Laboratory and preliminary clinical characterization of Vi capsular polysaccharide-protein conjugate vaccines.

To improve its immunogenicity for children and adults and to make it suitable for routine immunization of infants against typhoid fever, the capsular polysaccharide of Salmonella typhi (Vi) was bound to the B subunit of the heat-labile toxin (LT-B) of Escherichia coli or the recombinant exoprotein A (rEPA) of Pseudomonas aeruginosa. The conjugates elicited higher levels of antibodies (micrograms per milliliter of serum) in mice and in guinea pigs than did Vi and, unlike Vi alone, elicited booster antibody responses in both species. In adult volunteers, Vi-LT-B and Vi-rEPA, respectively, elicit

Protective immunity induced by 67 K outer membrane protein of phase I Coxiella burnetii in mice and guinea pigs.

A 67 K outer membrane protein (OMP) isolated from phase I Coxiella burnetii QiYi strain was purified with monoclonal antibodies (MoAb) coupled to CNBr-Sepharose 4B. Chemical analyses of the 67 K protein showed that it contained seventeen kinds of amino acids and no LPS. The immunogenicity and protectivity of the 67 K protein against C. burnetii was evaluated in mice and guinea pigs by in vitro lymphocyte proliferation assay, delayed-type skin test, antibody conversion rate, and immunization and challenge tests. Intraperitoneal injection of the 67 K protein resulted in antibody production again

Protein/AS01B vaccination elicits stronger, more Th2-skewed antigen-specific human T follicular helper cell responses than heterologous viral vectors.

Interactions between B cells and CD4+ T follicular helper (Tfh) cells are key determinants of humoral responses. Using samples from clinical trials performed with the malaria vaccine candidate antigen Plasmodium falciparum merozoite protein (PfRH5), we compare the frequency, phenotype, and gene expression profiles of PfRH5-specific circulating Tfh (cTfh) cells elicited by two leading human vaccine delivery platforms: heterologous viral vector prime boost and protein with AS01B adjuvant. We demonstrate that the protein/AS01B platform induces a higher-magnitude antigen-specific cTfh cell respons

Differential Evolutionary Selection and Natural Evolvability Observed in ALT Proteins of Human Filarial Parasites.

The abundant larval transcript (ALT-2) protein is present in all members of the Filarioidea, and has been reported as a potential candidate antigen for a subunit vaccine against lymphatic filariasis. To assess the potential for vaccine escape or heterologous protection, we examined the evolutionary selection acting on ALT-2. The ratios of nonsynonymous (K(a)) to synonymous (K(s)) mutation frequencies (ω) were calculated for the alt-2 genes of the lymphatic filariasis agents Brugia malayi and Wuchereria bancrofti and the agents of river blindness and African eyeworm disease Onchocerca volvulus

[Culture of rabbit parotid acinar cells and evaluation of its excretion function in vitro].

Shigellosis is a global human health problem. Four species of Shigella i.e. S. dysenteriae, S. flexneri, S. boydii and S. sonnei are able to cause the disease. These species are subdivided into serotypes on the basis of O-specific polysaccharide of the LPS. Shigella dysenteriae type 1 produces severe disease and may be associated with life-threatening complications. The symptoms of shigellosis include diarrhoea and/or dysentery with frequent mucoid bloody stools, abdominal cramps and tenesmus. Shigella spp. cause dysentery by invading the colonic mucosa. Shigella bacteria multiply within colon

cGAMP-Loaded M2e Nanovaccine Elicits Cross-Reactive Immunity and Mitigates H6N1 Avian Influenza Infection in Chickens.

INTRODUCTION: Avian influenza virus (AIV) is a rapidly evolving zoonotic pathogen that causes significant economic loss in the poultry industry and threatens public health. In the development of a broadly protective influenza vaccine, the extracellular domain of matrix protein 2 (M2e) is a promising antigen candidate as it is highly conserved across most influenza A subtypes. M2e-induced immune protection is primarily mediated through antibody-dependent cellular cytotoxicity (ADCC), which enables the clearance of infected cells before viral release. However, the immunogenicity of M2e is limite

Presentation of the protective epitope in a Chlamydia muridarum MOMP vaccine is dependent on γ-interferon-inducible lysosomal thiol reductase (GILT).

