This communication reviews most of the important findings related to venom components isolated from scorpions and spiders, mainly by means of gene cloning and expression. Rather than revising results obtained by classical biochemical studies that report structure and function of venom components, here the emphasis is placed on cloning and identification of genes present in the venomous glands of these arachnids. Aspects related to cDNA library construction, specific or random ESTs cloning, transcriptome analysis, high-throughput screening, heterologous expression and folding are briefly discus

Dengue virus, an arbovirus, is the causal factor of thousands of deaths around the globe, although the long-term consequences of the same have not been elucidated in great detail. The virus encodes for a polyprotein, which is later cleaved to form multiple proteins, of which NS1 protein oligomerization domain, 2k, and capsid anchor are three aggregation-prone peptides, as have been previously seen. A relatively under-studied angle of these peptides is the role of these peptides in cross-aggregation with certain human aggregation-prone proteins. Here, we have tried to shed light on this cross-a

Angiotensin-converting enzyme inhibitory peptides (ACEIPs) from natural food sources are effective alternatives for treating hypertension. This study aims to screen highly active ACEIPs from yellow tuna peptides, and to elucidate their inhibitory mechanism, stability, transport mechanism and potential antihypertensive mechanism by a multiple approach. Two novel ACEIPs STHPHF (IC50 = 30.71 μg/mL) and LTFSY (IC50 = 20.72 μg/mL) were screened by bio-activity guided approach and de novo sequencing. Inhibition kinetics and molecular docking indicated that STHPHF demonstrated a mixed inhibitor, wher

The EPICC peptides are a family of peptides that have been developed from the sequence of the capsid protein of human astrovirus type 1 and previously shown to inhibit the classical and lectin pathways of complement. The EPICC peptides have been further optimized to increase aqueous solubility and identify additional mechanisms of action. Our laboratory has developed the lead EPICC molecule, PA-dPEG24 (also known as RLS-0071), which is composed of a 15 amino acid peptide with a C-terminal monodisperse 24-mer PEGylated moiety. RLS-0071 has been demonstrated to possess other mechanisms of action

This article aims to identify the epitope corresponding to SARS-CoV spike protein specific neutralizing monoclonal antibody (MAb) 2C5. The antibody was used as the target and three rounds of bio-panning were conducted with a phage-displayed peptide library. After the third panning, 20 phage-plaque clones were randomly picked and analyzed for the binding ability with the MAb 2C5 by ELISA. The displayed sequence analysis demonstrated that among the 20 phage clones, eight clones displayed the same seven-peptide TPEQQFT. All these eight phage-clones showed strongest binding activity with 2C5 in th

Four monoepitopic MAPs (MAP A, B, C and E) and one bis-diepitopic MAP B-E derived fromthe primary sequence of Schistosoma mansoni glyceraldehyde 3-phosphate dehydrogenase, previously tested in BALB/c mice, were examined for their immunogenicity and protective capacity in C57BL/6 mice. Despite multimerization into MAPs, MAP Aand MAP C were poorly immunogenic. In contrast toBALB/c mice, MAP E was non-immunogenic in C57BL/6 mice. Peptide B in the form of MAP B orbis-diepitopic MAPB-E elicited immune responses in C57BL/6 mice that were associated with a significant decrease in worm burden. The MAP

Metal-peptide superhelices formed from 12/10-helical β-peptide foldamers exhibit programmable hierarchical assembly. A single metal-coordinated superhelix organizes into distinct parallel or antiparallel arrangements without conformational distortion. This adaptability arises from a minimized macrodipole and dynamic folding propensity, establishing general design principles for peptide-based supramolecular materials.

Solid phase peptide synthesis is a method used to generate peptides through the sequential addition of amino acids attached to an immobilized resin. Due to the procedure followed, excess reagents and byproducts are removed while synthesis is ongoing, resulting in a final product that is reproducibly obtained at high purity. Herein, we describe the synthetic procedure of peptides derived from the pleotropic protein macrophage migration inhibitory factor (MIF). Notably, this protocol is valid for the generation of peptides regardless of their sequence and length. Whereas MIF has diverse activiti

Type 1 "blue" copper proteins such as plastocyanin are of interest due to their unusual active site geometries, elevated redox potentials, and rapid electron-transfer rates. Previously, the Cu-coordination properties of small peptide models of the Poplar plastocyanin active site metal binding loop, containing His/Cys/Met putative ligands ('BCPA' peptides), were found to exhibit slight shifts towards the tetragonal distortion supported in the protein active site. Here, several additional Cu-peptide models are explored with spectroscopic and electrochemical measurements and computational methods

Cardiovascular diseases (CVDs) are the leading global cause of death, accounting for more than 17.6 million deaths per year in 2016, a number that is expected to grow to more than 23.6 million by 2030. While many technologies are currently under investigation to improve the therapeutic outcome of CVD complications, only a few medications have been approved. Therefore, new approaches to treat CVD are urgently required. Peptides regulate numerous physiological processes, mainly by binding to specific receptors and inducing a series of signals, neurotransmissions or the release of growth factors.

