Multi-Omics Analysis of the Gut-Brain Axis Elucidates Therapeutic Mechanisms of Guhong Injection in the Treatment of Ischemic Stroke.

Guhong injection (GH) is a compound preparation widely utilized in the treatment of cerebrovascular diseases. Accumulating evidence indicates that the gut microbiota is implicated in the development of ischemic stroke (IS). However, although the therapeutic potential of GH in IS may be mediated through the gut microbiota, the intricate relationships among the gut-brain axis, biomarkers, and target proteins remain to be completely explained. A rat model of middle cerebral artery occlusion (MCAO) was utilized to investigate the impact of GH on IS. Our 16S rRNA sequence analysis revealed that GH

BACKGROUND: Sarcopenia contributes to all-cause mortality in the elderly; however, there is no specific treatment. Mesenchymal stromal cells (MSCs) ameliorate age-related muscle loss and dysfunction and are potential therapeutic candidates for sarcopenia. However, their activity is easily affected by the surrounding environment and they are prone to replicative senescence during in vitro culture. Therefore, a drug that delays aging and enhances its function is required. Here, we investigated whether nicotinamide adenine dinucleotide (NAD+) pretreatment enhances the therapeutic efficacy of MSCs

Identifying genetic contributions of 6q21 loci and PPAR pathway to antipsychotic-induced metabolic syndrome: a Sex-Stratified Multi-Omics study.

Antipsychotics-induced metabolic syndrome (APs-induced MetS) is a common side-effect of antipsychotics, significantly increasing the risk of cardiovascular diseases and mortality. However, the genetic risk factors underlying APs-induced MetS remain poorly understood. Thus, we conducted a sex-stratified genome-wide association study (GWAS) in 3067 patients from Schizophrenia In Non-Occidental participants (SINO) trial, and significant results were validated in an independent cohort (all samples = 200) and proteomic data. Post-GWAS analyses were used to further explore the genetic mechanisms inv

Cross-tissue dual-omics analysis reveals molecular programs linked to myopia susceptibility and progression.

Myopia is a global cause of vision impairment, yet its molecular mechanisms remain unclear. We applied a multi-omics approach to the retina, choroid, and sclera of guinea pigs (GPs), a model of spontaneous myopia. Integrated transcriptomic and proteomic analyses suggest a two-state molecular framework. In a myopia-prone state, the choroid and sclera exhibited widespread transcript-protein discordance, with RNA and protein changes in remodeling pathways not aligned in direction, alongside metabolic down-regulation and suppressed immune transcription, a primed yet quiescent state preceding elong
