Role of the serine/threonine kinase polo in pupal-adult metamorphosis and oogenesis of Lasioderma serricorne female.

The serine/threonine-protein kinase polo plays a crucial role in regulating the cell cycle and mitosis in insects. While the cigarette beetle, Lasioderma serricorne, is a globally distributed pest threatening stored products, the functions of its polo gene remain largely unexplored. This study investigates the polo gene to elucidate its role in the development and reproduction of L. serricorne. The Lspolo gene, which encodes a protein containing a catalytic serine/threonine kinase domain and polo box domains, was identified and cloned. Spatio-temporal expression analyses indicated that Lspolo

Regulated apoptosis is a conserved mechanism pausing female reproduction and establishes the sterile worker caste in the eusocial wasp, Polistes.

Female reproduction is an energetically expensive process, so species evolve to balance survival with reproductive output. Many female organisms can temporarily pause their reproduction, including egg development, in response to physiological stress. The cellular mechanisms initiating and maintaining a stress-induced pause in oogenesis have been most extensively studied in Drosophila melanogaster. While the molecular control of paused oogenesis in response to starvation have been well characterized in flies, it remains unknown if these mechanisms are shared by other species with regulated paus

Bacterial Communities Associated with Whole Eggs and Gut Tissues of Heortia vitessoides Moore (Lepidoptera: Crambidae) Across Developmental Stages and Sexes.

Bacterial communities may affect insect nutrition and development, but their variation across complete metamorphosis remains poorly understood in many forest lepidopteran pests. Heortia vitessoides is an important defoliator of Aquilaria trees, which produce agarwood. In this study, we used 16S rRNA gene sequencing to characterize bacteria associated with whole eggs and gut tissues from larvae, pupae, and adults of H. vitessoides. Whole egg and larval gut samples generally showed higher bacterial richness and diversity than pupal and adult gut samples. Bacterial community composition differed

Targeting DOPA decarboxylase disrupts immune-metabolic coordination and renders Spodoptera frugiperda susceptible to bacterial infection.

DOPA decarboxylase (DDC) is a key enzyme that connects melanin production and innate immunity in insects, yet its regulatory network in major lepidopteran pests remains poorly understood. We combined CRISPR/Cas9 gene editing with multi-omics analysis to explore DDC's role in immune responses in the invasive pest Spodoptera frugiperda. Knocking out DDC caused complete albinism, severe developmental delays, pronounced developmental defects, and increased susceptibility to bacterial challenge. Following the E. coli challenge, DDC-mutant larvae showed substantially lower final survival than infect

Acute heat exposure induces lifetime shifts in non-fertilising sperm allocation in a sperm-heteromorphic insect.

Ectotherm reproduction is vulnerable to acute heat stress, yet the longitudinal impacts of transient thermal events on male reproduction remain poorly understood. Using the stored-product pest Plodia interpunctella, a sperm-heteromorphic moth, we investigated how acute thermal stress (4 h at 40°C) impacts males' lifetime reproductive resource allocation and offspring fitness. Heat-exposed adult males and controls (26°C) were paired daily with non-stressed females. We found that transient heat stress did not significantly impact pre-courtship duration, courtship duration, copulation latency, lo

Beyond toxicity: Systems-level virulence of entomopathogenic fungi in termite social immunity.

Termites live in dense colonies where frequent social contact can facilitate pathogen transmission, but coordinated social immunity can also limit disease spread. Entomopathogenic fungi (EPF) are particularly relevant to termite pathology because they infect primarily through the cuticle, using adhesion, enzymatic degradation, penetration, and internal proliferation to overcome host barriers. This review synthesizes current knowledge on EPF virulence in termites and argues that fungal pathogenicity should be understood not only as direct toxicity (lethal effects of pathogen-derived compounds)

Combined imidacloprid and chlorpyrifos disrupt gut microbiota-host interactions and reduced performance and survival in honey bees.

Gut microbiota plays a central role in insect physiology and health. However, how agrochemical exposure perturbs gut microbiota of the host and its downstream functional consequences remains understudied. Here, we employed an integrative multi-omics framework combining microbiome profiling, transcriptomics, and metabolomics to investigate how field-realistic exposures to the insecticides chlorpyrifos (CPF), imidacloprid (IMD), and their combination (IC) affect gut microbial composition as well as sucrose consumption, body weight dynamics, and survival in bees. We found that sucrose consumption

The Genetic Basis of Color Polymorphism in the Orb-Web Spider Gasteracantha cancriformis.

Animal coloration is a complex trait useful for studying adaptive evolution. In consequence, a major goal in evolutionary biology is to understand the genetic and developmental basis underlying coloration in animals and thus establish direct links between genetic and phenotype variability. Genomics tools have contributed the identification the specific genes and variants responsible for color variation in nonmodel animals, but the evidence is biased toward some taxa, while groups such as arachnids have been neglected. In this study, we aimed to identify genes that underlie coloration in spider

Heat shock protein genes Hsp70 and DnaJA1 are key contributors to thermotolerance and development in predatory ladybeetle Propylaea japonica.

Global climate change has increased the frequency of extreme high-temperature events, underscoring the need to elucidate thermotolerance mechanisms in natural enemy insects for effective biological control. The ladybird Propylaea japonica is a predominant predator in South-East Asia and a successful biocontrol agent, yet the molecular basis of its heat resistance remains unknown. Here we cloned full-length cDNAs of three heat shock protein genes (pjhsp70, pjDnaJA1, and pjhsp21) from P. japonica using transcriptomic data. We characterized their sequences, inferred phylogenetic relationships, an

Ovarian transcriptome analyses indicate that weak juvenile hormone signaling underlies the molecular basis of oogenesis deficiencies in mosquitoes.

