Irisin-integrin αV/β5 coupling of α-synuclein phagocytosis and clearance.

Parkinson's disease-associated cognitive impairment (PD-CI) is closely linked to α-synuclein (α-syn) accumulation and synaptic dysfunction, yet effective disease-modifying strategies remain limited. Irisin is an exercise-inducible myokine with neuroprotective potential, but its receptor mechanisms and its role in α-syn clearance in PD-CI are poorly defined. Here, we observed that aerobic exercise markedly increased circulating irisin levels, reduced serum α-syn levels, and improved cognitive performance in a cohort of 21 PD patients. In addition, irisin signals through integrin αV/β5 to enhanc

α-Synuclein Seeds Amplification Assays for Parkinson's Disease and Related Synucleinopathies.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by the pathological misfolding and aggregation of α-synuclein (α-syn), which leads to dopaminergic neuronal loss and multisystem dysfunction. Conventional clinical diagnosis is often delayed until after substantial neuronal degeneration has occurred. Recent advances in α-syn seed amplification assays (α-syn SAAs), including real-time quaking-induced conversion (RT-QuIC) and protein misfolding cyclic amplification (PMCA), have revolutionized PD biomarker research. These assays exploit the prion-like seeding and p

Compartment-specific correlation of pathological α-synuclein in prodromal Parkinson's disease.

INTRODUCTION: Neuronal α-synuclein dyshomeostasis and aggregation are essential features of early Parkinson's disease, as seen in isolated REM sleep behavior disorder (iRBD). The link between these pathologies across body compartments remains unclear. Our aim was to assess correlations between α-synuclein aggregates in stool and urine with α-synuclein content in neuronally derived L1CAM extracellular vesicles (L1EVs) from serum in iRBD. METHODS: We conducted a cross-sectional study, analyzing concurrent biobank samples from 46 individuals with iRBD to quantify α-synuclein aggregates in stool

Optimization of α-Synuclein and Tau Detection by Immunoblot in Enteroendocrine Cell Lines.

BACKGROUND: Enteroendocrine cells (EECs) are dispersed along the intestinal mucosa and transduce luminal stimuli into hormonal signals. EECs exhibit neuron-like features and express both α-synuclein and tau, two proteins pathologically and genetically linked to Parkinson disease (PD). These observations support the hypothesis that EECs may be involved in disease development in the "body-first" PD subtype. Cellular models represent invaluable tools for studying the role of α-synuclein and tau in PD pathogenesis. However, the sensitivity and specificity of commercial antibodies for detecting α-s

Glucosylceramide-induced ectosomes propagate pathogenic α-synuclein in Parkinson's disease.

The intercellular transmission of α-synuclein contributes to Parkinson's disease pathology. Yet, the mechanisms of α-synuclein spread are not fully understood. Here we used live-cell microscopy to examine the impact of Parkinson's disease associated lipid alterations on α-synuclein release. We discovered that increased glucosylceramides as a consequence of reduced β-glucocerebrosidase activity induce ectosome shedding from primary neurons and from dopaminergic neurons derived from induced pluripotent stem cells of a patient with Parkinson's disease harbouring mutations in GBA1 (N370S, L444P an

Fluorescence Detection of Alpha-Synuclein Aggregates in the Gut Using a Peptide Probe.

Parkinson's disease (PD) is diagnosed clinically by motor symptoms, with no clinical molecular diagnostic test currently available. By the time motor symptoms manifest, significant irreversible neurodegeneration would have already occurred, limiting the effectiveness of the therapies. Recent identification of pathological α-synuclein (α-syn) aggregates in the gastrointestinal tract of prodromal PD patients offers a potential avenue for early diagnosis. This study aims to explore specific fluorescence labeling of α-syn aggregates in the gastrointestinal tract using a peptide-based probe for ear

The α-synuclein proteostasis network and its translational applications in Parkinson's disease.

