Multi-Omics Integration Highlights the Role of Ubiquitination in CCl4-Induced Liver Fibrosis.

Liver fibrosis is the excessive accumulation of extracellular matrix proteins that occurs in chronic liver disease. Ubiquitination is a post-translational modification that is crucial for a plethora of physiological processes. Even though the ubiquitin system has been implicated in several human diseases, the role of ubiquitination in liver fibrosis remains poorly understood. Here, multi-omics approaches were used to address this. Untargeted metabolomics showed that carbon tetrachloride (CCl4)-induced liver fibrosis promotes changes in the hepatic metabolome, specifically in glycerophospholipi…

Abnormal level of CUL4B-mediated histone H2A ubiquitination causes disruptive HOX gene expression.

BACKGROUND: Neural tube defects (NTDs) are common birth defects involving the central nervous system. Recent studies on the etiology of human NTDs have raised the possibility that epigenetic regulation could be involved in determining susceptibility to them. RESULTS: Here, we show that the H2AK119ub1 E3 ligase CUL4B is required for the activation of retinoic acid (RA)-inducible developmentally critical homeobox (HOX) genes in NT2/D1 embryonal carcinoma cells. RA treatment led to attenuation of H2AK119ub1 due to decrease in CUL4B, further affecting HOX gene regulation. Furthermore, we found th

TRIM17 and TRIM28 antagonistically regulate the ubiquitination and anti-apoptotic activity of BCL2A1.

BCL2A1 is an anti-apoptotic member of the BCL-2 family that contributes to chemoresistance in a subset of tumors. BCL2A1 has a short half-life due to its constitutive processing by the ubiquitin-proteasome system. This constitutes a major tumor-suppressor mechanism regulating BCL2A1 function. However, the enzymes involved in the regulation of BCL2A1 protein stability are currently unknown. Here, we provide the first insight into the regulation of BCL2A1 ubiquitination. We present evidence that TRIM28 is an E3 ubiquitin-ligase for BCL2A1. Indeed, endogenous TRIM28 and BCL2A1 bind to each other …

Parkin targets HIF-1α for ubiquitination and degradation to inhibit breast tumor progression.

Mutations in E3 ubiquitin ligase Parkin have been linked to familial Parkinson's disease. Accumulating evidence suggests that Parkin is a tumor suppressor, but the underlying mechanism is poorly understood. Here we show that Parkin is an E3 ubiquitin ligase for hypoxia-inducible factor 1α (HIF-1α). Parkin interacts with HIF-1α and promotes HIF-1α degradation through ubiquitination, which in turn inhibits metastasis of breast cancer cells. Parkin downregulation in breast cancer cells promotes metastasis, which can be inhibited by targeting HIF-1α with RNA interference or the small-molecule inhi…

KLHL3 regulates paracellular chloride transport in the kidney by ubiquitination of claudin-8.

A rare Mendelian syndrome--pseudohypoaldosteronism type II (PHA-II)--features hypertension, hyperkalemia, and metabolic acidosis. Genetic li … e permeability. Mechanistically, KLHL3 was directly bound to claudin-8, and this binding led to the ubiquitination and degradation of claudin-8. The dominant PHA-II mutation in KLHL3 impaired claudin-8 binding, ubiquitination, and degradation. These findings have attested to the concept that the paracellular pathway is physiologically regulated through the ubiqu …

Our findings suggest that NEMO Lysine392, a well-established K63-linked polyubiquitination site, is an important mediator of PMMA-induced osteolysis. Therefore, this NEMO motif should be considered as a target to combat PMMA particle-induced osteolysis.

PCNA mono-ubiquitination and activation of translesion DNA polymerases by DNA polymerase {alpha}.

Translesion DNA synthesis (TLS) involves PCNA mono-ubiquitination and TLS DNA polymerases (pols). Recent evidence has shown that the mono-ubiquitination is induced not only by DNA damage but also by other factors that induce stalling of the DNA replication fork. We studied the effect of spontaneous DNA replication errors on PCNA mono-ubiquitination and TLS induction. In the pol1L868F strain, which expressed an error-prone pol alpha, PCNA was spontaneously mono-ubiquitinated. Pol alpha L868F had a rate-limiting step at the extension from mismatched primer termini. Electron microscopic observati…

Differential ubiquitination of stallion sperm proteins: possible implications for infertility and reproductive seasonality.

