Potential Antifungal Effects of Calcium Oxide/Zinc Oxide Nanosuspension on Mycelial Growth of Neoscytalidium dimidiatum (Ascomycota, Botryosphaeriaceae) Associated With Pistachio Dieback.

The study aimed to synthesise and evaluate the efficacy of calcium oxide (CaO) and zinc oxide (ZnO) nano-suspensions, in conjunction with adjuvants, on the mycelial growth of Neoscytalidium dimidiatum, a notable fungal pathogen impacting pistachio trees in Iran. The study tackles a significant agricultural challenge by exploring eight different treatments, including both nano and pure forms of calcium oxide (CaO) and zinc oxide (ZnO), as well as polyethylene glycol, peracetic acid, and copper oxychloride. The results indicated a notable reduction in mycelial growth, particularly with the zinc

In vitro assessment of entomopathogenic potential of Clonostachys rosea (Ascomycota: Hypocreales) against Ixodes scapularis (Acari: Ixodidae) and Dermacentor variabilis (Acari: Ixodidae) ticks.

This study isolated and identified a Clonostachys rosea strain from field-collected tick samples in Nova Scotia, Canada, and investigated its entomopathogenic potential against Ixodes scapularis and Dermacentor variabilis. Ticks were collected from April to June 2023 through drag sampling and the fungus was isolated from dead or mycotic tick specimens. The isolate obtained from D. variabilis was subcultured from a single colony to obtain pure cultures. The fungus species was characterized through morphological and molecular analysis. The efficacy of C. rosea conidia was evaluated against lab-r

Ophiostoma breviusculum sp. nov. (Ophiostomatales, Ascomycota) is a new species in the Ophiostoma piceae complex associated with bark beetles infesting larch in Japan.

A new species of the Ophiostoma piceae-complex was isolated from bark beetles and the insect galleries of Larix kaempferi in Japan. This species was described as Ophiostoma breviusculum. The fungus was morphologically similar to O. piceae and O. quercus. However the average length of the perithecial necks and synnemata were shorter than for O. piceae and O. quercus. Synnemata morphological characteristics also differentiated O. breviusculum from the other species of the Ophiostoma piceae-complex isolated from conifers. Mating tests demonstrated that this fungus did not produce perithecia with

Mucorales spores induce a proinflammatory cytokine response in human mononuclear phagocytes and harbor no rodlet hydrophobins.

Mucormycoses are life-threatening infections in immunocompromised patients. This study characterizes the response of human mononuclear cells to different Mucorales and Ascomycota. PBMC, monocytes, and monocyte derived dendritic cells (moDCs) from healthy donors were stimulated with resting and germinated stages of Mucorales and Ascomycota. Cytokine response and expression of activation markers were studied. Both inactivated germ tubes and resting spores of Rhizopus arrhizus and other human pathogenic Mucorales species significantly stimulated mRNA synthesis and secretion of proinflammatory cyt…

Analysis of Cyp51 protein sequences shows 4 major Cyp51 gene family groups across fungi.

Azole drugs target fungal sterol biosynthesis and are used to treat millions of human fungal infections each year. Resistance to azole drugs has emerged in multiple fungal pathogens including Candida albicans, Cryptococcus neoformans, Histoplasma capsulatum, and Aspergillus fumigatus. The most well-studied resistance mechanism in A. fumigatus arises from missense mutations in the coding sequence combined with a tandem repeat in the promoter of cyp51A, which encodes a cytochrome P450 enzyme in the fungal sterol biosynthesis pathway. Filamentous members of Ascomycota such as A. fumigatus have ei…

Enhancement of TCE removal by a static magnetic field in a fungal biotrickling filter.

A fungal biotrickling filter (BTF) was employed to treat trichloroethylene (TCE) gas under different magnetic field intensities (MFIs). When … antly affected the fungal community and significantly improved the relative abundance of the phylum Ascomycota, achieving the highest abundance of Ascomycota at MFI 20.0. These results indicated that a lower MFI can efficiently improve TCE removal performance in a fungal BTF.

The gut microbiome but not the resistome is associated with urogenital schistosomiasis in preschool-aged children.

