Olfactory relationships of the telencephalon and diencephalon in the rabbit. II. An autoradiographic and horseradish peroxidase study of the efferent connections of the anterior olfactory nucleus.

The efferent connections of the anterior olfactory nucleus in the female albino rabbit have been studied using the autoradiographic and horseradish peroxidase methods for tracing axonal pathways. Following a unilateral injection of 3H-leucine into the olfactory peduncle, radioactively labeled efferent projections from the anterior olfactory nucleus were traced into all layers of the ipsilateral main olfactory bulb beneath the olfactory nerve layer and through the ipsilateral anterior limb of the anterior commissure and plexiform layer of the medial side of the cerebral hemisphere to the deep h

Modulating salience network connectivity through olfactory nerve stimulation.

Depression is associated with reduced functional connectivity within the brain's salience network and its strengthened interactions with the default mode network (DMN). Modification of this clinical pattern is challenging. Leveraging the direct neural pathways from olfactory processing regions to the salience network, we explored the effects of electrical stimulation of the olfactory mucosa on brain connectivity. In a randomized, blinded within-subject design, 45 healthy individuals received olfactory or trigeminal nerve stimulation followed by resting-state fMRI. Olfactory stimulation resulte

Differential targeting of olfactory epithelium and respiratory epithelium in nose-to-brain drug delivery.

INTRODUCTION: Nose-to-brain drug delivery provides a promising noninvasive route to bypass the blood-brain barrier through direct nasal cavity-brain connections. Therapeutic agents reach the central nervous system via systemic circulation or olfactory/trigeminal nerve pathways. Only the olfactory epithelium enables direct brain transport through olfactory neurons, bypassing the blood-brain barrier, while the respiratory epithelium primarily supports systemic absorption before CNS access via trigeminal nerves. AREAS COVERED: This review examines anatomical and functional differences between ol

Gray matter alteration in isolated congenital anosmia patient: a voxel-based morphometry study.

Decreased volume of gray matter (GM) was observed in olfactory loss in patients with neurodegenerative disorder. However, GM volume has not yet been investigated in isolated congenital anosmia (ICA) people. We herewith investigated the volume change of gray matter of an ICA boy by morphometric analysis of magnetic resonance images (voxel-based morphometry), and compared with that of 20 age-matched healthy controls. ICA boy presented a significant decrease in GM volume in the orbitofrontal cortex, anterior cingulate cortex, middle cingulate cortex, thalamus, insular cortex, cerebellum, precuneu

Expanded endoscopic endonasal resection of an olfactory schwannoma.

Olfactory schwannomas are rare tumors of the anterior skull base that are possibly derived from ectopic Schwann cells, perivascular neural tissue, or sensory nerves of the meninges. The authors report the case of a 14-year-old boy with an olfactory schwannoma that extended inferiorly through the cranial base and superiorly into the frontal lobe. Because of the growth characteristics of the tumor and the significant overlying frontal lobe edema, the lesion was approached via an endonasal endoscopic route, as a strategy to minimize brain retraction. This tumor was characterized radiographically

Charting plasticity in the regenerating maps of the mammalian olfactory bulb.

The anatomical organization of a neural system can offer a glimpse into its functional logic. The basic premise is that by understanding how something is put together one can figure out how it works. Unfortunately, organization is not always represented purely at an anatomical level and is sometimes best revealed through molecular or functional studies. The mammalian olfactory system exhibits organizational features at all these levels including 1) anatomically distinct structural layers in the olfactory bulb, 2) molecular maps based upon odorant receptor expression, and 3) functional local ci

Olfactory interneurons in the brain of the larval sphinx moth Manduca sexta.

1. The physiology and morphology of olfactory interneurons in the brain of larval Manduca sexta were studied using intracellular recording and staining techniques. Antennal olfactory receptors were stimulated with volatile substances from plants and with pure odorants. Neurons responding to the stimuli were investigated further to reveal their response specificities, dose-response characteristics, and morphology. 2. We found no evidence of specific 'labeled-lines' among the odor-responsive interneurons, as none responded exclusively to one plant odor or pure odorant; most olfactory interneuron

[Electric stimulation of the human olfactory nerve--an approach to short-term memory?].

Electrical stimulation of the human olfactory mucosa was performed by means of an electrode, which was attached to a rhinoscope. Stimulations of the nasal mucosa did not evoke the sensation of smell, but suppressed smell sensations of presented odorants. When electrical stimulation followed the exposure to an odorant within a certain interval, the stimulus recalled the faded sensation of the preceding odorant. Electrical stimulation without prior natural stimulation produced unpleasant sensations in three patients with a history of temporal lobe seizures and olfactory auras, but not in patient

An electrophysiological study of the accessory olfactory bulb in the rabbit--I. Analysis of electrically evoked potential fields.

Following electrical stimulation of the vomeronasal nerves, the primary olfactory nerves, the lateral olfactory tract and the corticomedial amygdala, we have made a study of evoked potentials in the rabbit accessory olfactory bulb. Vomeronasal nerve stimulation evoked a complex field potential consisting of a compound action potential followed by 4 negative waves (N1, N2, N3, N4). In contrast to the field potential elicited in the main olfactory bulb following primary olfactory nerve stimulation, there was either no evoked wave or only a weak positive component of the field in the accessory bu

Time-course study of genetic changes in periodontal ligament regeneration after tooth replantation in a mouse model.

