Development of antibody-dependent cell-mediated cytotoxicity in the respiratory tract after natural infection with respiratory syncytial virus.

Antibody-dependent cell-mediated cytotoxicity (ADCC) was measured in nasopharyngeal secretions collected from 42 infants and young children at various intervals after primary or secondary infection with respiratory syncytial virus. ADCC was determined by specific immune release of 51Cr from respiratory syncytial virus-infected HEp-2 cell culture monolayers, with lymphocytes from adult volunteers as effector cells. Specific ADCC responses in nasopharyngeal secretions after primary infection were observed as early as 3 days after the onset of clinical symptoms, and peak activity was observed 14

The use of cough-nasal swabs in the rapid diagnosis of respiratory syncytial virus infection by the fluorescent antibody technique.

Thirty-five consecutive infants admitted into hospital in Newcastle upon Tyne with acute respiratory disease had cough/nasal swabs and nasopharyngeal secretions taken. Both types of specimens were examined by the fluorescent antibody technique for respiratory syncytial virus; isolation techniques were also used. Twenty-eight specimens of nasopharyngeal secretion were positive, as were 26 of the corresponding cough/nasal swab preparations. Respiratory syncytial virus was isolated from all but one.Sixteen consecutive children who were only suitable for examination by cough/nasal swab preparation

Small Airway Disease in Pre-COPD and Early COPD: Insights Into the Pulmonary "Silent Zone".

Chronic obstructive pulmonary disease (COPD) is increasingly recognized as a life-course disorder with origins rooted in early-life insults. However, the current diagnostic reliance on the FEV1/FVC < 0.7 threshold often fails to capture the early phases of disease evolution, particularly within the pulmonary "Silent Zone." This narrative review examines small airway disease (SAD) in pre-COPD and early COPD by integrating physiological, structural, molecular, and diagnostic evidence. Findings from imaging and pathology studies indicate that terminal bronchiole loss, mucus plugging, alveolar att

N-acetylcysteine in paediatrics: a review of efficacy, safety and dosing strategies in respiratory care.

N-acetylcysteine (NAC) is widely used for its mucolytic, antioxidant, anti-inflammatory and synergistic antibacterial properties in the treatment of respiratory diseases. NAC and other mucolytics and mucoactive medications are frequently employed in the adult population and in paediatric settings to improve mucus clearance in conditions such as cystic fibrosis, bronchiolitis, pneumonia, and both chronic and acute bronchitis, with varying degrees of success. This narrative review evaluates the efficacy and safety of NAC in paediatric acute and chronic respiratory diseases, synthesizing data fro…

Histologic patterns of chronic interstitial lung disease in dogs.

Chronic interstitial lung disease (cILD) is uncommon in dogs and little is known of the pathogenesis, apart from the condition in West Highland White Terriers. This study aimed to characterize histologic lesions of canine cILD, compare the lesions and clinical features, and classify the histopathologic patterns according to criteria used in humans. The study included 24 postmortem cases of subacute or chronic ILD in >6-month-old dogs with respiratory signs. Histologic lung lesions included attenuated bronchiolar epithelium, alveolar edema, type II pneumocyte proliferation, fibrosis of alveolar

Review: Knowledge Gained and Gaps in Understanding in the 25 Years Since Human Metapneumovirus Was First Identified as a Cause of Human Disease.

Human metapneumovirus (hMPV) is a nonsegmented, single-stranded, negative-sense RNA virus belonging to the Pneumoviridae family. It was first identified in 2001 in the nasopharyngeal secretions of 28 Dutch children with bronchiolitis collected over a 20-year period. hMPV exhibited paramyxovirus-like morphology with many genetic similarities to respiratory syncytial virus. hMPV has 1 serotype with 2 major subgroups (A and B) and 5 sublineages (A1, A2a, A2b, B1, and B2). In the wake of its discovery, a wealth of observational research has demonstrated global circulation of hMPV causing a wide sp…

Features of internal absorbed dose microdistribution in biological tissue irradiated by 31SiO2 microparticles compared with dose microdistribution from exposure to 56MnO2 particles.

