Plasma disposition, concentration in the hair, and anthelmintic efficacy of eprinomectin after topical administration in donkeys.

OBJECTIVE: To investigate plasma disposition, concentration in the hair, and anthelmintic efficacy of eprinomectin after topical administration in donkeys. ANIMALS: 12 donkeys naturally infected with strongyle nematodes. PROCEDURES: The pour-on formulation of eprinomectin approved for use in cattle was administered topically to donkeys at a dosage of 0.5 mg/kg. Heparinized blood samples and hair samples were collected at various times between 1 hour and 40 days after administration. Samples were analyzed via high-performance liquid chromatography with fluorescence detection. Fecal strongyle

Heligmosomoides polygyrus reduces infestation of Ixodes ricinus in free-living yellow-necked mice, Apodemus flavicollis.

Free-living animals are usually inhabited by a community of parasitic species that can interact with each other and alter both host susceptibility and parasite transmission. In this study we tested the prediction that an increase in the gastrointestinal nematode Heligmosomoides polygyrus would increase the infestation of the tick Ixodes ricinus, in free-living yellow-necked mice, Apodemus flavicollis. An extensive cross-sectional trapping survey identified a negative relationship between H. polygyrus and I. ricinus counter to the prediction. An experimental reduction of the nematode infection

Immunological diagnosis of human angiostrongyliasis due to Angiostrongylus cantonensis (Nematoda: Angiostrongylidae).

Angiostrongylus cantonensis-associated eosinophilic meningitis in humans has been commonly reported worldwide. However parasitologically confirmed cases are not common, as the parasite has been recovered only infrequently from the cerebrospinal fluid of patients. The potential value of immunodiagnosis is therefore self-evident. Immunological tests can also help in the differential diagnosis of parasitic (particularly helminths) infections that cause eosinophilic meningitis. This paper summarizes the state of and advances in the immunological diagnosis of human angiostrongyliasis due to Angiost

[Development of PCR assay for detection of Angiostrongylus cantonensis in Pomacea canaliculata].

OBJECTIVE: To establish a PCR assay for detecting the third-stage larvae of Angiostrongylus cantonensis in Pomacea canaliculata. METHODS: Polymerase chain reaction primers were designed by the software Lasergene, based on the specific cDNA of the third-stage larvae of A.cantonensis in Genbank. The total RNA was prepared from the third-stage larvae of A.cantonensis and of the snails by TRIzol one-step protocol. Amplification by RT-PCR was carried out following the kit protocol. RESULTS: RT-PCR assay revealed a clear differentiation between infected and negative snails. When a mixture of the t

Host immune status affects maturation time in two nematode species--but not as predicted by a simple life-history model.

In theory, the age at which maturation occurs in parasitic nematodes is inversely related to pre-maturational mortality rate, and cross-species data on mammalian nematodes are consistent with this prediction. Immunity is a major source of parasite mortality and parasites stand to gain sizeable fitness benefits through short-term adjustments of maturation time in response to variation in immune-mediated mortality. The effects of thymus-dependent immune responses on maturation in the nematode parasites Strongyloides ratti and Nippostrongylus brasiliensis were investigated using congenitally thym

Lung parasites of shrews from Pennsylvania.

We examined lung parasites of three species of soricids, Sorex cinereus (n = 58), Sorex fumeus (n = 23) and Blarina brevicauda (n = 45) collected from Pennsylvania (USA), from 1990 to 1995. Yeast-like cells of Hisfoplasma capsulatum var. capsulatum were found in lung sections stained with Grocott's modification of Gomori's methenamine silver, periodic acid-Schiff, Giemsa, and hematoxylin-eosin in two (3%) S. cinereus, eight (35%) S. fumeus and two (4%) B. brevicauda. The number of spores of H. capsulatum in the lungs was low and no inflammatory reaction was evident. The infection was not disse

Infectious complications of human T cell leukemia/lymphoma virus type I infection.

