Effects of In Vivo Gluten Challenge on PBMC Gene Expression Profiles in Diet Treated Celiac Disease.

The pathological mechanisms that lead to the onset and reactivation of celiac disease (CD) remain largely unknown. While gluten free diet (GFD) improves the intestinal damage and associated clinical symptoms in majority of cases, it falls short of providing full recovery. Additionally, late or misdiagnosis is also common as CD presents with a wide range of symptoms. Clear understanding of CD pathogenesis is thus critical to address both diagnostic and treatment concerns. We aimed to study the molecular impact of short gluten exposure in GFD treated CD patients, as well as identify biological p

Genetic Relatedness of NDM-Producing Klebsiella pneumoniae Co-Occurring VIM, KPC, and OXA-48 Enzymes from Surveillance Cultures from an Intensive Care Unit.

Klebsiella pneumoniae is an opportunistic bacterial pathogen most commonly associated with nosocomial infections, especially in intensive care unit (ICU) patients. Routine surveillance cultures for carbapenemase-producing Enterobacteriaceae have become a common practice for hospital infection prevention. The objective of this study was to investigate the genetic relatedness of carbapenem-resistant K. pneumoniae carrying blaNDM gene across multilocus sequence typing (MLST) scheme. Surveillance rectal swabs from 4,463 ICU patients admitted to the Rio de Janeiro hospital (March 2016-2017) were sc

New and Preliminary Evidence on Altered Oral and Gut Microbiota in Individuals with Autism Spectrum Disorder (ASD): Implications for ASD Diagnosis and Subtyping Based on Microbial Biomarkers.

Autism Spectrum Disorder (ASD) is a complex neurological and developmental disorder characterized by behavioral and social impairments as well as multiple co-occurring conditions, such as gastrointestinal abnormalities, dental/periodontal diseases, and allergies. The etiology of ASD likely involves interaction between genetic and environmental factors. Recent studies suggest that oral and gut microbiome play important roles in the pathogenesis of inflammation, immune dysfunction, and disruption of the gut-brain axis, which may contribute to ASD pathophysiology. The majority of previous studies

Indole, a Signaling Molecule Produced by the Gut Microbiota, Negatively Impacts Emotional Behaviors in Rats.

Gut microbiota produces a wide and diverse array of metabolites that are an integral part of the host metabolome. The emergence of the gut microbiome-brain axis concept has prompted investigations on the role of gut microbiota dysbioses in the pathophysiology of brain diseases. Specifically, the search for microbe-related metabolomic signatures in human patients and animal models of psychiatric disorders has pointed out the importance of the microbial metabolism of aromatic amino acids. Here, we investigated the effect of indole on brain and behavior in rats. Indole is produced by gut microbio

Comparison of geographic distributions of Irritable Bowel Syndrome with Inflammatory Bowel Disease fail to support common evolutionary roots: Irritable Bowel Syndrome and Inflammatory Bowel Diseases are not related by evolution.

Irritable Bowel Syndrome (IBS) shares overlapping symptoms and some features of pathogenesis with Inflammatory Bowel Diseases (IBD: Crohn's disease [CD], and Ulcerative Colitis [UC]). Geographic markers such as latitude/sunshine and more recently lactase population distributions are found to be correlated with IBD. As a result of clinical and pathogenic similarities between the 2 conditions, some authorities questioned whether a connection exists between them. We compare IBS directly with IBD, and indirectly with geographic markers associated with IBD, in order to evaluate possible evolutionar

Hypothesis of demodicidosis rosacea flushing etiopathogenesis.

Most of the patients with erythematotelangiectatic rosacea are characterized by flushing, oedema and telangiectasia. The etiopathogenesis of the flushing in rosacea patients is unknown. Clinically the flushing in rosacea is similar to the "Asian flushing syndrome". Most Asians have an overactive alcohol dehydrogenase (ADH) that tends to break down alcohol into acetaldehyde faster. People with "Asians flushing syndrome" have a genetic disorder with the Aldehyde Dehydrogenase 2(∗)2 (ALDH2(∗)2) allele. This is the reason why they do not metabolize very well the acetaldehyde that comes from the al

Fertility of ram semen frozen in Bioexcell and used for cervical artificial insemination.

The current use of ingredients of animal origin, such as egg yolk, in semen extenders presents a risk of microbial contamination, and has led to the search for alternatives. Such an extender is commercially available for bull semen (Bioexcell), IMV, L'Aigle, France), and it has previously been tested in vitro for freezing ram semen, with satisfactory results. The aim of the present study was to compare the fertility results of ewes in Uruguay, after cervical insemination with ram semen that was frozen in Bioexcell versus semen frozen in a conventional milk-egg yolk extender (control). Association of early life anti-pathogen immune responses with infant food sensitisation and allergy: An elastic-net stability-selection approach.

BACKGROUND: The development of food sensitisation (FS) and food allergy (FA) reflects complex interactions between genetic, environmental and immunological factors, which conventional univariate approaches may fail to capture. This exploratory study applied penalised regression to determine whether early-life immune responses to common pathogens are associated with infant FS and FA. METHODS: At 1 year of age, FS was determined by skin prick test and FA by allergy/tolerance history and oral food challenge. Elastic net regression (ENR) was applied to cytokine response data (13 cytokines) from w

Engineered probiotics that sequester arsenite in a mouse gastrointestinal system.

Chronic exposure to inorganic arsenic remains a major global health concern, as arsenite is frequently present in contaminated food and drinking water and readily absorbed through the gastrointestinal (GI) tract. Once internalized, arsenite accumulates in tissues and contributes to long-term health effects, including cancer, organ dysfunction, and neurological disorders. Despite extensive efforts to reduce environmental contamination, there are currently no practical strategies to prevent dietary arsenite from entering the human body during digestion. Here, we report a synthetic biology-based

"Non-habit associated oral squamous cell carcinoma: A systematic review".

