Life cycle of Amblyomma integrum (Acari: Ixodidae) under laboratory conditions.

Amblyomma integrum is a hard tick infesting mainly buffalo and cattle and has been identified as an agent of human otoacariasis in Sri Lanka. Data on the life cycle pattern of A. integrum were collected by experimental infestation on New Zealand white rabbits under laboratory conditions. Wild-caught females laid 55-7389 eggs for 2-35 days after spending a latent period of 10-25 days. Egg incubation period was 31-105 days and the newly emerged larvae started feeding after 4-11 days. Larvae dropped off after feeding and they moulted into nymphs after 10-16 days. Nymphs actively fed on rabbits fo…

Population genetics of Amblyomma americanum (Acari: Ixodidae) collected from Arkansas.

Lone star ticks, Amblyomma americanum L. (Acari: Ixodidae),infest multiple hosts such as birds, and mammals of various sizes (rodents to white-tailed deer) and can harbor human pathogens such as Borrelia lonestari and Ehrlichiosis chaffeensis. The population structure of 251 A. americanum ticks, collected from canines and two white-tailed deer in six Arkansas ecoregions, was examined using DNA sequences of a 247-bp region of the mitochondrial DNA ribosomal RNA 16S gene. Of the 247 nucleotide characters, 26 were variable. Thirty-three haplotypes were identified of which 25 haplotypes occurred o…

Amblyomma cajennense ticks induce immediate hypersensitivity in horses and donkeys.

Since host immune reaction to ticks interferes with tick-borne pathogen transmission, it is important to recognize naturally occurring tick-host immune relationships to better understand the epidemiology of such infectious diseases. Amblyomma cajennense is an important tick-borne disease vector in the Neotropical region and horses maintain it in domestic environments. In the present work intradermal testing of A. cajennense tick exposed horses and donkeys using crude tick antigens was used to evaluate the type of hypersensitivity induced by infestations. Animals sensitized by A. cajennense inf…

Density and behavior of capybara (Hydrochoerus hydrochaeris) ticks (Acari: Ixodidae) Amblyomma sculptum and Amblyomma dubitatum with notes on Rickettsia bellii infection: Assessing human exposure risk.

In several urban and peri‑urban areas of Brazil, populations of Amblyomma sculptum and Amblyomma dubitatum ticks are maintained by capybaras (Hydrochoerus hydrochaeris). In some of these areas, this host and these tick species are associated with Brazilian spotted fever (BSF), a lethal human disease caused by the bacterium Rickettsia rickettsii. In this work, we evaluated the risk of human exposure to these tick species using four collection techniques to discern host-seeking behavior. The study was carried out in 10 urban sites inhabited by capybaras in Uberlândia, a BSF-free municipality in …

Seasonality of the Rickettsia vectors Amblyomma ovale and Amblyomma sculptum on dogs in Atlantic Forest and Cerrado biomes of Minas Gerais, Brazil: Hypotheses for human rickettsiosis seasonality.

Free-ranging rural dogs in Brazil are parasitized by ticks from indigenous ixodofauna including Amblyomma sculptum and Amblyomma ovale, vectors of Rickettsia rickettsii and Rickettsia parkeri strain Atlantic Rainforest (ARF), respectively. Therefore, dogs are important sentinels and hosts for ticks and tick-borne diseases of public health relevance. This study evaluated abundance, and seasonal dynamics of these tick species infesting free-ranging rural dogs in two municipalities of Minas Gerais State, southeastern Brazil: Araguari (Cerrado biome remnants) and Guaxupé (Atlantic Forest remnants)…

Further insights into the sialome switch of Amblyomma americanum adult females.

BACKGROUND: The lone star tick, Amblyomma americanum, is a three-host species widely distributed across North America and a vector for multiple human pathogens. Bites from this tick is also associated with alpha-gal syndrome, underscoring its growing public health importance. Adult female ticks undergo a prolonged feeding cycle divided into preparatory, slow-feeding, and rapid-feeding phases. Tick salivary glands are essential for successful blood feeding, producing a complex mixture of molecules that modulate host hemostasis, immunity, and tissue repair. The dynamic remodeling of the salivary…

Amblyomma sculptum: genetic diversity and rickettsias in the Brazilian Cerrado biome.

Amblyomma sculptum (Ixodida: Ixodidae) Berlese, 1888 is the most important tick vector in Brazil, transmitting the bioagent of the most severe form of spotted fever (SF) in part of the Cerrado (in the states of Minas Gerais and São Paulo). In another part of the Cerrado (Central-West region of Brazil), a milder form of SF has been recorded. However, neither the rickettsia nor the vector involved have been characterized. The aim of the current study was to analyse genetic variation and the presence of rickettsia in A. sculptum in Cerrado, from silent areas and with the milder form of SF. Sample…

Discovery of filarial nematode DNA in Amblyomma americanum in Northern Virginia.

