Dementia with Lewy bodies in an elderly Greek male due to alpha-synuclein gene mutation.

We report the case of an elderly man of Greek background who presented with progressive cognitive decline and motor parkinsonism on a background of a strong family history of Parkinson's disease. Associated symptoms included visual hallucinations, excessive daytime drowsiness, recurrent falls, orthostatic hypotension and urinary incontinence. His major clinical symptoms and signs fulfilled consensus criteria for a clinical diagnosis of dementia with Lewy bodies. An alpha-synuclein gene mutation analysis for the G209A substitution was positive. We conclude that the alpha-synuclein (G209A) gene …

Alpha-synuclein accumulates in Purkinje cells in Lewy body disease but not in multiple system atrophy.

Alpha-synuclein has an important role in the pathogenesis of Parkinson disease (PD), dementia with Lewy bodies (DLB), and multiple system atrophy (MSA), comprising a new disease concept, that of alpha-synucleinopathies. Cerebellar degeneration with Purkinje cell depletion is present in the majority of MSA cases. By contrast, cerebellar pathology has not been demonstrated unequivocally in either PD or DLB. Recent immunohistochemical studies using anti-alpha-synuclein antibodies have shown that LB-type degeneration in PD and DLB is more widespread than previously recognized. To determine whether…

Behavioral and neurochemical effects of wild-type and mutated human alpha-synuclein in transgenic mice.

Human alpha-synuclein (halpha-SYN) is implicated in the Parkinson's disease phenotype (PDP) based on a variety of studies in man, animal models, and in vitro studies. The normal function of halpha-SYN and the mechanism by which it contributes to the PDP remains unclear. We created transgenic mice expressing either wild-type (hwalpha-SYN) or a doubly mutated (hm2alpha-SYN) form of halpha-SYN under control of the 9-kb rat tyrosine hydroxylase promoter. These mice expressed halpha-SYN in cell bodies, axons, and terminals of the nigrostriatal system. The expression of halpha-SYN in nigrostriatal t…

Analysis of the coding and the 5' flanking regions of the alpha-synuclein gene in patients with Parkinson's disease.

Missense mutations of the alpha-synuclein gene have been reported to explain a few kindreds with autosomal dominant Parkinson's disease (PD). In order to identify mutations in our PD patients, we have screened the coding region and 5'flanking region of the gene. DNA samples from 50 patients with familial PD were screened via single-strand conformation polymorphism (SSCP) for mutations in the alpha-synuclein gene. The 5' flanking region was examined in 117 additional PD patients (27 patients with unclear family history for PD, and 90 patients without family history) and in 169 control subjects.…

Tear fluid as a novel specimen for detection of alpha-synuclein seeding activity in Parkinson's disease.

Parkinson’s disease (PD) is a clinically heterogeneous neurodegenerative disorder characterized by intraneuronal accumulation of misfolded α-synuclein. Real-time quaking-induced conversion (RT-QuIC) detects seeding-competent αSyn species but commonly relies on invasive matrices. We evaluated tear fluid (TF) as a non-invasive biofluid for αSyn RT-QuIC in 44 PD patients and 32 matched healthy controls (HCs). ROC analyses were performed on participant-level readouts (mean of four technical replicates), and optimal cut-offs were selected using the Youden index before being applied to each technica

Alpha-synuclein at the crossroads of host-virus interactions: immunological roles beyond the nervous system.

Alpha-synuclein (α-syn) is best known as a presynaptic protein that supports synaptic vesicle dynamics and neurotransmission. Conversely, misfolded or aggregated α-syn represents a hallmark of synucleinopathies, including Parkinson's disease. Beyond the nervous system, α-syn has been detected in peripheral compartments, including blood cells and selected epithelial tissues, although the robustness and context dependence of expression outside neuronal and erythroid lineages remain under active investigation. Also, it can be released extracellularly through unconventional secretion or cell damag…

Alpha- and Gamma-Synucleins Regulate Energy Metabolism and Xanthine Oxidase Activity in Brain Cells.

The process of signal transmission and transformation in the central nervous system requires active energy metabolism with high consumption of glucose and oxygen. Reactive oxygen species (ROS) produced as a result of these processes participate in intracellular signaling, but their overproduction leads to oxidative stress. Oxidative stress and α-synuclein aggregation are recognized as activators of neuronal death in Parkinson's disease. However, much less is known about the physiological role of monomeric synucleins. Using acute brain slices and primary co-cultures of cortical neurons and glia

A Mendelian randomization study on association of alpha-synuclein and GPNMB with Parkinson's disease risk and progression.

The prevalence of Parkinson's disease (PD) is increasing due to the aging population. Early diagnosis and prognosis of PD remain challenging, suggesting that seeking appropriate biomarkers for PD is crucial. Glycoprotein nonmetastatic melanoma protein B (GPNMB) and alpha-synuclein (α-syn) have been reported to contribute to PD pathogenesis and are correlated with PD onset and disease progression. We utilized Mendelian randomization (MR) analysis to elucidate the association of GPNMB and α-syn with PD and its disease progression. Six MR approaches were employed, and inverse variance weighted wa…

The extent of alpha-synuclein aggregate accumulation in neurons varies depending on the type of neurotransmitter they release.

