted laminar patterns of 5-HT distribution were observed. There were also species-related differences in the pattern of 5-HT distribution, in each layer. The most notable species difference was apparent in the

[Activation of afferent pathways and of control frog olfactory bulb and extracellular field potentials].

It has been recorded the extracellular field potentials of the frog's olfactory bulb produced by electrical stimulation of the commissura anterior and small branches of the olfactory nerve. The activation of the commissural system obtain a positive, long-lasting field potential generated at the soma of the granular cells. The olfactory nerve stimulation produce also a positive surface wave bath at the end of bulbar reply: this effect can be attributed to the activation of the bulbar component of commissural system at the NOA. The recognizable electrical effects of the commissural system allow

Effects of nicotine and tobacco smoke on the electrical activity of the cerebral cortex and olfactory bulb.

1. Effects of nicotine, cigarette smoke and carbon monoxide have been compared in the cat encéphale isolé preparation, exhibiting a synchronized electrocorticogram (ECoG) and behavioural sleep.2. 2 ml samples of smoke, containing approximately 7 mug nicotine (approximately 2 mug nicotine/kg for a 3 kg cat), introduced into the lungs at 30 s intervals from a smoking simulator, caused desynchronization of the ECoG and behavioural arousal.3. Effects of smoke were matched in the same experiments by intravenous injections of nicotine, 2 mug/kg every 30 s.4. The use of specific nicotine antagonists,

Selective retrograde transport of tritiated D-aspartate from the olfactory bulb to the anterior olfactory nucleus, pyriform cortex and nucleus of the lateral olfactory tract in the rat.

After an injection of [3H]D-aspartate into the olfactory bulb of the rat, retrogradely labeled cells were detected bilaterally in the anterior olfactory nucleus (AON), and ipsilaterally in the pyriform cortex (PC) and nucleus of the lateral olfactory tract (NLOT). These results suggest a certain selective retrograde transport of this amino acid, and are discussed in relation to transmitter candidates in the olfactory bulb.

Case report of Parkinson's disease in isolated congenital anosmia with absent olfactory bulbs.

Olfactory dysfunction is an early, prodromal feature of Parkinson's disease (PD), often preceding motor symptoms by years. This has fueled the hypothesis that PD pathology may originate in the olfactory bulb (OB), where early Lewy-type α-synucleinopathy is frequently observed and from which pathology could propagate to neural systems. In this context, Arshamian et al. (2022) hypothesized that isolated congenital anosmia (ICA), a rare lifelong absence of smell typically associated with bilateral OB aplasia, might confer immunity to PD. They argued that identifying a single individual with both

The human olfactory bulb in Huntington's disease: sex-specific differences in volume and gliosis.

Huntington's disease is a multisystemic neurodegenerative disorder for which increasing evidence suggests early involvement of brain regions beyond the primarily affected motor areas of the brain, such as the olfactory bulb, which is affected by multiple neurodegenerative diseases. However, the involvement of the olfactory bulb in Huntington's disease and the possible effects of sex on olfactory bulb characteristics in Huntington's disease patients remain poorly understood. In this study, volumetric and glial alterations in the olfactory bulbs of male and female Huntington's disease patients w
