Acetic acid enhances attraction to and colonization of wood bolts by ambrosia beetles (Coleoptera: Curculionidae: Scolytinae).

Certain invasive ambrosia beetles are attracted to and colonize physiologically stressed trees emitting ethanol. Xylosandrus germanus (Blandford) and Xylosandrus crassiusculus (Motschulsky) are attracted to trees emitting ethanol and are 2 of the most damaging invasive ambrosia beetles in North America. Acetic acid has been detected in stressed trees emitting ethanol, but whether it has a role in ambrosia beetle ecology is unknown. Sections of tree stems (bolts) were soaked in various dilutions of acetic acid alone and mixed with a 5% dilution of ethanol, then deployed in a field near a woodla…

Natural history, landscape ecology, and management of ambrosia beetles (Coleoptera: Curculionidae) as pests of nursery crops.

Ambrosia beetles (Coleoptera: Curculionidae: Scolytinae) are economically important pests of ornamental nursery crops because the presence of <5 attacks can render thin-barked deciduous trees unmarketable. In North America, Xylosandrus crassiusculus (Motschulsky), Xylosandrus germanus (Blandford), Xylosandrus compactus (Eichhoff), and Cnestus mutilatus (Blandford) are among the most destructive ambrosia beetle pest species in nursery systems. Anisandrus maiche (Stark) is a new emerging pest in North America. These species preferentially colonize physiologically stressed hosts by orienting to e…

Natural terpenoids from Ambrosia species are active in vitro and in vivo against human pathogenic trypanosomatids.

Among the natural compounds, terpenoids play an important role in the drug discovery process for tropical diseases. The aim of the present work was to isolate antiprotozoal compounds from Ambrosia elatior and A. scabra. The sesquiterpene lactone (STL) cumanin was isolated from A. elatior whereas two other STLs, psilostachyin and cordilin, and one sterol glycoside, daucosterol, were isolated from A. scabra. Cumanin and cordilin were active against Trypanosoma cruzi epimastigotes showing 50% inhibition concentrations (IC50) values of 12 µM and 26 µM, respectively. Moreover, these compounds are a…

Modifiable Risk Factors for Common Ragweed (Ambrosia artemisiifolia) Allergy and Disease in Children: A Case-Control Study.

Ragweed allergy is a major public health concern. Within Europe, ragweed is an introduced species and research has indicated that the amounts of ragweed pollen are likely to increase over Europe due to climate change, with corresponding increases in ragweed allergy. To address this threat, improving our understanding of predisposing factors for allergic sensitisation to ragweed and disease is necessary, specifically focusing upon factors that are potentially modifiable (i.e., environmental). In this study, a total of 4013 children aged 2⁻13 years were recruited across Croatia to undergo skin p

Bark and Ambrosia Beetle (Curculionidae: Scolytinae) Diversity Found in Agricultural and Fragmented Forests in Piracicaba-SP, Brazil.

Land use changes and forest fragmentation result in biodiversity loss and displacement, with insects among the most affected groups. Among these, bark beetles (Curculionidae: Scolytinae) occupy a prominent position due to their close ties to food resources, i.e., trees, and importance as primary decomposers in forest ecosystems. Therefore, our study aimed to document scolytine biodiversity associated with landscape components that vary based on their physical or botanical composition. Bark beetle diversity was sampled monthly for 12 mo in an Atlantic forest remnant and five adjacent vegetation

Synergistic Effects of Acetic Acid and Ethanol on Offspring Production and Gallery Expansion by Fungus-Farming Ambrosia Beetles.

Wood-boring ambrosia beetles and their offspring obligately depend on cultivated fungi that they maintain within host trees. Stressed trees produce at least two potent antimicrobials, ethanol and acetic acid, which typically suppress the growth of most fungi. Ethanol facilitates the growth of ambrosia beetles' fungal mutualists and thus aids in host colonization; however, acetic acid's effects are unknown. Here, we evaluated the effects of acetic acid on tunneling and offspring production by two exotic ambrosia beetle species, Xylosandrus germanus and Anisandrus maiche. Cut wood stems (i.e., b…

Geosmithia associated with hardwood-infesting bark and ambrosia beetles, with the description of three new species from Poland.

