New pyridazinone-4-carboxamides as new cannabinoid receptor type-2 inverse agonists: Synthesis, pharmacological data and molecular docking.

In the last few years, cannabinoid type-2 receptor (CB2R) selective ligands have shown a great potential as novel therapeutic drugs in several diseases. With the aim of discovering new selective cannabinoid ligands, a series of pyridazinone-4-carboxamides was designed and synthesized, and the new derivatives tested for their affinity toward the hCB1R and hCB2R. The 6-(4-chloro-3-methylphenyl)-2-(4-fluorobenzyl)-N-(cis-4-methylcyclohexyl)-3-oxo-2,3-dihydropyridazine-4-carboxamide (9) displayed high CB2-affinity (KiCB2 = 2.0 ± 0.81 nM) and a notable selectivity (KiCB1/KiCB2 > 2000). In addition,

Ligand activation of cannabinoid receptors attenuates hypertrophy of neonatal rat cardiomyocytes.

: Endocannabinoids are bioactive amides, esters, and ethers of long-chain polyunsaturated fatty acids. Evidence suggests that activation of the endocannabinoid pathway offers cardioprotection against myocardial ischemia, arrhythmias, and endothelial dysfunction of coronary arteries. As cardiac hypertrophy is a convergence point of risk factors for heart failure, we determined a role for endocannabinoids in attenuating endothelin-1-induced hypertrophy and probed the signaling pathways involved. The cannabinoid receptor ligand anandamide and its metabolically stable analog, R-methanandamide, sup

Oral cannabinoid for the prophylaxis of chemotherapy-induced nausea and vomiting-a systematic review and meta-analysis.

INTRODUCTION: Chemotherapy-induced nausea and vomiting (CINV) is a burdensome adverse event frequently associated with chemotherapy treatment of cancer. Evidence suggests that cannabinoid CB2 receptors are present in brainstem neurons, and thus, there may exist a role for cannabinoids to counter CINV. The aim of this paper is to conduct a systematic review and meta-analysis of the efficacy and safety of oral cannabinoids compared with other treatments as documented in randomized controlled trials (RCTs). METHODS: A literature search was conducted using Ovid MEDLINE up until December 31, 2018;

Identification and Investigation of the Intrinsic Receptor Activation Potential and Metabolization of the New Oxo-Pyridyl Synthetic Cannabinoid Receptor Agonist CH-FUBBMPDORA.

CH-FUBBMPDORA (CHO-4'Me-5'Br-FUBOXPYRA), a recent addition to the recreational drug market, bypasses the Chinese generic ban (2021) on synthetic cannabinoid receptor agonists (SCRAs) due to its new 5-bromo-4-methylpyridin-2(1H)-one core. Its pharmacological properties are currently undefined, and it is yet to be found in biological samples. However, it is unclear whether this is due to low prevalence or hampered detection. The aim of this study was twofold. First, we used a powder seized by customs as a case study to evaluate the utility of low-field nuclear magnetic resonance (LF-NMR) to uneq

Antidepressant-like effects of β-caryophyllene on restraint plus stress-induced depression.

Chronic stress is depressogenic by altering neurotrophic and neuroinflammatory environments of the organism. The endocannabinoid system controls cognitive and emotional responses related with stress through the interaction with endocannabinoid receptors. β-Caryophyllene (BCP) is a CB2 agonist that exhibited anti-inflammatory, analgesic effects but minimal psychoactive effects. To test if BCP exhibits antidepressant-like action, animals were chronically restrained with additional stressors for 28 days, and BCP (25, 50, 100 mg/kg) was intraperitoneally injected once a day during the stress infli

Cannabinoids in preclinical research of sepsis: a scoping review.

BACKGROUND: Sepsis is a global health problem that ends millions of lives and costs billions of dollars in treatment and management every year. This disease is responsible for one in every five deaths worldwide, and is the third leading cause of death in hospitals. Despite decades of research, no current specific treatment or cure are available, only supportive and symptomatic care, and few preclinical studies reach human trials. Since the discovery of the endocannabinoid system (ECS), cannabinoids have been researched as a potential treatment for various diseases, including sepsis. Our review

Cannabinoids for Symptom Management in Patients with Kidney Failure: A Narrative Review.

People with kidney failure can experience a range of symptoms that lead to suffering and poor quality of life. Available therapies are limited, and evidence for new treatment options is sparse, often resulting in incomplete relief of symptoms. There is growing interest in the potential for cannabinoids, including cannabidiol and tetrahydrocannabinol, to treat symptoms across a wide range of chronic diseases. As legal prohibitions are withdrawn or minimized in many jurisdictions, patients are increasingly able to access these agents. Cannabinoid receptors, CB1 and CB2, are widely expressed in t

Decoding the Toxicity of Synthetic Cannabinoids: From Receptor Activation to Multiorgan Dysfunction.

