Promethazine affects optokinetic but not vestibular responses in monkeys.

BACKGROUND: Promethazine is used to treat motion sickness including Space Adaptation Syndrome, but there is incomplete information about how it affects vestibular and optokinetic responses. METHODS: Vestibular and optokinetic nystagmus, recorded with eye coils, were characterized in monkeys after administration of promethazine at dosages approximately equivalent to those used by humans in space. RESULTS: The initial increase of horizontal eye velocity during optokinetic nystagmus (OKN) was reduced after receiving the drug. Consequently, it took a longer time for eye velocity to rise to 60% o

Physiological and biomechanical considerations for a human Mars mission.

Evolving on Earth has made humans perfectly adapted, both physiologically and biomechanically, to its gravity and atmospheric conditions. Leaving the Earth and its protective environment, therefore, results in the degradation of a number of human systems. Long-duration stays on the International Space Station (ISS) are accompanied by significant effects on crew's cardiovascular, vestibular and musculoskeletal systems. Bone loss and muscle atrophy are experienced at a rate of 1-3% and 5% per month respectively, while VO2 (oxygen consumption) measurements are reduced by approximately 25% after a

Changes in the central nervous system during long-duration space flight: implications for neuro-imaging.

The purpose of this paper is to review the potential functional and morphological effects of long duration space flight on the human central nervous system (CNS) and how current neuroimaging techniques may be utilized to study these effects. It must be determined if there will be any detrimental changes to the CNS from long term exposure to the space environment if human beings are to plan interplanetary missions or establish permanent space habitats. Research to date has focused primarily on the short term changes in the CNS as the result of space flight. The space environment has many factor

Biomedical results of the Space Shuttle orbital flight test program.

On July 4, 1982, the Space Shuttle Columbia landed at Edwards Air Force Base, CA, thus successfully completing the fourth and last in a series of Orbital Flight Tests (OFT) of the Space Transportation System (STS). The primary goal of medical operations support for the OFT was to assure the health and well-being of flight personnel during all phases of the mission. To this end, crew health status was evaluated preflight, inflight, and postflight. Biomedical flight test requirements were completed in the following areas: physiological adaptation to microgravity, cabin acoustical noise, cabin at

Analysis of human microcirculation in weightlessness: Study protocol and pre-study experiments.

BACKGROUND: In weightlessness, alterations in organ systems have been reported. The microcirculation consists of a network of blood vessels with diameters of a few μm. It is considered the largest part of the circulatory system of the human body and essential for exchange of gas, nutrients and waste products. An investigation of the microcirculation in weightlessness seems warranted but has not yet been performed. OBJECTIVE: In this paper, we outline a study in which we will investigate the possible interrelations between weightlessness and microcirculation. We will induce weightlessness in t

Autonomic reaction to vestibular damage.

The vestibular system provides inputs to many neurons in the brain stem that participate in autonomic control. This multiplicity of vestibular-autonomic connections plays a variety of roles. Whereas it has been known for decades that unilateral vestibular lesions can result in motion sickness, recent data suggest that the vestibular system participates in making adjustments in blood pressure and respiration that are necessary to maintain homeostasis during movement and changes in posture. Animals with bilateral vestibular lesions are more susceptible to posturally related hypotension than vest

Localization of scopolamine induced electrocortical brain activity changes, in healthy humans at rest.

To prevent the participants of parabolic flights from nausea they are optionally treated with subcutaneously injected antiemetic scopolamine. The range of side effects of this substance include reductions of the α-2 frequency band. Previous studies, however, have given no information as to which generator centers are responsible for this effect. The objective of this study therefore, is to identify the cortex area that may be responsible for this effect. Six participants were subcutaneously administered 0.7 mg of scopolamine. EEG was recorded for 10 minutes before to 20 minutes after injection

The effects of vestibular stimuli on brain cognitive processing: an ERP study.

Cognition impairment caused by space motion sickness often is a risk of spaceflight. So the dynamic changes of pattern of brain cognitive processing under varied vestibular stimuli was explored using event-related potentials. Vestibular stimulation was produced by rotary chair and varied linearly, i.e. control, constant 10 degrees/s rotation and constant 0.6 degrees/s, 0.8 degrees/s, 1.0 degrees/s, 1.2 degrees/s acceleration. Thirty-three subjects participated in the study and performed same auditory selective attention task in which the odd numbers in a randomly mixed series of odd and even n…

Relative bioavailability of scopolamine dosage forms and interaction with dextroamphetamine.

The NASA Reduced Gravity Office (RGO) uses scopolamine (SCOP) alone and in combination with dextoamphetamine (DEX) to treat motion sickness symptoms during DC-9 parabolic flights. The medications are sometimes dispensed as custom dosage forms in gelatin capsules for convenience. Reports of treatment failure during flights by the flight surgeons suggest that these formulations may be less efficacious for the treatment of motion sickness due to unreliable and inadequate bioavailability. We estimated bioavailability of four different oral formulations used by the NASA RGO physicians for the treat

Optical and gravito-inertial contributions to the perception and control of height in a simulated Low-Altitude Flight context.

Low-Altitude Flight (LAF) is a flight formation consisting of rapid close ground flight. Perception and control of self-motion, allowing for optimal information collection and rapid adaptation, are of fundamental importance during LAF, but remain largely unexplored. This study aimed to analyse the impact of visuo-vestibular stimuli on the monitoring of height in a motion-based simulated LAF context. Thirteen non-pilots were tested in different environmental conditions, in which optical and gravito-inertial (GI) information were manipulated. The visual environment, displayed with a VR headset,

Hypergravity studies in the Netherlands.

It looks like that with the utilization phase of the International Space Station (ISS) scientists will have the possibility to perform long duration and more sophisticated microgravity experiments than could be performed previously. In preparation for these spaceflight studies, ground based experiment tools for simulated (or real) microgravity and hypergravity are important. To provide the infrastructure and user support necessary to perform these ground based studies we have setup the Dutch Experiment Support Center, DESC. This paper will focus on the three Dutch centrifuge facilities. It is

Clinical verification of a unilateral otolith test.

In a previous study we reported promising results for a new test to differentiate in vivo unilateral otolith functions. That study pointed to a need for further validation on known pathological cases. In this presentation we will detail the results gathered on a group of clinically verified vestibular defectives (verum) and a normal (control) group. The subjects in the verum group were former patients of the ENT clinic of the university hospital. These subjects had usually suffered from neurinoma of the VIIth cranial nerve or inner ear infections. All had reguired surgical intervention includi

The Canadian Forces Airsickness Rehabilitation Program, 1981-1991.

Airsickness is a significant obstacle in the training of some student pilots. When conventional therapy fails, desensitization therapy may be indicated. Using experience gained by the RAF and USAF, the Canadian Forces (CF) began such a program in 1981. This paper reports program results from 1981-1991. Following subject identification, treatment consists of three phases: biofeedback relaxation therapy, ground-based desensitization training and in-flight desensitization therapy with a pilot-flight surgeon. Employing a definition of cure used by the RAF, success was compared with that of the RAF
