Construction of a Saccharomyces Cerevisiae Cell Factory for Shikimate Production through Precise Regulatory Strategies.

Shikimate, a key intermediate in the biosynthesis of diverse high-value aromatic compounds, has significant applications in the food and pharmaceutical industries. However, efficient microbial production remains limited by complex pathway regulations and stringent feedback inhibition. Here, an optimized shikimate biosynthetic route was established in Saccharomyces cerevisiae through promoter engineering and rational pathway redesign. Specifically, replacement of the native shikimate kinase with EcaroK, together with promoter optimization, enabled the shikimate accumulation under non-auxotrophi…

Metabolic engineering of Saccharomyces cerevisiae to enhance resistance toward microbial toxicity in agave extracts.

Agave, which thrives in arid regions of North Americas, is a promising alternative feedstock for biofuel production due to its high sugar content and drought tolerance. A major challenge to using agave as a feedstock for bioprocessing is its toxicity. Saponins, natively produced glycosides, disrupt cell membranes and are highly toxic to Saccharomyces cerevisiae even at low concentrations. To enable agave as a feedstock, we combined rational engineering with adaptive laboratory evolution (ALE) to create new S. cerevisiae strains with high tolerance to agave extracts. Heterologous overexpression…

Sporulation generates stress-dependent phenotypic variability in the heterothallic industrial Saccharomyces cerevisiae Ethanol Red.

The industrial yeast Saccharomyces cerevisiae Ethanol Red is widely used in large-scale bioethanol production and is generally considered phenotypically stable because of its heterothallic diploid background. Here, we investigated whether extensive sporulation of this highly domesticated strain could generate functional diversification without genetic engineering. Thirty monosporal cultures (MSCs) were randomly selected and profiled for growth kinetics, CFU-based cultivability and Fourier-Transform Infrared (FTIR) fingerprints under three industrially relevant stressors, namely furfural (3 g L…

Lipidomic Insight Across 48 Saccharomyces cerevisiae Wine Yeast Strains Revealed Novel N-Acyl Lysophosphatidylethanolamines.

Saccharomyces cerevisiae (SC), an important unicellular yeast species in the biotechnology, beverage, and food industries, has remarkable applications in ethanol production. Lipids in SC play a crucial role in stress management during the fermentation process. Despite the importance of SC yeasts, comprehensive lipid profiling across multiple yeast strains remains limited. This study aimed to elucidate the comprehensive lipidomic profiles of 48 wine yeast strains of the SC species using a nontargeted liquid chromatography-mass spectrometry approach. A total of 135 lipid species were identified,…

Heat stress promotes pre-tRNA capping to modulate cap-dependent translation in Saccharomyces cerevisiae.

Methylguanosine capping, a post-transcriptional modification of the 5' terminus of RNA polymerase II transcripts, influences RNA function, stability, and protein synthesis. Previously, we identified the cap modification in specific transfer RNA (tRNA) precursors (pre-tRNAs) transcribed by RNA polymerase III in Saccharomyces cerevisiae, and demonstrated that pre-tRNA capping prevents 5' exonucleolytic degradation of pre-tRNAs. Herein, we comprehensively mapped pre-tRNA capping and found that nearly all pre-tRNAs undergo cap modification. This analysis enabled precise determination of transcript…

Metabolic and transport engineering enable efficient de novo biosynthesis of patchoulol in Saccharomyces cerevisiae.

Patchoulol is a valuable sesquiterpene for fragrance and cosmetics, but plant extraction is slow and resource-intensive. Here, we combined enzyme, pathway and transporter engineering to build a yeast chassis for high-titer patchoulol production. Semi-rational engineering of patchoulol synthase (PTS) increased catalytic efficiency; together with multi-copy PTS expression, the shake-flask titer rose from 0.40 ± 0.05 mg/L (PT01) to 258.72 ± 27.35 mg/L. Strengthening glycolysis and the mevalonate pathway and down-regulating ERG9 redirected flux to FPP. Guided by docking and transcript analysis, we

Transcriptomic Analysis Reveals How PHO4 Gene Modulates Growth and Ethanol Fermentation in Saccharomyces cerevisiae.

