A light and electron microscopic study of the limb long bones perichondral ossification in the quail embryo (Coturnix coturnix japonica).

The perichondral ossification of the limb long bones in the quail embryo is investigated, in this study, by means of light and electron microscopy. Longitudinal sections of the humerus, radius, ulna, femur, tibia and fibula stained with haematoxylin-eosin were examined by the light microscope. Ultrathin cross sections were selected for the electron microscope as well. Light microscopic analysis showed that the ossification began at the same time in the long bones of the wing and leg. At the embryonic day 6, all the cartilaginous rudiments consisted of three zones. The central zone composed of …

Distinct roles for hindbrain and paraxial mesoderm in the induction and patterning of the inner ear revealed by a study of vitamin-A-deficient quail.

The hindbrain and cranial paraxial mesoderm have been implicated in the induction and patterning of the inner ear, but the precise role of the two tissues in these processes is still not clear. We have addressed these questions using the vitamin-A-deficient (VAD) quail model, in which VAD embryos lack the posterior half of the hindbrain that normally lies next to the inner ear. Using a battery of molecular markers, we show that the anlagen of the inner ear, the otic placode, is induced in VAD embryos in the absence of the posterior hindbrain. By performing grafting and ablation experiments in …

Regulation of the expression of serotonin N-acetyltransferase gene in Japanese quail (Coturnix japonica): II. Effect of vitamin A deficiency.

Levels of serotonin N-acetyltransferase [arylalkylamine N-acetyltransferase (AANAT); EC2.3.1.87] mRNA, AANAT activity, and melatonin display a rhythmic pattern in both the pineal gland and the retina. It has been shown that vitamin A is required to maintain the rhythm of melatonin synthesis in the pineal gland of Japanese quail. To understand the mechanism underlying the direct relationship among these factors, we developed an assay system sensitive enough to determine AANAT mRNA, AANAT activity, and melatonin content from a single pineal gland of Japanese quail. Positive direct relationships …

A comparison of the effects of hexachlorobenzene, beta-naphthoflavone, and phenobarbital on cytochrome P-450 and mixed-function oxidases in Japanese quail.

Hexachlorobenzene (HCB), beta-naphthoflavone (BNF), or phenobarbital (PB) was administered to Japanese quail to determine their effects on hepatic porphyrin levels and drug-metabolizing enzymes. While HCB increased porphyrin levels, PB slightly reduced them, and BNF had no effect. HCB was an excellent inducer in quail, increasing the specific content of cytochrome P-450 to levels similar to those produced by BNF. Additional similarities between HCB- and BNF-treated quail included a comparable hypsochromic absorption shift in the CO-reduced difference spectra of cytochrome P-450 and similar eff…

Ascorbic Acid as a Dual Therapeutic and Preventive Agent against Lead and Cadmium Toxicity: Evidence from Controlled Quail Experiments.

Lead (Pb) and cadmium (Cd) are pervasive toxicants that bioaccumulate and cause multisystem injury. Two controlled experiments were conducted in quails, subdivided into co‑exposure (ascorbic acid (AA) given concurrently with Pb or Cd for four weeks) and post‑exposure (AA administered for three weeks after one week of metal exposure). Birds received Pb acetate or Cd chloride (500 ppm) with AA (1000 ppm). In the Pb experiment, co‑exposure with AA significantly reduced tissue burdens compared with Pb alone: liver (16.7 ± 0.8 vs. 29.0 ± 1.3 ppb), thigh (3.2 ± 0.6 vs. 10.6 ± 1.3 ppb), brisket (2.4 …

Clinico-behavioral, hepato-renal, erythrocytic, and genotoxicity induced by sub-chronic oral doses of acetochlor in male Japanese quail.

