Tumor cells expressing the herpes simplex virus-thymidine kinase gene in the treatment of Walker 256 meningeal neoplasia in rats.

A promising strategy in the treatment of neoplastic meningitis involves the use of herpes simplex virus-thymidine kinase (HSV-tk)-modified cells. In these experiments the authors used cells expressing HSV-tk to treat meningeal carcinomatosis in the rat Walker 256 model. Intrathecal injection of 2 x 10(5) Walker cells resulted in a median survival time of 15 days. Up to 80% of animals implanted with HSV-tk-modified Walker cells (Walker-tk+) and treated with ganciclovir showed long-term survival (120 days or more), whereas the remaining animals died from tumor growth between 37 and 44 days after

Genome-based characterization of a multifunctional plant growth-promoting and heavy metal-resistant Escherichia coli FACU2024 isolated from Jatropha rhizosphere.

Whole-genome sequencing (WGS) of microbial isolates is a valuable tool for mapping the genomes of novel organisms and is helpful for understanding plant-bacteria interactions. The close relationships between bacteria and plants are essential for maintaining healthy ecosystems, whether the bacteria reside the plant or in the rhizosphere surrounding its roots. In this study, isolation, characterization, and WGS were performed to identify promising plant growth-promoting rhizobacteria (PGPR) using the rhizospheric soil sample of jatropha tree roots. Out of 100 isolates, six (FACU 2024, 2, 3, 4, 5

FODMAP intake, gut microbiota, and gastroesophageal reflux disease: a triangulated study of Mendelian randomization and clinical validation.

BACKGROUND: Dietary factors related to fermentable oligosaccharides, disaccharides, monosaccharides and polyols (FODMAPs) and gut microbiota may influence gastroesophageal reflux disease (GERD), but causal evidence and clinical consistency remain uncertain. OBJECTIVE: We tested whether FODMAP-related and GERD-relevant dietary traits influence GERD risk using Mendelian randomization (MR), and whether gut microbiota mediate these associations. METHODS: We integrated UK Biobank dietary GWAS, MiBioGen microbiota GWAS, and FinnGen GERD summary statistics using inverse-variance weighted MR, multiv

Poria cocos polysaccharide alleviates multidrug-resistant Klebsiella pneumoniae-induced acute lung injury by modulating the gut-lung axis via gut microbiota remodeling.

The emergence and spread of multidrug-resistant Klebsiella pneumoniae (MDR-KP), which leads to acute lung injury (ALI) and severe pneumonia, pose a huge challenge to clinical treatment and public health. Non-antibiotic strategies have recently garnered significant attention as promising avenues for combating antibiotic resistance. In this study, whole-genome sequencing of an MDR-KP strain isolated from a deceased goat revealed key genetic features underlying its pathogenicity, including 31 resistance genes mediating broad-spectrum resistance to 14 antibiotic classes, 71 pathogen-host interacti

Association of Staphylococcus aureus virulence factors with clinical and subclinical bovine mastitis.

Staphylococcus aureus is a major pathogen responsible for both clinical and subclinical bovine mastitis. Its ability to persist within the mammary gland and evade host immune responses makes it particularly significant for herd health and dairy production. So, this study aimed to evaluate whether the presence and diversity of S. aureus virulence genes mastitis-causing are associated with the onset and severity of mastitis in dairy cows. We analyzed 50 S. aureus isolates obtained from milk samples of cows with subclinical mastitis (SM) and 101 isolates from clinical mastitis (CM), including 28

Close interactions between prokaryotes and plasmids or viruses highlight a pivotal role of horizontal gene transfer in shaping antibiotic/metal(loid) resistome and their prokaryotic supercarriers in untreated hospital sewage.

Unveiling horizontal gene transfer (HGT) of antibiotic (ARGs) and metal(loid) resistance genes (MRGs) in hospital sewage is critical for surveilling antimicrobial resistance (AMR) mobility that poses huge threats to public health. Using metagenomic shotgun sequencing, we provided an integrate insight into AMR characters and the relevant HGT in untreated sewage from one of the world's largest comprehensive hospitals from Oct 2022 to Aug 2023. We uncovered higher richness and diversity of ARGs or MRGs than mobile genetic elements (MGEs), while MGEs exhibited the highest abundance, suggesting gre

The role of MorI/MorR quorum sensing in Methylobacterium oryzae CBMB20: modulating bacterial functions for enhanced adaptability.

UNLABELLED: The quorum-sensing (QS) system in Methylobacterium oryzae CBMB20, an endophytic bacterium associated with rice (Oryza sativa L.), plays a critical role in regulating bacterial behaviors essential for plant growth promotion and adaptation. This study aimed to elucidate the functional role of the MorI/MorR QS system in M. oryzae CBMB20 and its potential application as a bioinoculant. We identified and characterized two QS signals, 3-OH-C12-HSL and 3-oxo-C12-HSL, synthesized by the MorI enzyme. The MorR receptor was found to preferentially respond to 3-OH-C12-HSL, indicating a high de

Enabling malic acid production from corn-stover hydrolysate in Lipomyces starkeyi via metabolic engineering and bioprocess optimization.

BACKGROUND: Lipomyces starkeyi is an oleaginous yeast with a native metabolism well-suited for production of lipids and biofuels from complex lignocellulosic and waste feedstocks. Recent advances in genetic engineering tools have facilitated the development of L. starkeyi into a microbial chassis for biofuel and chemical production. However, the feasibility of redirecting L. starkeyi lipid flux away from lipids and towards other products remains relatively unexplored. Here, we engineer the native metabolism to produce malic acid by introducing the reductive TCA pathway and a C4-dicarboxylic ac

Molecular characterization of vancomycin-resistant Staphylococcus aureus isolated from bovine milk.

Vancomycin-resistant Staphylococcus aureus (VRSA) is a zoonotic life-threatening pathogen. Vancomycin exhibits anti-bacterial activity by inhibiting peptidoglycan synthesis by binding to the D-ala-D-ala terminus of the peptidoglycan. But in VRSA, D-ala-D-ala is replaced by D-ala-D-lactate due to the presence of vanA, vanB or vanD genes. This study was intended to identify the molecular prevalence of VRSA in 768 bovine milk samples, risk factor association, antibiogram profile and bioinformatics analysis of VRSA by targeting vanB gene. Out of a total of 248 S. aureus isolates from mastitic milk