Infection with human T cell leukemia/lymphoma virus type I (HTLV-I) has been etiologically associated with two diseases: adult T cell leukemia and HTLV-I-associated myelopathy/tropical spastic paraparesis. Increasing evidence suggests that HTLV-I infection may be associated with immunosuppression and, as a consequence, affect the risk and expression of several other infectious diseases, of which the best studied are strongyloidiasis, tuberculosis, and leprosy. In strongyloidiasis, coinfection with HTLV-I appears to result in a higher rate of chronic carriage, an increased parasite load, and a

Eosinophilic meningitis caused by Angiostrongylus cantonensis: case report in a patient with false-positive immunological test result for Neisseria meningitides.

Eosinophilic meningitis caused by Angiostrongylus cantonensis is an uncommon disease in Brazil that occurs by ingesting slugs, water or food contaminated with the parasite. Here we report a case of a 64-year-old patient with neck stiffness, headache, fever, and peripheral and cerebrospinal fluid (CSF) eosinophilia, compatible with eosinophilic meningitis. He had a false-positive result for Neisseria meningitides and received unsuccessful treatment with antibiotics. After extensive investigation, DNA and anti-A.cantonensis antibodies were detected and the patient was successfully treated with p

Diagnostic Investigation of Two Cases of Oslerus osleri Infection in Dogs: Microscopic Challenges and Molecular Characterisation.

BACKGROUND: The diagnosis of certain lungworm infections in dogs relies heavily on bronchoalveolar lavage (BAL) cytology or lesion histopathology, especially in situations in which faecal diagnostics and bloodwork may have limited value. Differentiating morphologically similar parasitic larvae on BAL cytology, such as Oslerus osleri or Filaroides hirthi, may be challenging, and molecular approaches are not yet widely available. OBJECTIVE: Description of two cases of O. osleri infection in young dogs, including (1) representative examples of BAL cytology and lesion-specific histopathologic fin

When pooling fails: low-intensity Angiostrongylus cantonensis infections in Theba pisana result in false-negativity.

Pooling of gastropod samples is frequently used in surveys of the neurotropic nematode Angiostrongylus cantonensis to reduce cost and processing time. Here we highlight a potential limitation of this technique using the snail Theba pisana as an example. Pooled samples tested negative using a high-sensitivity qPCR assay, whereas subsequent individual testing revealed positive cases. This pattern is consistent with a dilution effect, where low parasite DNA concentrations could lead to false-negative results. These findings suggest that pooling may reduce diagnostic sensitivity in regions where i

The developmental lipidome of Nippostrongylus brasiliensis.

BACKGROUND: Nippostrongylus brasiliensis-a nematode of rodents-is commonly used as a model to study the immunobiology of parasitic nematodes. It is a member of the Strongylida-a large order of socioeconomically important parasitic nematodes of animals. Lipids are known to play essential roles in nematode biology, influencing cellular membranes, energy storage and/or signalling. METHODS: The present investigation provides a comprehensive, untargeted lipidomic analysis of four developmental stages/sexes (i.e. egg, L3, adult female and adult male stages) of N. brasiliensis utilising liquid chrom

New data on the life cycle of Nippostrongylus brasiliensis (Travassos, 1914) (Nematoda: Heligmosomidae): development of eggs and larval stages in the intestine of naturally infected Rattus norvegicus (Berkenhout, 1769).

Nippostrongylus brasiliensis is an intestinal parasite of rodents with a worldwide distribution. Due to the similarities between its life cycle and that of the human hookworms Ancylostoma duodenale and Necator americanus, N. brasiliensis has been widely used as a model in experimental research. We provide new data on the embryonation and hatching processes of N. brasiliensis eggs, not in the soil, but in the large intestine of naturally infected Rattus norvegicus. The parasite was investigated in 109 rats trapped in urban and periurban areas of Valencia, Spain. Rats were preserved at - 20 °C.

