Using polymerase chain reaction for rapid sex-determination of a case of sirenomelia.

A rare case of sirenomelia with its autopsy findings is reported. The 1188 g infant was delivered by cesarean section at the 36th week of gestation, and died at 4 h of life. Physical and pathologic findings included sirenomelia (a single lower extremity mass), lung hypoplasia, bilateral renal agenesis, blind-ending colon, severe deformity of the bony pelvis, an imperforate anus, absence of the bladder, and a single umbilical artery. Because of the difficulty in determining the sex of the infant, PCR directed against the SRY gene was performed. The PCR-based sex determination is a rapid techniq

[Ways to decrease inhibition of the polymerase chain reaction by the components of the clinical specimens].

Inhibition of the polymerase chain reaction (PCR) by components of the analyzed specimen is an important problem in analysis of clinical material by this method. The inhibitory activity of clinical material can be decreased by additional purification of DNA by gel filtration on microcolumns. The method was developed using blood-containing specimens and tried on many clinical specimens. Commercial PCR kit for detecting Mycobacterium tuberculosis was used. The proposed method for DNA purification notably improves the efficacy of detecting the agents by PCR.

The value of the polymerase chain reaction in the diagnosis of cutaneous T-cell infiltrates.

The distinction between reactive and neoplastic cutaneous T-cell infiltrates is difficult and requires good clinicopathologic correlation. Many cases manifest changes that are at the borderline between the two. The polymerase chain reaction (PCR) has been reported to detect monoclonality in 52-90% of cutaneous T-cell lymphomas and may be of use in the diagnosis of histologically borderline lesions. We have investigated the use of PCR in a series of borderline lesions including borderline biopsy samples from patients who subsequently developed cutaneous lymphoma. PCR amplification of T-cell rec…

Standardisation of polymerase chain reaction for the detection of Salmonella typhi in typhoid fever.

To improve the diagnosis of Salmonella typhi infection, a polymerase chain reaction (PCR) assay was developed for the amplification of the dH flagellin gene of S typhi. Primers were designed from dH flagellin gene sequence which will give an amplification product of 486 base pairs. In tests to study the specificity of the assay, no amplification was seen in non-salmonella strains or salmonella strains with flagellar gene other than "d". Sensitivity tests determined that 28 pg of S typhi target DNA or 3 x 10(2) target bacteria could be detected by the PCR assay. Subsequently, the PCR technique …

[The detection of Legionella pneumophila DNA with polymerase chain reaction in water samples].

Legionella pneumophila has been known as an important pathogen causing Legionnaires Disease (LD), responsible for more than 80 per cent of clinical LD cases and majority of the LD outbreaks. In order to detect the existence of Legionella pneumophila strains in water samples, we have developed a ploymerase chain reaction method, using two primers synthesized according to the reported Legionella pneumophila mip gene nucleotide sequence. This method was proved to be highly sensitive (18 cfu/ml) and specific. Two water samples were detected positive. This newly developed method provided a quick, a

A polymerase chain reaction assay for the detection of Leptospira spp. in bovine semen.

A rapid and specific method for the detection of pathogenic Leptospira spp. in bovine semen using the polymerase chain reaction (PCR) is described. The primers used were derived from an EcoR1/BamH1 fragment that hybridized strongly to chromosomal DNA from the hardjobovis serovar. Three different extraction methods were evaluated in this study: phenol-chloroform extraction method, proteinase K (PK) in 1% SDS, followed by phenol-chloroform, and phenol-chloroform followed by 1% cetyltrimethylammonium bromide (CTAB). A PCR product of approximately 500 base pairs (bp) in length was obtained when DN…

Comparison of the polymerase chain reaction and serology for the diagnosis of HTLV-I infection.

Human T-lymphotropic virus type I (HTLV-I) is associated with tropical spastic paraparesis (TSP) and adult T-cell leukaemia/ lymphoma (ATL). HTLV-I seroconversion may not take place for many years after infection and some patients develop relatively low concentrations of antibodies that are difficult to detect by means of conventional assays. Some serologic tests also yield high rates of false-positive results. We therefore decided to determine whether the polymerase chain reaction (PCR) might be more sensitive than serology for the diagnosis of HTLV-I infection. Samples of serum and of periph…

Use of the polymerase chain reaction for detecting Trypanosoma cruzi in triatomine vectors.

Determination of the rate of Trypanosoma cruzi infection in its triatomine vectors is an element in control programmes directed at reducing transmission of the organism to humans. Traditionally, T. cruzi has been detected in these insects by microscopical examination of intestinal contents or excreta. The sensitivity of this laborious process has not been defined because of the lack of a bench-mark method against which microscopical examination could be compared. The purpose of this study was to compare the sensitivity of a polymerase chain reaction (PCR) assay with that of microscopical exami…

Clinical significance of a polymerase chain reaction assay for the detection of Mycobacterium tuberculosis.

Various investigators have reported rapid detection of Mycobacterium tuberculosis (MTB) in clinical samples by polymerase chain reaction (PCR). To improve the specificity and efficiency of PCR, the authors adopted a variety of conditions, then analyzed sputum specimens from 217 patients clinically suspected of having MTB. Sputum samples were sonicated to obtain MTB DNA. The DNA was subjected to PCR using primer sets from the region of 650-900 in MTB. The PCR product was detected by direct gel electrophoresis and Southern blot hybridization using digoxigenin-labeled MTB-specific probe. The resu…