BACKGROUND: Oral squamous cell carcinoma (OSCC), the most common oral malignancy, accounts for 80-90% of oral cancers. India bears the highest global burden, with one-third of cases worldwide. A significant proportion of OSCC in the Indian subcontinent arises in individuals without traditional risk factors such as tobacco or alcohol use, suggesting the involvement of alternative etiologic factors that remains poorly defined. OBJECTIVES: To systematically review and synthesize evidence on the demographics, lesion distribution, and potential non-habit-related risk factors of OSCC, aiming to cla

The Gut Microbiota in Hematologic Malignancies: Mechanisms, Clinical Associations, and Translational Opportunities.

Hematologic malignancies arise and progress within a systemic ecosystem in which the gut microbiota is an increasingly recognized, partially modifiable component. Across acute leukemias, chronic lymphocytic leukemia, plasma cell disorders, lymphomas, and clonal myeloid neoplasms, human studies consistently report reduced microbial diversity, depletion of barrier-supportive, short-chain fatty acid-producing commensals, and enrichment of Gram-negative, pro-inflammatory, or hospital-adapted taxa. These alterations are associated with pre-leukemic clonal expansion, adverse genetic and immunologica

Microbial community structure, functional potential, probiotic signatures, and MAG reconstruction of fermented bamboo shoots from Northeast India.

Fermented bamboo shoot (FBS) products are widely consumed traditional foods across the Northeast region (NER) of India, yet their microbiome structure, functional capacity, biosynthetic potential, and safety attributes remain insufficiently explored. Here, comparative shotgun metagenomics of ten traditional FBS products from six NER states was used to address these gaps integrating previously generated metagenomic data from Tripura with newly generated datasets from Manipur, Meghalaya, Arunachal Pradesh, Nagaland, and Sikkim thereby bringing the total number of samples to 24. Taxonomic profili

Polyclonal antibodies and probiotics improve immune dysfunction in a mouse model of inflammatory bowel disease.

Inflammatory bowel disease (IBD) is a chronic digestive disorder caused by the overactivity of the mucosal immune system in response to natural intestinal bacteria and environmental factors, affecting genetically susceptible individuals. This research investigates the preventive effect of probiotics (P), specifically Lactobacillus fermentum, Lactobacillus L. reuteri, and polyclonal antibodies (AB), on morphological changes and markers of intestinal inflammation in the IBD mice model. Our study spanned 54 days. For the first 21 days, we observed microbial populations and immunoglobulin levels.

Bacteria synthesize a bacteriocin within the cell membrane.

UNLABELLED: Bacteria use bacteriocins to kill competitors and shape microbial communities. Many bacteriocins are ribosomally synthesized and post-translationally modified peptides (RiPPs), a diverse class of natural products and a source of new antimicrobials. The diversity of bacterial RiPPs stems from the combination of genetically encoded precursor peptides and biosynthetic proteins that post-translationally process the peptides. Characterized RiPP pathways involve soluble precursor peptides and biosynthetic proteins that modify them in the bacterial cytoplasm before secretion across the ce

The causal role of skin microbiota in rheumatoid arthritis via gut microbiota and immune cell mediation: A Mendelian randomization study.

Emerging evidence suggests that disturbances in skin microbiota homeostasis may contribute to rheumatoid arthritis (RA) development through systemic immunomodulatory pathways. However, the causal relationships and underlying intermediary mechanisms remain to be clarified. We utilized summary-level data from large-scale genome-wide association studies and the FinnGen database, including 150 skin microbiota taxa, 731 immune cell phenotypes, 473 gut microbiota taxa, and RA cases. Two-sample Mendelian randomization (TSMR) was applied to evaluate potential causal associations with RA. Mediation ana

Multidimensional regulatory mechanisms and translational potential of epigenetic networks in the rheumatoid arthritis disease course.

Rheumatoid arthritis (RA) is an autoimmune disease characterized by chronic synovitis that may progress to irreversible joint destruction and disability, thereby substantially impairing quality of life. RA results from complex interactions among genetic predisposition, environmental exposures, and immune dysregulation; however, current therapies are not curative, and many patients continue to experience pain, morning stiffness, and recurrent inflammation. In recent years, epigenetic mechanisms have emerged as key modulators of RA heterogeneity and disease persistence. Reversible regulatory lay

Gut bacterial and fungal signatures in relation to human leukocyte antigen-DQ2/DQ8 in children with celiac disease and siblings.

BACKGROUND: The presence of human leukocyte antigen (HLA)-DQ2 and/or HLA-DQ8 alleles is necessary but not sufficient for the development of celiac disease (CeD). This suggests that additional environmental and biological factors, including bacteria and, above all, the still rarely studied fungal gut microbiota, play key roles in disease onset and progression. AIM: To characterize and compare the intestinal bacteriobiota and mycobiota profiles of children with newly diagnosed CeD and their unaffected siblings, in comparison with a healthy control group. METHODS: The study included children an

The inhibition of lactate/Gpr81 signalling driven by moderate-intensity continuous training protects against osteoarthritis.

Osteoarthritis (OA) is among the most prevalent chronic diseases worldwide. However, currently employed pharmacological therapies for OA are associated with various reported side effects. Exercise therapy represents a potentially safer and more effective strategy for OA treatment, although its underlying mechanism remains unclear. Here, we demonstrate that compared with high-intensity continuous training, moderate-intensity continuous training (MICT) has greater therapeutic potential for OA. Notably, these beneficial effects of MICT were abrogated following gut microbiota depletion. Moreover,