Antibodies against ubiquitin, a universal proteolytic marker, show increased cross-reactivity with defective spermatozoa in men and bulls. We investigated sperm ubiquitination in the stallion, a seasonally polyestrous mammal. Immunofluorescence and immunoelectron microscopy demonstrated that anti-ubiquitin antibodies bind to the surface of both membrane-intact and aldehyde-fixed spermatozoa. Cross-reactivity to the ubiquitin-conjugating enzyme E2 was also detected in sperm. Immunohistochemistry showed that ubiquitinated spermatozoa were first detected in the caput epididymis, coincident with a…

Atypical E3 ligase HOIL1 promotes MDA5 signaling and ester-linked ubiquitination of LGP2.

The RIG-I-like receptors, RIG-I and MDA5, are innate sensors of RNA virus infections that are critical for mounting a robust antiviral immune response. Heme-oxidized IRP2 ligase 1 (HOIL1), a component of the linear ubiquitin chain assembly complex (LUBAC), is essential for interferon induction in response to viruses sensed by MDA5, but not viruses sensed by RIG-I; however, the underlying mechanism remains unclear. LUBAC contains two atypical E3 ubiquitin ligases, HOIL1 and HOIP. While HOIP generates "linear" polyubiquitin chains, HOIL1 catalyzes ester-linked ubiquitination of serine and threon…

FBXO6 regulates colon cancer migration and invasion via ITGB1 ubiquitination and downstream signaling.

Colorectal cancer (CRC) is the third most common malignant tumor worldwide, with high recurrence and metastasis rates significantly impacting outcomes. This study explores the role of FBXO6, a ubiquitination-related protein, in regulating CRC malignancy, particularly cell migration and invasion. Our analysis reveals that higher FBXO6 expression correlates with better prognosis in CRC patients, although its expression decreases in advanced-stage tumors. Functional studies demonstrate that FBXO6 overexpression suppresses the invasive and migratory abilities of HCT116 and RKO cells and reduces si…

SUMOylation and ubiquitination reciprocally regulate SMCHD1 antiviral activity against herpes simplex virus 1.

Host restriction factors serve as intrinsic barriers against viral infection, and are frequently counteracted by viral antagonists. Previous … contrast, during herpes simplex virus 1 (HSV-1) infection, the viral E3 ligase ICP0 induces SMCHD1 ubiquitination and proteasomal degradation, thereby relieving viral restriction. Loss of ICP0 stabilizes SMCHD1 and leads to marked accumulation of SUMOylated SMCHD1, rendering ICP0-deficient HSV-1 more sensitive to SMCHD1-mediated inhibition. Together, our findin …

CUL7-mediated KEAP1 ubiquitination promotes the progression of colon cancer via NRF2 signaling.

Cullin7 (CUL7) mediates cancer progression across multiple cancer types through its regulatory role in protein ubiquitination. However, the biological function and molecular mechanisms underlying CUL7 in colon cancer remain poorly defined. Here, we demonstrate that high CUL7 expression correlates with poor prognosis in patients. Ablation of CUL7 inhibited cell proliferation and migration in colon cancer cells, whereas CUL7 overexpression exhibited oncogenic properties. Moreover, Cul7-deplete mice exhibit a lower level of tumor growth. Mechanistically, CUL7 interacts with Kelch-like ECH-associa…

Ubiquitination modifications as central regulators of metabolic dysfunction in type 2 diabetes mellitus.

Type 2 diabetes mellitus (T2DM) is a multifactorial metabolic disorder characterized by insulin resistance, progressive pancreatic β-cell dysfunction, and multi-organ metabolic dysregulation. Among various post-translational modifications, ubiquitination has emerged as a central regulator of cellular homeostasis, with E3 ubiquitin ligases and deubiquitinating enzymes (DUBs) governing the stability and activity of key signaling proteins. This review systematically elucidates the pivotal role of ubiquitination in the pathogenesis of T2DM. We first outline the fundamental principles of the ubiqui…

Palmitoylation-Mediated Ubiquitination of SRPK1 Regulates Ferroptosis in High-Fat-Induced Erectile Dysfunction.

Serine-arginine protein kinase 1 (SRPK1) is a major protein kinase involved in mRNA splicing, cell cycle, and endothelial function. Recent studies have highlighted a close relationship between palmitic acid (PA) and endothelial cell ferroptosis. Here, we demonstrate that PA promotes the ubiquitination-dependent degradation of SRPK1 mediated by the E3 ubiquitin ligase mindbomb 1 (MIB1) at lysine 494. Moreover, SRPK1 S-palmitoylation is catalyzed by zinc-finger DHHC S-acyltransferase 24 (ZDHHC24) at cysteines 188/502/647 and deacylated by acyl protein thioesterase 1 (APT1). The dynamic S-palmito…
