Proteome analysis of Leptospira interrogans virulent strain.

Leptospirosis is a worldwide zoonotic infection of human and veterinary concern. Caused by pathogenic spirochetes of the genus Leptospira, the disease presents greater incidence in tropical and subtropical regions. The identification of proteins that could be involved in the bacteria host interactions may facilitate the search for immune protective antigens. We report the proteomic analysis of Leptospira interrogans serovar Pomona virulent strain LPF cultured from kidney and liver of infected hamsters. Total protein extracts were separated by two-dimensional gel electrophoresis (2-DE), 895 spo

Virulence factors in brucellosis: implications for aetiopathogenesis and treatment.

Brucella species are responsible for the global zoonotic disease brucellosis. These intracellular pathogens express a set of factors - including lipopolysaccharides, virulence regulator proteins and phosphatidylcholine - to ensure their full virulence. Some virulence factors are essential for invasion of the host cell, whereas others are crucial to avoid elimination by the host. They allow Brucella spp. to survive and proliferate within its replicative vacuole and enable the bacteria to escape detection by the host immune system. Several strategies have been used to develop animal vaccines aga…

[Virulence determinants and its evolution of the genus Listeria].

The genus Listeria consists of six species: L. monocytogenes, L. innocua, L. welshimeri, L. ivanovii, L. seeligeri and L. grayi. Two of the species, L. monocytogenes and L. ivanovii are pathogenic. The heterogeneity of remaining species, previously assumed to be nonpathogenic, regarding their capability of acquiring virulence-associated genes may reflect their potential ability to be causative agents of diseases, especially in immunocompromised mannals. Virulence determinants involved in environmental tolerance, adhesion and invasion of eukaryotic cells and intracellular life function interact…

Intersection of fungal fitness and virulence in Cryptococcus neoformans.

The use of insertional mutagenesis to discover genes that impact laccase activity has resulted in the identification of multiple cellular processes that affect the fitness of Cryptococcus neoformans. Fitness has been defined as the ability of an organism to propagate and evolve within a given environment. Because the human host is an evolutionary dead-end for an opportunistic pathogen, we have defined pathogenic fitness here as the capability to successfully propagate within the stressful environment of the host, causing disease by expression of virulence traits that damage the host. In this r…

[Advances in research on virulence factors of Entamoeba histolytica].

The parasitic protozoan Entamoeba histolytica is an important pathogen responsible for intestinal and extraintestinal amebiasis in human, claiming up to 100 000 victims every year. This parasite is characterized by its high potential for invading and destroying human tissues. Invasion past the mucous barrier of the colonic epithelium leads to the development of colitis and amebic dysentery. Dissemination of the trophozoite through blood stream permits the invasion of other tissues, resulting in abscesses in extracolonic location. Several virulence factors have been proposed to be involved in E…

Unlocking the DEAD-box: a key to cryptococcal virulence?

The DEAD-box RNA helicases are enzymes involved in many critical aspects of RNA metabolism within both eukaryotic and prokaryotic organisms. Several studies have shown that these proteins may have important functions in mediating microbial pathogenesis. A new study in this issue of the JCI identifies the first DEAD-box RNA helicase in the pathogenic fungus Cryptococcus neoformans and proposes novel roles for this family of proteins in the development and progression of cryptococcosis.

Is plasminogen deployed as a Streptococcus pyogenes virulence factor?

Streptococcus pyogenes (group A streptococcus) causes human skin and throat infections as well as highly invasive diseases including necrotizing fasciitis. Group A streptococcal infections and invasive disease have made a resurgence in developed countries during the past two decades. S. pyogenes use multiple pathways for the acquisition and activation of human plasminogen, securing potent proteolytic activity on the bacterial cell surface. Recent experimental evidence using a humanized transgenic mouse model suggests a crucial role for human plasminogen in the dissemination of S. pyogenes in v

Role of laccase in the biology and virulence of Cryptococcus neoformans.

Laccase is an important virulence factor for the human pathogen, Cryptococcus neoformans. In this review, we examine the structural, biological and genetic features of the enzyme and its role in the pathogenesis of cryptococcosis. Laccase is expressed in C. neoformans as a cell wall enzyme that possesses a broad spectrum of activity oxidizing both polyphenolic compounds and iron. Two paralogs, CNLAC1 and CNLAC2, are present in the fungus, of which the first one expresses the dominant enzyme activity under glucose starvation conditions. Regulation of the enzyme is in response to various environ…

Surgical treatment of virulent descending necrotizing mediastinitis.

We report a 58-year-old male treated with surgical drainage by mediansternotomy using a pedicled omental flap for descending necrotizing mediastinitis (DNM). The patient recovered from DNM after five months of mechanical respiratory support. In deciding upon the most appropriate surgical approach for mediastinal drainage, the level of infection is a good landmark and should be investigated by CT scan. We also review the 43 cases of successful surgical treatment of DNM reported since 1989 in Japan, including our own patient, who were diagnosed with DNM by CT scan according to the classification

Virulence of Malaysian isolates of Orientia tsutsugamushi in mice.

