Renalase deficiency in heart failure model of rats--a potential mechanism underlying circulating norepinephrine accumulation.

BACKGROUND: Sympathetic overactivity and catecholamine accumulation are important characteristic findings in heart failure, which contribute to its pathophysiology. Here, we identify a potential mechanism underlying norepinephrine accumulation in a rat model of heart failure. METHODOLOGY/PRINCIPAL FINDINGS: Initially, we constructed a rat model of unilateral renal artery stenosis (n = 16) and found that the expression of renalase, a previously identified secreted amine oxidase, was markedly reduced in the ischemic compared to the non-ischemic kidney (protein: 0.295±0.085 versus 0.765±0.171, p

[Differential diagnosis and treatment of complex renal cysts detected by ultrasound screening of the abdominal cavity organs].

Ultrasound screening of the abdominal cavity organs was performed in 98 patients, and renal cysts were revealed in 31 patientsare. 11 (26,6%) of 31 patients had renal cysts with complex structure. In 4 patients, complex cysts were located in parapelvic zone, in 7 patients - in different parts of the renal parenchyma. Pharmaco-Doppler sonography and computed tomography with bolus contrast enhancement in 7 patients with complex parenchymal cysts had revealed indirect signs of a malignant process - septums, thickening of the walls of cysts and septums, foci of calcination, increased blood circula

[Fibrous dysplasia of renal arteries (terminology, classification, clinical aspects and diagnosis)].

The examination was conducted in 72 patients with renovascular hypertension caused by fibromuscle dysplasia of the renal arteries. According to the authors, fibromuscle dysplasia of the renal arteries stands third after atherosclerosis and non-specific aorto-arteritis. Among the preliminary diagnostic measures of importance are such as intravenous urography, isotope renography, scintigraphy, but their value is much lower than in cases of atherosclerotic stenosis or stenosis due to non-specific aorto-arteritis, which is attributed to a good collateral circulation in the kidney. The diagnosis is

Renal perfusion and metabolism in experimental endotoxin shock.

Central and renal hemodynamics, renal oxygenation, renal uptake of glucose, lactate, fats, renal carnitine metabolism, arterial atrial natriuretic factor (ANF) and catecholamine release were studied in sixteen adult beagle dogs during pentobarbital anesthesia. Renal cortical oxygen tension was recorded by means of a Silastic tonometer. Twelve animals underwent acute circulatory shock induced by intravenous Escherichia coli endotoxin 0.5 mg/kg. Four control dogs received normal saline. The endotoxin infusion resulted in decreased cardiac function, renal blood flow and renal cortical PO2. The re

Alterations in regional blood flow during positive end-expiratory pressure ventilation.

PEEP improves the gas-exchange abnormalities that accompany adult respiratory distress syndrome (ARDS). However, since PEEP decreases cardiac output, it may also alter regional blood flow and therefore, substrate delivery to specific organs. To test this hypothesis, radiolabeled 15-mu microspheres were used to directly quantify the effects of mechanical ventilation with PEEP on regional blood flow to individual organs in animals. Mechanical ventilation alone produced a -21.2 +/- 3.6% and a -28.1 +/- 5.2% decrease in cardiac output at 30 and 60 min, respectively. The addition of 14 cm H2O PEEP

Near-drowning: new perspectives for human hypoxic acute kidney injury.

Concepts regarding hypoxic acute kidney injury (AKI) are derived from widely used warm ischemia-reflow (WIR) models, characterized by extensive proximal tubular injury and associated with profound inflammation. However, there is ample clinical and experimental data indicating that hypoxic AKI may develop without total cessation of renal blood flow, with a different injury pattern that principally affects medullary thick limbs in the outer medulla. This injury pattern likely reflects an imbalance between blood and oxygen supply and oxygen expenditure, principally for tubular transport. Experime

Impaired blood flow in acute kidney injury: pathophysiology and potential efficacy of intrarenal vasodilator therapy.

