Growth-limiting intracellular metabolites in yeast growing under diverse nutrient limitations.

Microbes tailor their growth rate to nutrient availability. Here, we measured, using liquid chromatography-mass spectrometry, >100 intracellular metabolites in steady-state cultures of Saccharomyces cerevisiae growing at five different rates and in each of five different limiting nutrients. In contrast to gene transcripts, where approximately 25% correlated with growth rate irrespective of the nature of the limiting nutrient, metabolite concentrations were highly sensitive to the limiting nutrient's identity. Nitrogen (ammonium) and carbon (glucose) limitation were characterized by low intrace

The effect of a diet containing 70% protein from plants on mineral metabolism and musculoskeletal health in chronic kidney disease.

BACKGROUND: Chronic Kidney Disease (CKD) is associated with alterations in phosphorus excretion, and increases in fibroblast growth factor (FGF23) and parathyroid hormone (PTH). Plant protein-based phytate-bound phosphorus, is less bioavailable than that from animal sources. Our one-week study that was conducted previously showed that a nearly 100% plant protein-based diet benefits mineral metabolism in CKD; however, this diet may not be acceptable to patients. Here we hypothesize that a diet containing 70% protein from plants has similar efficacy and is tolerated by CKD patients. METHODS: Th

Foliar spraying of salicylic acid and melatonin enhances the nutritional value of Arugula (Eruca vesicaria subsp. sativa mill.).

BACKGROUND: Arugula (Eruca vesicaria subsp. Sativa Mill.) is a leafy vegetable rich in nutritional value, vitamins, minerals, and antioxidants. While previous studies showed that foliar applications of melatonin and salicylic acid influence plant nutrient levels, research on the individual and combined effects of these two compounds in Arugula is Limited. Therefore, this study aims to assess both the individual and combined effects of salicylic acid and melatonin on the concentrations of essential elements, such as nitrogen, phosphorus, potassium, and various micronutrients in Arugula. 31P chemical shift tensors for canonical and non-canonical conformations of nucleic acids: a DFT study and NMR implications.

31P chemical shift anisotropy (CSA) tensors have been calculated for a set of selected DNA and RNA backbone conformations using density functional theory. The set includes canonical A-RNA, A-DNA, BI-DNA, BII-DNA, ZI-DNA, and ZII-DNA as well as four A-RNA-type, seven non-A-RNA-type, and three non-canonical DNA conformations. Hexahydrated dimethyl phosphate has been employed as a model. The 31P chemical shift tensors obtained are discussed in terms of similarities in the behavior observed for gauche-gauche (gg) and gauche-trans (gt) conformations around the P-O bonds. We show that torsion angles

Efficacy of calcium glycerophosphate vs conventional mineral salts for total parenteral nutrition in low-birth-weight infants: a randomized clinical trial.

To test the efficacy of calcium glycerophosphate (CaGlyP) vs the conventional mineral salts, calcium gluconate plus KH2PO4 + K2HPO4 (CaGluc + P), in promoting mineral retention, 72-h mineral balance, biochemical status, net acid excretion, and growth were assessed in 16 low-birth-weight infants receiving total parenteral nutrition (TPN) containing approximately 1.5 mmol Ca and P.kg-1.d-1 for 5 d. Net retentions of calcium (1.2 +/- 0.2 vs 1.0 +/- 0.2 mmol.kg-1.d-1, means +/- SD) and phosphorus (1.1 +/- 0.3 vs 0.8 +/- 0.3 mmol.kg-1.d-1) from CaGluc + P vs CaGlyP, respectively, were similar, as w

Zwitterionic Phosphorus(I) Compounds Supported by a Bis(carbene)borate: Synthesis, Protonation, and Thermodynamics of P-H Bonds.

Carbones are carbon(0) species stabilized by two donor ligands and are strong neutral bases; yet, heavier analogues and the thermodynamics of the E-H bonds of their protonated species remain largely unexplored. We report the synthesis and structural characterization of zwitterionic, two-coordinate phosphorus(I) complexes [(Ph,RBCB)P] 1a (R = tBu) and 1b (R = Me), supported by an anionic bis(carbene)borate ligand. These can be viewed as charge-neutral phosphorus(I) analogues of carbone derivatives. The Brønsted basicity of the P(I) center has been quantified for the first time: the pKa values f

In-situ potassium hydroxide catalysis improves bio-oil composition and bone-char quality during pyrolysis of chicken meat-bone waste.

The poultry industry generates significant amounts of chicken meat-bone waste (MBW). This study analysed the MBW pyrolysis products and evaluated the effects of potassium hydroxide (KOH) as a catalyst. Both untreated and KOH-impregnated MBW were pyrolysed at 400-800 °C. KOH addition increased biochar and gas yields but reduced bio-oil yield, and significantly improved bio-oil quality by reducing the yield of oxygenated and nitrogenous compounds. At 500 °C, KOH addition reduced the production of oxygenated compounds, mostly acids, in bio-oil by 85 % compared to non-catalysed pyrolysis. It also

Rapid Growth and Metabolism of Uropathogenic Escherichia coli in Relation to Urine Composition.

Uropathogenic Escherichia coli (UPEC) strains cause a majority of urinary tract infections (UTIs). Since UPEC strains can become antibiotic resistant, adjunct or alternate therapies are urgently needed. UPEC strains grow extremely rapidly in patients with UTIs. Thus, this review focuses on the relation between urine composition and UPEC growth and metabolism. Compilation of urinary components from two major data sources suggests the presence of sufficient amino acids and carbohydrates as energy sources and abundant phosphorus, sulfur, and nitrogen sources. In a mouse UTI model, mutants lacking

Influence of xylem fluid chemistry on planktonic growth, biofilm formation and aggregation of Xylella fastidiosa.