Geosmithia species (Hypocreales, Ascomycota) are associates of bark beetles and other arthropods. One species, Geosmithia morbida, is a virulent tree pathogen of Juglans nigra. To date, 10 Geosmithia spp. from conifer-infesting, and at least 23 species from hardwood associated bark beetles have been reported from Europe. The aim of this study was to survey Geosmithia spp. associated with 18 bark and ambrosia beetle species in hardwood ecosystems in Poland. In addition, we evaluated the pathogenicity of the six Geosmithia species by inoculating Acer, Fagus, Quercus, Tilia and Ulmus seedlings. O…

Fungal symbiont of an ambrosia beetle possesses high nutrient content and suppresses competing fungi with antimicrobial compounds.

Wood-colonizing beetles are associated with a diversity of microbes many of which are thought to act as mutualists with their beetle hosts, but the evidence is usually anecdotal. The ship-timber beetle Elateroides dermestoides, one of the few fungus-farming but nonsocial ambrosia beetles, is described to have a mutualistic relationship with the yeast-like fungus Alloascoidea hylecoeti. Here, we tested the hypothesis that A. hylecoeti has a high nutrient content thus allowing it to function as a valuable food source for the solitary larvae of E. dermestoides, which bore into the wood of dead tr…

Sesquiterpene Lactone Derivatives of Ambrosia artemisiifolia Act as Agonists of the Transient Receptor Potential Ankyrin 1 Ion Channel.

Background: Transient receptor potential ankyrin 1 (TRPA1) and vanilloid 1 (TRPV1) are key nociceptive ion channels involved in chemosensation, pain signaling, and neurogenic inflammation. Several sesquiterpene lactones target TRPA1, but the activity profile of these secondary metabolites of the common ragweed (Ambrosia artemisiifolia) remains uncharacterized. Purpose: The purpose of this study was to explore the activity of five ragweed sesquiterpene lactones on TRPA1 and TRPV1, respectively. Methods: We investigated the effects of five ragweed-derived sesquiterpene lactones-acetoxydihydrodam…

Carrying the seeds of your crop: fungal genetic networks activated during colonization of the ambrosia beetle symbiotic organ.

BACKGROUND: Myco-cultivation by ambrosia beetles involves unique symbiotic partnerships with select fungi. These beetles grow and tend their fungal gardens as food for larvae and adults. This close association has led to the evolution of specialized structures, termed mycangia, on or within the beetle to house fungal cells as "seeds" for establishing and maintaining their gardens. However, little is known concerning fungal gene networks involved in mediating the symbiotic colonization of the mycangia of their host beetles. Here, we report on global gene expression changes that occur in the fun…

Partnerships Between Ambrosia Beetles and Fungi: Lineage-Specific Promiscuity Among Vectors of the Laurel Wilt Pathogen, Raffaelea lauricola.

Nutritional mutualisms that ambrosia beetles have with fungi are poorly understood. Although these interactions were initially thought to be specific associations with a primary symbiont, there is increasing evidence that some of these fungi are associated with, and move among, multiple beetle partners. We examined culturable fungi recovered from mycangia of ambrosia beetles associated with trees of Persea humilis (silk bay, one site) and P. americana (avocado, six commercial orchards) that were affected by laurel wilt, an invasive disease caused by a symbiont, Raffaelea lauricola, of an Asian…

Characterization of a novel R98Q mutation that confers resistance to sulfentrazone in common ragweed (Ambrosia artemisiifolia) populations from Michigan.

BACKGROUND: In 2023, soybean growers in Eaton County, Michigan, USA, reported repeated failures to control common ragweed (Ambrosia artemisiifolia L.) with protoporphyrinogen IX (PPO)-inhibiting herbicides, sulfentrazone and fomesafen, in soybean plants. This study aimed to investigate resistance levels and mechanisms of resistance in two suspected resistant populations (R1 and R2). RESULTS: Dose-response assays revealed resistance to sulfentrazone in both populations, with median lethal dose (LD50) values 36- and 24-fold higher than the susceptible population for R1 and R2, respectively, and…

Seasonal changes in population structure of the ambrosia beetle Xylosandrus compactus and its associated fungi in a southern Mediterranean environment.