PURPOSE OF REVIEW: Acute kidney injury (AKI) is a common complication of hospitalized patients and associated with significant morbidity and mortality. Numerous studies have documented that acute reductions in glomerular filtration rates are associated with significant in-hospital mortality. Moreover, patients progressing to dialysis-dependent AKI can have mortality rates that exceed 60%. The pathophysiology of AKI is unknown, but marked reductions in corticomedullary blood flow leads to significant reductions in glomerular filtration rate during early phases of the disease. The recognition th

Treatment of vascular calcification.

A variety of potential therapies for vascular calcification, based either on the underlying biology or physical chemistry or solely on empiric observations in patients, may be effective but lack rigorous testing. Pasch et al. provide convincing evidence that sodium thiosulfate prevents medial vascular calcification in uremic rats. Although this provides some scientific basis for the clinical use of thiosulfate, uncertainty about mechanism of action and safety still remains.

Influences of diurnal bright or dim light exposure on urine volume in humans.

We investigated with eight healthy females if 8 hr diurnal (0700 to 1500 h) bright rather than dim light (5,000 vs. 80 lx) influenced urine volume. Environmental illuminance was made identical at all other times besides 07:00 to 15:00 h. The participants spent time at strictly regulated schedules in a bioclimatic chamber (26 degrees C, relative humidity 60%) for 57 h. Blood was drawn (2 ml) just before lunch in order to calculate Creatinine clearance (Ccr). Urine volume was significantly higher during wakefulness and the 8-h sleep period with bright rather than dim light. Ccr was significantly

Role of hypoxia in the pathogenesis of renal disease.

Despite a high overall oxygen supply, the tissue oxygen tensions in the kidney are comparatively low and render the kidneys prone to hypoxic injury. However, the role of hypoxia in the pathogenesis of different types of renal disease remains incompletely understood. The importance of hypoxic cell injury is most obvious in renal vascular disease, in which occlusion of the renal artery or one of its branches can induce tissue necrosis. In acute renal failure, circumstantial evidence suggests that hypoxic injury to the renal medulla plays a significant role. In addition, chronically impaired oxyg

Frequency domain of renal autoregulation in the conscious dog.

The dynamic range in which renal blood flow (RBF) autoregulation occurs was determined in eight conscious foxhounds chronically catheterized in the abdominal aorta and implanted with a transit-time flow probe over the renal artery. Sinusoidal driving pressures (amplitude of 10 mmHg) were forced on the renal arterial pressure at different frequencies by a servo-control device, and transfer functions were calculated. Only one frequency range was found below which the gain of the transfer function declined and in which the phase angle increased (n = 8). This indicates the presence of a potent mec

[Assessment of split renal function before and after angioplasty or bypass grafting in the patients with renovascular hypertension].

Two different methods to assess the change of split renal function following angioplasty or bypass grafting were studied in a total of 12 patients with renovascular hypertension. The studies were performed before and within seven days after the therapeutic intervention. Split effective renal plasma flow (ERPF) and glomerular filtration rate (GFR) after injections of I-131 hippuran and Tc-99m DTPA were measured using kidney counting corrected for depth and dose, described by Schlegel and Gates. In six of 10 patients with unilateral renal artery stenosis and in two patients with bilateral renal

Renal effects of antihypertensive drugs.

Antihypertensive drugs have disparate effects on renal haemodynamics, tubular function, plasma electrolytes, and hormonal responses. Calcium entry blockers and angiotensin-converting enzyme (ACE) inhibitors are unique in that they may increase glomerular filtration rate (GFR) and renal blood flow in patients with hypertension. Both classes of drugs are distinctive in that they prevent salt retention because of their inhibitory effect on tubular sodium reabsorption. In addition to these attributes, which are desirable in terms of lowering systemic blood pressure, these 2 classes of drugs exert

Renovascular hypertension. Relationship between hepatic blood flow and plasma renin activity.

Hepatic blood flow and peripheral plasma renin activity were determined in 15 true renovascular hypertensive patients and in 13 patients with essential hypertension. In the renovascular hypertensives, plasma renin activity and hepatic blood flow were negatively correlated ( p less than 0.01). In contrast, no relationship was observed in the essential hypertensives. The study suggests that hyperactivity of the renin-angiotensin system could induce splanchnic arteriolar constriction in patients with renovascular hypertension.