Xylella fastidiosa is the causal agent of Pierce's disease in grapevines. The mechanisms of pathogenicity are largely due to occlusion of xylem vessels by aggregation of X. fastidiosa and biofilm formation. Xylella fastidiosa was subjected to xylem fluids with varying chemistries to examine the effects of nutritional components on bacterial growth in vitro. The exposure of X. fastidiosa to xylem fluids collected from different Vitis genotypes resulted in highly significant differences in both planktonic growth and biofilm formation. Planktonic growth of X. fastidiosa in Vitis xylem fluid was c

Rh-catalyzed asymmetric hydrogenation of prochiral olefins with a dynamic library of chiral TROPOS phosphorus ligands.

A library of 19 chiral tropos phosphorus ligands, based on a flexible (tropos) biphenol unit and a chiral P-bound alcohol (11 phosphites) or secondary amine (8 phosphoramidites), was synthesized. These ligands were screened, individually and as a combination of two, in the rhodium-catalyzed asymmetric hydrogenation of dehydro-alpha-amino acids, dehydro-beta-amino acids, enamides and dimethyl itaconate. ee values up to 98% were obtained for the dehydro-alpha-amino acids, by using the best combination of ligands, a phosphite [4-P(O)2O] and a phosphoramidite [13-P(O)2N]. Diversity analysis of phytase-producing bacteria from coastal seawater and sediment and characterization of their phytases.

Phytic acid, also known as inositol hexaphosphate (IP6), is one of the most abundant organophosphorus compounds in nature. Its degradation by phytase plays a key role in the natural phosphorus cycle. In addition, phytases are widely used in livestock and poultry feed to enhance phosphorus utilization. While most reported and commercial phytases are derived from terrestrial organisms, relatively few originate from marine microorganisms, and information on the diversity of phytase-producing marine bacteria remains limited. In this study, following enrichment with sodium phytate, we analyzed the

Microbially mediated phosphine emission.

There is still a lot of controversy in literature concerning the question whether a biochemical system exists enabling micro-organisms to reduce phosphate to phosphine gas. The search for so-called 'de novo synthesised' phosphine is complicated by the fact that soils, slurries, sludges, etc., which are often used as inocula, usually contain matrix bound phosphine (MBP). Matrix bound phosphine is a general term used to indicate non-gaseous reduced phosphorus compounds that are transformed into phosphine gas upon reaction with bases or acids. A study was carried out to compare the different dige

Effect of a monensin controlled release capsule on metabolic parameters in transition dairy cows.

A prospective field study involving 251 Holstein cows and heifers from five dairy farms near Guelph, Ontario, Canada, was conducted to measure the effect of monensin delivered in a controlled release capsule 3 wk precalving on metabolic function in dairy cows immediately pre- and postcalving. At 3 wk before expected calving, cows were randomly assigned to receive either a controlled release capsule containing monensin or to serve as negative controls. Cows were blood sampled once per week precalving and once in the week following calving, at the same time of day and the same day of the week. S

Renal function in full-term neonates with hyperbilirubinemia.

The purpose of this study was to investigate the effect of unconjugated hyperbilirubinemia on endogenous creatinine clearance and urinary excretion of sodium, phosphorus, lysozyme, and amino acids in full-term infants. Thirty-seven healthy, breast-fed newborns who were not exposed to phototherapy were studied on their third to fifth day of life. Twenty had neonatal hyperbilirubinemia with a mean indirect bilirubin value of 16.4 mg/dl, whereas 17 who were used as controls had a mean indirect bilirubin value of 7.8 mg/dl. Urine was collected, and samples were taken for examination of creatinine,

Dynorphin A-(1-17) induces alterations in free fatty acids, excitatory amino acids, and motor function through an opiate-receptor-mediated mechanism.

The endogenous opioid dynorphin A-(1-17) (Dyn A) has been implicated as a mediator of tissue damage after traumatic spinal cord injury (TSCI) and causes hindlimb paralysis when administered intrathecally. Motor impairment following intrathecal Dyn A is attenuated by antagonists of excitatory amino acids (EAAs); whether opioid receptors mediate such injury has been questioned. TSCI causes various biochemical changes associated with secondary tissue damage, including alterations in tissue amio acids, phospholipids, and fatty acids. Such changes reflect injury severity and correlate with motor dy

Long-term PFOA and cadmium Co-contamination alters soil carbon, nitrogen, and phosphorus cycling: Insights from metagenomics and metabolomics.

The co-existence of perfluorooctanoic acid (PFOA) and cadmium (Cd) in soil poses a combined threat to microbial communities. However, the ecological effects and underlying mechanisms of their long-term combined exposure remain poorly understood. This study conducted a 90-day soil microcosm experiment to systematically investigate the effects of individual and combined effects of PFOA and Cd on microbial communities. Our results demonstrated that combined pollution of PFOA and Cd significantly affected four soil enzyme activities associated with carbon, nitrogen, and phosphorus cycling. It also

Experience of compounding total parenteral nutrition admixtures for preterm infants in a hospital pharmacy: evidence of calcium and phosphate compatibility problem.

OBJECTIVE: Parenterally fed preterm newborn infants require large amounts of calcium and phosphate in a low volume of solution. The lower the volume of solution, the higher is the possibility of precipitation of calcium hydrogen phosphate (CaHPO4). Precipitation could cause respiratory distress and pulmonary embolism, and the use of organic salts of calcium and phosphorus may reduce the likelihood of this problem. To date, no previous work on the stability of solutions with organic salts has been published in the literature. This study aims to evaluate the visible precipitation of calcium and