During high-concentration ethanol fermentation, Saccharomyces cerevisiae often faces multiple stresses, such as high osmotic pressure, ethanol toxicity, and nutrient limitation. These factors collectively limited the production of ethanol. To identify novel targets related to fermentation performance, we employed SHPERM- bCGHR strategy (a marker free allele replacement strategy based on comparative genomics and homologous recombination). We replaced the endogenous PHO4 of the high-producing strain MF01 with the PHO4 allele from MC15, thereby constructing a novel engineered strain MF01-PHO4. Un…

Engineered Probiotic Saccharomyces boulardii Produces Functional Insulin With Long-Acting Effect in Mice Model.

Diabetes mellitus affects over 500 million people globally, with current insulin therapies relying on subcutaneous injections that compromise the liver's natural role in glucose regulation and suffer from poor patient compliance. Oral insulin delivery offers a physiological alternative but faces challenges, including gastrointestinal degradation and poor absorption. Here, we engineered Saccharomyces boulardii, a probiotic yeast with established gastrointestinal survival capabilities, to secrete a long-acting Insulin-Fc fusion protein (Ins/Fc). Using strong constitutive promoters (TDH3 and TEF1…

Acid-Mediated Antimicrobial Activity and Probiotic Potential of an SKS1-knockout Saccharomyces Cerevisiae Strain.

Bacterial infections pose a significant threat to human health, and the prudent use of antibiotics remains a critical component of disease treatment and control. The increase in drug resistance of pathogenic bacteria poses a huge challenge to the world. Probiotics have become a promising approach to combating pathogenic bacterial infections. In this research, we found that the SKS1-KO Saccharomyces cerevisiae showed effective antibacterial activity against common clinical pathogenic bacteria in vitro, which has been confirmed through the determination of the minimum inhibitory concentration (M…

Characterizing probiotic properties of the Saccharomyces cerevisiae ABO1 strain isolated from toddy palm nectar.

Cardiovascular diseases are strongly associated with high cholesterol levels, which also cause hypertension and diabetes. It is important to have optimal cholesterol levels for general well-being. This research is intended to screen, isolate, and identify probiotic yeast strains with cholesterol-reducing ability from toddy palm nectar. Out of 63 isolates, five strains ABO1, ABO11, ABO13, ABO20, and ABO21 had probiotic characteristics. Among the five isolates, ABO1 showed the highest probiotic traits and was characterized as Saccharomyces cerevisiae ABO1 by molecular-level characterization. Cho…

Non-Saccharomyces yeasts enhance yield and flavor in industrial xiaoqu light-flavor baijiu production.

This study investigated the effects of adding Saccharomycopsis fibuligera (SF) and Pichia kudriavzevii (PK) on microbial communities and flavor substances in industrial xiaoqu light-flavor baijiu production. The result showed that the highest acidity was found in the control group (CK: Saccharomyces cerevisiae and Rhizopus) at the end of fermentation. SF and PK promoted the growth of Rhizopus while decreasing the abundance of S. cerevisiae during the saccharification stage. Metagenomic sequencing showed that S. cerevisiae and Lactobacillus helveticus were the dominant fungus and bacterium duri…

Automation protocol for high-efficiency and high-quality genomic DNA extraction from Saccharomyces cerevisiae.

Although many protocols have been previously developed for genomic DNA (gDNA) extraction from S. cerevisiae, to take advantage of recent advances in laboratory automation and DNA-barcode sequencing, there is a need for automated methods that can provide high-quality gDNA at high efficiency. Here, we describe and demonstrate a fully automated protocol that includes five basic steps: cell wall and RNA digestion, cell lysis, DNA binding to magnetic beads, washing with ethanol, and elution. Our protocol avoids the use of hazardous reagents (e.g., phenol, chloroform), glass beads for mechanical cel

Acquired resistance to severe ethanol stress-induced inhibition of proteasomal proteolysis in Saccharomyces cerevisiae.

