Relationships between the characteristics of endogenous luteinizing hormone surge and the degree of ovarian hyperstimulation during superovulation induction in women.

Ovarian hyperstimulation was induced in 17 normally cycling women undergoing in-vitro fertilization (IVF) and embryo transfer with clomiphene (9 cycles), clomiphene followed by pulsatile hMG (12 cycles) or clomiphene followed by pulsatile FSH (11 cycles). Hyperstimulation was greater with the combined treatments than with clomiphene alone. In all 32 cycles an endogenous LH surge occurred. The peak values and the duration of the LH surge showed significant negative correlations with the plasma oestradiol levels, the number of the follicles and the total follicular fluid volume aspirated at lapa

[Positive chronotropic and inotropic effects of stimulation of the vagus nerve centers in the frog heart].

In frogs, positive inotropic effects are transmitted catecholaminergically as they have been blocked with propranolol but not with atropine. Positive chronotropic effects were in some experiments blocked with atropine and not with propranolol (a "cholinergic type" of of positive chronotropic effect), the opposite occurred in other experiments (an "adrenergic type"). The cholinergic positive chronotropic effects were observed with the initial slow heart rate and were accompanied by negative inotropic effects; the chronotropic effect itself appeared suddenly (with no gradual acceleration of hear

The feminisation of gonadotrophin responses in intact male transsexuals.

In castrated animals and hypogonadal men other workers have shown an LH surge following oestrogen administration similar to that found in the normal female. However, there is no evidence that this can be achieved in intact males. We have therefore studied male transsexual patients before and after chronic oestrogen therapy given for at least three months to feminise the body habitus before undergoing plastic surgery. Subjects were assessed for the acute effects of oestradiol valerate on sex steroid hormone levels and gonadotrophin responses to gonadotrophin releasing hormone. The results showe

Electrical Stimulation Elicits Neural Stem Cells Activation: New Perspectives in CNS Repair.

Researchers are enthusiastically concerned about neural stem cell (NSC) therapy in a wide array of diseases, including stroke, neurodegenerative disease, spinal cord injury, and depression. Although enormous evidences have demonstrated that neurobehavioral improvement may benefit from NSC-supporting regeneration in animal models, approaches to endogenous and transplanted NSCs are blocked by hurdles of migration, proliferation, maturation, and integration of NSCs. Electrical stimulation (ES) may be a selective non-drug approach for mobilizing NSCs in the central nervous system. This technique i

[Characteristics of esophageal antiperistaltic activity in sheep].

Oesophageal motility was investigated in standing conscious sheep. A manometric method was used in four animals; two sheep, fitted with permanent electrodes at four levels, were used for oesophageal electromyography. The occurrence of retrograde peristalsis (mean speed 71,5 cm.s-1) at the beginning of every rumination cycle is reasserted. The characteristics of this activity are pointed out, with reference to normal peristalsis during the deglutition of saliva. Different types of stimulation have been compared. Retrograde peristalsis (mean speed 66 cm.s-1) can be triggered readily in non-rumin

Arabidopsis thaliana triggers Phelipanche ramosa germination via isothiocyanates under negative strigolactone regulation.

Parasitic weeds are major agricultural threats through fine-tuned chemical interactions with their hosts. We investigated the germination-stimulating metabolites released by Arabidopsis thaliana germinating seeds into the spermosphere, the zone surrounding the seeds and showed that host-parasite chemical recognition can be initiated at the earliest stages of host seed germination. Through co-germination assays, biochemical fractionation of host germinating seed exudates and a mutant approach targeting the glucosinolate (GSL) and strigolactone (SL) pathways, we demonstrated that germinating A.

High-throughput cytokine detection platform for evaluation of chemical induced microglial activation.

Environmental chemical exposure, such as pesticides and heavy metals, may contribute to neurodegenerative disorders through neuroinflammation. This study aims to identify suitable in vitro microglial models for assessing cytokine responses to potential neurotoxicants, particularly focusing on human induced pluripotent stem cell-derived microglia (hiMG). In this study, we evaluated the cytokine secretion profiles of four microglial cell types-hiMG, HMC3, IM-HM, and BV2-upon stimulation with lipopolysaccharides (LPS) using cytokine arrays. Our findings showed cytokine response patterns in hiMG c

Antinociceptive Effect of Spirocyclopiperazinium Salt Compound DXL-A-24 and the Underlying Mechanism.

The antinociceptive effects of spirocyclopiperazinium salt compound DXL-A-24 on neuropathic pain and chemical-stimulated pain were investigated in this study. After the administration of DXL-A-24, the paw withdrawal latency (PWL) and mechanical withdrawal threshold (MWT) were increased in rats suffering from neuropathic pain (chronic constriction injury, CCI) on days 1, 3, 5, 7 and 14 after surgery, and pain responses were inhibited in mice stimulated with chemicals (formalin or acetic acid). In the analysis of antinociceptive targets, the effect of DXL-A-24 was blocked by a peripheral nicotin

[Critical role of intracellular calcium in plasticity mechanisms of defense behavior command neurons LPl1 and PPl1 of Helix lucorum during nociceptive sensitization].

