Evaluation of different immunization schedules for oral cholera B subunit-whole cell vaccine in Swedish volunteers.

Different immunization schedules for oral B subunit-whole cell (B-WC) cholera vaccine were evaluated in Swedish volunteers to obtain information for recommendations of vaccine use in non-endemic areas. Two peroral doses of B-WC vaccine were as effective as three doses in inducing IgA and IgG antitoxin as well as vibriocidal antibody responses in serum. Administration of two vaccine doses either at 7, 14 or 28-42 day intervals resulted in comparable antitoxin responses in serum, whereas a 3-day immunization interval resulted in significantly lower titre increases. Vibriocidal antibody responses

Safety and immunogenicity of the oral, whole cell/recombinant B subunit cholera vaccine in North American volunteers.

A newly formulated, oral, inactivated whole cell plus recombinant B subunit (WC/rBS) cholera vaccine was evaluated in US military personnel. In the first study, 74 subjects were given two doses 14 days apart. In the second study, 186 subjects were randomized into four groups; two groups received vaccine with either full (4 g) or half (2 g) strength bicarbonate buffer, and two groups received either full or half strength buffer without vaccine. Mild gastrointestinal symptoms were associated with full buffer (P = .02) but not with the vaccine. In the first study, 36% of all subjects and 55% with

Immune responses following killed whole vibrio-B subunit oral cholera vaccine in human volunteers.

Immunogenicity of killed whole vibrio and B subunit oral cholera vaccines in American and Thai volunteers were analysed in terms of significant rise of antibody titre. Three doses of 2 x 10(11) killed vibrios and 5 mg of cholera toxin B subunit were given at two-week intervals. There were no differences in the percent of volunteers with significant rise of serum immunoglobulin G and secretory immunoglobulin A (sIgA) to cholera toxin. However, the percent with significant rises of serum antibody to whole cell V. cholerae Inaba measured by vibriocidal titre and serum immunoglobulin G, and secret

Synthetic TRAC478 emulsion and saponin based adjuvant systems enhance humoral and cellular immune responses elicited by computationally optimized H1 and H3 hemagglutinin subunit vaccines.

BACKGROUND: Current seasonal influenza vaccines elicit sub-optimal immune responses, allowing the virus to continually spread and cause disease from season to season. The inclusion of wild-type vaccine antigens and classical egg-based manufacturing practices often lead to the production of influenza vaccines that are strain specific and not well-matched to actively circulating viruses. METHODS: Computationally optimized broadly reactive antigens (COBRAs) for influenza were expressed as recombinant hemagglutinin (HA) proteins for use as subunit vaccines. These COBRA HA vaccines were adjuvanted

Fasciola gigantica vaccine construct: an in silico approach towards identification and design of a multi-epitope subunit vaccine using calcium binding EF-hand proteins.

Continuous attempts have been made to pinpoint candidate vaccine molecules and evaluate their effectiveness in order to commercialise such vaccines for the treatment of tropical fascioliasis in livestock. The pathophysiology of fascioliasis can be related to liver damage brought on by immature flukes that migrate and feed, as well as immunological reactions to chemicals produced by the parasites and alarm signals brought on by tissue damage. Future research should, in our opinion, concentrate on the biology of invasive parasites and the resulting immune responses, particularly in the early sta

A novel influenza subunit vaccine composed of liposome-encapsulated haemagglutinin/neuraminidase and IL-2 or GM-CSF. I. Vaccine characterization and efficacy studies in mice.

The aim of this study was to improve the potency of the currently used influenza subunit vaccines, which are of relatively low efficiency in high-risk groups. Influenza A virus (Shangdong/9/93) haemagglutinin/neuraminidase (H3N2), granulocyte-macrophage colony-stimulating factor (GM-CSF) and interleukin-2 (IL-2) were encapsulated, each separately or combined, in multilamellar vesicles composed of dimyristoyl phosphatidylcholine. BALB/c mice were immunized once, i.p. or s.c., with 0.05-2.0 microg HN administered either as free antigen (F-HN), adsorbed to aluminum hydroxide (Al-HN), or encapsula

A Comparative Study on Immune Protection Efficacy: An HSV-1 Trivalent Antigen Subunit Vaccine Formulated with a Cellular Immunity-Inducing Adjuvant Versus an mRNA Vaccine.

