Half-sandwich iridium complexes bearing bidentate urea-phosphorus ligands were found to catalyze the direct reductive amination of aromatic and aliphatic ketones under mild conditions at 0.5 mol % loading with high selectivity towards primary amines. One of the complexes was found to be active in both the Leuckart-Wallach (NH4 CO2 H) type reaction as well as in the hydrogenative (H2 /NH4 AcO) reductive amination. The protocol with ammonium formate does not require an inert atmosphere, dry solvents, as well as additives and in contrast to previous reports takes place in hexafluoroisopropanol (H…

3-Aminoquinolines are privileged heterocyclic motifs widely used in medicinal chemistry and functional materials. However, their efficient and regioselective synthesis remains highly challenging due to the intrinsic electronic bias of the quinoline ring and compatibility issues of the reaction system. Direct C3-H amination of quinolines represents one of the most straightforward and efficient strategies, yet practical and effective methods for this transformation remain rare. Against these challenges, we report a thermodynamically driven direct meta-C-H amination of quinolines. With quinoline …

Trace amine-associated receptor 1 (TAAR1), the founding member of a nine-member family of trace amine receptors, is responsible for recognizing a range of biogenic amines in the brain, including the endogenous β-phenylethylamine (β-PEA)1 as well as methamphetamine2, an abused substance that has posed a severe threat to human health and society3. Given its unique physiological role in the brain, TAAR1 is also an emerging target for a range of neurological disorders including schizophrenia, depression and drug addiction2,4,5. Here we report structures of human TAAR1-G-protein complexes bound to

Isohexide-derived diamines are considered preferred precursors for the production of biobased polyurethanes and polyamides. However, current synthesis methods from isohexides suffer from serious issues concerning selectivity and the recyclability of the process auxiliaries (e. g. homogeneous catalysts), which renders a translation to the industry highly unlikely. Here, we report on the production of such diamine building blocks, via a tandem electrooxidation - reductive amination process in which the process auxiliaries can be easily recycled. The application of (immobilized) TEMPO in combinat…

The Ru(dppbsa)-catalyzed reductive amination of ketones with nitroarenes and nitriles using H2 as the environmentally benign hydrogen surrogate is developed in this study. Cross-experiments demonstrated that both reactions are initiated by the reduction of nitroarenes or nitriles to the corresponding amines, followed by condensation with ketones to give imines and thereafter hydrogenation. However, the route to the formation of an amino-ligated Ru complex during the reduction of nitroarenes or nitriles, followed by in situ nucleophilic C-N coupling, cannot be completely excluded. This newly de…

Reactive amine compounds are critical for a vast array of useful chemicals in society, yet a limited number of them are derived from renewable resources. This study developed an efficient route to obtain aminated building blocks from phenolic resources derived from nature, such as lignin and tannic acid, for enhancing their utility in applications such as epoxy resins, nylons, polyurethanes, and other polymeric materials. The reaction utilized a carbon storage compound, 2-oxazolidinone as a solvent and as a reagent circumventing the need of hazardous chemistry of conventional amination routes …

Reported herein, for the first time, is the selective ruthenium-catalyzed reductive alkoxylation and amination of phthalimides/succinimides. Notably, this novel methodology avoids hydrogenation of the aromatic ring and allows methoxylation of substituted imides with good to excellent selectivity for one of the carbonyl groups. The reported method opens the door to the development of new processes for the selective synthesis of various functionalized N-heterocyclic compounds. As an example, intramolecular reductive couplings to afford tricyclic compounds are presented for the first time.

Modern organic synthesis relies heavily on toxic, nonrenewable organic solvents that often generate large volumes of waste, particularly in pharmaceutical processes, where solvents account for over 80% of chemical waste. Conventional cross-coupling reactions frequently require activated electrophiles, produce unwanted byproducts, and involve complex purification procedures. Herein, we report a solid-supported ruthenium catalyst that enables dehydrative allylic amination in water, directly converting allyl alcohols and amines into allylic amines. The 2-mercaptopyridine-ruthenium complex immobil…

Highly enantioselective nucleophilic amination and kinetic resolution of α-bromo-acylpyrazoles with primary aromatic amine nucleophiles are promoted by a stoichiometric amount of Ag2CO3 and a catalytic amount of a chiral PyIPI ligand. The reaction proceeds via an SN2-type kinetic resolution process, which not only provides access to enantioenriched α-amino amide derivatives (up to 95% ee) but also allows for the recovery of the chiral α-bromo-acylpyrazole starting materials (up to 98% ee).

