Increased Asialoglycoprotein Receptor 1 Level in Granulosa Cell as a Potential Biomarker for Polycystic Ovary Syndrome.

The study investigated whether asialoglycoprotein receptor 1 (Asgr1) can serve as a biomarker for polycystic ovary syndrome (PCOS). It is known that Asgr1 loss-of-function variants are associated with lower cholesterol levels and reduced cardiovascular disease risk, but its role as a PCOS biomarker was unclear. In this clinical translational study involving 60 women undergoing IVF at the Assisted Reproductive Center of Northwest Women's and Children's Hospital in Xi'an, China (including controls and PCOS patients), Asgr1 levels in granulosa cells were significantly higher in PCOS women compare…

Asialoglycoprotein receptor subunit Asgr1a loss results in attenuated cholesterol absorption in zebrafish fed a western diet.

One of the major pathways to clear glycoproteins from circulation is via the liver-specific asialoglycoprotein receptor. Loss of asialoglycoprotein receptor 1 (ASGR1), the major subunit of asialoglycoprotein receptor in humans, was found to correlate with lower levels of plasma apolipoprotein B-containing lipoproteins and a profound reduction in cardiovascular disease risk. However, the cell and molecular biology underlying this effect was unclear. Given that rodents carry their cholesterol largely in High Density Lipoprotein, we selected the zebrafish model to better understand the mechanism(…

Development and preclinical evaluation of an asialoglycoprotein receptor 1-targeted antibody-drug conjugate in hepatic inflammation.

Hepatic inflammation leads to progressive conditions such as metabolic dysfunction-associated steatohepatitis, cirrhosis, liver cancer, or failure. Due to the high prevalence rates of hepatitis-induced serious liver pathologies, effective interventions are needed. In this study, we developed an antibody-drug conjugate (ADC) from an in-house generated and well characterized monoclonal antibody targeting asialoglycoprotein receptor 1 (ASGPR1) in hepatocytes. The mAb showed strong affinity for ASGPR1 protein with a KD value of 4.2 ± 0.55 nM and high specificity for ASGPR1 in primary hepatocytes a…

Development of novel high-throughput biochemical competition assays to identify ligands of human asialoglycoprotein receptor 1.

Hepatocyte-specific Asialoglycoprotein receptor (ASGPR) and its native ligand N-acetylgalactosamine (GalNAc) have been actively exploited for targeted delivery of therapeutic and diagnostic agents to the liver. Identification of new potent ligands of ASGPR is of high interest to advance this field and expand to new applications in drug discovery. However, success of novel potent ASGPR ligand discovery has been limited due to the lack of robust high-throughput assays amenable to High-Throughput Screening (HTS). Here, we describe the design and development of two novel biochemical competition bi…

Autoantibodies to asialoglycoprotein receptor (ASGPR) measured by a novel ELISA--revival of a disease-activity marker in autoimmune hepatitis.

BACKGROUND: The liver-specific ASGPR is an autoantigen in autoimmune hepatitis (AIH) patients. Anti-ASGPR antibody correlates with disease activity, however, only in-house assays have been reported so far. METHODS: Rabbit ASGPR was purified by affinity chromatography on galactose-Sepharose and used for standardised detection of anti-ASGPR by ELISA. Anti-ASGPR IgG was measured in sera from 45 patients with AIH, PBC (n=43), alcoholic liver disease (n=13), HBV infection (n=35), HCV infection (n=53), and 118 blood donors. Anti-ASGPR was correlated with biochemical parameters of disease activity i

Asialoglycoprotein receptor 1 positively regulates PTGS2-induced inflammatory response in polycystic ovary syndrome via ERK1/2 pathway.