Acetochlor is one of the well-known and frequently used herbicides that acts as a potent endocrine disruptor. Still, there is a scarcity of studies examining the adverse effects of acetochlor on birds. Therefore, this study evaluated the behavioral alterations, oxidative stress, antioxidant enzymes, genotoxicity and histopathological impacts of subchronic oral doses of acetochlor in quails. A total of 80 male quails were procured and reared in four groups (A-D) for 45 days. Group A served as the control, while the birds reared in groups B, C and D received 20 mg/kg, 30 mg/kg and 40 mg/kg aceto…

Molecular detection of CTX-M ESBL-encoding genes in Escherichia coli isolated from quail cloacal swabs in Surabaya markets.

BACKGROUND: Escherichia coli (E. coli)is a common commensal bacterium in poultry and a key indicator of antimicrobial resistance (AMR). The emergence of extended-spectrum β-lactamase (ESBL)-producing strains, particularly those carrying cefotaximase (CTX-M) genes, poses a significant One Health concern. However, data on CTX-M-producing E. coli in quails from traditional markets in Surabaya are limited. AIM: This study aimed to detect the CTX-M gene in E. coli isolates obtained from quail cloacal swabs from traditional markets in Surabaya and to assess their antimicrobial resistance profiles. …

Apparent and standardized ileal digestibility of calcium in wheat and soybean meal for growing quail chicks using direct method.

Calcium (Ca) is necessary for bone health and metabolic functions in poultry, however, the extent to which it can be utilized varies among feed ingredients. The goal of this study was to determine the apparent ileal digestibility (AID) and standardized ileal digestibility (SID) of calcium in wheat and soybean meal (SBM) in young quail chicks using a direct method. Three dietary treatments were used in the experiment: a calcium-free basal diet to determine endogenous calcium losses, and two diets with either wheat or SBM as the primary calcium sources. Titanium dioxide was incorporated as an in…

Structural gonadal lesions observed in Japanese quail (Coturnix coturnix japonica) following exposure during puberty to the neonicotinoid pesticide, imidacloprid.

Exposure to the neonicotinoid insecticide, imidacloprid (IMI), causes reproductive toxicity in mammals and reptiles. However, reports on the effects of IMI on the gonads in birds are grossly lacking. Therefore, this study investigated the effects of pubertal exposure to IMI on the histology, ultrastructure, as well as the cytoskeletal proteins, desmin, smooth muscle actin and vimentin, of the gonads of Japanese quail (Coturnix coturnix japonica). Quails were randomly divided into four groups at 5 weeks of age. The control group was given only distilled water, whereas, the other three experimen…

Tryptophan and/or canthaxanthin in quail diets: effects on performance, carcass traits, hematology, blood chemistry and hepatic antioxidant capacity.

To enhance the health and performance of poultry, novel approaches have to be created. Using appropriate nutritional interventions to enhance body physiology and thus enhance productivity is one of these approaches. The purpose of the present investigation intended to examine how growing quail physiology and growth is affected by supplementing diets with tryptophan (Trp) and/or canthaxanthin (CX). The sum of 200 unsexed, 1-week-age Japanese quails (Coturnix coturnix japonica), with a nearly similar body weight (BW) of 33.50 ± 1.20 g, were assigned, in random, to four experimental groups. Construction of MC1R and ASIP Eukaryotic Expression Vector and its Regulation of Plumage Color in Japanese Quail (Coturnix japonica).

The Japanese quail expresses polymorphism in plumage colors, including black, yellow, white, wild-type (maroon), and various intermediate colors through hybridization of quail with different plumage colors. The expression levels of MC1R and ASIP play important roles in the regulation of plumage colors in birds. In this study, the eukaryotic expression vector of pcDNA 3.1 + was used to analyze the effects of forced expression of MC1R and ASIP on the plumage colors of Japanese quail embryos. The constructed eukaryotic expression vectors of pcDNA 3.1 (+)-MC1R and pcDNA 3.1(+)-ASIP were transfecte…

Di (2-ethyl hexyl) phthalate (DEHP)-induced spleen toxicity in quail (Coturnix japonica) via disturbing Nrf2-mediated defense response.

