Sequential release of bioactive factors from functionalized metal-organic framework hydrogel enhances interfacial osseointegration of 3D-printed titanium alloy porous scaffolds.

Rationale: Repairing bone defects in osteoporotic patients presents a significant clinical challenge due to inadequate osseointegration, persistent inflammation, and elevated oxidative stress. To overcome these barriers, this study proposes the development of a functionalized 3D-printed titanium alloy porous scaffold capable of sequentially releasing therapeutic agents to modulate the immune environment and enhance bone regeneration. Methods: A thermosensitive collagen hydrogel was integrated with a zeolitic imidazolate framework (ZIF-8) to construct a dual-release platform capable of deliveri

Biomimetic Urethra-on-a-Chip Platform for Modelling Fibrosis: 3D-Printing and Near-Field Electrospinning in BAM-Functionalized Microenvironment.

Urethral stricture, a prevalent urological disorder characterized by fibrosis of periurethral tissues, severely compromises urinary function and patient quality of life. Despite various clinical interventions, recurrence remains frequent, largely due to the lack of physiologically relevant in vitro models for mechanistic investigation and drug screening. Here, we present a biomimetic urethra-on-a-chip platform that integrates microfluidics, three-dimensional (3D) printing, and near-field electrospinning to recapitulate the structural and biochemical complexity of the native urethra. The device

Enhancing the efficiency of bone tissue regeneration by using a 3D printed scaffold optimized with heparan sulfate proteoglycan 2.

Craniofacial bone deficiencies caused by trauma or disease pose clinical challenges as the shape of the damaged area varies between people. Although bone grafts are effective, they face issues such as poor drug retention and potential immune responses. PLA scaffolds possess therapeutic potential owing to their size, mechanical properties, stability, and biocompatibility. However, PLA scaffolds inherently lack bioactive molecules necessary to promote osteogenesis. HSPG2, also known as perlecan (Pln), are a basement membrane-specific GAG-containing core protein. Pln is a reservoir for heparin-bi

Tobramycin-laden 3D-printed UV-cured hyaluronic acid-PVA-based contact lens-like patches for improved antibiofilm activity and corneal healing in bacterial keratitis.

Delivering therapeutic agents through 3D-printed contact lenses like patches (CLLPs) is a promising strategy to achieve notable bioavailability while minimizing ocular drainage. In this study, 3D-printed CLLPs (15 mm in diameter, 0.3 mm in thickness, and 4 mm in height) were fabricated using a unique combination of polyvinyl alcohol methacrylate (PVAMA) and hyaluronic acid methacrylate (HAMA) in an optimal ratio, which was further UV-crosslinked (365 nm) and loaded with tobramycin to form a drug-eluting contact lens-like patch to relief bacterial keratitis (BK). The bio-ink utilized for 3D pri

Knee biomechanics during walking in individuals with anterior cruciate ligament repair: The role of a custom 3D-printed knee brace.

BACKGROUND: Anterior cruciate ligament (ACL) injuries frequently lead to altered gait biomechanics and muscle activation patterns, increasing the risk of osteoarthritis. Knee braces are commonly used to address these issues although a lack of consensus remains regarding their clinical benefits. The most recent 3D-printed braces, lighter and personalized, could improve rehabilitation. OBJECTIVES: To evaluate the effect of a novel custom-made 3D-printed knee brace (Provoke™) in individuals after unilateral ACL reconstruction during walking. The brace incorporates an asymmetrical hinge system ai

Mechanical and biological properties of 3D-printed porous titanium scaffolds coated with composite growth factors.

BACKGROUND: Osseointegration is considered a prerequisite for predicting implant success, and structure, biocompatibility, and properties of the implant are important parts of the factors that influence osseointegration. The focus of current research is on how to increase the strength of osseointegration on the implant and shorten the osseointegration time. METHODS: In this research, different porous scaffolds, including uniform, radial-gradient, and axial-gradient porous, were designed and fabricated. Their mechanical properties and biocompatibility were comprehensively evaluated through mec

Infill-Modulated, Bioerodible, and Biocompatible Ocular Inserts for Tunable Acyclovir Release via Direct Powder Extrusion 3D Printing.

