African trypanosomiasis alters prostaglandin production by murine peritoneal macrophages.

Many factors contribute to the severe immunosuppression associated with African trypanosomiasis. Macrophages have been shown to be important target cells which after uptake of parasites, mediate immune dysfunction in vivo. We observed that infection of mice with Trypanosome brucei brucei (clone NIM 6) induces profound changes in arachidonic acid metabolism and prostaglandin (PG) secretion by macrophages. Normal macrophages release more PGI2 than PGE2 and production of both these prostaglandins is stimulated equally by endotoxin (LPS). Macrophages taken from NIM 6 infected mice at the peak of t

[Importance of familial contamination in Trypanosoma brucei gambiense trypanosomiasis].

Referring to the sociological structure of the family in the Bateke ethnic group which most of the inhabitants of Kwamouth (Bandundu region, Zaïre) are descendants, the author defines the concept to family contamination in trypanosomiasis induced by Trypanosoma brucei gambiense. The calculated incidence in the selected population according to the different types of family and vicinity contamination was found to be at least five times higher than the incidence obtained for the global population. To explain this considerable difference, the author suggests the mechanical transmission of the tryp

Suppression of cell-mediated immunity in experimental African trypanosomiasis.

Adult New Zealand white rabbits were experimentally infected with a parasitic African hemoflagellate, Trypanosoma congolense, and were subsequently tested for in vivo and in vitro aspects of cell-mediated immune function. Chronically infected rabbits were sensitized to mycobacterial protein and skin-tested with purified protein derivative; all infected animals demonstrated much milder skin-test responses to antigen than control groups. Similarly, peripheral blood lymphocyte responses in vitro to purified protein derivative and, as well, to phytohemagglutinin were markedly suppressed. Supernata

Clinical occurrence of trypanosomiasis in Arabian horses from Ahvaz.

Trypanosoma evansi (T. evansi) is a hemoprotozoan parasite affecting camels and equids, such as horses, mules, and donkeys, and is known to cause surra disease in these animals. Despite the worldwide distribution of T. evansi infections in equids, surra has not been reported in Arabian horses in Khuzestan Province for over 60 years. In September 2018, a 7-year-old Arabian mare was referred from a 10-horse farm in the suburbs of Ahvaz City. The mare presented with a history of weight loss, poor appetite, and proximity to a camel herd. Physical examination revealed a poor body condition score (B

Animal Trypanosomiasis: Challenges and Prospects for New Vaccination Strategies.

Animal trypanosomiasis, such as nagana, surra, and dourine, represent a significant challenge to animal health and economic development, especially in tropical and subtropical regions where livestock production is an essential component of a country's economy. Despite advances in the control of human trypanosomiasis, animal diseases caused by several species of trypanosomes remain neglected. The lack of funding for the development of new treatments and vaccines contributes to sustaining the severe economic impacts these diseases have on the farming industry, especially in low-income rural area

Long-term sequelae of congenital gambiense human African trypanosomiasis.

BACKGROUND: The clinical presentation of gambiense human African trypanosomias (gHAT) is generally considered to be the same among children and adults. In general, when describing the clinical presentation of children with gHAT, no differentiation is made between congenital gHAT and gHAT acquired later. There is a lack of knowledge regarding the signs and symptoms attributable to congenital gHAT and its long-term sequelae. METHODS: Following an evaluation of the hospital register for gHAT, the authors observed that six children born to mothers with gHAT during their pregnancies still had sequ

Bovine trypanosomiasis an emerging disease in Maranhão State-Brazil.

In South American countries, bovine trypanosomiasis is caused mainly by Trypanosoma vivax. Among the infected animals, bovines are considered more susceptible, and this susceptibility varies among breeds. To determine the occurrence of T. vivax in Maranhão State, Brazil, a total of 559 cattle blood samples were collected for molecular and parasitological studies. On São Luís Island, no parasites were observed in the parasitological exam; however, with the polymerase chain reaction (PCR) method, three samples (1.06%) were positive. In four municipalities that compose the Pedreiras Basin, 3.39%

Human African trypanosomiasis in non-endemic countries (2000-2010).

