Potentiation of the actions of acetylcholine, epibatidine, and nicotine by methyllycaconitine at fetal muscle-type nicotinic acetylcholine receptors.

Methyllycaconitine (MLA) is a norditerpenoid alkaloid found in high abundance in toxic Delphinium (larkspur) species. It is a potent and selective antagonist of α(7)-nicotinic acetylcholine receptors, but has not been well investigated for activity aside from receptor antagonism. The aim of this study was to investigate the effects of MLA alone and in combination with acetylcholine, epibatidine, nicotine, and neostigmine for actions other than receptor antagonism in TE-671 cells expressing (α(1))(2)β(1)γδ nicotinic acetylcholine receptors. Ligand activity was assessed through measurements of m…

B cell-derived acetylcholine mitigates skin inflammation in mice through α9 nicotinic acetylcholine receptor-mediated signaling.

Chronic inflammatory skin disorders are characterized by keratinocyte hyperproliferation and hyperactivation as well as immune cell infiltration. We investigated whether immune cell-derived acetylcholine (ACh) is a modulator of skin inflammation in mice. Here, we identify skin epithelial B cells as a key source of ACh that damps down inflammation. We used imiquimod (IMQ) to induce inflammatory skin disease (ISD) in mice lacking ACh production specifically in B cells (ChATfl/fl;Mb1-Cre mice). Increased keratinocyte proliferation, epidermal thickening, and elevated levels of proinflammatory cyto…

Ethanol inhibits dorsomedial striatum acetylcholine release.

Alcohol use disorder (AUD) has severe adverse health and economic impacts totaling over $240 billion annually. Despite this, FDA approved treatments for AUD are limited. For this reason, further understanding of the neurobiological mechanisms in AUD is required for new treatments. An aspect of AUD involves deficits in behavioral flexibility that is similarly seen in animal models with depletion of dorsal striatal cholinergic interneurons (CINs). We found that acute EtOH (40 mM) inhibits the firing rate of dorsal striatal CINs, which are the primary source of acetylcholine (ACh) in the dorsal s…

Characterization of ganglionic acetylcholine receptor autoantibodies.

In myasthenia gravis (MG), autoantibodies bind to the alpha1 subunit and other subunits of the muscle nicotinic acetylcholine receptor (AChR). Autoimmune autonomic ganglionopathy (AAG) is an antibody-mediated neurological disorder caused by antibodies against neuronal AChRs in autonomic ganglia. Subunits of muscle and neuronal AChR are homologous. We examined the specificity of AChR antibodies in patients with MG and AAG. Ganglionic AChR autoantibodies found in AAG patients are specific for AChRs containing the alpha3 subunit. Muscle and ganglionic AChR antibody specificities are distinct. Ant…

Acetylcholine receptors in small cell carcinomas.

The presence of muscarinic and nicotinic acetylcholine receptors (m- and nAChR, respectively) in small cell carcinomas (SCC) of the lung was assessed by measurement of specific binding of (-)[3H]quinuclidinyl benzilate ([3H]QNB) and 125I-alpha-bungarotoxin, respectively. Of five SCC studied, four were originally derived from patients with the Lambert-Eaton myasthenic syndrome, an autoimmune disease of neuromuscular transmission, and one was from a patient without evidence of neurological disease. There was no evidence of nAChR, but all tumors bound (-)[3H]QNB in a saturable and specific manner…

Three acetylcholine receptors in Aplysia neurones.

1. In the pleural ganglion of Aplysia californica, groups of cells were identified that respond differently to an iontophoretic injection of ACh. The anterior neurones are excited by ACh, whereas the medial cells are inhibited. The inhibitory response is biphasic, consisting of a short-latency, rapid component and a longer-latency, slow component. Homologous responses (e.p.s.p.s in the anterior cells and a two-component inhibitory response in the medial cells) are evoked in these same cell groups by stimulation of an identifiable presynaptic neurone.2. The e.p.s.p. and the corresponding ACh po

ACETYLCHOLINE-LIKE ACTIVITY IN SCIATIC NERVE.

Extracts of sciatic nerve exhibit acetylcholine-like activity that is only partly attributable to acetylcholine. The extracts show relatively greater activity on the rectus abdominis muscle of the frog than on the ileum of the guinea pig. To prevent the action of the extract on the frog rectus abdominis muscle, a greater concentration of d-tubocurarine is required than is necessary to prevent the action of known acetylcholine.

Muscarinic acetylcholine receptor M1 and M3 subtypes mediate acetylcholine-induced endothelium-independent vasodilatation in rat mesenteric arteries.