BACKGROUND: Juvenile hormone (JH) is synthesized by the corpora allata (CA) and controls development and reproduction in insects. We recently used CRISPR/Cas9 to establish a line lacking the enzyme that catalyzes the final step of JH biosynthesis in mosquitoes, a P450 epoxidase. The CA of the epox-/- mutants do not synthesize epoxidized JH III but methyl farneosate (MF), a weak agonist of the JH receptor. Female epox-/- mosquitoes have reduced JH signaling and show a substantial loss of reproductive fitness. To understand the molecular basis of this loss of fitness, we constructed ovarian mRNA

Comparative analysis of grape berry microbiota uncovers sour rot associates from a Maryland vineyard.

Grape sour rot (GSR) is a disease complex involving fungi and bacteria that can cause significant yield losses of susceptible varieties. It is widely spread in the eastern U.S. and other grape-growing regions globally. Previous studies suggest that damaged fruit skin and feeding insects like Drosophila spp. are required for the disease to occur. Current control strategies for the management of sour rot are not sustainable, and research on the implications of chemical management of the disease on microbiome diversity is scarce. Our aim was to: i) investigate the effect of insecticide applicatio

DEET Metabolite and Hearing Loss in United States Adults.

Hearing loss (HL) causes numerous challenges for individuals. N,N-diethyl-meta-toluamide (DEET) is a common ingredient in insect repellants and some sunscreens. M-(diethylcarbamoyl) benzoic acid (DCBA) is a DEET metabolite measured in the urine of National Health and Nutrition Examination Survey participants. This study examines potential associations between HL and urinary DCBA in U.S. adults. Speech-frequency hearing loss (SFHL) was defined as an average hearing threshold above 25 dB across four frequencies (0.5, 1, 2, and 4 kHz). In comparison, high-frequency hearing loss (HFHL) was based o

Wolbachia-mediated reproductive manipulation in rice planthoppers.

Rice planthoppers, including brown (Nilaparvata lugens), small brown (Laodelphax striatellus), and white-backed (Sogatella furcifera) planthoppers, are major agricultural pests in China and severely affect rice production and food security. The endosymbiotic bacterium Wolbachia is commonly found in these insects, where it regulates reproduction through mechanisms such as cytoplasmic incompatibility (CI) and increased fertility. In this review, we discuss the strain-specific effects of Wolbachia: wLug (in N. lugens, < 50% infection) increases fecundity without CI; wStri (in L. striatellus, 99%

Functional role of TaGSTs1 in the detoxification of spinetoram and development of Tuta absoluta.

Tuta absoluta has evolved resistance to many biological insecticides, resulting in significant annual agricultural and economic losses. Glutathione S-transferases (GSTs) are one of the major insect detoxification enzyme systems. However, the detoxification metabolism of GSTs in T. absoluta against biological insecticides remains poorly understood. In this study, We identified five key GST genes (TaGSTs1, TaGSTs2, TaGSTe1, TaGSTe3, and TaGSTd1) by screening from the comparative transcriptomes of two regional populations of T. absoluta in Xinjiang, China. Among the five GSTs, TaGSTs1 exhibited a

Effects of surfactin stress on gene expression and pathological changes in Spodoptera litura.

Spodoptera litura (S. litura) is a polyphagous pest of the family Lepidoptera, which causes damage and yields losses to many crops. The long-term use of chemical pesticides for control not only seriously threatens environmental health, but also causes S. litura to develop drug resistance. Therefore, there is an urgent need to develop environmentally safe and friendly biogenic pesticides. However, the mechanism of action of the secondary metabolite (surfactin) of Bacillus Vélezensis (B. vélezensis) on lepidopteran pests (S. litura) has not been reported yet. We found that several metabolites an

Neurobiology of egg-laying behavior in Drosophila: neural control of the female reproductive system.

Egg-laying is one of the key aspects of female reproductive behavior in insects. Egg-laying has been studied since the dawn of Drosophila melanogaster as a model organism. The female's internal state, hormones, and external factors, such as nutrition, light, and social environment, affect egg-laying output. However, only recently, neurobiological features of egg-laying behavior have been studied in detail. fruitless and doublesex, two key players in the sex determination pathway, have become focal points in identifying neurons of reproductive significance in both central and peripheral nervous

Characterization of Brazilian Cordyceps fumosorosea isolates: Conidial production, tolerance to ultraviolet-B radiation, and elevated temperature.

Cordyceps fumosorosea is an entomopathogenic fungus with a global distribution and is used for the biological control of agricultural pests. High conidial productivity and tolerance to abiotic stresses such as elevated temperature and ultraviolet radiation (UV-B) are desired characteristics in candidate isolates for commercial products. Our goal in this study was to characterize promising isolates of C. fumosorosea from five Brazilian biomes regarding conidial production, tolerance to UV-B, and elevated temperature (45°). Seventy-two isolates out of 172 were chosen visually, based on growth an

Rapid but narrow - Evolutionary adaptation and transcriptional response of Drosophila melanogaster to toxic mould.

Insects have adapted to a multitude of environmental conditions, including the presence of xenobiotic noxious substances. Environmental microorganisms, particularly rich on ephemeral resources, employ these noxious chemicals in a chemical warfare against predators and competitors, driving co-evolutionary adaptations. In order to analyse how environmental microbes may be driving such evolutionary adaptations, we experimentally evolved Drosophila melanogaster populations by exposing larvae to the toxin-producing mould Aspergillus nidulans that infests the flies' breeding substrate. To disentangl