Background: Herpes simplex virus (HSV) is a neurotropic virus that can be categorized into two serotypes: HSV-1 and HSV-2. HSV-1 causes symptoms such as herpes labialis, herpetic keratitis, genital ulcers, and encephalitis, and primarily establishes latent infection in the trigeminal ganglion. The complexity of membrane fusion mechanisms and potential infection in nerves allow HSV to easily evade recognition and clearance by host immune cells. Therefore, developing a vaccine that can prevent both primary and reactivated HSV-1 infection is critical. Currently, no preventive or therapeutic HSV-1

Gilt Vaccination with a Mixed Administration of a PRRS MLV and a PPV1 Subunit Vaccine Protects against Heterologous PRRSV1 Infection and Prevents Detrimental Effects on Piglet Performance.

The efficacy of the combined administration of a porcine reproductive and respiratory syndrome (PRRS) modified live virus (MLV) vaccine and a porcine parvovirus 1 (PPV1) subunit vaccine in gilts was addressed in two experiments. Experiment A aimed to establish a 4-week onset of immunity (OOI). Gilts were randomly distributed in three treatment groups: non-vaccinated control animals (group 1), animals vaccinated with the combined vaccine (group 2), and a third group that consisted of animals vaccinated with the PRRS MLV vaccine alone (group 3). Four weeks after the first vaccination, gilts were

Protection against Congenital CMV Infection Conferred by MVA-Vectored Subunit Vaccines Extends to a Second Pregnancy after Maternal Challenge with a Heterologous, Novel Strain Variant.

Maternal reinfection of immune women with novel human cytomegalovirus (HCMV) strains acquired during pregnancy can result in symptomatic congenital CMV (cCMV) infection. Novel animal model strategies are needed to explore vaccine-mediated protections against maternal reinfection. To investigate this in the guinea pig cytomegalovirus (GPCMV) model, a strictly in vivo-passaged workpool of a novel strain, the CIDMTR strain (dose, 1 × 107 pfu) was used to infect dams that had been challenged in a previous pregnancy with the 22122 strain, following either sham-immunization (vector only) or vaccinat

Novel nanoemulsion adjuvant stabilized by TPGS possesses equivalent physicochemical properties, Turbiscan stability, and adjuvanticity to AS03 for eliciting robust immunogenicity of subunit vaccines in mice.

Emulsion-based antigen delivery systems have emerged as a novel approach to enhance the effectiveness of subunit vaccines. This study presents the development of a newly formulated oil-in-water (o/w) nanoemulsion adjuvant (NEA) composed of squalene oil and α-tocopheryl polyethylene glycol 1000 succinate (TPGS), which serves dual roles as an emulsifier and an immunostimulator. In comparison to AS03, an FDA-approved emulsion adjuvant that includes α-tocopherol, squalene, and polysorbate 80, NEA is devoid of α-tocopherol and exhibits comparable physicochemical properties, including particle size,

A comparative study of cationic liposome and niosome-based adjuvant systems for protein subunit vaccines: characterisation, environmental scanning electron microscopy and immunisation studies in mice.

Vesicular adjuvant systems composing dimethyldioctadecylammonium (DDA) can promote both cell-mediated and humoral immune responses to the tuberculosis vaccine fusion protein in mice. However, these DDA preparations were found to be physically unstable, forming aggregates under ambient storage conditions. Therefore there is a need to improve the stability of such systems without undermining their potent adjuvanticity. To this end, the effect of incorporating non-ionic surfactants, such as 1-monopalmitoyl glycerol (MP), in addition to cholesterol (Chol) and trehalose 6,6'-dibehenate (TDB), on th

An immunoinformatics-driven multiepitope subunit vaccine targeting Oropouche virus: Molecular docking, immune and dynamics simulation approaches.

The Oropouche virus (OROV) is an emerging zoonotic orthobunyavirus responsible for recurring outbreaks of febrile illness and neurological disorders in South and Central America. Despite its growing public health issues, no FDA-approved vaccines or antiviral therapies currently exist. To address this gap, a multiepitope subunit vaccine was designed to target OROV glycoproteins G1 (Gn), G2 (Gc), and the nucleocapsid (N) protein using a reverse-vaccinology (RV) approach. For vaccine construction, suitable epitopes (MHC-I, MHC-II, and linear B-cell) were selected based on percentile rank, antigen

Circular Permutation of the E. coli Heat-Labile Enterotoxin Pentameric B Subunit for Mucosal Vaccine Adjuvant Design.