The present study investigated pharmacological characterizations of muscarinic acetylcholine receptor (AChR) subtypes involving ACh-induced endothelium-independent vasodilatation in rat mesenteric arteries. Changes in perfusion pressure to periarterial nerve stimulation and ACh were measured before and after the perfusion of Krebs solution containing muscarinic receptor antagonists. Distributions of muscarinic AChR subtypes in mesenteric arteries with an intact endothelium were studied using Western blotting. The expression level of M1 and M3 was significantly greater than that of M2. Endothel…

G alpha(q)-coupled muscarinic acetylcholine receptors enhance nicotinic acetylcholine receptor signaling in Caenorhabditis elegans mating behavior.

In this study, we address why metabotropic and ionotropic cholinergic signaling pathways are used to facilitate motor behaviors. We demonstrate that a G alpha(q)-coupled muscarinic acetylcholine receptor (mAChR) signaling pathway enhances nicotinic acetylcholine receptor (nAChR) signaling to facilitate the insertion of the Caenorhabditis elegans male copulatory spicules into the hermaphrodite during mating. Previous studies showed that ACh (acetylcholine) activates nAChRs on the spicule protractor muscles to induce the attached spicules to extend from the tail. Using the mAChR agonist Oxo M (o…

Nicotinic acetylcholine receptors in pain modulation.

Chronic pain is widely recognized as a major global health problem that affects approximately one-fifth of the adult population and is assoc … safer and more effective non-opioid analgesics remains an important unmet clinical need. Nicotinic acetylcholine receptors (nAChRs) have been identified as promising targets for the development of next-generation analgesics because of their roles in pain signal transmission, neuroinflammation, and immune-neural interactions. nAChRs are pentameric ligand-gated …

Autoimmunity to neuronal nicotinic acetylcholine receptors.

Nicotinic acetylcholine receptors (nAChRs) are widely expressed in many and diverse cell types, participating in various functions of cells, tissues and systems. In this review, we focus on the autoimmunity against neuronal nAChRs, the specific autoantibodies and their mechanisms of pathological action in selected autoimmune diseases. We summarize the current relevant knowledge from human diseases as well as from experimental models of autoimmune neurological disorders related to antibodies against neuronal nAChR subunits. Despite the well-studied high immunogenicity of the muscle nAChRs where…

Nicotinic acetylcholine receptor signaling in atherogenesis.

Smoking is a major risk factor for the development of atherosclerosis, stroke and myocardial infarction. Cigarette smoke consists of a complex mixture of about 4000 compounds. Out of these, polycyclic hydrocarbons, tobacco-specific nitrosamines, oxidizing agents and carbon monoxide have been implicated in the development of atherosclerosis. Recent studies have shown that nicotine (the addictive component of cigarettes) binds to high affinity cell-surface receptors and accelerates the atherogenic process. These receptors are called nicotinic acetylcholine receptors (nAChRs) and are expressed ub…

Muscarinic acetylcholine receptors and airway diseases.

Parasympathetic nerves provide the dominant autonomic innervation of the airways. Release of acetylcholine from parasympathetic nerves activates postjunctional muscarinic receptors present on airway smooth muscle, submucosal glands, and blood vessels to cause bronchoconstriction, mucus secretion, and vasodilatation, respectively. Acetylcholine also feeds back onto prejunctional muscarinic receptors to enhance or inhibit further acetylcholine release. In asthma and chronic obstructive pulmonary disease, bronchoconstriction and mucus secretion is increased and the airways are hyperresponsive to …

Acetylcholine release from cat carotid bodies.

Hypoxia, hypercapnia and acidosis stimulate the carotid body (CB) sending increased neural activity via a branch of the glossopharyngeal ner … in response to hypoxia, and serve as the stimulating transmitter(s) for chemosensory nerve endings. Acetylcholine (ACh) has been proposed as such an agent from the outset, but this proposal has been, and remains, controversial. The present study tests two hypotheses: (1) The CB releases ACh under normoxic/normocapnic conditions; and (2) The amount released incr …

Who do barium ions imitate acetylcholine?

The action of 1-4 mM barium on bullfrog sympathetic ganglion cells was studied using voltage clamp. Barium imitated the action of muscarine, causing depolarization, increased input resistance, tendency to repetitive firing, and a specific inhibition of a slow small outward current termed the M current. Barium did not produce significant short-term inhibition of three other outward currents; the delayed rectifier, the calcium-dependent K current, and the A current.