Background We recently discovered a small endogenous peptide, peptide Lv, with the ability to activate vascular endothelial growth factor receptor 2 and its downstream signaling. As vascular endothelial growth factor through vascular endothelial growth factor receptor 2 contributes to normal development, vasodilation, angiogenesis, and pathogenesis of various diseases, we investigated the role of peptide Lv in vasodilation and developmental and pathological angiogenesis in this study. Methods and Results The endothelial cell proliferation, migration, and 3-dimensional sprouting assays were use

Anti-citrullinated protein antibodies (ACPAs) are a hallmark of rheumatoid arthritis (RA) and represent an important tool for the serological diagnosis of RA. In this study, we describe ACPA reactivity to overlapping citrullinated Epstein-Barr virus nuclear antigen-1 (EBNA-1)-derived peptides and analyze their potential as substrates for ACPA detection by streptavidin capture enzyme-linked immunosorbent assay. Using systematically overlapping peptides, containing a 10 amino acid overlap, labelled with biotin C-terminally or N-terminally, sera from 160 individuals (RA sera (n=60), healthy contr

Rheumatoid arthritis (RA) is one of the most common autoimmune diseases, affecting approximately 1-2% of the world population. One of the characteristic features of RA is the presence of autoantibodies. Especially the highly specific anti-citrullinated peptide antibodies (ACPAs), which have been found in up to 70% of RA patients' sera, have received much attention. Several citrullinated proteins are associated with RA, suggesting that ACPAs may react with different sequence patterns, separating them from traditional antibodies, whose reactivity usually is specific towards a single target. As A

A new cysteine-based disulfide linker for Fmoc solid phase peptide synthesis was developed (Fmoc-Cys(3-mercapto-3-methylbutanoic acid)OPp) that allows the on-resin assembly and side chain deprotection of cyclic peptides. Model peptides and a cyclic peptide library of the structure [a-a-x-x-a-a-c] composed of D-amino acids were assembled and the synthesis and cleavage conditions studied. The best cyclization results were obtained with PyBOP/HOAt/diisopropylethyl amine. Racemization rates of the cysteine in the analyzed model sequences were between 5.2 and 12.3%. Cleavage of the disulfide bond w

A 37-residue peptide has been purified from the endocrine pancreas of the anglerfish. The first 36 residues were sequenced by gas phase Edman degradation. The sequence at the carboxyl-terminus was determined by sequencing the carboxyl-terminal tryptic dipeptide. The sequence of the peptide, which is consistent with the amino acid composition, was determined to be: Y X P X P X K X P X E X T X P X G X S X N X A X S X P X E X D X W X A X S X Y X Q X A X A X V X R X H X Y X V X N X L X I X T X R X Q X R X Y X G. Herbivore elicitors, most of which identified thus far are from the oral secretions (OS) of herbivores, are important in herbivore-induced plant defense. However, whether and how larval OS of the striped stem borer (SSB), Chilo suppressalis, a devastating pest on rice, induce plant defenses remains largely unknown. We found that treating plants with SSB OS enhanced levels of phosphorylated OsMPK3/4/6, JA, JA-Ile, ethylene, and ABA in rice, which subsequently induced the production of defense compounds, such as trypsin protease inhibitors, and decreased the growth of SSB larvae. Digestion by pr

The growing number of antibiotic-resistant bacteria necessitates the search for new antimicrobial agents and the principles by which they work. We report that cell membrane-permeant rhodamine B (RhB)-conjugated peptides based on the phosphatidylinositol-4,5-bisphosphate binding site of gelsolin can kill the gram-negative organisms Escherichia coli and Pseudomonas aeruginosa and the gram-positive organism Streptococcus pneumoniae. RhB linkage to the QRLFQVKGRR sequence in gelsolin was essential for the antibacterial function, since the unconjugated peptide had no effect on the bacteria tested.

A synthetic adjuvant, N-acetylmuramyl-L-alanyl-D-isoglutamine (MDP), failed to produce polyarthritis with a wide dose range in a water-in-oil emulsion of mineral oil such as liquid paraffin, Drakeol, or heavy mineral oil. MDP, however, produced moderate to severe arthritis with almost 100% incidence in a water-in-oil emulsion made up of Difco incomplete adjuvant, which consists of Bavol F as an oil vehicle and Arlacel A as an emulsifier. N-acetylmuramyl-L-alanyl-L-isoglutamine did not produce arthritis, whereas 4,6-diacetyl-MDP produced the disease. Bacterial peptidoglycans, such as disacchari

Leukemia is one of the most prevalent malignancies worldwide that causes the unusual evolution of hematopoietic stem cells. The type of leukemia determines the optimal treatment plan and the patient's survival. However, finding safer and more effective medications and developing novel therapeutic strategies are still the most challenging research topics. Nowadays, over half of the medications used to treat cancer are derived from natural ingredients. Medicinal plants are a reliable natural source of anti-leukemic medications.
