Astronauts encounter several challenges on spaceflight missions, including space radiation and microgravity. Over the past several decades, studies conducted in the field of space medicine have uncovered broad impacts on human physiology, including the integumentary system, which serves as a protective barrier to environmental factors. Dermatological alterations in astronauts are commonly observed during spaceflight missions. Advancing our understanding of the intracellular mechanisms impacted in skin within the context of microgravity may uncover potential therapies for maintaining and improv

Microgravity (µg)-generated three-dimensional (3D) multicellular aggregates can serve as models of tissue and disease development. They are relevant in the fields of cancer and in vitro metastasis or regenerative medicine (tissue engineering). Driven by the 3R concept-replacement, reduction, and refinement of animal testing-µg-exposure of human cells represents a new alternative method that avoids animal experiments entirely. New Approach Methodologies (NAMs) are used in biomedical research, pharmacology, toxicology, cancer research, radiotherapy, and translational regenerative medicine. Vario

OBJECTIVE: This study aims to investigate the expression and possible role of autophagy in the retina of rats under microgravity.
METHODS: Adult Sprague-Dawley (SD) rats were randomly allocated to either the tail suspension group (TS) or the control group (CTRL). To simulate microgravity-induced redistribution of cephalad fluid observed in space, the rats in the TS group underwent tail suspension for a duration of 4 weeks. Optical coherence tomography angiography (OCTA) was applied to assess the ocular blood flow and thickness of the retina. Hematoxylin and eosin (H&E) staining, along with tr

BACKGROUND: Imaging plays a critical role in evaluating gynecologic emergencies. However, access to such technology is limited in austere environments such as spaceflight. Portable point-of-care ultrasonography (POCUS) offers rapid diagnostic capabilities potentially useful for supporting women's health. However, the feasibility of using portable POCUS for gynecological imaging in spaceflights characterized by shifts in weightlessness has not been previously evaluated.
INSTRUMENT: Transabdominal ultrasonography was undertaken to assess the feasibility of using portable POCUS for gynecologic s…

INTRODUCTION: As spaceflight missions become more frequent and prolonged, the effects of microgravity on the musculoskeletal system represent a critical concern for astronauts' health given their increased risk of spinal pain and injury. A better understanding of the adaptations induced by microgravity on neuromuscular control of the spine is essential to guide the development of effective countermeasures. Thus, this systematic review will aim to investigate the effects of microgravity on the neuromuscular control of the spine.
METHODS AND ANALYSIS: This protocol has been developed following

For more than 60 years, humans have travelled into space. Until now, the majority of astronauts have been professional, government agency astronauts selected, in part, for their superlative physical fitness and the absence of disease. Commercial spaceflight is now becoming accessible to members of the public, many of whom would previously have been excluded owing to unsatisfactory fitness or the presence of cardiorespiratory diseases. While data exist on the effects of gravitational and acceleration (G) forces on human physiology, data on the effects of the aerospace environment in unselected

Mast cell (MC)-specific proteases are of particular interest for space biology and medicine due to their biological activity in regulating t … s-vivarium control and anti-orthostatic suspension-included in the experiment simulating effects of weightlessness in the ground-based conditions. After a space flight, there was a decreased number of MCs in the studied organs combined with an increased proportion of chymase-positive MCs and MCs with a simultaneous content of tryptase and chymase; the secretion …

INTRODUCTION: Space Motion Sickness (SMS) is a syndrome that affects around 70% of astronauts and includes symptoms of nausea, dizziness, fatigue, vertigo, headaches, vomiting, and cold sweating. Consequences range from discomfort to severe sensorimotor and cognitive incapacitation, which might cause potential problems for mission-critical tasks and astronauts and cosmonauts' well-being. Both pharmacological and non-pharmacological countermeasures have been proposed to mitigate SMS. However, their effectiveness has not been systematically evaluated. Here we present the first systematic review

