Hereditary spherocytosis (HS) is a common variant of inherited hemolytic anemia, in which abnormalities of red blood cells (RBC) structural proteins lead to loss of erythrocyte membrane surface area, resulting in spherical, hyperdense, weakly deformable RBCs. It is characterized by the presence of osmotically fragile erythrocytes, known as spherocytes in peripheral blood smear (PBS). The mutations that cause HS occur in genes encoding for red blood cell membrane/cytoskeletal proteins, predominantly, ankyrin-1, α- and β-spectrins, band-3, and protein 4.2. This results in defective cell membrane

Hereditary Spherocytosis (HS) is a type of hemolytic anemia characterized by a defect in the membrane of red blood cells, which causes them to assume a spherical shape, become osmotically fragile, and be prone to early hemolysis. While it is relatively well understood in the medical field, there is limited evidence regarding the dental management of patients with this condition. Therefore, this report aims to present a case involving the extraction of permanent teeth before splenectomy in a child with HS who had been hospitalized. Additionally, treatment guidelines for these patients are propo

Adducins are a family of cytoskeleton proteins encoded by three genes (alpha, beta, gamma). In a comprehensive assay of gene expression, we show the ubiquitous expression of alpha- and gamma-adducins in contrast to the restricted expression of beta-adducin. beta-adducin is expressed at high levels in brain and hematopoietic tissues (bone marrow in humans, spleen in mice). To elucidate adducin's role in vivo, we created beta-adducin null mice by gene targeting, deleting exons 9-13. A 55-kDa chimeric polypeptide is produced from the first eight exons of beta-adducin and part of the neo cassette

OBJECTIVE: To determine the zeta potential of erythrocyte and its association with morphological changes during preeclampsia so as to use it … icantly less than that of control pregnant volunteers along with structural deformity and increased osmotic fragility of erythrocytes.
CONCLUSIONS: The data suggest that as pregnancy progresses there is increase in ZP values whereas in preeclampsia there is significant decrease in the ZP values below the normal level. This decrease in ZP values can be seen in earl …

We studied the functional and metabolic changes of erythrocytes in systemic vasculitis (SV). The below was detected: an increasing intensity of blood chemiluminescence, a higher concentration of TBA-positive substances in erythrocyte membranes, inhibited Na+, K+ - and Ca2+ -activated and Mg2+ -related ATPases in erythrocytes and a reduced osmotic and acidic resistance of erythrocytes. An enhanced aggregation ability of erythrocytes and their inhibited desagregation were observed in hemorrhagic vasculitis an especially in microscopic polyangiitis. The revealed changes characterize the significa

The clinical suspicion of hereditary spherocytosis (HS) must be confirmed at the clinical laboratory. The osmotic fragility test (OFT) and the autohemolysis test (AHT) are the worldwide accepted assays to establish a definite diagnosis of HS; however, they have some disadvantages. We describe herein our experience with the cryohemolysis test (CHT) as a tool to confirm the HS diagnosis. We included four groups of subjects, namely, patients with clinical HS, patients with mechanical heart valve prosthesis, malignant hematological diseases and healthy blood donors. CHT was carried out in all the …

We conclude that both cellular and membrane thiols bind beta-93 sulfhydryls of CHBHA hemoglobins as mixed disulfides. Concomitantly, heme avidity to beta-92 lessens, suggesting that degradation of the resulting excessively freed …

BACKGROUND: Transfusion of blood at the limits of approved storage time is associated with lower red blood cell (RBC) post-transfusion recovery and hemolysis, which increases plasma cell-free hemoglobin and iron, proposed to induce endothelial dysfunction and impair host defense. There is noted variability among donors in the intrinsic rate of storage changes and RBC post-transfusion recovery, yet genetic determinants that modulate this process are unclear.
METHODS: We explore RBC storage stability and post-transfusion recovery in murine models of allogeneic and xenogeneic transfusion using b

BACKGROUND: Hereditary spherocytosis is autosomal dominant inherited extravascular hemolytic disorder and is the commonest cause of inherited hemolysis in northern Europe and the United States. The classical clinical features of hereditary spherocytosis are anemia, jaundice, and splenomegaly. However, all of these classical features are not always revealed in the case of mild hemolysis or when hemolysis is well compensated. Patients with hereditary spherocytosis may remain undiagnosed for years if their hemolysis is mild.

We investigated the resistance of erythrocytes from rat brain venous blood to acid hemolysis in the dynamics of brain ischemic period (15, 30, 45 and 60 min), as well as in the early (5 min) and distant (24h) period of brain reperfusion. Brain ischemia-reperfusion was made in rats that received ecdysterone (standartized extract of Serratula coronata) within 18 days (per os, 1 mg/kg). Analysis of the kinetic curves of acid hemolysis showed a pronounced (60 times, from 1.45 to 85.85% at 60 min of brain ischemia and at 5 min of brain reperfusion, respectively) increase of unstable erythrocytes th

OBJECTIVES: To evaluate the effect of haematological alterations resulting from antiretroviral therapy (ART) on the diagnosis of thalassaemia carriers in HIV-1-infected Thai patients.
METHODS: Complete blood cell counts, osmotic fragility (OF) test and haemoglobin (Hb)-A(2) values were measured in blood samples of 52 antiretroviral-treated and 14 untreated HIV-1-infected patients. Data were analysed according to thalassaemia type and ART.
RESULTS: Sixteen patients carried at least one of the investigated thalassaemia types and most of them (87.5%) received ART. Their red cell indices [mean c…
