Ubiquitin-Mediated Proteolysis as a Regulator of the Plant Defense-Growth Balance.

To survive in challenging environments, plants must rapidly activate immune responses while maintaining developmental plasticity and reproductive success. This requires continuous negotiation of limited energy and metabolic resources between growth, development, and defense. Ubiquitin-mediated proteolysis has emerged as a versatile regulatory mechanism that may integrate immune responses with plant developmental programs. In this review, we summarize accumulating evidence that ubiquitination shapes immune responses at multiple regulatory levels. Many of these immune-regulatory mechanisms depen

Immunogenic profile of a plant-produced nonavalent African horse sickness viral protein 2 (VP2) vaccine in IFNAR-/- mice.

A safe, highly immunogenic multivalent vaccine to protect against all nine serotypes of African horse sickness virus (AHSV), will revolutionise the AHS vaccine industry in endemic countries and beyond. Plant-produced AHS virus-like particles (VLPs) and soluble viral protein 2 (VP2) vaccine candidates were developed that have the potential to protect against all nine serotypes but can equally well be formulated as mono- and bi-valent formulations for localised outbreaks of specific serotypes. In the first interferon α/β receptor knock-out (IFNAR-/-) mice trial conducted, a nine-serotype (nonava

Prior exposure of Arabidopsis seedlings to mechanical stress heightens jasmonic acid-mediated defense against necrotrophic pathogens.

BACKGROUND: Prolonged mechanical stress (MS) causes thigmomorphogenesis, a stress acclimation response associated with increased disease resistance. What remains unclear is if; 1) plants pre-exposed to a short period of repetitive MS can prime defence responses upon subsequent challenge with necrotrophic pathogens, 2) MS mediates plant immunity via jasmonic acid (JA) signalling, and 3) a short period of repetitive MS can cause long-term changes in gene expression resembling a stress-induced memory. To address these points, 10-days old juvenile Arabidopsis seedlings were mechanically stressed f…

Distinct targeting of 5mC and 6mA DNA methylation during the onset of Flg22-induced immunity between Arabidopsis and rice.

Cytosine methylation drives transcriptional reprogramming during plant innate and acquired immunity. Beyond RNA adenosine methylation, DNA adenine methylation (6mA) has recently emerged as a candidate regulator of gene expression, though its role in plant immunity is unknown. Using the bacterial elicitor Flg22, we examined genome-wide DNA methylation responses of Arabidopsis thaliana and Oryza sativa to identify shared epigenetic responses across divergent genomes. High-resolution Oxford Nanopore sequencing enabled simultaneous detection of 5-methylcytosine (5mC) and 6mA DNA modifications. In …

The miPEP408-miR408 module regulates plant susceptibility to Rhizoctonia solani through LAC3-mediated lignin deposition in Arabidopsis.

Plant microRNAs (miRNAs) function as critical regulators of various developmental and physiological processes by downregulating the expression of their target genes. miRNA-encoded peptides (miPEPs) represent an additional regulatory layer by enhancing the transcription of their cognate miRNAs. . Although miPEP408 has been implicated in heavy-metal tolerance through regulation of sulfur assimilation, its role in plant immunity remains unknown. Here, we investigate the role of the miPEP408-miR408-Laccase3 (LAC3) regulatory cascade in defense against the necrotrophic fungus Rhizoctonia solani AG-…

Single transcription factor SlYABBY5b synchronizes gibberellin-mediated stature and basal immunity in tomato.

Optimizing plant architecture without compromising disease resistance is a fundamental challenge in crop breeding, particularly for space-efficient systems such as vertical farming. Here, we identify the YABBY-family transcription factor SlYABBY5b as a pivotal regulator that synchronizes these traits in tomato. Through a genome-wide association study, we establish SlYABBY5b as a major locus governing natural variation in plant height. Functional analyses reveal that SlYABBY5b directly orchestrates gibberellin (GA) metabolism by activating the inactivation gene SlGA2ox2 and repressing the biosy

The Phytophthora infestans effector Pi05910 suppresses and destabilizes host glycolate oxidase StGOX4 to promote plant susceptibility.