Asialoglycoprotein receptor 1 regulates the inflammatory response in granulosa cells associated with polycystic ovary syndrome. In the granulosa cells of individuals with polycystic ovary syndrome, elevated levels of asialoglycoprotein receptor 1 may activate the extracellular signal-regulated kinase 1/2 (ERK1/2) signaling pathway, which in turn enhances the expression of prostaglandin-endoperoxide synthase 2. Abstract This study evaluated whether asialoglycoprotein receptor 1 (ASGR1) can regulate the expression of prostaglandin-endoperoxide synthase 2 (PTGS2) in granulosa cells (GCs) in polyc…

Asialoglycoprotein receptor-1 mediated targeted drug delivery to liver cancer cells using stimuli responsive galactosylated electrospun nanofibers.

Synthesis of core-shell nanofiber (DOX@Gal-PVA/NSC) with galactose decorated poly(vinyl alcohol) as shell and doxorubicin loaded N-succinyl chitosan as core was achieved via coaxial electrospinning for site-specific targeting of liver cancer cells. Nanofibers of diameter 200 nm decorated with galactose were realised by in-situ photopolymerization during electrospinning. The galactose confers specific binding to asialoglycoprotein receptor-1 (ASGPR-1), overexpressed on hepatocellular carcinoma cells. The DOX loaded NSC core enable pH-responsive drug release, with higher rate of release to tumor…

Asialoglycoprotein receptor-mediated delivery of copper to hepatic tumors exerted inhibitory effect on tumor growth and progression.

Hepatocellular carcinoma (HCC) remains a major contributor to global cancer mortality and its rising incidence underscores the urgent need to explore novel therapeutic targets. Cancer is often characterized by dysregulated copper metabolism, which plays a crucial role in modulating tumor cell properties like cell proliferation, angiogenesis and metastasis. Therefore, exploiting their metabolic vulnerability using copper overload-based anticancer strategies has emerged as a novel approach. Despite the significant therapeutic potential of copper, its application in anticancer therapy has been li

Autoantibodies against the human asialoglycoprotein receptor: effects of therapy in autoimmune and virus-induced chronic active hepatitis.

The hepatic asialoglycoprotein receptor (ASGPR) was recently identified as a target antigen for both humoral and cellular immune response in inflammatory liver diseases. Thereby anti-ASGPR autoantibodies directed against human substrate were closely associated with autoimmune chronic active hepatitis. The present study compares the occurrence, titer and immunoglobulin classification of anti-human(h-)-ASGPR antibodies in 23 patients with newly diagnosed autoimmune chronic hepatitis before and after initiation of immunosuppressive therapy to 22 patients with autoimmune hepatitis in remission. Ad…

Efficient gene transfer to hepatoblastoma cells through asialoglycoprotein receptor and expression under the control of the cyclin A promoter.

Specific gene delivery into hepatoma cells by liposomes and specific gene expression under the control of the cyclin A promoter were examined in HepG2 cells, a hepatoblastoma cell line that overexpresses cyclin A. A plasmid carrying the luciferase gene under the cyclin A promoter sequence was condensed with poly-L-lysine and encapsulated into anionic asialofetuin-labeled liposomes (AF-liposomes), which were preferentially taken up by hepatocytes through the action of the asialoglycoprotein receptor (AgpR). AF-liposomes delivered plasmids to the hepatoma cells by receptor-mediated endocytosis t…

Radiosynthesis and biological evaluation of an fluorine-18 labeled galactose derivative [18F]FPGal for imaging the hepatic asialoglycoprotein receptor.

The asialoglycoprotein receptor (ASGPR) is abundantly expressed on the surface of hepatocytes where it recognizes and endocytoses glycoproteins with galactosyl and N-acetylgalactosamine groups. Given its hepatic distribution, the asialoglycoprotein receptor can be targeted by positron imaging agents to study liver function using PET imaging. In this study, the positron imaging agent [18F]FPGal was designed to specifically target hepatic asialoglycoprotein receptor and its effectiveness was assessed in in vitro and in vivo models. The radiosynthesis of [18F]FPGal required 50 min with total radi…

Asialofetuin-Coupled Lipid-Based nanosystems to target the Asialoglycoprotein receptor: Delivering genes to hepatocytes for the treatment of Fabry disease.

