Decabromodiphenyl ethane (DBDPE), a novel brominated flame retardant, has been widely detected in various environmental media and exhibits significant bioaccumulation potential. This study first analyzed the distribution characteristics of DBDPE in zebrafish tissues using a toxicokinetic model. The results revealed that DBDPE preferentially accumulated in the brain, with the concentration order being brain > liver > gonads, indicating a potential neurotoxic risk. Further research demonstrated that zebrafish chronically exposed to environmentally relevant concentrations of DBDPE exhibited abnor

Pets can have accidental, intentional, or malicious exposure to illicit drugs. It is a growing concern over the last decade because there is an increase in usage of illicit drugs in humans and diagnosis is difficult. Owners are often not aware of exposure, or they are reluctant to admit possession of recreational drugs in the household due to potential legal consequences. In addition, illicit drugs sold on the black market are often adulterated with other substances resulting in non-specific clinical presentation and aggravation of symptoms. There are affordable onsite diagnostic tests on the

PPD10558 is an orally active, lipid-lowering 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase inhibitor (statin) being developed as a treatment for hypercholesterolemia in patients who have not been able to tolerate statins because of statin-associated myalgia. We have studied the potential developmental toxicity effects of PPD10558 in pregnant rats and rabbits given daily oral doses during the period of organogenesis. Rats were dosed with 0, 20, 80, or 320 mg/kg/day from Gestation Day (GD) 6 to 17 and rabbits received dose levels of 0, 12.5, 25, or 50 mg/kg/day from GD 6 to 18. Addit

Although the three most commonly used large mammal species in the safety assessment of drugs remain the dog, the macaque and the marmoset, swine, especially minipigs, have also been widely used over the years in many toxicological studies. Swine present a number of interesting biological and physiological characteristics. Similarities in skin properties with humans have led to extensive in vitro and in vivo studies. There is a specific interest in cardiovascular research, as well as in anaesthesiology and critical care medicine due to common features of swine and human physiology. Although kno

Renal toxicity is a hallmark of uranium exposure, with uranium accumulating specifically in the S3 segment of the proximal tubules causing tubular damage. As the distribution, concentration and dynamics of accumulated uranium at the cellular level is not well understood, here, we report on high-resolution quantitative in situ measurements by high-energy synchrotron radiation X-ray fluorescence analysis in renal sections from a rat model of uranium-induced acute renal toxicity. One day after subcutaneous administration of uranium acetate to male Wistar rats at a dose of 0.5 mg uranium kg(-1) bo

High-resolution accurate mass non-targeted analysis (NTA) is a useful discovery tool for metabolite characterization of in vivo dosing studies since it enables detection of both predicted and unexpected biotransformation products. We used NTA to investigate biotransformation of perfluorohexanesulfonamide (PFHxSA) in plasma and liver from male and female Sprague Dawley rats after a 5-day repeat exposure study. PFHxSA is an emerging per- and polyfluoroalkyl substance (PFAS) with unknown toxicity and a potentially reactive headgroup. NTA revealed the presence of predicted in vivo biotransformatio

Although there are a number of guidance documents and frameworks for evaluation of carcinogenicity, none of the current methods fully reflects the state of the science. Common limitations include the absence of dose-response assessment and not considering the impact of differing exposure patterns (e.g., intermittent, high peaks vs. lower, continuous exposures). To address these issues, we have developed a framework for risk assessment of dietary carcinogens. This framework includes an enhanced approach for weight of evidence (WOE) evaluation for genetic toxicology data, with a focus on evaluat

Aluminium (Al) is the most ubiquitous metal in the Earth's crust. Even though its toxicity is well-documented, the role of Al in the pathogenesis of several neurological diseases remains debatable. To establish the basic framework for future studies, we review literature reports on Al toxicokinetics and its role in Alzheimer's disease (AD), autism spectrum disorder (ASD), alcohol use disorder (AUD), multiple sclerosis (MS), Parkinson's disease (PD), and dialysis encephalopathy (DE) from 1976 to 2022. Despite poor absorption via mucosa, the biggest amount of Al comes with food, drinking water,

The detrimental footprint of mycotoxins in agriculture and on animal production has been widely recognized, especially in swine. Despite an increased number of research evaluating the toxicokinetics of mycotoxins in animal organisms, the absorption, distribution, metabolization, and excretion (ADME) patterns of zearalenone (ZEN) need further understanding. Furthermore, in vivo bioindicator for ZEN exposure in individual pigs has yet to be characterized. This study explored the ADME of ZEN in Bama Aroma pigs, a Chinese miniature pig breed, that has been used herein as a swine model. The finding

