PIWIL2, a member of PIWI/AGO family, is expressed in germline stem cells and precancerous stem cells, but not in adult somatic cells. PIWIL2 plays an important role in tumor development. It is considered as a cancer–testis antigen (CT80). It has been reported that the spliced fragment of PIWIL2, PL2L60, was widely expressed in cancer cell lines. In this study, HLA-A2-restricted epitopes from PL2L60 were predicted by online tools. To improve the activity of the native epitope, a candidate peptide P281 with potent binding affinity was chosen to investigate the modification strategy. A series of

Alcohol consumption is an established risk factor for breast cancer, with a particularly strong association with estrogen receptor-positive (ER+) disease. Although the estrogenic activity of alcohol is well recognized, how alcohol-induced ER signaling is coupled to growth factor receptor pathways that promote tumor cell growth and progression remains incompletely understood. Here, we identify neuregulin-1 (NRG1) as a functional mediator, linking alcohol-induced ER activity to ErbB3 receptor tyrosine kinase signaling in ER+ breast cancer cells. Under estrogen-depleted conditions, alcohol induce

As part of our ongoing efforts to develop N-benzylated oxindole derivatives as multi-target anticancer agents, novel N-benzyl oxindole indoles (4a-z) were designed, synthesized and structurally characterized. Their cytotoxic activities were evaluated against HCT-116, HT-29, A549, MIA PaCa-2, A498, CAKI-1, MDA-MB-231, and MCF-7 cancer cells, as well as Beas2b noncancerous cells. Most compounds exhibited significantly stronger cytotoxicity and selectivity than the reference drug sunitinib malate. Mechanistic studies were performed on compounds 4a, 4b, 4d, 4g, 4i, 4q, and 4y, which displayed subm

Breast cancer remains the most common cause of mortality among women globally. Although improvements in early detection and systemic therapies have enhanced patient outcomes, conventional treatment strategies, including free drug administration, are often plagued by significant limitations such as poor selectivity, systemic toxicity, rapid drug degradation, and low therapeutic indices. Paclitaxel (PTX), a commonly used chemotherapeutic drug, suffers from poor aqueous solubility and nonspecific distribution, resulting in severe off-target effects and reduced clinical efficacy. The present study

The development of novel anticancer agents capable of modulating pathways associated with tumor progression remains a critical strategy, particularly for aggressive breast cancer subtypes such as triple-negative breast cancer (TNBC). In this study, a new series of hydrazide-hydrazone/thiourea derivatives (MIH-31-46) was rationally designed, synthesized, and evaluated as potential anticancer agents targeting vascular endothelial growth factor receptor-2 (VEGFR-2). Structural characterization was performed using 1H NMR, 13C NMR, and HR-MS. The antiproliferative activities of the synthesized deri

In this work, the biological potency of nitazoxanide (NTZ) was enhanced through coordination with transition metal ions Cu(II), Ni(II), and Zn(II). Initially, complexes with a ligand-metal stoichiometry of 2:1 were successfully synthesized and characterized by spectroscopic techniques and thermogravimetric methods. Measurement of the infrared spectrum revealed the bidentate nature of the ligand and excluded the possibility of the metal ion-amide group interaction. Nuclear magnetic resonance spectra showed a reduction in the NH- intensity signal and integration, indicating the possibility of en

Three series of new benzimidazole hybrids were designed and synthesized as promising human TopoII inhibitors. They were characterized by different spectroscopic techniques (1H, 13C NMR, ESI-MS and IR). All hybrids (6-23) were screened for their in vitro antiproliferative activity against five human cancer cell lines namely; HepG-2, MCF-7, PC-3, HCT-116 and Hela. Compound 21 showed the most potent anticancer activity against all cancer cell lines, with IC50 range of 2.82 to 12.59 µM, while proving safe towards normal cells WI-38 (IC50 = 31.89 µM) compared to the reference drug doxorubicin (IC50

Previous studies show that dopamine D2-like receptor (D2DR) antagonist sulpiride (SUL) enhances the antitumor efficacy of dexamethasone (DEX) in drug-resistant breast cancer involving cancer stem-like cells (CSCs). In this study, we investigated the pharmacokinetic (PK) properties of SUL in nude mice and developed a semi-mechanism PK/PD model to quantitatively characterize the synergistic effect of DEX and SUL in preclinical breast cancer xenografts. After nude mice received oral administration of a single dose of SUL (50 mg/kg, ig), plasma concentrations were assessed using LC-MS/MS. A two-co

In this article, a hybrid TiO2/Au/graphene layer-based surface plasmon resonance (SPR) sensor with improved sensitivity and capability for cancer detection is presented. The finite element method (FEM) was used for numerical analysis. The proposed SPR biosensor was structured based on the angular analysis of the attenuated total reflection (ATR) method for the detection of various types of cancer using the refractive index component. The resonance angle shifted owing to the increment of normal and cancerous cells' refractive index, which varied between 1.36 and 1.401 for six different types of

Cyclin-dependent kinase 4/6 (CDK4/6) plays a crucial role in cell cycle regulation, is overexpressed in various cancers and is an important target in the development of radiotracers for tumour imaging. Despite the increasing recognition of CDK4/6 inhibitors in cancer therapy, their application is limited by the lack of suitable biomarkers. Herein, we developed a series of technetium-99m-labelled CDK4/6 radiotracers and utilized a linker optimization strategy to reduce their abdominal uptake and enhance their imaging properties. Chromosomal amplifications are among the most common genetic alterations found in human cancers. However, experimental systems to study the processes that lead to specific, recurrent amplification events in human cancers are lacking. Moreover, some common amplifications, such as that at 8p11-12 in breast cancer, harbor multiple driver oncogenes, which are poorly modeled by conventional overexpression approaches. We sought to develop an experimental system to model recurrent chromosomal amplification events in human cell lines. Our strategy is to use homologous-recombination-mediated gene targe