Conventional treatment of herpetic keratitis via eye drops or ointments is limited by rapid precorneal elimination, frequent dosing requirements, and poor patient adherence. To overcome these challenges, this study explores the use of single-step direct powder extrusion (DPE) 3D printing for the fabrication of sustained-release, dissolvable ocular inserts composed of acyclovir, hydroxypropyl methylcellulose acetate succinate-high flow (HPMCAS-HF), and polyethylene glycol (PEG) 6000. Inserts were printed with varying infill densities (30%, 60%, and 90%) to modulate microstructural properties, d

3D printed titanium banana interbody cages versus titanium-coated PEEK bullet cages for TLIF.

INTRODUCTION: Lumbar degenerative spinal disease is a common, major cause of pain and disability. Titanium and polyetheretherketone (PEEK) are popular materials for interbody implants although evidence is mixed on which material is superior in terms of fusion and subsidence. The purpose of this study was to evaluate the clinical outcome of 3D printed titanium (3DPT) cages in patients undergoing TLIFs, as well as complication profiles based on widely used outcome metrics and reoperation events. METHODS: A retrospective review was conducted for patients receiving 1- or 2-level TLIF at an academ

Titanium patient-specific scaphoid replacement for unreconstructable nonunions: a case series study.

Although reconstruction is the preferred treatment option for symptomatic scaphoid nonunions, this may not be an option due to inadequate bone quality or degenerative changes. Existing salvage procedures tend to compromise function. This study introduces an alternative approach through the utilisation of a 3-D-printed titanium patient-specific scaphoid implant. In this retrospective case series, the outcomes of 12 patients treated for an unreconstructable scaphoid nonunion with a 3-D-printed prosthesis were evaluated. Patient-reported outcomes, range of motion, grip strength and plain radiogra

A xenogeneic extracellular matrix-based 3D printing scaffold modified by ceria nanoparticles for craniomaxillofacial hard tissue regeneration via osteo-immunomodulation.

Hard tissue engineering scaffolds especially 3D printed scaffolds were considered an excellent strategy for craniomaxillofacial hard tissue regeneration, involving crania and facial bones and teeth. Porcine treated dentin matrix (pTDM) as xenogeneic extracellular matrix has the potential to promote the stem cell differentiation and mineralization as it contains plenty of bioactive factors similar with human-derived dentin tissue. However, its application might be impeded by the foreign body response induced by the damage-associated molecular patterns of pTDM, which would cause strong inflammat

Bone Fracture-Treatment Method: Fixing 3D-Printed Polycaprolactone Scaffolds with Hydrogel Type Bone-Derived Extracellular Matrix and β-Tricalcium Phosphate as an Osteogenic Promoter.

Bone formation and growth are crucial for treating bone fractures. Improving bone-reconstruction methods using autologous bone and synthetic implants can reduce the recovery time. Here, we investigated three treatments using two different materials, a bone-derived decellularized extracellular matrix (bdECM) and β-tricalcium phosphate (β-TCP), individually and in combination, as osteogenic promoter between bone and 3D-printed polycaprolactone scaffold (6-mm diameter) in rat calvarial defects (8-mm critical diameter). The materials were tested with a human pre-osteoblast cell line (MG63) to dete

Piezoelectric needle sensor reveals mechanical heterogeneity in human thyroid tissue lesions.

Palpable thyroid lesions are common, and although mostly benign, lethal malignant nodules do occur and may be difficult to differentiate. Here, we introduce the use of a piezoelectric system called Smart-touch fine needle (or STFN) mounted directly onto conventional biopsy needles, to evaluate abnormal tissues, through quantitative real-time measurements of variations in tissue stiffness as the needle penetrates tissue. Using well-characterized biomaterials of known stiffness and explanted animal tissue models, we first established experimental protocols for STFN measures on biological tissues

3D titanium implant for orbital reconstruction after maxillectomy.

The surgical treatment of maxillary tumours often consists of an open subtotal or total maxillectomy with a subsequent significant defect. Reconstruction is, therefore, a major challenge for head and neck surgeons. Along with 3D printing development, titanium pre-bent implants have been created for orbital wall and floor reconstruction. The aim of this study was to evaluate the post-operative tolerance of these implants in patients who had undergone this procedure in our department. Implant tolerance was the primary endpoint, evaluated by whether or not surgery was required for infection or ex