Ticks collected in 2011 were screened for the presence of filarial nematode genetic material, and positive samples were sequenced for analysis. Monanema-like filarial nematode DNA was recently discovered in Amblyomma americanum in northern Virginia, marking the first time genetic material from this parasite has been discovered in ticks in the state. Phylogenetic analysis revealed that this material was directly related to a previously discovered filarial nematode in A. americanum populations in Maryland as well as recently identified parasites in Ixodes scapularis from southern Connecticut. Tick paralysis caused by Amblyomma maculatum on the Mexican Pacific Coast.

Tick paralysis is a rare entity in which it is necessary to identify the cause and remove the arthropod to have a rapid remission of symptoms. In the absence of an early diagnosis, the outcome can be fatal, as toxins are released from the tick's saliva as it feeds. To the best of the authors' knowledge, this is the first clinical report of the disease in Mexico and Latin America. A 22-year-old man from a rural area, who was in contact with cattle, developed ascending flaccid paralysis secondary to Amblyomma maculatum tick toxin. He presented flaccid paraplegia and arreflexia that progressed un…

Distribution and genetic variation of Amblyomma triste (Acari: Ixodidae) in Argentina.

A study was conducted to investigate the distribution of Amblyomma triste in Argentina under the hypothesis that this tick prevails in riparian localities along the Paraná River and adjacent humid environments from 34° 30' S to 25° 20' S, approximately. Ticks were collected from mammals and vegetation in those environments from November 2008 to October 2012. Additionally, genetic variation was tested from Argentinean, Brazilian, Chilean, and Uruguayan populations of A. triste by comparing sequences of 16S rDNA mitochondrial gene. The hypothesis was not confirmed because A. triste were collecte…

A case of tick bite from Amblyomma testudinarium on the glans penis.

A tick which bit the glans penis of a 40-year-old Japanese man was surgically excised together with the skin where the bite occurred. It was identified as Amblyomma testudinarium through the taxonomical investigation of its morphological characteristics. To our best knowledge, our patient is the sixth tick bite from this species of tick recorded in the Chugoku District of Japan.

Amblyomma americanum: a potential vector of Lyme disease in New Jersey.

Amblyomma americanum is a likely secondary vector of Lyme disease in New Jersey. Ticks of this species were removed from the site of the characteristic skin lesion known as erythema chronicum migrans on two patients with the disease, and the Lyme disease spirochete was isolated from nymphs and adults of this species. That A. americanum is a potential vector is supported by its similarities to Ixodes dammini, the known tick vector, in seasonal distribution and host utilization. The extensive range of A. americanum may have great implications for potential Lyme disease transmission outside known…

Comparing scapular morphology of Amblyomma sculptum and Amblyomma dubitatum nymphs allows a fast and practical differential diagnosis of ticks in highly infested areas with dominance of these two species.

Rickettsia rickettsii infection causes the highest human fatality rate among all Rickettsia species of the world and is endemic in Southeast Brazil. In this part of the country most human spotted fever cases are related to unnaturally high populations of ticks, usually a mix of two species, Amblyomma dubitatum Neumann and Amblyomma sculptum Berlese and their local host, capybara (Hydrochoerus hydrochaeris). At the same time, an increase in the number of SF notifications as well as its urbanization was observed, and a better characterization of disease epidemiology is mandatory for control meas…

Human otoacariasis by Amblyomma testudinarium identified with 16S rRNA gene sequencing.

Human otoacariasis caused by tick bites in the external auditory canal is rarely reported in Japan, despite the recent increase in the movement of wild animals, known hosts of ticks into human living areas due to global warming. We report a case of human otoacariasis caused by Amblyomma testudinarium, genetically identified using 16S rRNA gene sequencing. A 43-year-old man presented to an urban hospital in Tokyo with left-sided otalgia. An endoscopic examination revealed a tick anchored to the skin of the external auditory canal, which showed erythema and swelling. Initial attempts to remove t…

Time-resolved proteomic profile of Amblyomma americanum tick saliva during feeding.

Amblyomma americanum ticks transmit more than a third of human tick-borne disease (TBD) agents in the United States. Tick saliva proteins are critical to success of ticks as vectors of TBD agents, and thus might serve as targets in tick antigen-based vaccines to prevent TBD infections. We describe a systems biology approach to identify, by LC-MS/MS, saliva proteins (tick = 1182, rabbit = 335) that A. americanum ticks likely inject into the host every 24 h during the first 8 days of feeding, and towards the end of feeding. Searching against entries in GenBank grouped tick and rabbit proteins in…