Alpha-synuclein (aSyn) is a highly conserved 140-amino acid presynaptic protein involved in neuronal plasticity and recognized as the pathogenic protein in Parkinson's disease. Aggregated aSyn propagates transsynaptically between neurons in vivo, impairing the cellular function of affected cells. However, it remains unclear whether aggregated aSyn propagates similarly across all neuronal cell types. In this study, we injected sonicated aSyn preformed fibrils into the striatum of mice and used immunohistochemistry to observe the intracellular localization of aSyn aggregates at designated surviv…

ADVANCEMENT IN ALPHA-SYNUCLEIN PROTEOMICS: EXPLORING ANALYTICAL TECHNIQUES AND THEIR CLINICAL IMPLICATIONS IN PARKINSON'S DISEASE.

Parkinson's disease (PD) is a prevalent neurodegenerative disorder, affecting around 500,000 to 1 million Americans, with a significant portion diagnosed before age 50. Despite advances in treatments such as dopamine replacement therapy and deep brain stimulation, no therapies currently exist to halt or slow disease progression in advanced stages. Research is increasingly focused on identifying early biomarkers for PD to enable earlier intervention. Alpha-synuclein (α-Syn), a key protein implicated in PD pathology, is studied using various proteomics techniques like mass spectrometry, gel elec…

A Structural Proteomics Exploration of Synphilin-1 and Alpha-Synuclein Interaction in Pathogenesis of Parkinson's Disease.

Pathological significance of interaction of Synphilin-1 with mutated alpha-synuclein is well known to have serious consequences in causing the formation of inclusion bodies that are linked to Parkinson's disease (PD). Information extracted so far pointed out that specific mutations, A53T, A30P, and E46K, in alpha-synuclein promote such interactions. However, a detailed structural study of this interaction is pending due to the unavailability of the complete structures of the large protein Synphilin-1 of chain length 919 residues and the mutated alpha-synuclein having all the reported specific …

Visualizing alpha-synuclein and iron deposition in M83 mouse model of Parkinson's disease in vivo.

Abnormal alpha-synuclein (αSyn) and iron accumulation in the brain play an important role in Parkinson's disease (PD). Herein, we aim to visualize αSyn inclusions and iron deposition in the brains of M83 (A53T) mouse models of PD in vivo. The fluorescent pyrimidoindole derivative THK-565 probe was characterized by means of recombinant fibrils and brains from 10- to 11-month-old M83 mice. Concurrent wide-field fluorescence and volumetric multispectral optoacoustic tomography (vMSOT) imaging were subsequently performed in vivo. Structural and susceptibility weighted imaging (SWI) magnetic resona…

Overexpression of alpha synuclein disrupts APP and Endolysosomal axonal trafficking in a mouse model of synucleinopathy.

Mutations or triplication of the alpha synuclein (ASYN) gene contribute to synucleinopathies including Parkinson's disease (PD), Dementia with Lewy bodies (DLB) and multiple system atrophy (MSA). Recent evidence suggests that ASYN also plays an important role in amyloid-induced neurotoxicity, although the mechanism(s) remains unknown. One hypothesis is that accumulation of ASYN alters endolysosomal pathways to impact axonal trafficking and processing of the amyloid precursor protein (APP). To define an axonal function for ASYN, we used a transgenic mouse model of synucleinopathy that expresses

A review of the current research on in vivo and in vitro detection for alpha-synuclein: a biomarker of Parkinson's disease.

Parkinson's disease is a health-threatening neurodegenerative disease of the elderly with clinical manifestations of motor and non-motor deficits such as tremor palsy and loss of smell. Alpha-synuclein (α-Syn) is the pathological basis of PD, it can abnormally aggregate into insoluble forms such as oligomers, fibrils, and plaques, causing degeneration of nigrostriatal dopaminergic neurons in the substantia nigra in the patient's brain and the formation of Lewy bodies (LBs) and Lewy neuritis (LN) inclusions. As a result, achieving α-Syn aggregate detection in the early stages of PD can effectiv…

Erythrocytic alpha-synuclein as potential biomarker for the differentiation between essential tremor and Parkinson's disease.

INTRODUCTION: The differentiation between essential tremor (ET) and Parkinson's disease (PD) can be difficult because of the symptom overlaps. Erythrocytes are the major source of peripheral α-synuclein (α-syn), which is the most studied pathological molecular of PD. We have reported that erythrocytic α-syn levels in PD patients are significantly increased compared to those in healthy controls (HCs). However, little is known about the levels of erythrocytic α-syn species in ET patients. METHODS: This study includes 15 patients with ET, 64 patients with PD, and 49 age and sex matched HCs. A we

Exacerbation of sensorimotor dysfunction in mice deficient in Atp13a2 and overexpressing human wildtype alpha-synuclein.

Loss of function mutations in the gene ATP13A2 are associated with Kufor-Rakeb Syndrome and Neuronal Ceroid Lipofuscinosis, the former designated as an inherited form of Parkinson's disease (PD). The function of ATP13A2 is unclear but in vitro studies indicate it is a lysosomal protein and may interact with the presynaptic protein alpha-synuclein (aSyn) and certain heavy metals. Accumulation of aSyn is a major component of lewy bodies, the pathological hallmark of PD.