This research focused on analyzing gene expression changes in the periodontal ligament (PDL) after tooth re-plantation to identify key genes and pathways involved in healing and regeneration. Utilizing a mouse model, mRNA was extracted from the PDL at various intervals post-replantation for RNA sequencing analysis, spanning from 3 to 56 days. The results revealed significant shifts in gene expression, particularly notable on day 28, supported by hierarchical clustering and principal component analysis. Gene ontology (GO) enrichment analysis highlighted an upregulation in olfactory receptor and

Human Olfactory Receptors: Novel Cellular Functions Outside of the Nose.

Olfactory receptors (ORs) are not exclusively expressed in the olfactory sensory neurons; they are also observed outside of the olfactory system in all other human tissues tested to date, including the testis, lung, intestine, skin, heart, and blood. Within these tissues, certain ORs have been determined to be exclusively expressed in only one tissue, whereas other ORs are more widely distributed in many different tissues throughout the human body. For most of the ectopically expressed ORs, limited data are available for their functional roles. They have been shown to be involved in the modula

Primary olfactory fibres project to the ventral telencephalon and preoptic region in trout (Salmo trutta): a developmental immunocytochemical study.

We studied the development of the primary olfactory system of a teleost, the brown trout, with the aims of clarifying whether the caudal projection pertains to the olfactory or to the terminal nerve system, of identifying the brain regions receiving this projection, and of investigating its possible functional significance. As olfactory markers (OMs) we used two polyclonal antibodies (to substance P and to alpha-melanocyte-stimulating hormone) that were found to label the olfactory projection strongly after preadsortion of the antibody with the corresponding antigen (OMs), and as a terminal ne

Thalamocortical mechanisms in odor-guided behavior. II. Effects of lesions of the mediodorsal thalamic nucleus and frontal cortex on odor preferences and sexual behavior in the hamster.

The role of the thalamocortical "olfactory" pathway in odor preferences and sexual behavior was examined in the hamster, a species which depends crucially upon olfaction for mating. Before and after ablation of primary or secondary olfactory structures, male hamsters were tested for threshold, interest, and preference for the odor of an estrus female hamster and other odors. Sexual competency was also observed in daily mating sessions. Disruption of the primary olfactory pathway resulted in an absence of interest in odors and mating. Lesions of the mediodorsal thalamic nucleus or frontal neoco

Neuronal architecture of the mosquito deutocerebrum.

Mosquito behavior is heavily dependent on olfactory and mechanosensory cues, which are detected by receptor neurons on the antenna and on the palps. Recent progress in mosquito sensory genomics highlights the need for an up-to-date understanding of the neural architecture of the mosquito brain. Here we present a detailed description of the neural structure of the primary target of the majority of these neurons, the deutocerebrum, in the African malaria (Anopheles gambiae) and yellow fever (Aedes aegypti) mosquitoes. Special focus is made on the olfactory system, the antennal lobe (AL), where w

Olfactory structures in staged human embryos.

The olfactory region was investigated in 303 serially sectioned human embryos, 23 of which were controlled by precise graphic reconstructions. The following findings in the embryonic period are new for the human. (1) The nasal plates arise at the neurosomatic junction, as do also the otic placodes. (2) Crest comes from the nasal plates later (stage 13) than the maximum production in the neural folds (stage 10). (3) The crest arises and migrates during a much longer time (at least until the end of the embryonic period) than the neural crest of the head, where origin and migration end at stage 1

The vomeronasal organ of the cat.

The vomeronasal organ of the cat was studied macroscopically, by light microscopy and by immunohistochemical techniques. Special attention was paid to the general distribution of the various soft tissue components of this organ (duct, glands, connective tissue, blood vessels and nerves.) Examination of series of transverse sections showed that the wall of the vomeronasal duct bears 44 different types of epithelium: simple columnar in the caudal part of the duct, respiratory and receptor respectively on the lateral and medial walls of the middle part of the duct, and stratified squamous rostral

[Olfactory disorders in Alzheimer's disease and in Parkinson's disease. Review of the literature].

Olfactory disorders in Alzheimer's disease and Parkinson's disease have been the topic of a large body of work over the last decades. Work devoted to olfactory disorders in Alzheimer's disease includes over 300 papers providing clinical and fundamental data. Anatomy studies in Alzheimer's disease have demonstrated a specific concentration of lesions in peripheral and central olfactory structures (senile plaques, neurofibrillary degeneration) as well as lesions in layers II and III of the entorhinal cortex. These neuropathological findings led to the development of the hypothesis that olfactory

[Comparison of the effects of anosmia induced by either peripheral lesion or bulbectomy upon the feeding pattern of the rat (author's transl)].

In order to support the contention that the feeding pattern seen after olfactory bulb removal is due to a sensory loss, the feeding pattern … smia. Thus, the disruption of the feeding pattern after bulbectomy and after lesions of the central olfactory pathways is clearly the result of anosmia and not of the loss of other non-sensory functions of the olfactory bulbs.

Structural anomalies of the peripheral olfactory system in psychosis high-risk subjects.

BACKGROUND: Olfactory impairments are prominent in both schizophrenia and the preceding at-risk state. Their presence prior to illness predicts poor functional outcome. In schizophrenia, these impairments reflect peripheral olfactory structural abnormalities, which are hypothesized to arise during early embryonic development. If this is correct, then similar structural anomalies should be apparent among clinical high-risk subjects. METHODS: Thirty-nine clinical high-risk (CR) subjects (24M/15F) were compared to 36 low-risk (LR) subjects (19M/17F). Olfactory measures derived from 3T MRI scans