BACKGROUND: We have previously shown that the anti-tumor activity of human lymphocytes is diminished in vitro after 12-hours pre-exposure to simulated microgravity (SMG). Here we used an immunocompromised mouse model to determine if this loss of function would extend in vivo, and to also test the efficacy of IL-2 and zoledronic acid (ZOL) therapy as a potential countermeasure against SMG-induced immune dysfunction. We adoptively transferred human lymphocytes that were exposed to either SMG or 1G-control into NSG-Tg (Hu-IL15) mice 1-week after they were injected with a luciferase-tagged human c

BACKGROUND: Spaceflight places astronauts in multiple environments capable of inducing pathological changes. Alterations in the spine have a significant impact on astronauts' health during and after spaceflight. Low back pain is an established and common intra-flight complaint. Intervertebral disc herniation occurs at higher rates in this population and poses significant morbidity. Morphological changes within intervertebral discs, vertebral bodies, and spinal postural muscles affect overall spine function and astronaut performance. There remains a paucity of research related to spaceflight-in

The peripheral vestibular organs are sensors for linear acceleration (gravity and head tilt) and rotation. Further, they regulate various body functions, including body stability, ocular movement, autonomic nerve activity, arterial pressure, body temperature, and muscle and bone metabolism. The gravitational environment influences these functions given the highly plastic responsiveness of the vestibular system. This review demonstrates that hypergravity or microgravity induces changes in vestibular-related physiological functions, including arterial pressure, muscle and bone metabolism, feedin

INTRODUCTION: A majority of astronauts experience symptoms of headache, vomiting, nausea, lethargy, and gastric discomfort during the first few hours or days after entering a microgravity environment. Due to similarities in symptoms and their time evolution, it has been hypothesized that some of these conflicts are related to the development of benign intracranial hypertension in these individuals in microgravity.
METHODS: This hypothesis was tested using a validated mathematical model that embeds the intracranial system in whole-body physiology. This model was used to predict steady-state in

Arterial blood pressure (BP) is one of the four life signs in human beings, it could reflect cardiovascular function in some degrees. Thus, BP serves as an important cardiovascular parameter during various stages in foreign spaceflight activities. Although the reports about BP are not always in consistency during spaceflight, hypotension commonly occurs during orthostatic stress after spaceflight. The mechanisms of orthostatic intolerance after spaceflight are virtually the same as that of orthostatic hypotension after spaceflight. This paper simply reviewed the reports on BP during various st

Space flight exerts substantial effects on fluid volume control in humans. Cardiac distension occurs during the first 1-2 days of space flight relative to supine and especially upright 1g conditions. Plasma volume contraction occurs quickly in microgravity, probably as a result of transcapillary fluid filtration into upper-body interstitial spaces. No natriuresis or diuresis has been observed in microgravity, such that diuresis cannot explain microgravity-induced hypovolemia. Reduction of fluid intake occurs irrespective of space motion sickness and leads to hypovolemia. The fourfold elevation

Intensity of osteoclastic resorption and calcium content were investigated in intact limb bones of the newts flown on board of a biosatellite Cosmos-2229 after amputation of their forelimbs and tail. Using X-ray microanalysis it was shown an increase in calcium content in the bones on 20th day after operation. Histological study revealed an activation of osteoclastic resorption on endosteal surface of long bones. The newts exposed after surgery on a biosatellite had the same level of bone mineralisation as operated ground control ones, but the increase in number of polynuclear osteoclasts was

Ten selected strains of enterococci and staphylococci were isolated from the crop and caecum of Japanese quails exposed to microgravity conditions. Isolates were allotted to the species Enterococcus gallinarum, Ent. avium, Ent. faecium, Staphylococcus gallinarum and Staph. aureus. Isolated strains were facultatively anaerobic, non-motile, Gram-positive cocci, occurring in pairs, short chains or irregular clusters. Isolates utilized most of the soluble sugars tested and produced bacteriocin-like substances. Enterococci and staphylococci were resistant to monensin (50 mg/l) and sensitive to tylo
