Equine pulmonary mycosis due to Aspergillus niger and Rhizopus stolonifer.

Invasive pulmonary mycosis caused by Aspergillus niger and Rhizopus stolonifer is reported in a 2-year-old horse, one of three that died after being housed in a disused, uncleaned stable. Lesions were characterized by thrombosis of the blood vessels with haemorrhage and tissue necrosis. Fungal hyphae were observed both in thrombosed vessels and in adjacent necrotic tissue. In culture media inoculated with lung samples and samples from the bedding hay, two types of colony were recorded and identified as A. niger and R. stolonifer. This study is the first description of equine pulmonary mucormyc…

Biotransformation of the Streptomyces scabies phytotoxin thaxtomin A by the fungus Aspergillus niger.

Of several hundred microorganisms randomly selected from the environment, only a fungal isolate identified as Aspergillus niger van Tiegham var. niger was found to transform the phytotoxin thaxtomin A to much less toxic metabolites. The rate and extent of transformation of thaxtomin A was tested under a variety of conditions, including different growth media, biomass concentrations, incubation periods, and shaker speeds. Under optimum conditions the fungus converted thaxtomin A into two major and five minor metabolites. The two major metabolites and three of the five minor metabolites were ful…

Aspergillus niger infection in an immunosuppressed patient confined solely to the brain.

A 68-year-old woman with a background of hypertension, stroke and rheumatoid arthritis presented to her local hospital after a 4-week history of gradual deterioration and increasing confusion with new onset right-sided weakness. Her initial CT scan revealed a rim enhancing mass lesion with surrounding oedema in the left parietal lobe for which she underwent CT stealth-guided biopsy. Microbiology culture of the 2 biopsy samples yielded Aspergillus niger and she was started on the antifungal agent voriconazole. MRI 2 weeks after the procedure also demonstrated radiological findings consistent wi…

Fatal invasive aspergillosis caused by Aspergillus niger after bilateral lung transplantation.

Aspergillus niger is usually considered to be a low virulence fungus, not commonly reported to cause invasive infections. Invasive pulmonary aspergillosis due to Aspergillus niger was diagnosed in a 43-year-old woman following bilateral lung transplantation. Intravenous voriconazole failed to control progression of the disease. Despite salvage therapy with a combination of voriconazole and caspofungin for 23 days, the patient developed massive hemoptysis leading to death. The authors report the clinical features and treatment of this case.

Aspergillus niger aortitis after aortic valve replacement diagnosed by transesophageal echocardiography.

Aspergillus aortitis following cardiac surgery has an important role among the cardiac infections as almost all affected cases result in death. Survival of the patient with Aspergillus aortitis is dependent on early initiation of aggressive medical and surgical treatment. Transesophageal echocardiography proved very useful in the diagnosis of this uncommon case of aortitis. In this paper, we present a patient with aortitis caused by Aspergillus niger that hasn't been reported previously diagnosed by transesophageal echocardiography following cardiac surgery.

Biochemical characterization of recombinant Aspergillus niger GZUF36 acetylcholinesterase for pesticide monitoring.

An acetylcholinesterase from Aspergillus niger GZUF36 (AnAChE) was expressed in Escherichia coli BL21(DE3) and comprehensively characterized with respect to its biochemical properties and suitability for pesticide analysis. The recombinant enzyme (rAnAChE) exhibited typical AChE substrate specificity, with optimal activity at pH 7.0 and 30-35 °C. rAnAChE tolerated hydrophobic solvents and methanol, supporting its use in practical assays. The enzyme was highly sensitive to methomyl, carbofuran, trichlorfon, fenitrothion, and carbaryl. For the 12 pesticides tested, the limits of detection of mos…

Light driven Aspergillus niger-ZnS nanobiohybrids for degradation of methyl orange.

Inorganic-microbial hybrid systems have potential to be sustainable, efficient and versatile chemical synthesis platforms by integrating the light-harvesting properties of semiconductors with microbial cells. Here, we demonstrate light-driven photocatalytic semiconducting Aspergillus niger cells-ZnS nanoparticles for enhanced removal of the dye methyl orange. Chemically synthesized ZnS nanoparticles exhibited a zinc blende pattern in X-ray diffraction, had a dimension of 20-90 nm with a band gap (Ebg) of 3.4 eV at 1.83 × 1018 photons/second. Biologically synthesized ZnS nanoparticles of 40-90 …

Calcium oxalate crystal deposition in necrotizing otomycosis caused by Aspergillus niger.

