Bacillus thuringiensis (Bt) is a well-known entomopathogenic bacterium widely used in biopesticide formulations due to its diverse arsenal of insecticidal toxins and environmental adaptability. However, the genetic diversity and virulence potential of native Bt strains from South Asia remain underexplored. In this study, we performed whole-genome sequencing and comparative genomic analysis of B. thuringiensis strain JSd1, isolated from Bangladeshi soil. The high-quality draft genome, assembled at 75x coverage, comprises 5.39 Mb in 64 contigs with a GC content of 35.28% and 99.26% completeness.

Chemical pesticides have been used as a reliable method of controlling pests on crops. However, their applications have led to the development of resistance, environmental problems, and risks to human health. Therefore, new chemicals in nanoform such as chlorophyll derivatives could be a good choice for pest control through the induction of reactive oxygen species, liberating singlet oxygen when they are excited by photo exposure. The present work evaluated the toxicity of magnesium chlorophyllin (Mg-Chl) and magnesium chlorophyllin zinc oxide (Mg-Chl-ZnO NC) nanocomposite on Spodoptera frugip

Biological puncture systems span microorganisms, plants, invertebrates, and vertebrates, yet all must solve a common mechanical problem: creating a localized fracture in a protective barrier while avoiding failure of the penetrating structure and minimizing damage to the target. Living systems therefore provide diverse natural solutions to the same fundamental mechanical challenges faced by microneedles, making them useful comparators for bioinspired puncture design. This review examines representative biological puncture systems-including bacterial secretion needles, jellyfish nematocysts, in

INTRODUCTION: Insect bite hypersensitivity (IBH) is a seasonal, IgE-mediated allergic dermatitis of horses caused by salivary gland proteins of biting midges (Culicoides spp.). Current management relies on relief of clinical signs and on physical protection. In a previous pilot study, healthy horses were fed transgenic barley expressing a Culicoides allergen via a special spiral bit, which successfully induced allergen-specific antibody responses. Building on this concept, the present study aimed to evaluate a more practical feeding approach, delivering transgenic barley expressing the major C

INTRODUCTION: The indiscriminate use of pesticides poses a significant risk to human health and the environment. Plant-based biopesticides offer an alternative to insecticides for integration into insect pest management programs.
METHODS: The current study assessed the toxicological effects of leaf extracts from indigenous weed plants, Chenopodium murale and Achyranthes aspera, in albino rats, Rattus norvegicus. The extract-mixed diet was fed in three different doses (100 ppm, 150 ppm, and 250 ppm) for 28 days, while the Cypermethrin insecticide was used as a reference insecticide at the same

Bacillus thuringiensis (Bt) is a Gram-positive bacterium that during sporulation produces insecticidal Crystal (Cry) proteins, which play a major role in insect control today. Some Bt Cry proteins, e.g., Cry5Ba, target nematodes and, when given orally, can cure animals of gastrointestinal nematode (GIN) parasites. To eliminate concerns about treating humans and animals with spores and live bacteria, we developed an asporogenous system for scalable and safe Cry protein delivery called IBaCC (Inactivated Bacteria with Cytosolic Crystal(s)), which results in production of a bioactive crystal and …

BACKGROUND: The steroid hormone ecdysone, acting through its nuclear receptor (EcR), is a key regulator of insect development and reproduction and also contributes to immune regulation and gut microbiota homeostasis. In sand flies, both immunity and the gut microbiota are critical determinants for Leishmania establishment in the midgut. Phlebotomus perniciosus is the principal vector of Leishmania infantum in the western Mediterranean basin; however, the mechanisms affecting interactions among parasite, vector immune system, and resident microbiota remain poorly understood in this species.

BACKGROUND: The bites from infected female Anopheles mosquitoes lead to hundreds of thousands of deaths annually. To find a human, mosquitoes rely on their keen sense of smell. Repellents are chemicals that can target a mosquito's chemosensory senses. Therefore, targeting olfaction using repellents can be an effective way to prevent their attraction to humans and subsequent biting. However, the precise mechanisms underlying the mode of action for most commonly used repellents is surprisingly not well known. Recent evidence suggests that commonly used insect repellents target Anopheles mosquito

Aedes aegypti is the main vector of several arboviruses including chikungunya, dengue, yellow fever and Zika. Beyond arboviruses, Aedes aegypti harbours insect-specific viruses (ISVs), which can modulate mosquito's ability to transmit diseases by interfering with viral processes and triggering immune responses. Both arboviruses and ISVs can be transmitted vertically, where viruses are passed from parent to offspring. The lack of systematic molecular and entomological surveillance, has left the diversity of viruses in local Aedes aegypti populations largely unexplored. This study aimed to chara

BACKGROUND: Oropouche virus (OROV) is a neglected insect-borne orthobunyavirus that causes a febrile illness, neurological disease, and pregnancy complications in humans across an endemic area spanning South and Central America. The host factors associated with disease pathogenesis have nonetheless remained obscure, and little is known about the immune determinants of protection against OROV.
METHODS: We tracked morbidity, mortality, viral loads, and serum neutralisation in wild-type (WT), Rag1-/-, CD19-Cre+Ifnarfl, and CD19-Cre+MyD88fl mice and performed immunophenotyping experiments, passiv