The Chlamydia outer membrane complex has approximately 20 cysteine-rich proteins, including the major outer membrane protein (MOMP) with 8 cysteines. The enzyme γ-interferon-inducible lysosomal thiol reductase (GILT) is expressed constitutively in lysosomes of antigen presenting cells and reproductive tract epithelial cells. GILT catalyzes disulfide bond reduction during protein degradation that generates T cell epitopes. To evaluate the role that GILT plays in chlamydial infections, C57BL/6 WT mice and mice deficient in Ifi30 (GILT-KO) were challenged vaginally with Chlamydia muridarum (Cm).

ESAT-6-derived lipopeptide-based mucosal vaccine elicits distinct humoral immune responses concomitant with a reduction in mycobacterium infection.

Respiratory mycobacterial infections remain a major global health threat, yet current vaccine strategies, which focus primarily on cell-mediated immunity, have met with limited success. We previously demonstrated that a vaccine composed of five lipopeptides (LPs) encoding immunodominant ESAT-6 epitopes elicits protective cellular immune responses against Mycobacterium tuberculosis infection. In this study, we enhanced the ESAT-LPs vaccine with a novel mucosal immune modulator, heat-killed Caulobacter crescentus (HKCC), and examined both cellular and humoral immunity in a mouse model (BALB/c) o

Prediction and validation of potent peptides against herpes simplex virus type 1 via immunoinformatic and systems biology approach.

The human herpes simplex virus type 1 (HSV-1) is an extremely rampant human pathogen, and its infection could cause life-long diseases, including the central nervous system disorders. The glycoproteins of HSV-1 such as glycoprotein B, glycoprotein C, glycoprotein D, glycoprotein H, and glycoprotein L are highly involved in mediating the viral attachment and infection of the host cell. Therefore, immunoinformatic approaches followed by molecular dynamics simulation and systems biology has been used to analyze these glycoproteins in order to propose effective peptide-based vaccine candidates aga

Molecular characterization and immunoinformatics-based design of a multi-epitope vaccine against Staphylococcus nepalensis.

Staphylococcus nepalensis has recently been identified in clinical settings, raising questions regarding its potential role as an emerging opportunistic pathogen. However, no vaccine-based preventive strategy has yet been proposed against this bacterium. The objective of this study was to characterize a clinical isolate using PCR-based molecular identification and to design a rational multi-epitope vaccine candidate against S. nepalensis through an integrated immunoinformatics-driven reverse vaccinology approach. Species-level identification was performed by PCR amplification and sequencing of

Nanoadjuvant and recombinant hemagglutinin protein-based mucosal influenza nanovaccine confers protection against lethal viral challenge in mice.

Influenza virus poses a constant pandemic threat and global health challenge. As the first line of defense at the virus entry portal, mucosal immunity is essential to provide cross-protection against influenza virus. Hemagglutinin (HA), a major influenza virus antigen, could not penetrate the nasal barriers and is prone to removal by mucociliary clearance when nasally administered alone. Appropriate mucosal adjuvants and delivery systems were necessitated to induce the mucosal immunity. In this study, a novel mucosal nanovaccine (rHA-GEP-cG) was prepared by assembly of HA with β glucan, polype

A novel peptide cocktail containing B and T cell epitopes confers robust protection against Staphylococcus aureus bacteremia in a murine model.

Previous Staphylococcus aureus vaccines have been unsuccessful, but it seems that including T cell immunity in the vaccine is essential for its success. This study used Metal ABC transporter substrate-binding protein (MABC), Nickel ABC transporter (NABC), and Phosphatidylinositol phosphodiesterase (PIc) B and T cell epitopes as a vaccine in a mouse bacteremia model. Mice immunized with PIc and epitope mixture groups showed robust humoral immunity and high survival rates in the bacteremia model. The highest protection level of mice was shown in the peptide mixture group, which was correlated wi

Characterization of the enterovirus 71 VP1 protein as a vaccine candidate.

Enterovirus 71 (EV71) is an important agent responsible for hand-foot-and-mouth disease (HFMD), which can cause severe neurological complications and death in children. However, there is no specific treatment for EV71 infection, and a safe and effective vaccine is needed urgently. In this study, an effective and economical method for the production of EV71-VP1 protein was developed, and the VP1 protein was evaluated in humoral and cellular immune responses as an EV71 vaccine. The results revealed that the VP1 protein induced high titers of cross-neutralizing antibodies for different EV71 subty

Investigation of carbohydrate-based molecules of Theileria parva parasites.