Exploiting the protein production capacity of hepatocytes for de novo expression of α-Galactosidase A (α-Gal A) by gene supplementation therapy represents one of the most promising strategies for the treatment of Fabry disease (FD). The asialoglycoprotein receptor (ASGPr) has proven to be one of the target receptors of choice for hepatocyte-directed nanomedicines, and natural glycoproteins such as asialofetuin (AF) can be used as specific ligands. Herein, we have developed AF-decorated solid lipid nanoparticles (SLNs), prepared by different techniques and cationic lipid compositions, for resto…

The rat hepatic lectin-1 subunit of the asialoglycoprotein receptor is upregulated by thyrotropin and downregulated by neoplastic transformation of thyroid cells.

We have recently shown that the rat hepatic lectin (RHL)-1 subunit of the asialoglycoprotein receptor (ASGPr) is expressed in the PC C13 differentiated thyroid cell line. To investigate in vivo the expression of RHL-1 and the ability of thyrotropin (TSH) to modulate its expression, reverse-transcriptase polymerase chain reaction (RT-PCR) and Western blot assays have been performed on thyroid extracts from rats treated with thyroxine (T4) or propylthiouracil (PTU), each of which modulates TSH levels. It is shown that RHL-1 expression is down-regulated by T4 (which decreases serum TSH) and upreg…

Hyperasialoglycoproteinemia in patients with chronic liver diseases and/or liver cell carcinoma. Asialoglycoprotein receptor in cirrhosis and liver cell carcinoma.

The pathogenesis of hyperasialoglycoproteinemia in cirrhosis and liver cell carcinoma was investigated by measuring the amount of asialoglycoprotein receptor in hepatic tissues from 30 patients with either cirrhosis or a malignancy. The asialoglycoprotein receptor activity in the cirrhotic liver was 770 +/- 423 U/g liver, i.e., 28% of the control value (2770 +/- 731 U/g liver). Receptor activity was negligible in tissues from patients with either liver cell carcinoma or metastatic tumor. The receptor levels in noncirrhotic and cirrhotic tissues in close proximity to liver cell carcinoma were 2…

Interaction between desialylated hepatitis B virus and asialoglycoprotein receptor on hepatocytes may be indispensable for viral binding and entry.

The cellular receptor for hepatitis B virus (HBV) infection has not yet been identified. The purpose of this study was to address the possibility of participation by desialylated HBV and the asialoglycoprotein receptor (ASGP-R) exclusively expressed on liver parenchymal cells, in infection. Assays for viral binding and entry were performed by culturing a hepatoblastoma cell line, HepG2, and HBV particles derived from the HBV carrier in the presence or absence of neuraminidase (NA). Viral binding and entry were clearly enhanced in the presence of NA, and the enhancement of the binding could be …

Differences in the abundance of variably spliced transcripts for the second asialoglycoprotein receptor polypeptide, H2, in normal and transformed human liver.

The human hepatic asialoglycoprotein receptor comprises two homologous polypeptides designated H1 and H2. Two distinct complementary DNA clones encoding these receptor subunits have been previously isolated from the human hepatoblastoma cell line HepG2. We discovered that multiple variants of H2 transcripts exist both in HepG2 cells and in the normal human liver that, at least in part, appear to be the result of alternative splicing events. We have found that (a) the complementary DNA clone for H2 previously isolated from HepG2 cells, characterized by a 57-nucleotide insertion within the 5' en…

Suppressive effect of ethanol on the expression of hepatic asialoglycoprotein receptors augmented by interleukin-1beta, interleukin-6, and tumor necrosis factor-alpha.

Blood levels of inflammatory-related cytokines, including interleukin (IL)-1beta, IL-6, and tumor necrosis factor (TNF)-alpha, are elevated in patients with alcoholic liver diseases. We investigated the effects of these cytokines and ethanol on the expression of hepatic asialoglycoprotein receptors (AGPRs) in a human hepatoblastoma cell line, HepG2. An [125I]-asialo-orosomucoid binding assay showed significant increases in surface AGPR numbers in HepG2 cells by treatment with IL-1beta, IL-6, and TNF-alpha, to levels which were approximately 130% of the values in untreated control cells.