Neonicotinoids, a relatively new widely used class of insecticide is used in agriculture to control insect populations. We examined the capacity of ancestral exposure to the neonicotinoid thiacloprid (thia) to induce transgenerational effects on thyroid tissue. Pregnant outbred Swiss female mice were exposed to thia at embryonic days E6.5 to E15.5 using 0, 0.6, and 6 mg/kg/day doses. Thyroid paraffin sections were prepared for morphology analysis. We apply ELISA method to measure T4 and TSH levels, RT-qPCR for gene expression analysis, ChIP-qPCR techniques for sperm histone H3K4me3 analysis, a

In all vertebrates, visual signals from each visual field project to the opposite midbrain tectum (called the superior colliculus in mammals). The tectum/colliculus computes visual salience to select targets for context-contingent visually guided behavior: a frog will orient toward a small, moving stimulus (insect prey) but away from a large, looming stimulus (a predator). In mammals, visual signals competing for behavioral salience are also transmitted to the visual cortex, where they are integrated with collicular signals and then projected via the dorsal visual stream to the parietal and fr

Equine influenza (EI) is a highly contagious acute respiratory disease of equines caused by the H3N8 subtype of Influenza A virus i.e. equine influenza virus (EIV). Vaccination is an important and effective tool for the control of EI in equines. Most of the commercial influenza vaccines are produced in embryonated hen's eggs which has several inherent disadvantages. Hence, subunit vaccine based on recombinant haemagglutinin (HA) antigen, being the most important envelope glycoprotein has been extensively exploited for generating protective immune responses, against influenza A and B viruses. Lead is considered a common old chronic toxicant around the world due to expanded environmental pollution, it is likely an inevitable contaminant in food, dairy products, air etc. Also, fipronil is a wide-ranging effective N-phenyl pyrazole insecticide which used commonly in agriculture and public health insect control, but until now no adequate data are available on the oxidative stress, cytotoxicity, and mutagenic influence of fipronil and lead or their mixture subchronic exposure. Both xenobiotics (lead and fipronil) exert a harmful impact on reproduction, prompting the exploration of vario…

UNLABELLED: The adaptation of Yersinia pestis, the flea-borne plague agent, to fluctuating environmental conditions is essential for the successful colonization of the flea vector. A previous comparative transcriptomic analysis showed that the Cpx pathway of Y. pestis is up-regulated in infected fleas. The CpxAR two-component system is a component of the envelope stress response and is critical for maintaining the integrity of the cell. Here, a phenotypic screening revealed a survival defect of the cpxAR mutant to oxidative stress and copper. The measured copper concentration in the digestive

BACKGROUND: Dirofilariasis, the disease caused by Dirofilaria spp., and in particular by Dirofilaria immitis and Dirofilaria repens in canines, occurs frequently in canids and felids, and occasionally in humans, in temperate, sub-tropical and tropical regions globally. Although highly effective, safe and convenient preventive medicines have been available for the treatment of dirofilariasis for the past three decades, the disease remains a major veterinary and public health concern in endemic areas. The insect vectors, host-parasite relationships and interactions of Dirofilaria spp. have recei

Infectious bronchitis (IB) is a highly contagious, acute respiratory disease in chickens, with a severe economic impact on poultry production globally. The rapid emergence of regional variants of this Gammacoronavirus warrants new vaccine approaches that are more humane and rapid to produce than the current embryonated chicken egg-based method used for IB variant vaccine propagation (chemically-inactivated whole viruses). The production of virus-like particles (VLPs) expressing the Spike (S) glycoprotein, the major antigen which induces neutralizing antibodies, has not been achieved in planta

BACKGROUND: The flesh fly, Sarcophaga peregrina (Diptera: Sarcophagidae), is an important hygiene pest, that causes myiasis in humans and other mammals, typically livestock, and as a vector for various parasitic agents, including bacteria, viruses, and parasites. The role of long non-coding RNAs (lncRNAs) in regulating gene expression during metamorphosis of the flesh fly has not been well established.
METHODOLOGY/PRINCIPAL FINDINGS: In this study, we performed genome-wide identification and characterization of lncRNAs from the early pupal stage (1-days pupae), mid-term pupal stage (5-days pu

INTRODUCTION: Field trials and modelling studies suggest that elimination of dengue transmission may be possible through widespread release of Aedes aegypti mosquitoes infected with the insect bacterium Wolbachia pipientis (wMel strain), in conjunction with routine dengue control activities. This study aimed to develop a modelling framework to guide planning for the potential elimination of locally acquired dengue in Yogyakarta, a city of almost 400 000 people in Java, Indonesia.
METHODS: A scenario-tree modelling approach was used to estimate the sensitivity of the dengue surveillance system