Phytophthora infestans is a notorious oomycete pathogen that causes potato late blight. It secretes numerous effector proteins to manipulate … mmune function. Our results revealed that the core RXLR effector of P. infestans Pi05910 suppresses plant immunity by targeting StGOX4, which results in decreased enzymatic activity and protein accumulation, leading to enhanced plant susceptibility.

Oral immunization of mice using transgenic tomato fruit expressing VP1 protein from enterovirus 71.

Enterovirus 71 (EV71) causes seasonal epidemics of hand-foot-and-mouth disease associated with fatal neurological complications in young children, and several major outbreaks have occurred recently. This study developed an effective antiviral agent by transforming the gene for VP1 protein, a previously defined epitope and also a coat protein of EV71, into tomato plant. VP1 protein was first fused with sorting signals to enable it to be retained in the endoplasmic reticulum of tomato plant, and its expression level increased to 27 microg/g of fresh tomato fruit. Transgenic tomato fruit expressi

Altering expression of benzoic acid/salicylic acid carboxyl methyltransferase 1 compromises systemic acquired resistance and PAMP-triggered immunity in arabidopsis.

Methyl salicylate (MeSA), which is synthesized in plants from salicylic acid (SA) by methyltransferases, has roles in defense against microbial and insect pests. Most of the MeSA that accumulates after pathogen attack is synthesized by benzoic acid/SA carboxyl methyltransferase 1 (AtBSMT1). To investigate the role of AtBSMT1 in plant defense, transgenic Arabidopsis with altered AtBSMT1 function or expression were assessed for their ability to resist pathogen infection. A knockout mutant (Atbsmt1) failed to accumulate MeSA following pathogen infection; these plants also failed to accumulate SA

Plant species-specific recognition of long and short β-1,3-linked glucans is mediated by different receptor systems.

Plants survey their environment for the presence of potentially harmful or beneficial microbes. During colonization, cell surface receptors perceive microbe-derived or modified-self ligands and initiate appropriate responses. The recognition of fungal chitin oligomers and the subsequent activation of plant immunity are well described. In contrast, the mechanisms underlying β-glucan recognition and signaling activation remain largely unexplored. Here, we systematically tested immune responses towards different β-glucan structures and show that responses vary between plant species. While leaves …

Salicylic Acid Mediates Chitosan-Induced Immune Responses and Growth Enhancement in Barley.

Chitosan (CS), derived from the partial deacetylation and hydrolysis of chitin, varies in the degree of deacetylation, molecular weight, and origin, influencing its biological effects, including antifungal properties. In plants, CS triggers immune responses and stimulates biomass growth. Previously, we found that the antifungal activity of CS was strongly dependent on its physicochemical properties. This study revealed that the chitosan batch CS_10 with the strongest antifungal activity also effectively activated plant immune responses and promoted biomass growth. Barley treated with CS_10 exh

Recognition of a salivary effector by the TNL protein RCSP promotes effector-triggered immunity and systemic resistance in Nicotiana benthamiana.

Insects secret chemosensory proteins (CSPs) into plant cells as potential effector proteins during feeding. The molecular mechanisms underlying how CSPs activate plant immunity remain largely unknown. We show that CSPs from six distinct insect orders induce dwarfism when overexpressed in Nicotiana benthamiana. Agrobacterium-mediated transient expression of Nilaparvata lugens CSP11 (NlCSP11) triggered cell death and plant dwarfism, both of which were dependent on ENHANCED DISEASE SUSCEPTIBILITY 1 (EDS1), N requirement gene 1 (NRG1) and SENESCENCE-ASSOCIATED GENE 101 (SAG101), indicating the act…