BACKGROUND: Human African trypanosomiasis (HAT) can affect travelers to sub-Saharan Africa, as well as migrants from disease endemic countries (DECs), posing diagnosis challenges to travel health services in non-disease endemic countries (non-DECs). METHODS: Cases reported in journals have been collected through a bibliographic research and complemented by cases reported to the World Health Organization (WHO) during the process to obtain anti-trypanosome drugs. These drugs are distributed to DECs solely by WHO. Drugs are also provided to non-DECs when an HAT case is diagnosed. However, in non

Late-stage human African trypanosomiasis in a Sudanese refugee.

A 19-year-old Sudanese woman, who had lived for about a decade in Ugandan refugee camps, was referred for investigation of a 12-month history of a generalised rash. Two months later, her condition had deteriorated to include cachexia and drowsiness. Despite initial negative findings on investigation, human African trypanosomiasis (HAT) was suspected, and parasites were found in a double-centrifuged sample of cerebrospinal fluid. Eflornithine, the appropriate drug for treatment of late-stage disease, was obtained through the World Health Organization. This case highlights the diagnostic and the

Tsetse fly control and trypanosomiasis in Africa, quo vadis?

National and international efforts to eradicate tsetse fly-borne human and animal trypanosomiasis are critically evaluated, and possible reasons for their failure in many cases are discussed. Some formerly performed campaigns in specific areas with positive results cannot be taken as examples to solve the main problems. In future, a significant reduction of trypanosomiasis cases will be possible to achieve only if a concerted long-term Pan-African approach, based on financial security, the continuity of expert staff, and a well-planned, ecologically sound land use, is generally accepted.

The continuing problem of human African trypanosomiasis (sleeping sickness).

Human African trypanosomiasis, also known as sleeping sickness, is a neglected disease, and it continues to pose a major threat to 60 million people in 36 countries in sub-Saharan Africa. Transmitted by the bite of the tsetse fly, the disease is caused by protozoan parasites of the genus Trypanosoma and comes in two types: East African human African trypanosomiasis caused by Trypanosoma brucei rhodesiense and the West African form caused by Trypanosoma brucei gambiense. There is an early or hemolymphatic stage and a late or encephalitic stage, when the parasites cross the blood-brain barrier t

Human African trypanosomiasis transmission, Kinshasa, Democratic Republic of Congo.

To investigate the epidemiology of human African trypanosomiasis (sleeping sickness) in Kinshasa, Democratic Republic of Congo, 2 entomologic surveys were conducted in 2005. Trypanosoma brucei gambiense and human-blood meals were found in tsetse fly midguts, which suggested active disease transmission. Vector control should be used to improve human African trypanosomiasis control efforts.

Comparison of cytokine plasma levels in human African trypanosomiasis.

BACKGROUND: Immunological studies suggest that human African trypanosomiasis (HAT) is associated with inflammatory responses. A better understanding of the complex cytokine interactions regulating HAT infections is essential to elucidate the mechanisms of generalized immunosuppression. METHOD: We determined levels of interleukin (IL)-2, IL-4, IL-6, IL-10, tumour necrosis factor (TNF)-alpha and interferon (IFN)-gamma protein levels in plasma samples from three groups of individuals from the Democratic Republic of Congo: (i) HAT cases; (ii) seropositive individuals for whom parasite detection w

Candidate protein selection for diagnostic markers of African trypanosomiasis.

The ability to accurately diagnose the presence of an infective micro-organism is not only important for individual human and animal health and wellbeing, but is also central to surveillance programmes. Effective and sustainable control of many diseases in the developing world depends on the availability of field applicable diagnostics that are cheap, reliable, simple in design and application, and which provide immediate results. This review examines how the genome sequences can be used in the selection of potential candidate proteins for developing new serodiagnostics for African trypanosomi
