Unique adaptations in sulfatases underpin colonic mucin degradation by Akkermansia muciniphila.

Excessive foraging of colonic mucin glycans by gut bacteria is associated with diseases such as inflammatory bowel disease. Although Akkermansia muciniphila is an important mucin degrader, the role of carbohydrate sulfatases that facilitate digestion of these heavily sulfated glycans remains unclear. Combining in vitro digestion assays, proteomics and structural biology, we show that A. muciniphila sulfatases, such as Amuc1755 and Amuc0953, have rare adaptations targeted towards known sulfated mucin structures. They show larger degrees of modularity, including a previously unknown mucin-bindin…

Akkermansia muciniphila inhibits jejunal lipid absorption and regulates jejunal core bacteria.

Insufficiency of Akkermansia muciniphila (Akk) has been implicated in the pathogenesis of metabolic diseases, and administration or restoration of Akk has ameliorated these disorders. Recently, Pasteurized Akk (PA-Akk) has been approved as a functional food. However, the impact of Akk on lipid absorption in the proximal intestine, which is directly exposed to orally administered Akk, remains largely unexplored. In this study, we orally administered Akk and PA-Akk to mice and investigated the subsequent lipid absorption. Long-term administration of Akk resulted in reduced lipid deposits in the …

Akkermansia muciniphila: A key player in gut microbiota-based disease modulation.

Akkermansia muciniphila (A. muciniphila), a mucin-degrading bacterium residing in the gut's mucus layer, has emerged as a key modulator of host physiology with significant implications for health and disease. Growing evidence shows that A. muciniphila influences host metabolism, strengthens gut barrier integrity, modulates microbial composition, and regulates immune responses. This review synthesizes current literature on A. muciniphila, emphasizing its role in conditions such as metabolic disorders, inflammatory bowel disease (IBD), Clostridioides difficile infection (CDI), cancer, cardiovasc…

Oral administration of Akkermansia muciniphila elevates systemic antiaging and anticancer metabolites.

The presence of Akkermansia muciniphila (Akk) in the human gut is associated with good health, leanness and fitness. Mouse experimentation has demonstrated positive effects for Akk, which counteracts aging, mediates antiobesity and antidiabetic effects, dampens inflammation and improves anticancer immunosurveillance. Clinical trials have confirmed antidiabetic effects for Akk. Here, we investigated the time-dependent effects of oral administration of Akk (which was live or pasteurized) and other bacteria to mice on the metabolome of the ileum, colon, liver and blood plasma. Metabolomics was pe…

Different subtype strains of Akkermansia muciniphila abundantly colonize in southern China.

AIM: This study investigates the colonization rate of Akkermansia muciniphila in the gastrointestinal tracts of people living in southern China and applies a modified method for the isolation and subtyping of A. muciniphila strains from faecal samples. METHODS AND RESULTS: Fresh faecal samples were collected and bacterial DNA was extracted from these samples for real-time PCR analysis. Strains were separated using a culture-dependent sPCR-directed method and classified using an enterobacterial repetitive intergenic consensus (ERIC-PCR) DNA fingerprinting method. The colonization rate for the …

Solubilized lignin promotes the growth of Akkermansia muciniphila and improves insulin sensitivity.

Solubilized lignin (SL) is a water-soluble derivative of lignin, which is typically classified as an insoluble dietary fiber and is predicted to possess structural and physicochemical properties that are distinct from those of conventional dietary fibers. In this study, we evaluated the effects of SL intake on gut microbial composition, intestinal barrier function, and insulin sensitivity in mice. SL supplementation significantly increased the abundance of Akkermansia muciniphila in both lean and diet-induced obese (DIO) mice. In vitro, SL supported the growth of A. muciniphila, suggesting a u…

Advancements in understanding the relationship between Akkermansia muciniphila and the development of ulcerative colitis.

