Calcium-dependent pathway as a primary cause of hypoxic RGC damage in monkey retinal explants.

PURPOSE: Retinal ganglion cells (RGCs) loss or degeneration in the retina is a hallmark of many sight-threatening diseases, including glauco … ith propidium Iodide (PI) to assess RGC damage and with an antibody specific for calpain-specific α-spectrin break down product 150 kDa (SBDP150). Immunoblotting assays were performed for α-spectrin and calpains. RESULTS: The cathepsin inhibitors were confirmed to be specific to cathepsin without calpain inhibitory effects in vitro. The number of PI-posit …

Activation of mu-calpain in developing cortical neurons following methylmercury treatment.

In order to examine the possible involvement of mu-calpain in methylmercury (MeHg)-induced neurotoxicity in developing cortical neurons, we performed biochemical and immunohistochemical studies utilizing two antibodies which specifically recognize the 150-kDa mu-calpain-specific alpha-spectrin breakdown product (SBDP) and the active form of mu-calpain in rats on postnatal day 16. Soluble fractions of the cerebral cortex from control rats exhibited slight immunoreactivity for SBDP. Although the amount of SBDP in the cerebral cortex was only slightly increased the day after the final treatment o…

A novel homozygous splice-site mutation in the SPTBN4 gene causes axonal neuropathy without intellectual disability.

Mutations in spectrin beta non-erythrocytic 4 (SPTBN4) have been linked to congenital hypotonia, intellectual disability and motor neuropathy. Here we report on two siblings with a homozygous splice-site mutation in the SPTBN4 gene, lacking previously reported features of the disorder such as seizures, feeding difficulties, respiratory difficulties or profound intellectual disability. Our findings indicate that muscular hypotonia, myopathic facies with ptosis and axonal neuropathy can be the core clinical features in the SPTBN4 disorder and suggest that SPTBN4 mutation analysis should be consi…

Shapes of Red Blood Cells: Comparison of 3D Confocal Images with the Bilayer-Couple Model.

Cells and organelles are shaped by the chemical and physical forces that bend cell membranes. The human red blood cell (RBC) is a model syst … l model including the MS and real cells, (ii) that weakening the MS, by using urea (which denatures spectrin), leads to the theoretically predicted gradual decrease in spicule number of echinocytes, (iii) that the theory predicts that the MS is essential for stabilizing the discocyte morphology against changes in lipid composition, and that without it, the …

Presence of allele alphaLELY in an Amazonian Indian population.

Allele alphaLELY is a low-expression allele of the erythroid spectrin alpha-chain that is characterized by a C --> G mutation at position alpha1857 in exon 40 and a C --> T (nt -12) mutation in intron 45. This second mutation is probably responsible for the partial skipping of exon 46. This exon is essential for the nucleation of the alpha-chains by the beta-chains during erythropoeisis. Although allele alphaLELY remains asymptomatic in both heterozygotes and homozygotes, it enhances the expression of deleterious alpha-alleles that occur and, as such, has clinical importance. In this study, th…

Improvement of mouse beta thalassaemia by hydroxyurea.

The present report provides evidence that hydroxyurea (HU) improves the beta thalassaemic phenotype in mice receiving 200 mg/kg/d for 30 d. The haematocrit rose from 29 +/- 3% at day 0 to 37 +/- 4% at day 30 (P < 0.05), despite myelosuppression and decreased reticulocyte counts. The beta minor/alpha ratio of globin chain synthesis increased from 0.78 at day 0 to 0.97 at day 30 (P < 0.001). Membrane defects improved: the proportion of bound alpha chains decreased, the proportion of spectrin and ankyrin increased and red cell deformability also increased.

[Hereditary spherocytosis].

Hereditary spherocytosis (HS) is a congenital hemolytic anemia characterized by microspherocytosis, increased osmotic fragility of erythrocytes and a favorable response to splenectomy. It is now clear that the molecular basis of HS is heterogeneous and that several membrane proteins (spectrin, ankyrin, band 3, and band 4.2) are involved in the pathogenesis of HS. A number of gene abnormalities have recently been identified but mutations specific for clinical phenotypes and racial backgrounds are still unclear. In this report, the clinical features of HS, including its diagnosis and treatment, …

Reduced expression of Ankyrin-G and E-cadherin in duodenal mucosal biopsy of subjects with celiac disease.

Confirmatory diagnosis of celiac disease (CD) include histopathology of duodenal biopsy and tissue trans-glutaminase-IgA. Identification of tissue-specific histological markers is warranted to improve the diagnosis. A genetic study in CD identified the association of ankyrin-G that connects E-cadherin with β2-spectrin in epithelial cells of the duodenal tissue. We attempted to investigate the differential expression of ankyrin-G, E-cadherin and β2-spectrin in duodenal biopsy of CD subjects compared to non-CD controls. Duodenal tissue was collected from 83 study participants, of which 50 were C…

Phosphoinositide signalling system in human erythrocyte and its role in cell pathology.

Human erythrocytes contain 217.2 +/- 70.5 pmole of inositol 1,4,5-trisphosphate per ml of packed cells (n = 14). The increased generation of Ins 1,4,5P3, was induced by 1 microM A 23187, 20 microM Pb2+, and by AlF4-. Ins 1,4,5P3, Pb2+ and AlF4- evoke shape changes, disorganisation of spectrin network and vesiculation. In erythrocytes of patients with hereditary spherocytosis the level of Ins 1,4,5P3 was increased to 556.7 +/- 374 pmole per ml of packed cells. We suggest that activation of the phosphoinositide signalling system may represent a common denominator for many divergent stimuli and d…