Exotic ambrosia beetles are increasing in Europe due to global trade and global warming. Among these xylomycetophagous insects, Xylosandrus compactus (Eichhoff) (Coleoptera: Curculionidae) is a serious threat for several Mediterranean host plants. Carob trees growing in Sicily (Italy) have been extensively attacked by beetles leading to rapid tree decline. Although X. compactus has been found in Europe for several years, most aspects of its ecology are still unknown. We thus studied the population structure and dynamics of X. compactus, together with its twig size preference during a sampling …

Spore Acquisition and Survival of Ambrosia Beetles Associated with the Laurel Wilt Pathogen in Avocados after Exposure to Entomopathogenic Fungi.

Laurel wilt is a disease threatening the avocado industry in Florida. The causative agent of the disease is a fungus vectored by ambrosia beetles that bore into the trees. Until recently, management strategies for the vectors of the laurel wilt fungus relied solely on chemical control and sanitation practices. Beneficial entomopathogenic fungi (EPF) are the most common and prevalent natural enemies of pathogen vectors. Laboratory experiments demonstrated that commercial strains of EPF can increase the mortality of the primary vector, Xyleborus glabratus, and potential alternative vectors, Xylo…

Pyrethroids demonstrate effective insecticidal activity against ambrosia beetles (Coleoptera: Curculionidae: Scolytinae) within tree fruit, nut, and ornamental production systems.

The ambrosia beetles Xylosandrus crassiusculus (Motschulsky) and Xylosandrus germanus (Blandford) (Coleoptera: Curculionidae) are major pests in fruit, nut, and ornamental tree nurseries. Adult females tunnel into stressed trees, creating galleries in the sapwood and heartwood to cultivate their nutritional fungal mutualists, which are associated with branch dieback and tree death. The current management approach relies on trunk applications of permethrin and bifenthrin to decrease infestation risk in the United States. However, the efficacy of other insecticides has not been thoroughly evalua…

Comparing the effects of verbenone, methyl salicylate, and permethrin on attacks of ambrosia beetles (Coleoptera: Curculionidae Scolytinae) in ornamental nurseries.

The granulate ambrosia beetle, Xylosandrus crassiusculus (Motschulsky), and the black stem borer, Xylosandrus germanus (Blandford) are important pests in ornamental nurseries in the eastern USA. These beetles are managed mainly using preventative trunk applications of pyrethroids, such as permethrin or bifenthrin when females typically fly out of woodlots and attack young trees in the spring. Verbenone and methyl salicylate are potential phytochemicals reported as repellants but not completely validated in ornamental nurseries for ambrosia beetle management as an alternative option. Thus, this…

Fusarium kuroshium is the primary fungal symbiont of an ambrosia beetle, Euwallacea fornicatus, and can kill mango tree in Japan.

This study identifies fungi associated with Euwallacea fornicatus and determines whether these fungal species play the role of primary symbiont. E. fornicatus adults that emerged from the branches of infested trees in Okinawa main island, Japan, were collected and used to isolate fungi. Fusarium kuroshium and Penicillium citrinum were the most dominant fungal associates of females and males, respectively. F. kuroshium was much more frequently isolated from the head, including mycangia (fungus-carrying organs), of females than any other body parts. We inoculated healthy mango saplings with F. k

Acute effect of an extract of Ambrosia paniculata (Willd.) O. E. Schultz (mugwort) in several models of experimental epilepsy.

The acute effect of Ambrosia paniculata was studied in several animal models of epilepsy. Intraperitoneal injections (0.01 mL/g body wt) of a decoction of the dry leaves significantly enhanced the latency to the first convulsion and survival time in mice injected with picrotoxin (7 mg/kg) or isoniazid (210 mg/kg). Epileptic spikes were induced by topical application of penicillin through a glass electrode filled with a penicillin-agar-saline mixture and recorded in sensorimotor and occipital cortices, in rats immobilized with d-tubocurarine. The plant decoction reduced significantly the spike …