Synthetic cannabinoids (SCBs) represent a rapidly expanding and chemically diverse class of new psychoactive substances, deliberately designed to mimic the effects of natural cannabis. However, their high potency and full agonism at cannabinoid receptors lead to severe public health risks. The primary mechanism of SCBs' action involves the robust activation of CB1 and CB2 receptors, which in turn trigger a cascade of downstream events. This includes the profound dysregulation of key neurotransmitter systems, critical ion channel functions, and essential intracellular signaling pathways, culmin

Obesity as a Condition Determined by Food Addiction: Should Brain Endocannabinoid System Alterations Be the Cause and Its Modulation the Solution?

Obesity is a complex disorder, and the number of people affected is growing every day. In recent years, research has confirmed the hypothesis that food addiction is a determining factor in obesity. Food addiction is a behavioral disorder characterized by disruptions in the reward system in response to hedonic eating. The endocannabinoid system (ECS) plays an important role in the central and peripheral control of food intake and reward-related behaviors. Moreover, both obesity and food addiction have been linked to impairments in the ECS function in various brain regions integrating peripheral

The endocannabinoid system, eating behavior and energy homeostasis: the end or a new beginning?

The endocannabinoid system (ECS) consists of two receptors (CB(1) and CB(2)), several endogenous ligands (primarily anandamide and 2-AG), and over a dozen ligand-metabolizing enzymes. The ECS regulates many aspects of embryological development and homeostasis, including neuroprotection and neural plasticity, immunity and inflammation, apoptosis and carcinogenesis, pain and emotional memory, and the focus of this review: hunger, feeding, and metabolism. This mini-review summarizes the main findings that supported the clinical use of CB1 antagonists/inverse agonists, the clinical concerns that h

The Role of Endocannabinoids in Physiological Processes and Disease Pathology: A Comprehensive Review.

The endocannabinoid system is a complex communication system involved in maintaining homeostasis in various physiological processes, including metabolism, immune response, pain modulation, and neuroprotection. Endocannabinoids, mainly anandamide and 2-arachidonoylglycerol, are natural ligands of the cannabinoid receptors CB1 and CB2, which are widely distributed throughout the central nervous system and peripheral tissues. Their biosynthesis, degradation, and interaction with other signaling pathways play crucial roles in both health and disease. This article provides a comprehensive overview

Placental transfer and vasoactivity of cannabidiol: beware of rapid oxidation.

INTRODUCTION: Cannabis use is increasing among pregnant women, although such use associates with fetal growth restriction and preeclampsia. While most research focuses on Δ9-tetrahydrocannabinol (THC), many pregnant women use cannabidiol (CBD)-containing products like CBD-oils. It is unknown whether CBD is transferred across the placenta. METHODS: CBD transfer was studied using an ex vivo placental perfusion model, making use of healthy term placentas. CBD was added to the maternal side. Placentas were perfused for 180 min. Using liquid chromatography-tandem mass spectrometry, CBD was quantif

Structural analogs of pyrazole and sulfonamide cannabinoids: effects on acute food intake in mice.

Obesity contributes to a multitude of serious health problems. Given the demonstrated role of the endogenous cannabinoid system in appetite regulation, the purpose of the present study was to evaluate structural analogs of two cannabinoids, rimonabant (cannabinoid CB(1) receptor antagonist) and O-2050 (sulfonamide analog of Δ(8)-tetrahydrocannabinol), that showed appetite suppressant effects in previous studies. Structure-activity relationships of these two lead compounds were examined in several assays, including cannabinoid CB(1) and CB(2) receptor binding, food intake, and an in vivo test b

Endocannabinoid System: A Multi-Facet Therapeutic Target.

Cannabis sativa is also popularly known as marijuana. It has been cultivated and used by man for recreational and medicinal purposes since many centuries. Study of cannabinoids was at bay for very long time and its therapeutic value could not be adequately harnessed due to its legal status as proscribed drug in most of the countries. The research of drugs acting on endocannabinoid system has seen many ups and downs in the recent past. Presently, it is known that endocannabinoids has role in pathology of many disorders and they also serve "protective role" in many medical conditions. Several di

Identification of Potential Multitarget Directed Ligands for Alzheimer's Disease by Coupling Virtual Screening and Experimental Validation.

Alzheimer's disease (AD) is a neurodegenerative disorder (NDD) associated with the accumulation of beta-amyloid plaques (βA), oxidative stress, and a decrease in cholinergic activity among other pathologies. Given the limitations of current treatments, multitarget strategies present a promising alternative. In this study we prioritized six AD-related protein targets: acetylcholinesterase (AChE), beta-secretase 1 (BACE-1), cannabinoid receptor type 2 (CB2), glycogen synthase kinase 3 beta (GSK-3β), monoamine oxidase A (MAO-A), and the neuronal acetylcholine receptor subunit alpha-7 (nAChR7).