Radiobiological studies are ongoing to understand the consequences of internal exposure to neutron-activated radioactive microparticles, which were sprayed over experimental rats and mice. Special attention in these experiments is given to internal irradiation with radioactive microparticles with short-lived neutron-activated radionuclides 31Si (T1/2 = 2.62 h) and 56Mn (T1/2 = 2.58 h), which are among the main dose-forming factors from residual radioactivity activated in soils by neutrons in the first hours after atmospheric nuclear explosions. The presented work is devoted to microdosimetry p

Cripto-1 and Cripto-3 differential expression pattern in human placenta, umbilical cord and fetal tissues suggests new functions for these proteins.

Cripto-1 (CR1) is an oncofetal protein involved in EGF/TGFβ signal transduction, with important functions in development, stem cell biology and cancer. Its homolog, Cripto-3 (CR3), is almost identical, with the exception of six (out of 188) amino acids. Thus, until recently, no antibody could distinguish them. Here, we compare the immunostaining pattern for highly specific monoclonal antibodies against CR1 and CR3 in human samples of placenta, umbilical cord and fetal tissues. Immunoreactivity (IR) for both proteins was found in placenta, umbilical cord, fetal retina, testis, lung, esophagus,

Impact of CD11c+ cells in conducting airway lumen on Aspergillus fumigatus conidia deposition in neutropenic mice.

INTRODUCTION: Inhaled conidia of the opportunistic fungi Aspergillus fumigatus settle in the airway mucosa and in alveolar spaces. Different immune cells typically provide crucial defense against fungal germination. However, in immunocompromised patients, the lack of sufficient pro-inflammatory immune response often leads to invasive aspergillosis, with current treatments being limited by insufficient understanding of the precise conidial distribution patterns in the airways. METHODS: Therefore, we employed advanced imaging techniques, including immunohistochemistry, optical clearing, and con

Imaging findings of multislice computed tomography in 21 patients with airway invasive pulmonary aspergillosis.

BACKGROUND: Airway invasive aspergillosis (AWIA) poses a diagnostic challenge due to its nonspecific clinical manifestations. This study aimed to characterize the imaging findings of AWIA and explore the clinical characteristics that facilitate the diagnosis of AWIA. METHODS: A retrospective analysis was conducted on 21 patients clinically and pathologically diagnosed with AWIA. All subjects underwent chest multislice computed tomography (MSCT) scans, and their clinical data were collected. The computed tomography (CT) features were evaluated, and 17 patients received the follow-up of MSCT in

Exploring Clinical Predictors of Severe Human Metapneumovirus Respiratory Tract Infections in Children: Insights from a Recent Outbreak.

Human metapneumovirus (hMPV) is an important pathogen that causes both upper (URTIs) and lower respiratory tract infections (LRTIs) in children. The virus can be implicated in severe bronchiolitis and pneumonia, necessitating hospitalization, with certain cases requiring intensive care unit intervention. As part of a retrospective observational study, we aimed to identify indicators of severe hMPV respiratory tract infections in children referred to the University Children's Hospital Ljubljana and the Department of Infectious Diseases Ljubljana, Slovenia, during a recent outbreak. We analyzed …

Infectivity and transmissibility of an avian H3N1 influenza virus in pigs.

In 2019 a low pathogenic H3N1 avian influenza virus (AIV) caused an outbreak in Belgian poultry farms, characterized by an unusually high mortality in chickens. Influenza A viruses of the H1 and H3 subtype can infect pigs and become established in swine populations. Therefore, the H3N1 epizootic raised concern about AIV transmission to pigs and from pigs to humans. Here, we assessed the replication efficiency of this virus in explants of the porcine respiratory tract and in pigs, using virus titration and/or RT-qPCR. We also examined transmission from directly, intranasally inoculated pigs to
