Tick infestation and occurrence of Anaplasma phagocytophilum and piroplasms in cattle in the Republic of Serbia.

Ticks transmit important pathogens affecting cattle such as intracellular bacteria of the genus Anaplasma or protozoa of the order Piroplasmida. This study aimed at assessing tick species present on pastures and cattle and determining occurrence of the tick-borne pathogens Anaplasma phagocytophilum and Babesia spp. in cattle. During May and June 2013, ticks and EDTA blood were collected from 185 dairy cows at six locations in Serbia. Ticks were also collected directly from the pastures at four of these locations. The occurrence of A. phagocytophilum was investigated by serological (immunofluor

Sequence Polymorphism of Cytochrome b Gene in Theileria annulata Tunisian Isolates and Its Association with Buparvaquone Treatment Failure.

BACKGROUND: Buparvaquone (BW 720C) is the major hydroxynaphtoquinone active against tropical theileriosis (Theileria annulata infection). Previous studies showed that buparvaquone, similarly to others hydroxynaphtoquinone, probably acts by binding to cytochrome b (cyt b) inhibiting the electron transport chain in the parasite. Several observations suggested that T. annulata is becoming resistant to buparvaquone in many endemic regions (Tunisia, Turkey and Iran), which may hinder the development of bovine livestock in these areas. METHODOLOGY/PRINCIPAL FINDINGS: In the present study we sought

Molecular detection of tick-borne rickettsial and protozoan pathogens in domestic dogs from Turkey.

BACKGROUND: Canine tick-borne parasites have emerged in recent years, showing a wider geographic distribution and increased global prevalence. In addition to their veterinary importance, domestic dogs play an important role in the transmission cycles of some agents by acting as reservoirs and sentinels. This study investigated Babesia, Theileria, Anaplasma, and Ehrlichia species in asymptomatic dogs in ten provinces of Turkey. METHODS: DNA obtained from blood samples collected from 757 domestic dogs (243 stray, 351 shelter, 163 pet) of both sexes and various ages were evaluated using PCR and

Interference between Theileria orientalis and hemotropic Mycoplasma spp. (hemoplasmas) in grazing cattle.

Theileria orientalis and hemotropic Mycoplasma spp. (hemoplasmas) can cause anemia in cattle. Cattle infected with one of these two pathogens tend to resist infection by the other pathogen. This is called the "interference phenomenon". However, the detailed investigation of this phenomenon using molecular techniques has not been performed until now. We used PCR to analyze blood samples from cattle grazing in two pastures, and investigated interference between T. orientalis and hemoplasmas. Results indicate that single infection with T. orientalis or hemoplasmas was more common than co-infectio

Molecular detection of Theileria annae and Hepatozoon canis in foxes (Vulpes vulpes) in Croatia.

An epizootiological field study on tick-borne protozoan infections in foxes (Vulpes vulpes) was carried out in different parts of Croatia. Spleen samples of 191 carcasses of red foxes killed in sanitary hunting, were examined for the presence of hematozoa by polymerase chain reaction (PCR) and subsequent sequencing. The investigation revealed four species of hematozoa in 57 foxes (30%), namely Theileria annae, Theileria sp. 3182/05 and Hepatozoon canis. T. annae was found in 10 foxes (5%), Theileria sp. 3182/05 in a single animal (1%), H. canis in 44 (23%) and Hepatozoon sp. was detected in tw

A molecular phylogeny of the benign Theileria parasites based on major piroplasm surface protein (MPSP) gene sequences.

To investigate the phylogeny of benign Theileria parasites, we determined the complete major piroplasm surface protein (MPSP) gene sequences for 6 benign theilerial organisms, including the first from tick. Sequences were analysed alongside published sequences for 39 benign Theileria parasites, using Bayesian inference and maximum parsimony. All MPSP sequences were 852 nucleotides, except for Gansu, Wuchangbuf, VB01, and VB01; Gansu contained 873 nucleotides, and the other 3 had 855. Deduced amino acid sequences contained 284 residues, except for Gansu (291) and Wuchangbuf, VB01, and VB01 (285

Black rhinoceros (Diceros bicornis) populations in northwestern Namibia are apparently not infected with piroplasms.

Babesiosis is a potentially fatal disease in black rhinoceroses. Blood specimens collected from black rhinoceroses from Etosha National Park (n = 29) and Damaraland (n = 22), Namibia, were subjected to polymerase chain reaction using Theileria and Babesia genus-specific primers and reverse line blot, with negative results. The animals were sparsely infested with ticks. In the absence of suitable prophylactic measures, naïve rhinoceroses would be at risk if translocated to Babesia-endemic areas.

Sequence heterogeneity in the 18S rRNA gene within Theileria equi and Babesia caballi from horses in South Africa.

A molecular epidemiological survey of the protozoal parasites that cause equine piroplasmosis was conducted using samples collected from horses and zebra from different geographical locations in South Africa. A total of 488 samples were tested for the presence of Theileria equi and/or Babesia caballi using the reverse line blot hybridization assay. Ten percent of the samples hybridized to the Theileria/Babesia genus-specific probe and not to the B. caballi or T. equi species-specific probes, suggesting the presence of a novel species or genotype. The small subunit of rRNA gene (18S; approximat

Observations on the treatment of natural haemosporidia infections by total alkaloid of Peganum harmala L. in cattle.