Parkinson's disease (PD) is a debilitating neurodegenerative condition that results in a loss of mobility and muscle control. A neuropathological hallmark of PD is the presence of aberrant inclusions, known as Lewy pathology, of which α-synuclein (α-Syn) is a major component. The accumulation of α-Syn may be due to an imbalance in the proteostasis system regulating α-Syn. To investigate this hypothesis, we delineated the proteostasis network (PN) of α-Syn in the human substantia nigra at the proteomic and transcriptomic level. We then defined an α-Syn proteostasis activity score (PAS) that qua

Diffusion MRI and α-Synuclein Seed Amplification Status in Parkinson's Disease.

OBJECTIVE: Positive α-synuclein seed amplification assay (SAA) is a biomarker found in most people with Parkinson's disease (PD). We explored if free-water (FW) imaging detects microstructural differences in the brains of patients with early PD with SAA+ or SAA- status. METHODS: We studied patients with PD with baseline diffusion imaging and α-synuclein SAA data from the Parkinson's Progression Markers Initiative (PPMI). We compared FW, FW corrected fractional anisotropy (FAT), and clinical characteristics between SAA+ and SAA- groups. We also applied the Automated Imaging Differentiation for

An automated workflow for quantifying the formation of synuclein aggregates in human dopaminergic neurons.

Parkinson's disease (PD) is a neurodegenerative disorder characterized by alpha-synuclein (α-syn) aggregates termed Lewy bodies. To model PD pathology in vitro, preformed fibrils of α-syn (PFFs), which can be taken up by cells, provide a seed that drives misfolding and aggregation of endogenous α-syn, with new aggregates amplifying this process. External application of PFFs to dopaminergic neurons (DNs) increases aggregate formation, marked by α-syn phosphorylation at serine 129 (pS129-syn), a pathological PD marker. Building on this, we developed an automated synuclein seeding assay to quanti

α-Synuclein aggregation landscape from phase separation to neurotoxic intermediates.

The aberrant aggregation of α-synuclein (αS) into insoluble amyloid fibrils is a hallmark of Parkinson's disease. Despite recent advances in characterising the properties of mature αS fibrils, the transient and heterogeneous intermediates that underlie cellular toxicity remain largely elusive. Here, we review the mechanistic principles of αS aggregation, focussing on liquid-liquid phase separation (LLPS) as a critical intermediate step. We discuss how the structural evolution of αS within the condensed phase governs the subsequent patterns of cellular dysfunction and pathological propagation.

CHCHD2 links mitochondrial dysfunction and α-synuclein misfolding in Parkinson's disease.

Parkinson's disease comprises multiple biological subtypes and a heterogeneous clinical course. A recent study by Liao et al. identifies CHCHD2 mutations as a mitochondrial entry point that links metabolic dysfunction to α-synuclein pathology. These findings highlight how rare sporadiclike monogenic forms of Parkinson's disease may inform mechanistic and therapeutic stratification.

A Systematic Comparison of Alpha-Synuclein Seed Amplification Assays for Increasing Reproducibility.

Seed amplification assays (SAAs) enable ultrasensitive detection of misfolded α-synuclein across biofluids and tissues. Yet, heterogeneity in protocols limits cross-study comparability and clinical translation. Here, we review α-synuclein SAA methods and their performance across various biological matrices. We catalog key variables (e.g., human recombinant α-synuclein variants, buffers, salts, beads, shaking, incubation temperature) and outline pre-analytical factors that shape outcomes. We propose a prioritized framework of key analytical and pre-analytical variables most likely to influence

Molecular profiling of alpha-synuclein pathology and seeding activity in Parkinson's disease.

Parkinson's disease (PD) is neuropathologically characterized by the abnormal accumulation of fibrillar alpha-synuclein (aSyn) within selectively vulnerable neuronal populations. Although this pathological hallmark is shared across individuals with PD, the disease presents with marked clinical heterogeneity in age of onset, progression rate, and clinical symptoms, the molecular basis of which remains incompletely understood. In this study, we examined whether biochemical and seeding-related properties of aSyn vary across clinically defined PD subgroups. Using well-characterized, autopsy-confir

When Pathologies Collide: The Intersection of Tau and Alpha-Synuclein in Neurodegenerative Diseases.