Liver-draining portal lymph node responds to enteric nematode infection by generating highly parasite-specific follicular T helper and B cell responses.

INTRODUCTION: While research on the gut-liver axis in non-communicable liver diseases has expanded exponentially, few studies have investigated the liver-gut relationship in the context of gastrointestinal nematode infections. This study aimed to determine whether liver-draining lymph nodes (LLNs) contribute to the immune response against a strictly enteric nematode infection. METHODS: We analyzed the cellular and functional immune responses in the portal (PLN) and celiac (CLN) liver-draining lymph nodes following infection with the small intestinal nematode Heligmosomoides (polygyrus) bakeri

Efficacy of a PP2A vaccine for Angiostrongylus costaricensis against rat lungworm disease caused by Angiostrongylus cantonensis in wild-caught rats (Rattus rattus) in Hawaii.

The nematode Angiostrongylus cantonensis is a rat lungworm, a zoonotic pathogen that causes an emerging infectious disease known as neuroangiostrongyliasis or rat lungworm disease. This study evaluates the efficacy of a vaccine developed for a related species, Angiostrongylus costaricensis, to A. cantonensis in the definitive rat host. Wild-caught rats (Rattus rattus) (n = 28) were mated in captivity to produce uninfected F1 progeny. A total of 43 F1 rats were involved in this trial; 20 non-vaccinated, 21 vaccinated, and two unvaccinated, uninfected. F1 offspring in the vaccinated group were i

Heligmosomoides bakeri and Toxoplasma gondii co-infection leads to increased mortality associated with changes in immune resistance in the lymphoid compartment and disease pathology.

Co-infections are a common reality but understanding how the immune system responds in this context is complex and can be unpredictable. Heligmosomoides bakeri (parasitic roundworm, previously Heligmosomoides polygyrus) and Toxoplasma gondii (protozoan parasite) are well studied organisms that stimulate a characteristic Th2 and Th1 response, respectively. Several studies have demonstrated reduced inflammatory cytokine responses in animals co-infected with such organisms. However, while general cytokine signatures have been examined, the impact of the different cytokine producing lymphocytes on

Angiostrongylus cantonensis infection induces MMP-9 and causes tight junction protein disruption associated with Purkinje cell degeneration.

Angiostrongylus cantonensis causes a human central nervous system (CNS) infection characterized by eosinophilic meningitis or meningoencephalitis. Individuals infected with A. cantonensis exhibit unbalanced walking. The mechanism of extensive neurological impairments of hosts caused by A. cantonensis larvae remains unclear. Tight junction proteins (e.g., claudin-5 and zonula occludens-1) are the most important regulators of paracellular permeability and cellular adhesion. In a previous study, we found that increased matrix metalloproteinase-9 (MMP-9) activity may be associated with blood-CNS b

BALB/c mice infected with Angiostrongylus cantonensis: A new model for demyelination in the brain.

In this study, we present a new model for demyelination of the central nervous system (CNS). BALB/c mice were infected with Angiostrongylus cantonensis and analyzed 7, 14, and 21 days postinfection. Neurological scale evaluation, magnetic resonance imaging (MRI), histology, real-time quantitative polymerase chain reaction, and western blotting were all performed on days 7, 14, and 21. The results showed that the neurological functions and weight of A. cantonensis-infected mice decreased markedly after 21 days of infection. MRI showed subdural effusion and white high signals in the corpus callo

Managing anthelmintic resistance in cyathostomin parasites: Investigating the benefits of refugia-based strategies.

Selective anthelmintic therapy has been recommended as a sustainable strategy for cyathostomin control in horse populations for several decades. The traditional approach has been to determine strongyle fecal egg counts (FEC) for all horses, with treatment only recommended for those exceeding a predetermined threshold. The aims are to achieve a reduction of overall egg shedding, while leaving a proportion of the herd untreated, which lowers anthelmintic treatment intensity and reduces selection pressure for development of anthelmintic resistance. This study made use of the cyathostomin model to
