Endoplasmic reticulum stress: New insights into the pathogenesis and treatment of retinal degenerative diseases.

Physiological equilibrium in the retina depends on coordinated work between rod and cone photoreceptors and can be compromised by the expression of mutant proteins leading to inherited retinal degeneration (IRD). IRD is a diverse group of retinal dystrophies with multifaceted molecular mechanisms that are not fully understood. In this review, we focus on the contribution of chronically activated unfolded protein response (UPR) to inherited retinal pathogenesis, placing special emphasis on studies employing genetically modified animal models. As constitutively active UPR in degenerating retinas

Network and atomistic simulations unveil the structural determinants of mutations linked to retinal diseases.

A number of incurable retinal diseases causing vision impairments derive from alterations in visual phototransduction. Unraveling the structural determinants of even monogenic retinal diseases would require network-centered approaches combined with atomistic simulations. The transducin G38D mutant associated with the Nougaret Congenital Night Blindness (NCNB) was thoroughly investigated by both mathematical modeling of visual phototransduction and atomistic simulations on the major targets of the mutational effect. Mathematical modeling, in line with electrophysiological recordings, indicates …

[Diagnostic value of pseudoprotanomaly for the differential diagnosis between retinal and optic nerve diseases].

It has been known since the time of Koellner that some diseases of the retina produce a shift of the Rayleigh equation toward red on the anomaloscope. In 1951, at the suggestion of Engelking, this "pseudoprotanomaly" was analyzed using Helmholtz's color mixer at Heidelberg University Eye Clinic. Pseudoprotanomaly is pathognomonic for diseases accompanied by macular edema (central serous chorioretinitis) as well as for the early stage of Stargardt's macular degeneration. The various hypotheses regarding its origin are discussed. In cases of visual disturbance with just-recognizable scotoma, oph

Retinal capillary degeneration and blood-retinal barrier disruption in murine models of Alzheimer's disease.

Extensive effort has been made studying retinal pathology in Alzheimer's disease (AD) to improve early noninvasive diagnosis and treatment. Particularly relevant are vascular changes, which appear prominent in early brain pathogenesis and could predict cognitive decline. Recently, we identified platelet-derived growth factor receptor beta (PDGFRβ) deficiency and pericyte loss associated with vascular Aβ deposition in the neurosensory retina of mild cognitively impaired (MCI) and AD patients. However, the pathological mechanisms of retinal vascular changes and their possible relationships with

Nrf2/HO-1 signaling in retinal neurodegenerative diseases: redox homeostasis, cytoprotection, and therapeutic perspectives.

Retinal neurodegenerative diseases (RNDs) represent a major group of disorders leading to irreversible visual impairment. Persistent oxidative stress, mitochondrial dysfunction, inflammatory responses, lipid peroxidation, and disruption of neurovascular unit homeostasis are closely implicated in retinal neuronal and vascular injury. Nuclear factor erythroid 2-related factor 2 (Nrf2) and its downstream effector heme oxygenase-1 (HO-1) constitute a key regulatory axis in cellular antioxidant defense and adaptive stress responses. Current preclinical evidence suggests that Nrf2/HO-1 signaling may

Environmental enrichment as a mitochondria-targeting systems strategy across neurodegenerative diseases and retinal dystrophies.

Mitochondrial dysfunction is a central contributor to neurodegenerative disorders affecting both the central nervous system and the retina, where impaired energy metabolism, oxidative stress, and defective cellular resilience converge to drive progressive neuronal loss. Environmental enrichment (EE), a multimodal non-pharmacological paradigm, has emerged as a powerful modulator of brain and retinal plasticity in preclinical models, promoting adaptive responses that support mitochondrial function and neurotrophic signaling. This review synthesizes evidence indicating that EE influences mitochon

Retinal gene therapies for inherited ocular diseases: Translational delivery strategies from bench to bedside.

Inherited retinal diseases (IRDs), including retinitis pigmentosa, Leber congenital amaurosis, and Stargardt disease, represent a major cause of inherited visual impairment and blindness, particularly in working-age populations. These disorders are genetically heterogeneous and result in progressive degeneration of photoreceptors and the retinal pigment epithelium. The eye offers a uniquely favorable environment for gene therapy due to its anatomical compartmentalization, accessibility, and relative immune privilege; however, successful clinical translation remains constrained by multiple biol…

3D OCT-Based Retinal Biomarker Analysis for Automatic Regional-Wise Characterization of Neurodegenerative Diseases.

Neurodegenerative diseases (NDDs) such as Alzheimer's disease (AD), essential tremor (ET), multiple sclerosis (MS), and Parkinson's disease (PD) are complex disorders that often exhibit overlapping symptoms, leading to diagnostic challenges. Given the increasing interest in retinal imaging as a non-invasive biomarker for neurodegeneration, this study proposes a fully automated machine learning pipeline for disease characterization using optical coherence tomography (OCT). We analyze macular thickness patterns across three key and relevant retinal elements: retinal nerve fibre layer (RNFL), gan

Clinical and Socioeconomic Burden of Retinal Diseases: Can Biosimilars Add Value? A Narrative Review.

Retinal diseases, such as neovascular age-related macular degeneration, diabetic retinopathy, and retinal vein occlusion, pose a significant global burden on individuals, families, and healthcare systems. Intravitreal anti-vascular endothelial growth factor (anti-VEGF) therapy has become the standard treatment for retinal diseases, improving clinical outcomes, while delaying disease progression. Although effective, biologics are associated with high costs, which can lead to underutilisation and, consequently, suboptimal patient treatment outcomes, further contributing to healthcare costs. Addi…