Helminth parasites have been shown to have systemic effects in the host. Using shotgun metagenomic sequencing, we characterise the gut microbiome and resistome of 113 Zimbabwean preschool-aged children (1-5 years). We test the hypothesis that infection with the human helminth parasite, Schistosoma haematobium, is associated with changes in gut microbial and antimicrobial resistance gene abundance/diversity. Here, we show that bacteria phyla Bacteroidetes, Firmicutes, Proteobacteria, and fungi phyla Ascomycota, Microsporidia, Zoopagomycota dominate the microbiome. The abundance of Proteobacteri…

Leucine-Rich Repeat Domains in the Class III Adenylyl Cyclase of Fungi: Some Features of the Sequences and Structures.

Fungal adenylyl cyclase (AC) converts adenosine triphosphate (ATP) to the second messenger 3',5'-cyclic AMP (cAMP). Class III ACs contain multiple domains, including the adenylyl cyclase domain and a leucine-rich repeat domain (LRR), through which multiple signals are integrated into the cAMP-Protein Kinase A (PKA) signaling response. This review focuses on the sequences and structural features of the LRR domains. To this end, we analyzed LRR domain sequences from ACs in the phyla Ascomycota, Mucoromycota, Basidiomycota, Blastocladiomycota, Zoopagomycota, and Chytridiomycota. Protein structure…

A broadly conserved fungal alcohol oxidase (AOX) facilitates fungal invasion of plants.

Alcohol oxidases (AOXs) are ecologically important enzymes that facilitate a number of plant-fungal interactions. Within Ascomycota they are primarily associated with methylotrophy, as a peroxisomal AOX catalysing the conversion of methanol to formaldehyde in methylotrophic yeast. In this study we demonstrate that AOX orthologues are phylogenetically conserved proteins that are common in the genomes of nonmethylotrophic, plant-associating fungi. Additionally, AOX orthologues are highly expressed during infection in a range of diverse pathosystems. To study the role of AOX in plant colonization…

Genetic and Functional Diversity of Nitrilases in Agaricomycotina.

Nitrilases participate in the nitrile metabolism in microbes and plants. They are widely used to produce carboxylic acids from nitriles. Nitrilases were described in bacteria, Ascomycota and plants. However, they remain unexplored in Basidiomycota. Yet more than 200 putative nitrilases are found in this division via GenBank. The majority of them occur in the subdivision Agaricomycotina. In this work, we analyzed their sequences and classified them into phylogenetic clades. Members of clade 1 (61 proteins) and 2 (25 proteins) are similar to plant nitrilases and nitrilases from Ascomycota, respe…

Plastic-Colonizing Fungi in the Eastern Mediterranean Sea: Community Structure and Physiological and Transcriptional Responses of Aspergillus to Pristine and Weathered PET.

Marine plastic debris provides persistent artificial substrates for microbial colonization, yet the fungal component of the plastisphere remains poorly characterized. Using ITS metabarcoding and culture-based isolation approaches, we characterized the early mycobiome associated with polyethylene (PE), polypropylene (PP), polystyrene (PS), and polyethylene terephthalate (PET) plastic pellets deployed in an Eastern Mediterranean Sea (EMS) marina. The sequence-based communities were taxonomically heterogeneous, with only a limited effect of polymer type. Ascomycota, Zoopagomycota, and Basidiomyco…

The selective antifungal activity of Drosophila melanogaster metchnikowin reflects the species-dependent inhibition of succinate-coenzyme Q reductase.

Insect-derived antifungal peptides have a significant economic potential, particularly for the engineering of pathogen-resistant crops. However, the nonspecific antifungal activity of such peptides could result in detrimental effects against beneficial fungi, whose interactions with plants promote growth or increase resistance against biotic and abiotic stress. The antifungal peptide metchnikowin (Mtk) from Drosophila melanogaster acts selectively against pathogenic Ascomycota, including Fusarium graminearum, without affecting Basidiomycota such as the beneficial symbiont Piriformospora indica…

High ascomycete diversity and novel fungal associations found in expanding Epipactis helleborine populations outside their native range.

Epipactis helleborine is an orchid species that has been introduced to North America and continues to expand its range. Numerous studies have investigated the mycorrhizal specificity of these orchids in their native range and found broad associations with Ascomycota, particularly Wilcoxina. Similar studies in North America are rare. We assessed the fungal diversity associated with populations of E. helleborine, and the surrounding soil, in mixed habitats on Vancouver Island (Canada) to make comparisons with findings from the species native range. After collecting 45 rhizomes and 36 soil sample…
