Cascaded modulator architecture for WDM applications.

Integration density, channel scalability, low switching energy and low insertion loss are the major prerequisites for on-chip WDM systems. A number of device geometries have already been demonstrated that fulfill these criteria, at least in part, but combining all of the requirements is still a difficult challenge. Here, we propose and demonstrate a novel architecture consisting of an array of photonic crystal modulators connected by a dielectric bus waveguide. The device architecture features very high scalability and the modulators operate with an AC energy consumption of less than 1fJ/bit.

Oral biofilm architecture on natural teeth.

Periodontitis and caries are infectious diseases of the oral cavity in which oral biofilms play a causative role. Moreover, oral biofilms are widely studied as model systems for bacterial adhesion, biofilm development, and biofilm resistance to antibiotics, due to their widespread presence and accessibility. Despite descriptions of initial plaque formation on the tooth surface, studies on mature plaque and plaque structure below the gum are limited to landmark studies from the 1970s, without appreciating the breadth of microbial diversity in the plaque. We used fluorescent in situ hybridizatio

Liver architecture, cell function, and disease.

The liver is an organ consisting of the largest reticulo-endothelial cell network in the body and playing an important role in host defense against invading microorganisms. The organ is comprised of parenchymal cells and many different types of non-parenchymal cells, all of which play a significant role. Even biliary epithelial cells are not only the target in autoimmune liver diseases but also have central role in orchestrating several immune cells involved in both innate and acquired immunity. Tissue damage caused by various agents results in inflammation, necrosis, fibrosis, and, eventually

Genetic architecture of intrinsic antibiotic susceptibility.

BACKGROUND: Antibiotic exposure rapidly selects for more resistant bacterial strains, and both a drug's chemical structure and a bacterium's cellular network affect the types of mutations acquired. METHODOLOGY/PRINCIPAL FINDINGS: To better characterize the genetic determinants of antibiotic susceptibility, we exposed a transposon-mutagenized library of Escherichia coli to each of 17 antibiotics that encompass a wide range of drug classes and mechanisms of action. Propagating the library for multiple generations with drug concentrations that moderately inhibited the growth of the isogenic pare

Sleep architecture, cocaine and visual learning.

BACKGROUND: Disturbances in sleep associated with chronic cocaine use may underlie abstinence-related cognitive dysfunction. We hypothesized that sleep-related cognitive function would be impaired in chronic cocaine users, and that this impairment would be associated with abstinence-related changes in sleep architecture. METHODS: Twelve chronic cocaine users completed a 23-day in-patient study that included randomized, placebo-controlled, cocaine self-administration sessions. We report polysomnographic measurement of rapid eye-movement (REM) sleep and slow-wave activity, and performance on a

First cuneiform osteotomy alters hindfoot architecture.

UNLABELLED: Three-dimensional concepts underlie various congenital foot deformities in the midfoot. We wondered whether the first cuneiform osteotomy, usually indicated for forefoot correction, had an effect on the hindfoot. We retrospectively studied 18 patients (31 feet) with metatarsus adductus and varus who had a biplane first cuneiform osteotomy between 1994 and 2001. No patients had associated malformations or neuromuscular disease. All had persistent pain and functional complaints for at least 2 years before surgery, and all had a minimum followup of 2 years. Group A (13 feet) with fore

Genome architecture, rearrangements and genomic disorders.

An increasing number of human diseases are recognized to result from recurrent DNA rearrangements involving unstable genomic regions. These are termed genomic disorders, in which the clinical phenotype is a consequence of abnormal dosage of gene(s) located within the rearranged genomic fragments. Both inter- and intrachromosomal rearrangements are facilitated by the presence of region-specific low-copy repeats (LCRs) and result from nonallelic homologous recombination (NAHR) between paralogous genomic segments. LCRs usually span approximately 10-400 kb of genomic DNA, share >or= 97% sequence i

Epigenetic architecture of the developing brain.

This chapter reviews the evolution of DNA methylation programs during brain development, focusing on how DNA methylation patterns define cell identity and regulate gene expression. It traces early hypotheses about tissue-specific DNA methylation and highlights limitations of bulk tissue analysis. Advances in genome-wide mapping, cell sorting, and single-cell sequencing have clarified the roles of CG and non-CG methylation in neuronal and glial differentiation. The chapter details how enhancer and gene body methylation correlate with transcriptional activity and how transcription factors, like

The molecular architecture of tunneling nanotubes.

Tunneling nanotubes (TNTs) are thin intercellular bridges that mediate the exchange of proteins, organelles, and nucleic acids between neighboring cells. They are enriched in tumor cells, implicated in chemotherapy resistance, induced in models of aggregation-based diseases, and their formation is stimulated by viruses that use TNTs to enhance infection. Despite their broad relevance and therapeutic potential, TNT morphology and function remain poorly understood, owing to the absence of clear morphological criteria and the limitations of light microscopy. Here, we establish two complementary s

Natural and Synthetic Metabolic Architectures.

Metabolic engineering often treats microbial metabolism as an inventory of metabolites, reactions, and the enzymes that catalyze them. This Perspective argues that function emerges from metabolic architecture, the connectivities that bind reactions into stable regimes shaped, among other factors, by space and time. The Japanese Metabolism movement motivates an architectural view in which the same metabolites could lead to rather different phenotypes when cells reconfigure metabolic routing subjected to environmental constraints. Natural examples, including the native cyclic glycolytic wiring o

Lipid Architecture in the Zika Virus.