Ulcerative colitis (UC) is a chronic inflammatory bowel disease characterized by severe intestinal barrier dysfunction and immune dysregulation in patients, with limited clinical treatment options. Recent research highlights the important role of the gut bacterium Akkermansia muciniphila (AKK) in both the development and management of UC. AKK supports the integrity of the intestinal barrier by metabolizing mucins, enhances the production of tight junction proteins (Occludin-1/ZO-1), and influences immune responses by shifting macrophage polarization from M1 to M2, suppressing pro-inflammatory …

Akkermansia muciniphila and osteoporosis: emerging role of gut microbiota in skeletal homeostasis.

Osteoporosis (OP) is a prevalent age-related skeletal disease. It is marked by compromised bone strength and higher fracture risk. Emerging evidence ties gut dysbiosis to OP development. Yet, the exact role of specific commensal bacteria remains unclear. Here, we review how Akkermansia muciniphila (A. muciniphila) affects bone metabolism. This mucin-degrading bacterium acts through three well-documented mechanisms: metabolite signaling, immune modulation, and gut-bone axis crosstalk. We also discuss emerging factors, such as host metabolic status, mechanical loading, and biomaterial applicatio…

Akkermansia muciniphila as a novel powerful bacterial player in the treatment of metabolic disorders.

Akkermansia muciniphila (A. muciniphila) is a mucin-degrading bacterium that commonly lives in the intestinal mucus layer. It is normally detected in human faecal specimens and is one of the few bacteria potentially associated to obesity development. In this narrative review, possible mechanisms that support how A. muciniphila is implicated in the pathogenesis of obesity and metabolic-associated disease are described with the evaluation of its role as an intermediary or independent agent whose manipulation could be useful in the management of metabolic disorders. The ampleness of A. muciniphil…

Microbe on the move: Akkermansia in infectious diseases and emerging roles in gynecological health.

The genus Akkermansia was first described in 2004 following the identification of Akkermansia muciniphila, a Gram-negative, mucin-degrading bacterium of the intestine that constitutes 1-3% of the total adult fecal content. Since the interest in A. muciniphila in human health has increased over the past decade, an extensive amount of research examining the impact of A. muciniphila on metabolic disorders, non-communicable diseases, and during infection has been published. Furthermore, a rapidly evolving area of research is the role of A. muciniphila in gynecological health. Many studies have sho…

Faecalibacterium prausnitzii and Akkermansia muciniphila: driving the multiple sclerosis-a systematic review.

The gut microbiota (GM) is a complex microbial ecosystem, and its alteration contributes to the development of several diseases including multiple sclerosis (MS). Progressive research concerning neurodegenerative diseases and GM reveals that GM dysbiosis has been linked to fostering the development and progression of MS. Among existing bacteria, F.prausnitzii and A.muciniphila are key species implicated in this disease. Thus, the present study systematically reviewed and synthesized the research on the involvement of F. prausnitzii and A. muciniphila in MS patients versus healthy individuals.

Potential role of Akkermansia massiliensis in multiple sclerosis protection by the FcRL3 gene.

The contribution of human genetics in shaping the abundance of gut microbiota is small, about 5%; however, it can inform about disease etiopathogenesis, including multiple sclerosis (MS). Searching for MS-gut microbiota cross-comparison of reported genome-wide association studies (GWAS), we identified a coincident association of the disease with a commensal - Akkermansia massiliensis-in the selective Fc receptor of secretory IgA gene (FcRL3). Those two signals colocalized, supporting that lower abundance of A.massiliensis may contribute to MS predisposition. In cis, the FcRL3-MS predisposing s…

Characterization of antibiotic-resistance traits in Akkermansia muciniphila strains of human origin.

Akkermansia muciniphila, a commensal bacterium commonly found in healthy gut microbiota, is widely considered a next-generation beneficial bacterium candidate to improve metabolic and inflammatory disorders. Recently the EFSA's Panel on Nutrition, Novel food, and Food Allergens has declared that pasteurized A. muciniphila DSM 22959T (also MucT, ATCC BAA-835) can be considered safe as a novel food, opening the door to its commercialization as a food supplement. Despite its recognized health benefits, there is still little information regarding the antimicrobial susceptibility of this species an…
