BACKGROUND The anesthetic management of patients with Charcot-Marie-Tooth disease (CMT) requires special deliberation. Previous literature has suggested that patients with CMT may have increased sensitivity to non-depolarizing neuromuscular blocking agents, and hyperkalemia associated with the administration of succinylcholine has been reported. The potential risk of malignant hyperthermia and underlying cardiopulmonary abnormalities, such as pre-existing arrhythmias, cardiomyopathy, or respiratory muscle weakness, must also be considered in patients with CMT. CASE REPORT We describe a case of

We report a 60-year-old Chinese woman with Machado-Joseph disease (MJD/SCA3), initially managed as Charcot-Marie-Tooth disease due to distal sensory loss, pes cavus, and areflexia. Later cerebellar, bulbar, and ocular signs, together with ATXN3 CAG expansion, clarified the diagnosis and highlighted the need to revisit atypical hereditary neuropathy diagnoses.

BACKGROUND: The DNAjB2 gene encodes a co-chaperone protein that interacts with the heat shock protein (HSP) family to maintain protein quality control and preserve neuronal integrity. Variants in this gene have been associated with the axonal form of Charcot-Marie-Tooth Disease (CMT2). Recent literature has also suggested an association between DNAjB2 variants and neurodegenerative disorders such as Parkinson's disease (PD).
DESIGN/METHODS: Case Report.
CASE DESCRIPTION: We present a 36-year-old female patient initially diagnosed with CMT2 at the age of 28, who later developed symptoms of PD

Charcot-Marie-Tooth disease (CMT) is a genetically diverse hereditary disorder that affects the motor and sensory nerves, impacting about 1 in 2500 people. It can be inherited through autosomal dominant (AD), autosomal recessive (AR), or X-linked genetic patterns. CMT2, one of the primary subtypes, is characterized by axonal degeneration and commonly presents with muscle weakness, atrophy, foot deformities, and sensory loss. Aminoacyl-tRNA synthetases (aaRSs) play an important role in the genetic underpinnings of CMT2, with more than 60 disease-causing alleles identified across eight different

We report a case of a 75-year-old woman who has had poor physical performance since childhood and developed bilateral lower limb muscle weakness at age 54. At 64, she was diagnosed with Charcot-Marie-Tooth disease type 4C (CMT4C) due to a homozygous p.Arg77Trp variant in the SH3TC2 gene. At 74, she developed memory impairment. At 75, brain MRI revealed extensive cerebral white matter lesions with T2-weighted hyperintensity and linear high intensity signal along the corticomedullary junction on diffusion-weighted imaging. Suspecting neuronal intranuclear inclusion disease (NIID), we performed a

Spinal and bulbar muscular atrophy is an X-linked neuromuscular disease caused by a trinucleotide CAG repeat expansion in the androgen receptor gene; it is clinically characterized by adult-onset, slowly progressive weakness and atrophy mainly affecting proximal limb and bulbar muscles. Charcot-Marie-Tooth disease type 1A is an autosomal dominant polyneuropathy due to peripheral myelin protein 22 gene duplication and characterized by slowly progressive distal limb muscle weakness, atrophy and sensory loss with foot deformities. Here we report the co-occurrence of both neuromuscular genetic dis

BACKGROUND: The ganglioside-induced differentiation-associated protein 1 gene (GDAP1), which is involved in the Charcot-Marie-Tooth disease (CMT), the most commonly inherited peripheral neuropathy, encodes a protein anchored to the mitochondrial outer membrane. The phenotypic presentations of patients carrying GDAP1 mutations are heterogeneous, making it difficult to determine genotype-phenotype correlations, since the majority of the mutations have been found in only a few unrelated patients. Locus-specific databases (LSDB) established in the framework of the Human Variome Project provide pow

AIM: Clinical attachment loss (CAL) measured at retained teeth is used as an indicator of periodontal disease experience. Because CAL can be measured only on teeth present at examination, retained-dentition summaries are conditioned on the observable dentition. Where tooth loss is severity-dependent, severely affected teeth may be preferentially removed from observation, causing these summaries to diverge from cumulative disease experience across the original dentition. We hypothesised that CAL summaries more sensitive to severe attachment loss would show earlier and stronger attenuation at ol

BACKGROUND: The use of ankle-foot orthoses (AFOs) is a mainstay in the treatment of cavovarus deformity associated with Charcot-Marie-Tooth (CMT) disease, but their ability to reduce cavovarus deformity is poorly characterized. Similarly, associations between AFO design features and degree of deformity reduction are unknown. Weightbearing CT has been used to describe CMT disease-related cavovarus deformity through both two-dimensional (2D) radiographic measures and three-dimensional statistical shape modeling (SSM). However, neither method has been used to evaluate differences in foot posture