Mycotoxins are fungal secondary metabolites with bioaccumulation levels leading to their carry-over into animal fluids, organs, and tissues. As a consequence, mycotoxin determination in biological samples from humans and animals has been reported worldwide. Since most mycotoxins show toxic effects at low concentrations and considering the extremely low levels present in biological samples, the application of reliable detection methods is required. This review summarizes the information regarding the studies involving mycotoxin determination in biological samples over the last 10 years. Relevan

BACKGROUND: Bisphenol A (BPA) is an ubiquitous chemical contaminant that has recently been associated with adverse effects on human health. There is incomplete understanding of BPA toxicokinetics, and there are no established interventions to eliminate this compound from the human body. Using 20 study participants, this study was designed to assess the relative concentration of BPA in three body fluids-blood, urine, and sweat-and to determine whether induced sweating may be a therapeutic intervention with potential to facilitate elimination of this compound.
METHODS: Blood, urine, and sweat w

Salicylates are common exposures. We report an unusual case of salicylate ingestion, as salsalate, with resolution of symptoms and return of salicylate levels to non-toxic values, with a subsequent, unexpected recrudescence to toxic levels requiring reinstitution of therapy. A 31-year-old man ingested unknown amounts of salsalate, hydroxyzine, and a benzodiazepine. He was intubated and treated with IV sodium bicarbonate and two doses of oral activated charcoal. Eight hours after presentation, his serum salicylate concentration peaked at 55 mg/dL, and then decreased to a nadir of 5.6 mg/dL 38 h

Rose oxide (RO) is used in cosmetic, hygiene, laundry, cleaning, and household products as a synthetic fragrance. Synthetic rose oxide differs from naturally occurring rose oxide in its enantiomer composition. While natural RO consists mainly of the (-)-cis isomer, synthetic RO contains approximately equal parts of (-)-cis and (+)-cis isomers, as well as small amounts of (-)-trans and (+)-trans isomers. Since in this project, the exposure to industrially produced chemicals by human biomonitoring (HBM) was in the focus, the method development aimed at suitable urinary metabolites derived from t

Prenatal exposure to per- and polyfluoroalkyl substances (PFAS) represents a critical developmental concern because several PFAS cross the placenta and may perturb biological programming during sensitive windows. Although epidemiological and experimental studies have linked prenatal exposure to several measured PFAS, primarily legacy perfluoroalkyl acids such as perfluorooctanoic acid (PFOA), perfluorooctanesulfonic acid (PFOS), Perfluorohexane Sulfonic Acid (PFHxS), and Perfluorononanoic Acid (PFNA), to diverse offspring outcomes, the strength of evidence differs substantially across compound

Global production and widespread use of plastics are increasing dramatically. With current limited recycling and recovery options, microplastics and nanoplastics (MNPs) persist in the natural environment. Due to their ubiquity, human exposure to MNPs is inevitable. In addition to their inherent toxic effects, MNPs can adsorb harmful contaminants and act as vectors for microorganisms, compounding toxicological effects. After entering the body, bioaccumulation occurs in several tissues and organs, including the liver and the gastrointestinal (GI) tract. Proposed clinical effects of MNP absorptio

BACKGROUND: Cocaine intoxication and abuse is a worldwide problem that can be the cause of numerous acute medical complications, including severe acute hepatitis. Although these cases are scarce, they are extremely serious and may lead to liver transplantation or death. Management of toxic hepatitis, once the causative agent has been discontinued, is essentially symptomatic, based on clinical and biological monitoring and prevention of complications related to acute hepatitis.
CASE DETAILS: We present a case of a 28-year-old woman admitted to the emergency department for acute hepatitis due t

Mercury (Hg) is a widespread environmental contaminant, with methylmercury (MeHg) being its most toxic and bioavailable form. MeHg accumulates in aquatic food chains, leading to significant human exposure, particularly in Amazonian riverine populations where fish is a dietary staple. This study examined the impact of genetic polymorphisms on Hg exposure levels and oxidative stress biomarkers in 329 individuals from riverside communities in the Brazilian Amazon. Hg concentrations in blood, plasma, and hair were measured using inductively coupled plasma mass spectrophotometer, while TaqMan genot

Exposure to environmental pollutants and human microbiome composition are important predisposition factors for tumour development1,2. Similar to drug molecules, pollutants are typically metabolized in the body, which can change their carcinogenic potential and affect tissue distribution through altered toxicokinetics3. Although recent studies demonstrated that human-associated microorganisms can chemically convert a wide range of xenobiotics and influence the profile and tissue exposure of resulting metabolites4,5, the effect of microbial biotransformation on chemical-induced tumour developmen