The heat-labile enterotoxins (LTs) of Escherichia coli structurally belong to the AB5 toxin, which consists of an A subunit and a pentameric B subunit. LTs have been extensively reported for their potent mucosal adjuvant activities. The B subunit of type II LT (LT-IIb-B5) recognizes the GD1a ganglioside, which is associated with the internalization of the toxic A subunit and meanwhile involved in the recognition of the Toll-like receptor (TLR-2/1) and downstream NF-κB activation. In developing LT-IIb-B5 as a safe adjuvant, we intend to prevent concerns derived from the intrinsic toxicity of LT

Effectiveness and immunogenicity of a nanoemulsion protein subunit vaccine against Pseudomonas aeruginosa: investigation in diet-induced obese mice.

Pseudomonas aeruginosa (Pa) is an opportunistic pathogen threatening individuals with obesity, a condition associated with chronic meta-inflammation and altered immunity. In this study, we evaluate the immunogenicity and protective efficacy of a nanoemulsion-based subunit vaccine (L-PaF/ME/BECC) targeting Pa in a diet-induced obese mouse model. Mice fed a high-fat diet (HFD; 60% fat) were compared to those on a low-fat diet (LFD; 10% fat) or regular chow (3% to 4% fat) to assess differences in vaccine responsiveness. Our results show that while L-PaF induces robust immune responses across all

Evaluation of humoral and cellular immune responses induced by a novel YidRv subunit vaccine from hypervirulent Klebsiella pneumoniae.

BACKGROUND: Klebsiella pneumoniae (KP) has re-emerged as a prominent etiological agent of pneumonia and systemic infection, with extended-spectrum β-lactamase (ESBL)-producing strains posing considerable therapeutic challenges. Moreover, vaccines may have limited efficacy because of the emergence of variant strains. The yidRv gene encodes a protein linked to excessive adherence and is conserved in over 99% of KP isolates. The present study aimed to explore the ability of a rec-YidRv protein to stimulate humoral and cellular immune response in vitro and in vivo. MATERIALS AND METHODS: The capa

Immunogenicity and Protective Effects of an Ag85B Tuberculosis Subunit Vaccine Formulated with Synthetic TLR4 Agonists in BCG-Boosted Mice.

BACKGROUND/OBJECTIVES: Tuberculosis (TB) remains a major global health challenge, and the Bacillus Calmette-Guérin (BCG) vaccine has limited efficacy against adult pulmonary disease. Protein subunit vaccines are a promising alternative but require strong adjuvants to induce cell-mediated immunity. Synthetic agonists targeting toll-like receptor 4 (TLR4) and stimulators of interferon genes (STINGs) have emerged as effective immunostimulants. Therefore, we aimed to evaluate the immunogenicity and protective efficacy of Ag85B-based subunit vaccines formulated with synthetic TLR4 and STING agonist

Clostridium septicum Alpha Toxin-Based Recombinant Subunit Vaccine Protects Broiler Chickens Against Clostridial Dermatitis.

Clostridial dermatitis (CD), caused by Clostridium septicum, is an economically important disease of broiler chickens, specifically in flocks raised without antibiotics. Our previous work in turkeys identified a nontoxic domain no. 2 of C. septicum alpha-toxin (ntATX-D2) as a protective vaccine antigen. In the present study, we used a prime-boost immunization regimen to vaccinate broiler chickens subcutaneously with recombinant ntATX-D2 protein followed by a challenge with virulent C. septicum to assess protective immunity. Immunization led to clinical protection, as determined by a significan

Sustained Delivery of a Shingles Subunit Vaccine Overcomes Age-Related Declines in Humoral and Cellular Immunity Relative to Shingrix.

The global aging population faces heightened vulnerability to infectious diseases due to immunosenescence, which diminishes the potency, durability, and breadth of vaccine-induced immunity. While the leading shingles vaccine, Shingrix, provides protection against herpes zoster virus for adults aged 50 and older, it is often associated with severe local and systemic reactogenicity, which limits vaccine compliance. Here, we report an injectable polymer-nanoparticle (PNP) hydrogel platform for the sustained delivery of a shingles subunit vaccine to enhance immune responses while mitigating reacto

Preparation and evaluation of Escherichia coli-derived glycoprotein E for development of a subunit vaccine against varicella-zoster virus.

Glycoprotein E (gE) of varicella-zoster virus (VZV) is a validated target for subunit vaccine development. However, gE produced in Chinese hamster ovary (CHO) cells is associated with high manufacturing costs and glycosylation-related heterogeneity. Here, the ectodomain of gE (aa 25-539) was expressed in Escherichia coli, yielding a monomeric, non-glycosylated gE ectodomain (n-gE) that was efficiently prepared and comprehensively characterized. To recover properly folded protein from inclusion bodies, a two-stage refolding strategy was established, in which disulfide bond formation was promote