Eighty two cattle naturally infected with haemosporidians were treated with total alkaloid hydrochloride of Peganum harmale L. (0.5 mg/kg/day). Fifty eight cases with Theileria sergenti showed a cure rate of 86%; thirteen cases with Theileria annulata showed a cure rate of 85%; eight cattle infected with Babesia bigemina showed a cure rate of 88% and three cases of Anaplasma marginale were completely cured. The results suggested that the curative effect of total alkaloid of P. harmale was better than that of diminazene aceturate and produced minimal side effects. The alkaloid could also be adm

Prevalence and genetic diversity of Theileria and Anaplasma species infecting cattle in Paraguay.

Bovine theileriosis and anaplasmosis are major tick-borne diseases affecting cattle health and productivity worldwide. Although cattle farming significantly contributes to Paraguay's economy, limited epidemiological data on the causative agents hinder the formulation of disease control measures. To address this challenge, the present study investigated the prevalence, risk factors, and genetic diversity of Theileria annulata, Theileria orientalis, and Anaplasma marginale among cattle populations in Paraguay. Blood DNA samples from 326 cattle in nine departments of Paraguay were screened with t

Transovarial transmission of Babesia ovis in Rhipicephalus bursa, confirmed by multi-generational experiments.

Babesia ovis is a tick-transmitted intraerythrocytic apicomplexan parasite that causes ovine babesiosis, a disease with high economic impact in endemic regions. Although Rhipicephalus bursa is a known biological vector of B. ovis, the dynamics of vertical (transovarial) transmission are poorly understood. In the present study, the transovarial transmission of B. ovis was investigated experimentally in four continuous generations of R. bursa under laboratory controlled conditions. A total of 11 sheep were used during the study. An initial stabilate co-infected with B. ovis and Theileria ovis wa

Comparison of PCR-based methods for the detection of Babesia caballi and Theileria equi in field samples collected in Central Italy.

Equine piroplasmosis (EP) is a disease of equids caused by Theileria equi and Babesia caballi, members of the order Piroplasmida, transmitted by several species of ticks. As the disease is endemic in many countries, a clinical examination or a serological test are required prior to movement of horses to prove freedom from infection and to avoid the introduction of EP with its sanitary and economic impact, especially in areas where it is absent. Currently, numerous diagnostic PCR protocols are available, some of which are recommended by the World Organisation for Animal Health (OIE). In order t

Rapid isothermal duplex real-time recombinase polymerase amplification (RPA) assay for the diagnosis of equine piroplasmosis.

Equine piroplasmosis (EP) is a severe disease of horses caused by the tick-borne protozoa Theileria equi (T. equi) and Babesia caballi (B. caballi). Infectious carriers are not always symptomatic, meaning there is a risk to non-enzootic areas. Regulatory tests for EP include sero-epidemiological methods for equine babesiosis, but these lack specificity due to cross-reactivity with other Babesia species. In this study, we present a real-time quantitative recombinase polymerase amplification (qRPA) method for fast simultaneous detection of both T. equi and B. caballi. In this method, primers and

Epidemiological survey of hemoprotozoan parasites in cattle from low-country wet zone in Sri Lanka.

The diseases caused by hemoprotozoan parasites in cattle often result in economic losses. In Sri Lanka, previous studies found that the up-country wet zone, which is located in central Sri Lanka, was characterized by a high rate of Theileria orientalis and a low rate of Theileria annulata compared with the dry zone. In this study, DNA samples were prepared from the blood of 121 cattle in Galle, a coastal district located in low-country wet zone in Sri Lanka, and were PCR-screened for Babesia bovis, Babesia bigemina, T. annulata, T. orientalis, and Trypanosoma theileri. Problems in the Protection of Reintroduced Przewalski's Horses (Equus ferus przewalskii) Caused by Piroplasmosis.

The objectives of our research on equine piroplasmosis were to clarify the prevalence of the piroplasms Babesia caballi and Theileria equi among reintroduced Przewalski's horses (takhi, Equus ferus przewalskii) in comparison with horses (Equus caballus) of nomads from around Hustai National Park in Mongolia and to determine the mortality among takhi during 2012 to 2015. The prevalence of T. equi was high in 2014 at 80% among nomadic horses and 84% among takhi. We found no evidence of B. caballi infection among takhi. A homology comparison of the 18S rRNA base sequence of T. equi showed the sam

Diversity of Haemaphysalis-associated piroplasms of ruminants in Central-Eastern Europe, Hungary.

BACKGROUND: Increasing numbers of genetic variants are being recognized among piroplasms, but the precise taxonomical status, the tick vector and the geographical range of several species or genotypes are still unknown. Bovine piroplasmosis was reported to re-emerge in north-east Hungary. Because Theileria-infection was newly diagnosed in one cattle herd in the same region of the country, the aim of this study was to molecularly identify the relevant agent, to find its local vector tick species, and to examine the range of Babesia/Theileria spp. of ruminants in Haemaphysalis sp. ticks collecte