BACKGROUND: Although the budding yeast, Saccharomyces cerevisiae, produces ethanol via alcoholic fermentation, high-concentration ethanol is harmful to yeast cells. Severe ethanol stress (> 9% v/v) inhibits protein synthesis and increases the level of intracellular protein aggregates. However, its effect on proteolysis in yeast cells remains largely unknown. METHODS: We examined the effects of ethanol on proteasomal proteolysis in yeast cells through the cycloheximide-chase analysis of short-lived proteins. We also assayed protein degradation in the auxin-inducible degron system and the ub…

[URA3 affects artemisinic acid production by an engineered Saccharomyces cerevisiae in pilot-scale fermentation].

CRISPR/Cas9 has been widely used in engineering Saccharomyces cerevisiae for gene insertion, replacement and deletion due to its simplicity and high efficiency. The selectable markers of CRISPR/Cas9 systems are particularly useful for genome editing and Cas9-plasmids removing in yeast. In our previous research, GAL80 gene has been deleted by the plasmid pML104-mediated CRISPR/Cas9 system in an engineered yeast, in order to eliminate the requirement of galactose supplementation for induction. The maximum artemisinic acid production by engineered S. cerevisiae 1211-2 (740 mg/L) was comparable to…

Enhanced flavour profiles through radicicol induced genomic variation in the lager yeasts, Saccharomyces pastorianus.

The yeasts, Saccharomyces pastorianus, are hybrids of Saccharomyces cerevisiae and Saccharomyces eubayanus and have acquired traits from the combined parental genomes such as ability to ferment a range of sugars at low temperatures and to produce aromatic flavour compounds, allowing for the production of lager beers with crisp, clean flavours. The polyploid strains are sterile and have reached an evolutionary bottleneck for genetic variation. Here we describe an accelerated evolution approach to obtain lager yeasts with enhanced flavour profiles. As the relative expression of orthologous allel…

mDrop-Seq: Massively Parallel Single-Cell RNA-Seq of Saccharomyces cerevisiae and Candida albicans.

Advances in high-throughput single-cell RNA sequencing (scRNA-seq) have been limited by technical challenges such as tough cell walls and low RNA quantity that prevent transcriptomic profiling of microbial species at throughput. We present microbial Drop-seq or mDrop-seq, a high-throughput scRNA-seq technique that is demonstrated on two yeast species, Saccharomyces cerevisiae, a popular model organism, and Candida albicans, a common opportunistic pathogen. We benchmarked mDrop-seq for sensitivity and specificity and used it to profile 35,109 S. cerevisiae cells to detect variation in mRNA leve…

Can Saccharomyces cerevisiae keep up as a model system in fungal azole susceptibility research?

The difficulty of manipulation and limited availability of genetic tools for use in many pathogenic fungi hamper fast and adequate investigation of cellular metabolism and consequent possibilities for antifungal therapies. S. cerevisiae is a model organism that is used to study many eukaryotic systems. In this review, we analyse the potency and relevance of this model system in investigating fungal susceptibility to azole drugs. Although many of the concepts apply to multiple pathogenic fungi, for the sake of simplicity, we will focus on the validity of using S. cerevisiae as a model organism

Impact of Homologous Resistance Mutations from Pathogenic Yeast on Saccharomyces cerevisiae Lanosterol 14α-Demethylase.

Fungal infections frequently affect immunodeficient individuals and are estimated to kill 1.35 million people per annum. Azole antifungals target the membrane-bound cytochrome P450 monooxygenase lanosterol 14α-demethylase (CYP51; Erg11p). Mutations in CYP51 can render the widely used triazole drugs less effective. The Candida albicans CYP51 mutation G464S and the double mutation Y132F G464S (Y140F and G464S by Saccharomyces cerevisiae numbering) as well as the CYP51A G54E/R/W mutations of Aspergillus fumigatus (G73E/R/W by S. cerevisiae numbering) have been reproduced in a recombinant C-termin…
