CAG/CTG repeats are prone to expansion, causing several inherited human diseases. The initiating sources of DNA damage which lead to inaccurate repair of the repeat tract to cause expansions are not fully understood. Expansion-prone CAG/CTG repeats are actively transcribed and prone to forming stable R-loops with hairpin structures forming on the displaced single-stranded DNA (S-loops). We previously determined that damage by the Saccharomyces cerevisiae cytosine deaminase, Fcy1, was required for both fragility and instability of CAG/CTG tracts engaged in R-loops. To determine whether this mec

The role of spin orbit interaction in superconducting proximity effect is an area of intense research effort. Recent theoretical and experimental works investigate the possible role of spin-orbit interaction in generating spin-triplet pair correlations. In this work, we present an experimental survey of thin normal metal-superconductor-normal metal trilayers with Nb superconductor and Al, Ti, Cu, Pt, Ta, and Au normal metals, along with single layers of Nb as reference. We aim to probe the role of spin-orbit interaction and resistivity on the normal metal proximity effect through measurements

High-level ab initio CCSD(T) and spin-orbit icMRCI+Q calculations were used to predict potential energy curves (PECs) for the lowest-lying states of ZrO, ZrS, HfO, and HfS. The prediction of the ground state is basis set dependent at the icMRCI+Q level for ZrO and ZrS due to the small singlet-triplet splitting between the lowest 1Σ+ and 3Δ states. CCSD(T) with a spin orbit correction predicted the 1Σ+ ground state in agreement with experiment. New all-electron basis sets were developed for Hf to improve the results over those predicted by use of effective core potentials (ECPs) that subsume th

Ochratoxin A (OTA) is a type of mycotoxin found in various contaminated foods, and it is highly toxic to the livers and kidneys of humans. Herein, we develop a self-enhanced electrochemiluminescent (ECL) sensor based on tandem signal amplification and a self-luminescent europium covalent-organic polymer (Eu-COPTMT-BPA) for OTA assay. Eu-COPTMT-BPA is a self-enhanced ECL emitter that is obtained by using 2,4,6-trimethyl-1,3,5-triazine (TMT) and 2,2'-bipyridine-5,5'-dicarbaldehyde (BPA) as the first and second ligands to form a highly conjugated structure for sensitizing the Eu3+ luminescence. We studied a series of thermally activated delayed fluorescence (TADF) emitters based on phenothiazine-dibenzothiophene-S,S-dioxide (PTZ-DTO) electron donor-acceptor (D-A) dyads, using femtosecond/nanosecond transient absorption (fs/ns-TA) spectroscopy and pulsed laser-excited time-resolved electron paramagnetic resonance (TREPR) spectroscopy. In the analyzed compounds, a Se atom replaced the S atom in the phenothiazine (PTZ) unit to study the heavy atom effect on the reverse intersystem crossing (RISC). Moreover, oxidation of the PTZ to sulfoxide and sulfone was also used to tune the energy o

Artificial neural networks in drug discovery have shown remarkable potential in various areas, including molecular similarity assessment and virtual screening. This study presents a novel multimodal Siamese neural network architecture. The aim was to join molecular electrostatic potential (MEP) images with the texture features derived from reduced density gradient (RDG) diagrams for enhanced molecular similarity prediction. On one side, the proposed model is combined with a convolutional neural network (CNN) for processing MEP visual information. This data is added to the multilayer perceptron

Studying mutation in healthy somatic tissues is the key for understanding the genesis of cancer and other genetic diseases. Mutation rate varies from site to site in the human genome by up to 100-fold and is influenced by numerous epigenetic and genetic factors including GC content, trinucleotide sequence context, and DNAse accessibility. These factors influence mutation at both local and regional scales and are often interrelated with one another, meaning that predicting mutability or uncovering its drivers requires modelling multiple factors and scales simultaneously. Historically, most inve

Dysarthria, characterised by slurred speech, is a hallmark of many neurological disorders and brain trauma. Clinical assessment requires an audio-visual investigation by a trained healthcare expert, who evaluates criteria such as respiration, phonation, articulation, resonance, and prosody during speech. Quantitative assessment of dysarthria is challenging due to its complexity, variability, and the subjective nature of human-observation-based scoring methods. We present a novel machine-learning framework using transformers for stratifying and monitoring patient speech. Our framework integrate

BACKGROUND: Early continuous lenalidomide use for multiple myeloma (MM) treatment has led to more patients with lenalidomide-refractory disease at earlier lines of therapy (LOTs). Real-world treatment practices and outcomes in elderly patients with comorbidities are not well characterized.
MATERIALS AND METHODS: Using the Surveillance, Epidemiology, and End Results-Medicare database, we analyzed data from patients who were diagnosed with MM between 2014 and 2019, had 1 to 3 prior LOTs, including a proteasome inhibitor and an immunomodulatory drug, and were lenalidomide refractory. Patients we