Most drug development stories end predictably: promising compounds either succeed or fail. Occasionally, however, a therapy repeatedly returns from apparent extinction. PXT3003 for Charcot-Marie-Tooth disease type 1A (CMT1A) represents one such example. More than a decade after early promise, the program has survived formulation instability, interrupted clinical development, regulatory uncertainty, and controversy surrounding efficacy before re-emerging with renewed evidence of clinical benefit. Yet, as with many therapeutic development programs, interpretation of the available evidence is inf

BACKGROUND: Charcot-Marie-Tooth disease type 2Z (CMT2Z) is an uncommon inherited condition characterized by autosomal dominant inheritance. It is marked by axonal neuropathy, leading to progressive muscle weakness, cramps, and sensory loss. MORC2 is one of the candidate pathogenic genes for CMT2Z.
METHODS: Here, we enrolled two families affected by Charcot-Marie-Tooth (CMT) disease, with both families having probands who exhibit distal lower limb weakness. Whole-exome sequencing (WES) was used to explore the genetic lesion.
RESULTS: Two heterozygous variants (NM_001303256, c.34G > C;p.A12P;

Charcot-Marie-Tooth disease (CMT) is an inherited peripheral neuropathy characterized by sensory dysfunction and muscle weakness, manifesting in the most distal limbs first and progressing more proximal. Over a hundred genes are currently linked to CMT with enrichment for activities in myelination, axon transport, and protein synthesis. Mutations in tRNA synthetases cause dominantly inherited forms of CMT, and animal models with CMT-linked mutations in these enzymes display defects in neuronal protein synthesis. Rescuing protein synthesis in CMT-mutant neurons could offer exciting therapeutic

Lamin A/C (LMNA)-related laminopathies can involve adipose tissue, peripheral nerves, skeletal muscle, and the heart. We report a 65-year-old woman with a seven-year history of progressive distal sensorimotor neuropathy, burning pain in both feet, loss of hand dexterity, and worsening steppage gait. Physical examination demonstrated distal intrinsic hand and foot atrophy, absent reflexes, distal sensory loss, pes cavus, claw and hammer toe deformities, hindfoot varus, limb lipoatrophy, and relative cervicofacial/central adiposity. Electromyography and nerve conduction studies (EMG/NCS) showed

INTRODUCTION: Some patients with Charcot-Marie-Tooth disease (CMT) exhibit prolonged auditory brainstem response (ABR) latencies or abnormal waveforms, suggesting potential damage to the peripheral auditory nerve or central auditory pathways. Diffusion tensor imaging (DTI), a non-invasive neuroimaging technique, can detect the integrity and functional properties of white matter structures with high sensitivity. However, research on the association between DTI characteristics and ABR changes in patients with CMT remains relatively limited, and whether both modalities reflect synergistic damage

Central nervous system (CNS) demyelination is an uncommon observation in patients with Charcot-Marie-Tooth disease (CMT). Where it does occur, it is usually associated with X-linked CMT. We present a case of CMT type 1A with a likely de novo mutation who experienced initial symptoms, and subsequent exacerbation, of multiple sclerosis following respiratory infection. A review of the literature reveals that reports of CMT1A with CNS demyelination are rare. We propose that the mutations in the PMP22 gene result in an over-expression of PMP22 mRNA, which overcomes the normal suppression by miRNA s

Charcot-Marie-Tooth (CMT) is a hereditary neuromuscular syndrome associated with peripheral neuropathy. We describe a case of long-standing CMT disease who underwent general anesthesia for a total knee replacement and developed significant perioperative hypotension. Clinical assessment revealed vasoplegia due to underlying autonomic dysfunction as the most likely cause. Further review also revealed a similar episode of intraoperative hypotension during a previous anesthetic. After failure of conservative strategies, the patient was started on a vasopressor infusion. However, hypotension persis

BACKGROUND AND AIMS: Despite the known association of hip dysplasia and Charcot Marie Tooth disease (CMT), evidence is limited regarding its exact prevalence. Available studies pre-date genetic confirmation of CMT subtypes and current hip reconstruction surgical options. This study examined the prevalence of hip dysplasia in CMT in a tertiary neuromuscular center.
METHODS: This was a retrospective study of children with CMT who had at least one pelvic radiograph between 2000 and 2020. Reimer's migration percentage, acetabular index and lateral center edge angle were used to identify hip dyspl

A man in his 60s presented with acute, progressive right lower limb weakness, low back pain, and paraesthesia. Neuroimaging ruled out spinal cord and central pathology. Cerebrospinal fluid analysis revealed albumin-cytological dissociation, and nerve conduction studies confirmed Guillain-Barré syndrome (GBS) superimposed on Charcot-Marie-Tooth disease type 1A (CMT1A). There was no preceding infection or ganglioside antibody positivity. His weakness progressed rapidly, resulting in flaccid paralysis of the right leg. He received intravenous immunoglobulin with marked clinical improvement and re