Far from being passive barriers, viral membranes actively influence the mechanical properties and biological activity of viruses. Their lipid-protein composition forms a responsive interface that impacts how viruses assemble, remain stable, and interact with their surroundings. While the behavior of lipids and proteins in cellular membranes is well-described, their specific contributions within viral systems remain underexplored, largely due to nanoscale complexity and experimental limitations involved. Here, coarse-grained molecular dynamics simulations of Zika virus were analyzed to characte

Molecular architecture of Influenza A virions.

Influenza A viruses (IAV) are clinically important pathogens that cause seasonal epidemics and pandemics in humans. IAV produce pleomorphic, enveloped virions, which can range from a spherical or bacilliform morphology, the predominant form in the most commonly studied laboratory strains, to long filamentous virions which are characteristic of clinical and veterinary isolates. Understanding the structure and function of filamentous virions is crucial for clarifying their role in viral persistence and immune evasion, and for informing the development of therapeutics that target their entry and/

The genetic architecture of brainstem structures.

The brainstem houses numerous nuclei and tracts that serve vital functions. Genome-wide associations with brainstem substructure volumes have been explored in European individuals, yet other ancestries remain under-represented. Here, we conduct cross-ancestry genome-wide association meta-analyses in 103,098 individuals for brainstem and 78,062 individuals for eight substructure volumes, including 7094 Chinese Han individuals. We identify 713 locus-trait associations with brainstem and substructure volumes at P < 5.56 ×10-9, comprising 569 new associations. Two associations show different effec

Papillary architecture in the tayra tongue.

The tayra (Eira barbara) is a mammal belonging to the Mustelidae family that occurs in all Brazilian biomes. The present work aimed to describe the morphology of the tongue of these specimens highlighting their structures and particularities that will serve as a subsidy to elucidate the anatomy of the same and for comparative studies among other species of domestic and wild animals. Five adult male specimens of E. barbara were studied, which were fixed using 10% aqueous formaldehyde solution. The tongue was removed by opening the oral cavity and separating the maxillary/mandible bone complex.

Molecular architecture and function of the hemidesmosome.

Hemidesmosomes are multiprotein complexes that facilitate the stable adhesion of basal epithelial cells to the underlying basement membrane. The mechanical stability of hemidesmosomes relies on multiple interactions of a few protein components that form a membrane-embedded tightly-ordered complex. The core of this complex is provided by integrin α6β4 and P1a, an isoform of the cytoskeletal linker protein plectin that is specifically associated with hemidesmosomes. Integrin α6β4 binds to the extracellular matrix protein laminin-332, whereas P1a forms a bridge to the cytoplasmic keratin intermed

The genetic architecture of alopecia areata.

A major impetus to initiating the Human Genome Project was the belief that information encoded in the human genome would "accelerate progress in understanding disease pathogenesis and in developing new approaches to diagnosis, treatment, and prevention in many areas of medicine". Alopecia areata (AA) is a notable example of how understanding the genetic basis of a disease can have an impact on the care of patients in a relatively short time. Our first genome-wide association study in AA identified an initial set of common variants that increase risk of AA, some of which are shared with other a

The genetic architecture of multiple myeloma.

Based on the clinical features of myeloma and related malignancies of plasma cells, it has been possible to generate a model system of myeloma progression from a normal plasma cell through smouldering myeloma to myeloma and then plasma cell leukaemia. Using this model system we can study at which points the genetic alterations identified through whole-tumour molecular analyses function in the initiation and progression of myeloma. Further genetic complexity, such as intraclonal heterogeneity, and insights into the molecular evolution and intraclonal dynamics in this model system are crucial to

The architecture of cognitive control in schizophrenia.

Executive dysfunctions have long been considered a common feature of schizophrenia. However, due to their extreme heterogeneity, it is not clear whether these impairments take place at a particular level of executive functioning or non-specifically affect various aspects of behavioural control. To answer this question, we used an experimental paradigm based upon a multistage model of prefrontal executive function. This model postulates that cognitive control is organized in three hierarchically ordered control processes, operating with respect to the perceptual context (sensory and contextual

Neuronal architecture of the mosquito deutocerebrum.

Mosquito behavior is heavily dependent on olfactory and mechanosensory cues, which are detected by receptor neurons on the antenna and on the palps. Recent progress in mosquito sensory genomics highlights the need for an up-to-date understanding of the neural architecture of the mosquito brain. Here we present a detailed description of the neural structure of the primary target of the majority of these neurons, the deutocerebrum, in the African malaria (Anopheles gambiae) and yellow fever (Aedes aegypti) mosquitoes. Special focus is made on the olfactory system, the antennal lobe (AL), where w

Identification of architectural distortion requires a good practice of mammography. Prevalence is estimated at 6% of detected abnormalities in screening programs. Under this denomination are gathered focal architectural distortion with predictive positive value (PPV) of 10% and stellate images without central densification, which are more suspicious (PPV 50%). In order to establish a diagnosis, false images must be eliminated by other views. Minimal architectural distortion have to be investigated by other techniques (sonography, MRI percutaneous biopsy) in order to define the best strategy fo