Neurodegenerative diseases, including Alzheimer's disease (AD) and Parkinson's disease (PD), are becoming increasingly prevalent in today's aging population and with significant cost to society. While these diseases were traditionally defined by distinct protein aggregates, namely, tau tangles and amyloid-β (Aβ) plaques in AD and α-synuclein (α-syn) inclusions in PD, substantial evidence reveals frequent mixed pathologies, with tau, α-syn, and Aβ aggregates coexisting in patients. This copathology complicates understanding disease mechanisms, classification, and progression, highlighting the n

Alpha-Synuclein in Neurodegeneration: From Shared Biology to Disease-Specific Phenotypes.

Alpha-synuclein (αSyn) is one of the most abundant proteins in the nervous system and is currently associated with devastating synucleinopathies, yet its biology extends far beyond this. In this review, we suggest that αSyn-driven disease emerges within specific neural circuits through the combined effects of cell-type-specific roles, subcellular environments, post-translational modifications (PTMs), and co-pathology. These interacting and additive dimensions, rather than αSyn alone, generate the pathological diversity, shaping whether pathology manifests as Parkinson's disease (PD), Parkinson

Rebalancing α-Synuclein Clearance: Novel Therapeutic Frontiers in Parkinson's Disease.

Parkinson's disease (PD), the second most common neurodegenerative condition, develops because of abnormal protein misfolding and aggregation of α-synuclein with its subsequent intercellular spread. Such pathological changes lead to disruption of neuronal homeostasis and contribute to neuronal degeneration. During normal conditions, α-synuclein clearance is controlled by different types of lysosomal degradation, namely, macro autophagy, chaperone-mediated autophagy (CMA), micro autophagy, and the ubiquitin-proteasome system. Malfunction of these systems results in increased α-synuclein secreti

Enhancing heme synthesis attenuates pathological α-synuclein propagation in vivo.

Intraneuronal α-synuclein (αS) accumulation is a central event in the pathogenesis of Parkinson's disease and dementia with Lewy bodies. The spread of αS pathology throughout the central nervous system contributes to disease progression via seed-dependent propagation, yet disease-modifying therapies remain unavailable. We previously reported that porphyrin compounds inhibit αS aggregation in vitro; however, because of their high molecular weight, porphyrins poorly penetrate the blood-brain barrier (BBB). Heme, a porphyrin derivative, is synthesized from 5-aminolevulinic acid (5-ALA) via the he

Phosphorylation of α-synuclein directs trafficking of the v-SNARE synaptobrevin-2.

α-Synuclein has been extensively studied for its role in Parkinson's disease, however its native function remains unclear. Several lines of evidence suggest it modulates presynaptic function. As many presynaptic proteins have their functions regulated by phosphorylation, we sought to determine whether α-synuclein, a known phosphoprotein, might similarly have phospho-dependent roles. We identified two residues in α-synuclein, Ser42 and Thr81, that undergo activity-dependent phosphorylation in response to neuronal depolarisation. We employed a molecular replacement strategy, introducing α-synucl

Parkinson's disease physiopathology-beyond the α-synuclein aggregation: a narrative review.

BACKGROUND: Parkinson's disease (PD) is traditionally defined by dopaminergic degeneration and α-synuclein aggregation. However, mounting evidence supports a multifactorial and systemic pathophysiology that extends beyond the central nervous system. This narrative review explores the interconnected mechanisms underlying sporadic PD, including environmental exposures, gut dysbiosis, α-synuclein pathology and propagation, systemic and neuroinflammation, metabolic dysfunctions (notably insulin and lipid metabolism), sleep disturbances, glymphatic impairment, and proteostatic failure. RESULTS: Th

PLK2 disrupts autophagic flux to promote SNCA/α-synuclein pathology.

The aggregation and transmission of SNCA/α-synuclein (synuclein, alpha) is a hallmark pathology of Parkinson disease (PD). PLK2 (polo like kinase 2) is an evolutionarily conserved serine/threonine kinase that is more abundant in the brains of all family members, is highly expressed in PD, and is linked to SNCA deposition. However, in addition to its role in phosphorylating SNCA, the role of PLK2 in PD and the mechanisms involved in triggering neurodegeneration remain unclear. Here, we found that PLK2 regulated SNCA pathology independently of S129. Overexpression of PLK2 promoted SNCA preformed