The role of intracellular calcium in changes in excitability and responses of defense behavior command neurons LP11 and PP11 of Helix lucorum to sensory stimulation was investigated in semi-intact preparation of a snail during nociceptive sensitization. It was found that application of sensitizing stimuli onto the snail's head initiated membrane depolarization, increase in its excitability as well as depression of neural responses evoked by sensory stimuli in short-term period of sensitization and significant facilitation of neural responses in long-term period of sensitization. To elucidate t

Supplementation With Phytogenic Compounds Modulates Salivation and Salivary Physico-Chemical Composition in Cattle Fed a High-Concentrate Diet.

Saliva facilitates feed ingestion, nutrient circulation, and represents an important pH buffer for ruminants, especially for cattle fed high-concentrate diets that promote rumen acidification. This experiment evaluated the short-term effects of nine phytogenic compounds on salivation, saliva physico-chemical composition as well as ingested feed boli characteristics in cattle. A total of nine ruminally cannulated Holstein cows were used. Each compound was tested in four of these cows as part of a high-concentrate meal (2.5 kg of total mixed ration in dry matter basis for 4 h) in low or high dos

Chemotactic Behaviors of Vibrio cholerae Cells.

Vibrio cholerae, the causative agent of cholera, swims in aqueous environments with a single polar flagellum. In a spatial gradient of a chemical, the bacterium can migrate in "favorable" directions, a property that is termed chemotaxis. The chemotaxis of V. cholerae is not only critical for survival in various environments and but also is implicated in pathogenicity. In this chapter, we describe how to characterize the chemotactic behaviors of V. cholerae: these methods include swarm assay, temporal stimulation assay, capillary assay, and receptor methylation assay.

Simplified drug efficacy evaluation system for vasopressin neurodegenerative disease using mouse disease-specific induced pluripotent stem cells.

Familial neurohypophyseal diabetes insipidus (FNDI) is a degenerative disorder in which vasopressin-secreting neurons degenerate over time due to the production of mutant proteins. We have demonstrated therapeutic effects of chemical chaperones in an FNDI mouse model, but the complexity and length of this evaluation were problematic. In this study, we established disease-specific mouse induced pluripotent stem cells (iPSCs) from FNDI-model mice and differentiated vasopressin neurons that produced mutant proteins. Fluorescence immunostaining showed that chemical chaperones appeared to protect v

Mechanism of action of 4-substituted phenols to induce vitiligo and antimelanoma immunity.

Monobenzone is a 4-substituted phenol that can induce vitiligo and antimelanoma immunity. We investigated the influence of the chemical structure on the biological activity of a series of structurally related 4-substituted phenols. All phenols inhibited cellular melanin synthesis, and eight of ten phenols inhibited tyrosinase activity, using the MBTH assay. These phenols also induced glutathione (GSH) depletion, indicative of quinone formation and protein thiol binding, which can increase the immunogenicity of melanosomal proteins. Specific T-cell activation was found upon stimulation with phe

Furthering the debate on the role of interstitial cells of Cajal in enteric inhibitory neuromuscular neurotransmission.

The gut, a muscular organ, performs a critical role in transporting ingested contents, yet it is also controlled to periodically stop transport to maximize digestion and toxin detection. The complex intraluminal composition and rheology challenge the mechanistic requirements of inhibitory neuromuscular neurotransmission. The interstitial cells of Cajal (ICCs)-generated slow wave may tune the promiscuous luminal chemical environment, which prepares the smooth muscle membrane potential for a depolarizing or hyperpolarizing response as needed. Slow waves are abolished during stimulation-induced i

Antinociceptive effect of the monoterpene R-(+)-limonene in mice.

In the present study were studied the antinociceptives properties of monoterpene R-(+)-limonene (LM) in chemical and thermal models of nociception in mice. The R-(+)-limonene was administered, intraperitoneally (i.p.), at doses of 25 and 50 mg/kg. The results showed significant inhibition produced on chemical nociception induced by intraperitoneal acetic-acid and in the second phase of subplantar formalin test, but did not manifest a significant effect in hot-plate test. The R-(+)-limonene-induced antinociception in second phase of formalin test was insensitive to naloxone (1 mg/kg, s.c.). It

Synaptic mechanisms in the kindled epileptic focus: a speculative synthesis.

This chapter reviews the chemical kindling model of epilepsy and speculates on its significance. Both human and experimental epilepsies are extremely heterogeneous, and it is unlikely that a single molecular or cellular mechanism can account for such a diversity of behavioral manifestations. Recent studies of chemical kindling favor the view that in this model, epilepsy is a property of neuronal networks that can take place in a structurally intact brain and does not depend on the presence of gross or microscopic brain damage. Kindling can be obtained by daily injections of nanomolar amounts o

Vestibular functional assessment using the caloric test in mice with arsanilic acid-induced vestibular dysfunction.

The vestibular system contributes to postural control, eye movements, spatial perception, and autonomic regulation. Its dysfunction causes vertigo, nausea, postural instability, which gradually improve via central plasticity known as vestibular compensation. Experimental vestibular compensation studies commonly use unilateral vestibular neurectomy or chemical/labyrinthectomy models, with vestibular function assessed by vestibulo-ocular reflex (VOR) analysis, vestibular evoked myogenic potentials, and vestibular sensory evoked potentials. However, these methods require specialized equipment. In