Theileria parva (Tp), the causative agent of East Coast Fever, poses a major threat to Bos taurus cattle in sub-Saharan Africa. Progress towards a stable, affordable, cross-protective subunit vaccine covering more than one major histocompatibility complex (MHC)-presented Tp antigen has been hindered by the limited knowledge of antigens. Given the importance of glycoconjugates in protozoan pathogen biology, especially during the invasion process, we investigated the glycosylation potential of Tp. Screening of Tp Muguga (TpM) schizonts with C-type lectin (-like) receptors showed that ruminant, b

Vanishing White Matter Disease in Children: An Unusual Association, a Novel Mutation, and a Literature Review.

Vanishing white matter (VWM) disease is an autosomal recessive disorder caused by mutations in the gene EIF2B encoding the subunits 1-5 of eukaryotic initiation factor 2B. Although rare, with a reported prevalence of 1:80,000 (0.001%), it was considered as one of the most common leukodystrophies. However, the worldwide incidence and prevalence of this disease are not clear. In Bahrain, of 21 patients who were diagnosed with leukodystrophy, two patients were found to have VWM disease accounting for 9.5%. Vaccinations and infections were the trigger factors for this disease to manifest. Rapid ne

Neutralizing monoclonal antibodies against the Gc fusion loop region of Crimean-Congo hemorrhagic fever virus.

Crimean-Congo hemorrhagic fever virus (CCHFV) is a highly pathogenic tick-borne virus, prevalent in more than 30 countries worldwide. Human infection by this virus leads to severe illness, with an average case fatality of 40%. There is currently no approved vaccine or drug to treat the disease. Neutralizing antibodies are a promising approach to treat virus infectious diseases. This study generated 37 mouse-derived specific monoclonal antibodies against CCHFV Gc subunit. Neutralization assays using pseudotyped virus and authentic CCHFV identified Gc8, Gc13, and Gc35 as neutralizing antibodies.

Sweet complexity: O-linked protein glycosylation in pathogenic Neisseria.

The genus Neisseria, which colonizes mucosal surfaces, includes both commensal and pathogenic species that are exclusive to humans. The two pathogenic Neisseria species are closely related but cause quite different diseases, meningococcal sepsis and meningitis (Neisseria meningitidis) and sexually transmitted gonorrhea (Neisseria gonorrhoeae). Although obvious differences in bacterial niches and mechanisms for transmission exists, pathogenic Neisseria have high levels of conservation at the levels of nucleotide sequences, gene content and synteny. Species of Neisseria express broad-spectrum O-

[Bacillus Calmette-Guérin infection and chronic granulomatous disease due to new pathogenic variants in the NCF2 gene in the Mayan ethnic group. Report of two cases.].

INTRODUCTION: Chronic granulomatous disease (CGD) is an inborn error of immunity, characterized by abnormal susceptibility to bacterial and fungal infections and a lack of systemic inflammatory regulation. Pathogenic variants in the CYBB gene are transmitted in an X-linked pattern of inheritance; while the pathogenic variants present in the EROS, NCF1, NCF2, NCF4, or CYBA genes are transmitted with an autosomal recessive inheritance pattern. OBJETIVES: To describe the clinical, immunological, and genetic characteristics of two patients with CGD and BCG infection. METHODS: In peripheral blood

HR121 targeting HR2 domain in S2 subunit of spike protein can serve as a broad-spectrum SARS-CoV-2 inhibitor via intranasal administration.

The continuously emerging SARS-CoV-2 variants pose a great challenge to the efficacy of current drugs, this necessitates the development of broad-spectrum antiviral drugs. In the previous study, we designed a recombinant protein, heptad repeat (HR) 121, as a variant-proof vaccine. Here, we found it can act as a fusion inhibitor and demonstrated broadly neutralizing activities against SARS-CoV-2 and its main variants. Structure analysis suggested that HR121 targets the HR2 domain in SARS-CoV-2 spike (S) 2 subunit to block virus-cell fusion. Functional experiments demonstrated that HR121 can bin

Protective Human Anti-Poxvirus Monoclonal Antibodies Are Generated from Rare Memory B Cells Isolated by Multicolor Antigen Tetramers.

Smallpox, an epidemic disease caused by Orthopoxvirus variola, was eradicated worldwide through immunization. The immunization against smallpox was discontinued in 1980. However, incidences of monkeypox virus infection in humans have occurred sporadically, and there is also great fear that engineered forms of poxvirus could be used as biological weapons. Therefore, monoclonal antibodies against poxvirus are urgently needed for the detection and treatment of poxvirus infection. The vaccinia virus' extracellular envelope protein A33 is a potential candidate for a subunit vaccine. We used multi-f