Metal-ammine (NH3) and metal-amide (NH2) species are common intermediates in ammonia oxidation, typically designed with weak N-H bonds to facilitate the sequential breakage of N-H bonds. Since the σ-bonding interaction between a metal center and an NH3 ligand weakens the N-H bonds, M-NH3 complexes with high N-H bond dissociation free energy (BDFE) are rare. However, we are particularly interested in M-NH3 complexes with high BDFEN-H for their potential in the conversion of C(sp3)-H bonds to C(sp3)-NH2 bonds. Herein, we report the synthesis and characterization of copper(II) and formal copper(I

Deuterated amines are key building blocks for drug synthesis and the identification of metabolites of new pharmaceuticals, which drives the search for general, efficient, and widely applicable methods for the selective synthesis of such compounds. Here, we describe a multifunctional phosphorus-doped carbon-supported Fe catalyst with highly dispersed isolated metal sites that allow for tandem reductive amination-deuteration sequences. The optimal phosphorus-modified Fe-based catalyst shows excellent performance in terms of both reactivity and regioselectivity for a wide range of deuterated anil…

Palladium-catalyzed para-C-H bond amination of 2-aryl chloromethylbenzenes is described for the first time. The reactions of 2-aryl chloromethylbenzenes with cyclic amines proceeded smoothly in the presence of Pd(acac)2, tri(2-furyl)phosphine, and NaH in tetrahydrofuran at 40 °C to provide para-C-H bond aminated products in satisfactory to high yields with acceptable regioselectivity in most cases. The electronic property of the substituents linked to the benzene rings did not significantly influence the reactivity of the 2-aryl chloromethylbenzene substrates and the reaction regioselectivity.

A metal-controlled, regioselective intermolecular amination of unsaturated N-acylpyrazoles with azodicarboxylates is described. Under zinc catalysis, the N-acylpyrazole substrates undergo amination at the α-position of the N-acylpyrazole moiety. Conversely, with silver as the catalyst, the reaction gave γ-amination products. Both catalytic protocols provided alternative, convenient, and simple strategies for efficiently and regioselectively accessing structurally unique C-N-bond containing compounds. The synthetic utility of this method was illustrated by a gram-scale experiment and subsequent…

We report a copper-catalyzed deaminative sp3 C-N cross-coupling of primary alkyl amines with N-heteroarenes, enabled by a deaminating reagent that generates alkyl radicals in situ. The method features general conditions, a broad substrate scope, excellent functional group tolerance, and successful late-stage functionalization of bioactive compounds. Mechanistic studies support a radical cross-coupling pathway. This approach significantly expands the scope of alkyl radical precursors for copper-mediated sp3 C-N bond formation.

Amine cyanoboranes and amine carboxyboranes (boron analogs of alpha-amino acids) were shown to inhibit inflammation. The analogs effectively blocked general inflammation, induced arthritis, and the writhing reflex associated with inflammation pain, while the inflammation associated with pleurisy was marginally inhibited. The boron analogs were shown in vitro to inhibit the release of lysosomal enzymes from liver and polymorphonuclear neutrophils. Furthermore, prostaglandin synthesis was blocked by these agents at a low concentration, i.e., 10(-6) M. Liver oxidative phosphorylation processes al

Herein, an engineered amine dehydrogenase is developed as an exemplary closed-boundary catalytic system for addressing the substrate scope-enantioselectivity dilemma. Asymmetric reductive amination with essentially quantitative enantioselectivity is demonstrated, with a K68S, N261L, A113G, T134G mutant of Bacillus species REN16 Glu/Leu/Phe/Val amino acid dehydrogenase for structurally diverse ketones. Reciprocal conformational adaptation, with complementary matching of conformational flexibility and sterics on both the enzyme and substrate sides, is proposed as the working mechanism.