Numerous calcium oxalate crystals were present within fruiting heads of Aspergillus niger and among necrotic debris in a case of bilateral invasive otomycosis occurring in a diabetic female with end stage renal disease. This is the first report of in vivo calcium oxalate crystal deposition associated with Aspergillus niger at this anatomic site. The presence of localized oxalate crystals within necrotic tissue from the external auditory canal is presumptive of otomycosis caused by Aspergillus niger, and may serve as a diagnostic clue to the etiologic agent before histologic demonstration of hy…

Aspergillus niger, an Unusual Case With Fatal Massive Hemoptysis: A Case Report.

Aspergillus Niger is considered a low-virulence fungus. Primary and secondary immunocompromise represent the principal risk factors for developing this disease. There are only a few cases of invasive infection that have been documented, and only a few fatal cases of secondary invasive pulmonary aspergillosis as a cavitation by this agent have been published. We present the case of a 45-year-old man with immunocompromise who developed massive hemoptysis, which, despite treatment, led to his death. Pathological findings documented invasive pulmonary aspergillosis due to A. Niger and a co-infecti…

Thriving in salty environments: Aspergillus niger's halotolerance and BTEX biodegradation potential.

Benzene, toluene, ethylbenzene, and xylene (BTEX) can be found in marine and estuarine waters due to accidental spills of oil and derivatives, as well as in production water and effluents discharged from petrochemical plants. Addressing the bioremediation of these compounds in saline environments and effluents with elevated salinity levels is imperative. In this study, the halotolerance of Aspergillus niger was assessed by subjecting it to a stepwise increase in salinity, achieved through progressive addition of NaCl from 2 to 30‰ (v/v). The fungal strain exhibited optimal growth support up to…

Rutinosidase from Aspergillus niger: crystal structure and insight into the enzymatic activity.

Rutinosidases (α-l-rhamnosyl-β-d-glucosidases) catalyze the cleavage of the glycosidic bond between the aglycone and the disaccharide rutinose (α-l-rhamnopyranosyl-(1→6)-β-d-glucopyranose) of specific flavonoid glycosides such as rutin (quercetin 3-O-rutinoside). Microbial rutinosidases are part of the rutin catabolic pathway, enabling the microorganism to utilize rutin and related plant phenolic glycosides. Here, we report the first three-dimensional structure of a rutinosidase determined at 1.27-Å resolution. The rutinosidase from Aspergillus niger K2 (AnRut), a member of glycoside hydrolase

Clinical and immunological reactions to Aspergillus niger among workers at a biotechnology plant.

The workforce at a biotechnology plant producing citric acid by fermentation of molasses with a strain of Aspergillus niger was studied. A combination of a respiratory questionnaire and clinical assessment identified 18 subjects (4.9% of the workforce) with work related bronchospasm. In nine of these evidence of sensitisation to A niger was obtained by skin prick tests and radioallergosorbent test (RAST) using as an antigen an extract of the A niger culture fluid from the process. Of the 325 subjects without work related bronchospasm, only nine (2.7%) had a positive prick test. There were no s…

Endophytic Aspergillus niger and Aspergillus oryzae improve growth, antioxidant defense, and ion homeostasis in maize under salinity stress.

Salt stress limits maize productivity worldwide, and sustainable strategies are required for stress alleviation. The current study assessed the ability of endophytic fungi Aspergillus niger (MA2) and Aspergillus oryzae (MA5), isolated from Moringa oleifera seeds and identified by rDNA sequencing (GenBank: PV616998, PV616999), to mitigate salinity stress in maize. Both the isolates exhibited considerable production of indole-3-acetic acid, proline, phenolics, flavonoids, and sugars, which was enhanced under saline conditions. The application of 200 mM NaCl significantly (p ≤ 0.05) reduced shoot…