Developing high-resolution organic light-emitting diodes (OLEDs) faces significant challenges, particularly in the blue light region. Multiple resonance thermally activated delayed fluorescence (MR-TADF) has been recognized as a breakthrough in next-generation TADF and high-quality displays due to its narrow bandwidth, exceptional color purity, and high efficiency. Extensive efforts have been devoted to introducing auxiliary groups into the BBCz-SB scaffold to improve OLED efficiency; however, such modifications often lead to undesirable emission shifts from sky-blue to green. Herein, we prese

Phenalenyl is known for its highly delocalized radical structure, making it a fundamental building block in the construction of polyradical compounds. This study explores how different connection topologies between phenalenyl units via acetylenic bridges modulate the polyradical character, as well as the electronic and magnetic properties of the resulting systems. The connection type depends on the atom occupation pattern of the phenalenyl singly occupied orbital (SOMO). Three types of connections are defined that induce different π conjugation strength. Linear di- and tetra-phenalenyl chains

Friedreich ataxia (FA) is an autosomal recessive disease characterized by progressive damage to the nervous system and severe cardiac abnormalities. The disease is caused by a GAA•TTC triplet repeat expansion in the first intron of the FXN gene, resulting in epigenetic repression of FXN transcription and reduction in FXN (frataxin) protein which results in mitochondrial dysfunction. Factors and pathways that promote FXN repression represent potential therapeutic targets whose inhibition would restore FXN transcription and frataxin protein levels. Here, we performed a candidate-based RNAi scree

We present an implementation of the coupled frozen-density embedding (FDEc) formalism for the calculation of ground-state and excited-state properties, linear-response properties, and transition moments with the coupled cluster with the singles and approximate doubles (CC2) model. Following the general strategy introduced by Höfener and Visscher [J. Chem. Theory Comput.12, 549-557 (2016)], we derive the working equations needed for the evaluation of these properties and describe their implementation into our open-source quantum chemistry program, Serenity. Our implementation comprises both pro

CONTEXT: SOx emissions from diesel fuel necessitate the development of efficient and environmentally friendly desulfurization technologies. This study investigates the catalyst-free photochemical desulfurization mechanism of a diesel model compound, 2-decyl-7-(10-phenyldecyl)dibenzo[b,d]thiophene (D-PD-DBT), under red light irradiation, providing a theoretical foundation for experimentally observed phenomena. Experimental validation confirmed a desulfurization efficiency of up to 46.2%, which was accompanied by the degradation of aromatic structures as observed by FTIR. The proposed three-step

RNA-binding proteins (RBPs) regulate gene function by controlling RNA processing, transport, stability, and translation. Recent mechanistic and pre-clinical studies demonstrate that nociceptive sensitivity and plasticity are regulated by RNA-protein interactions. Investigating RBP function in sensory neurons may reveal new strategies to modulate nociceptor excitability and/or sensitivity and improve our understanding of mechanisms that contribute to pain chronification. We previously identified the RBP CUG triplet repeat binding protein (CUGBP) embryonic lethal abnormal vision (Elav)-like fami

Collagen, a vital matrix protein for various tissue and functions in animals, is widely applied in biomaterials. In type Ⅰ collagen, missense mutations of glycine (Gly) in the Gly-Xaa-Yaa triplet of the triple helix are a major cause of osteogenesis imperfecta (OI). Clinical manifestations exhibit marked heterogeneity, spanning a broad disease spectrum from mild skeletal fragility (Type Ⅰ) to severe limb deformities (Type Ⅲ) and perinatal lethal forms (Type Ⅱ). This study utilized recombinant collagen as a model to further elucidate whether Gly→Ala/Val mutations at the N-terminus of the integr

Understanding excited-state intramolecular proton transfer (ESIPT) is essential for designing organic molecules to enhance photophysical and luminophore properties in the development of optoelectronic devices. In this context, an attempt has been made to understand the impact of substituents on the ESIPT process of 2-(oxazolinyl)-phenol. Electron donating (EDG: -NH2, -OCH3, and -CH3) and electron withdrawing (EWG: -Cl, -Br, -COOH, -CF3, -CN, and -NO2) substitutions have been computationally designed and screened through density functional theory (DFT) and time-dependent density-functional theo

In recent years, metal-organic framework (MOF) materials with long persistent luminescence (LPL) have inspired extensive attention and presented various applications in security systems, information anticounterfeiting, and biological imaging fields. However, obtaining LPL materials with ultralong lifetime remains challenging. Halogen atoms, as nonmetallic elements existing in the frameworks, can not only induce the heavy-atom effect, effectively enhancing spin-orbit coupling and promoting intersystem crossing (ISC) processes, but also suppress non-radiative transition of the triplet states thr

Efficient photoredox chemical transformations are essential to the development of novel, cost-effective, and environmentally friendly synthetic methodologies. The concept of the entatic state in bioinorganic catalysis proposes that a preorganized structural configuration can reduce the energy barriers associated with chemical reactions. This concept provides one of the guiding principles to enhance catalytic efficiency by maintaining high-energy conformations close to the reaction's transition state. Copper(I)-based photocatalysts, recognized for their low toxicity and highly negative oxidatio