Di(2-ethylhexyl) phthalate (DEHP), as a widely used plasticizer, is reported to have widespread environmental and global health hazards. Trace amounts of phthalates in the environment are sufficient to disrupt ecological balance and affect human health. However, DEHP-induced splenic toxicity remains in an unknown state. Therefore, to explore the mechanism of DEHP-induced splenic toxicity, male quail were employed with 0, 250, 500 and 750 mg/kg body weight DEHP by daily gastric perfusion for 45 days. Notably, splenic corpuscular border and cell gap enlargement were observed in the spleen tissue…

Plasma Concentrations of Itraconazole, Voriconazole, and Terbinafine When Delivered by an Impregnated, Subcutaneous Implant in Japanese Quail ( Coturnix japonica ).

Aspergillosis is a common fungal infection in both wild and pet birds. Although effective antifungal medications are available, treatment of aspergillosis can require months of medication administration, which entails stressful handling one or more times per day. This study examined the delivery of the antifungal drugs itraconazole, voriconazole, and terbinafine to Japanese quail ( Coturnix japonica ) via an impregnated implant. Implants contained 0.5, 3, 8, or 24 mg of itraconazole, voriconazole, or terbinafine. The implants were administered subcutaneously over the dorsum and between the sca…

Impact of high and low anxiety trait on object habituation and discrimination: evidence from selected lines of Japanese quail.

Compared to rodents, the relationship between anxiety and cognitive performances has been less studied in birds. Yet, birds are frequently exposed to stimulations that constitute a potential source of anxiety and can affect their adaptation to their living conditions. The present study was aimed at evaluating, in birds, the relationship between levels of anxiety and object habituation and discrimination with the use of Japanese quail lines divergently selected for a fear response, tonic immobility. Previous studies demonstrated that the selection programme has modified the general anxiety trai…

Negative evidence for stachydrine or Galeopsis ladanum L. seeds as the causal agents of coturnism after quail meat ingestion.

Quail poisoning is known to produce an acute myoglobinuric syndrome called coturnism. The cause of this syndrome is still unknown, although it has been postulated that Galeopsis ladanum L. seeds, in particular lipidic compounds or stachydrine, are responsible for this toxicity. Thus, we aimed to study the implication of this plant in coturnism in order to explore the physiopathology of the disease, especially with regard to stachydrine and lipidic compounds extracted from seeds. For this purpose, Wistar rats were fed with G. ladanum seed extracts or with quail meat. However, the rhabdomyolysis…

Retinal TUNEL-positive cells and high glutamate levels in vitreous humor of mutant quail with a glaucoma-like disorder.

PURPOSE: To investigate whether retinal cell death observed in an avian glaucoma-like disorder occurs by apoptosis and whether an increase in excitotoxic amino acid concentration in the vitreous humor is associated temporally with cell death in the retina. METHODS: Presumptive retinal apoptotic nuclei were identified by histochemical detection of DNA fragmentation (by TdT-dUTP terminal nick-end labeling [TUNEL]), and vitreal concentrations of glutamate and several other amino acids were determined by high-pressure liquid chromatography with fluorometric detection in the al mutant quail (Cotur…

Mechanism controlling photostimulated luteinizing hormone secretion is different from preovulatory luteinizing hormone surge in Japanese quail (Coturnix coturnix japonica).

A so-called "night-interruption" experiment with a 15-min light pulse showed that a sensitive phase for the photoperiodic LH secretion in male Japanese quail extended over a period of 2 hr from 12.5 to 14.5 hr after dawn. Exposure of a 1-hr light pulse at this photosensitive phase to male quail kept under 8L:16D induced the increase of plasma LH concentrations just the same as quail transferred to 16L:8D. In the first few days of photostimulation either by night interruption or by long days, LH concentrations increased at several hours after the photosensitive phase and decreased to the basal …