The pathogenicity of Malaysian isolates of Orientia tsutsugamushi was investigated by a mouse virulence assay. The isolates could be differentiated as low (4 isolates), moderately (3 isolates) and highly virulent (2 isolates) based on the different responses in infected mice. No direct correlation between severity of human scrub typhus infections and virulence of the O. tsutsugamushi in mice was observed. Mice infected with virulent strains of O. tsutsugamushi showed splenomegaly, ascitis accumulation and enlargement of kidneys and livers whereas avirulent O. tsutsugamushi strains were asympto…

Virulence parameters in the characterization of strains of Entamoeba histolytica.

Differences in virulence of strains of Entamoeba histolytica have long been detected by various experimental assays, both in vivo and in vitro. Discrepancies in the strains characterization have been arisen when different biological assays are compared. In order to evaluate different parameters of virulence in the strains characterization, five strains of E. histolytica, kept under axenic culture, were characterized in respect to their, capability to induce hamster liver abscess, erythrophagocytosis rate and cytopathic effect upon VERO cells. It was found significant correlation between in vit…

Pyruvate oxidase, as a determinant of virulence in Streptococcus pneumoniae.

Pneumococcus has been shown to bind to epithelial cells of the nasopharynx and lung, and to endothelial cells of the peripheral vasculature. … ween the enzyme and the expression of bacterial adhesins. A defect in spxB corresponded to impaired virulence of the mutant in vivo. Compared to the parent strain, an spxB mutant showed reduced virulence in animal models for nasopharyngeal colonization, pneumonia, and sepsis. We propose that a mutation in spxB leads to down-regulation of the multiple adhesi …

Comparative virulence of periodontopathogens in a mouse abscess model.

OBJECTIVE(S): This report compares the virulence of selected strains of P. gingivalis, A. actinomycetemcomitans, C. rectus, F. nucleatum and T. denticola in a murine model as a measure of pathogenic potential of these oral microorganisms. The characteristics of the tissue destruction associated with these monoinfections were then related to a potential model for bacterial synergism in progressing periodontitis. DESIGN AND METHODS: All bacterial strains were grown to mid-logarithmic to early stationary growth phase, harvested and used at various doses to challenge BALB/c normal and BALB/c dexa…

[The connection of the antigenic activity of Leptospira to its virulence].

Virulence of Leptospira cultures isolated in leptospirosis centres of different regions of Ukraine has been studied. It is shown that certain strains of leptospiras with high virulence several times exceed in their antigenic activity the standard strains and their introduction into the set of strains for the reaction of microagglutination and use for making diagnostic and, probably, prophylactic preparations are promising.

[The virulence of L. leucocephala for goats in northeastern Mexico].

When fed 100% Leucaena during a period of 45 days goats with experience in L.-fodder took significantly (P less than 0.05) more dry matter (50.01 g/kg0.75) than those without or only possible experience (44.40 g/kg0.75 and 45.63 g/kg0.75). The uptake of Mimosine from animals with Mimosine experience (1.14 g/kg0.75) has also been significantly higher (P less than 0.05) than from two other groups with non or only possible experience (0.93 g/kg0.75 and 0.92 g/kg0.75). The Serum-Thyroxin level (T4) of the control group (5.12 micrograms/100 ml) and of the group with Leucaena experience (4.65 microg

[Populations of pathogenic Leptospira of varying virulence in nature].

The study of geographically remote populations of Norway rats (Rattus norvegicus) revealed that in one of these populations a highly virulent population of Leptospira copenhageni, serogroup icterohaemorrhagiae, and in another population of rats a faintly virulent population of these microorganisms circulated simultaneously. At the same time in vitro experiments with Leptospira cultures showed the absence of the constant probability of sharp changes in the level of their virulence in time.

The virulence of clinical and environmental isolates of Campylobacter jejuni.

The virulence of Campylobacter jejuni and C. coli isolated from various water sources was compared with that of clinical strains by in vitro assays of adhesion, invasion and cytotoxicity to HeLa cells. Variation in degree of attachment was observed, but this did not appear to be related to strain source, However, water strains were less invasive and less cytotoxic to HeLa cells than clinical strains as shown by immunofluorescence and electron microscopy. These differences were particularly evident between clinical and water isolates of the same serotype and biotype implicated in an outbreak of…

Haemolysis by urinary Escherichia coli and virulence in mice.

The influence of haemolysin production on virulence was studied in an experimental mouse model. Urinary strains of Escherichia coli can be divided into three virulence groups by determining their kinetics in the mouse kidney after intravenous injection. Virulent strains of groups II and III were more often haemolytic than avirulent group-I strains. Haemolytic virulent strains often caused haemoglobinuria in the mice, and killed the mice more rapidly than did non-haemolytic virulent strains. No relationship was found between alpha-haemolytic activity and virulence in wild-type haemolytic strain…
