MicroRNAs association with azoospermia, oligospermia, asthenozoospermia, and teratozoospermia: a systematic review.

Infertility is a major health problem across the world. One of the main reasons for male infertility are defects in sperm. Semen analysis is the most common test utilized to evaluate male fertility and since it suffers from multiple drawbacks, reproduction scientists have tried to find new molecular markers for detecting sperm defects. MicroRNAs (miRNAs) are small molecules in cells which take part in regulating gene expression. Various studies have confirmed miRNAs to have a role in defining multiple sperm characteristics, including sperm count, motility, and morphology. In this paper, we hav

Expression profiles and characteristics of human lncRNA in normal and asthenozoospermia sperm†.

Increasing studies have shown that specific mRNAs and miRNAs expressed in mature sperm may be related to sperm motility. However, the expression profiles and roles of lncRNAs in sperm remain unknown. In the present study, numerous lncRNAs were identified in human sperm, and some lncRNAs were expressed exclusively in sperm and testis. Compared with those in normal sperm, the lncRNA expression profiles in asthenozoospermia (AZS) sperm showed significant differences. Gene ontology and pathway analyses showed that function of differentially expressed lncRNA targets and mRNAs between AZS and normal

Association of DNAH11 gene polymorphisms with asthenozoospermia in Northeast Chinese patients.

Reduced or no progressive sperm motility in the fresh ejaculate defines asthenozoospermia as one of the major causes of male infertility. The axonemal heavy chain dynein type 11 (DNAH11) gene encodes for one of the axonemal dynein heavy chain (DHC) family members and participates in assembling respiratory cilia and sperm flagella. Given the high degree of conservation of DNAH11, mutations could give rise to primary ciliary dyskinesia (PCD) and asthenozoospermia. To date, few studies have reported on the association between variants in DNAH11 and asthenozoospermia. In the present study, 87 pati

Expression of TRPC5 is decreased in the sperm of patients with varicocele-associated asthenozoospermia.

The present study aimed to determine whether the expression of transient receptor potential channel 5 (TRPC5) protein is altered in spermatozoa of patients with varicocele-associated asthenozoospermia. TRPC5 expression in spermatozoa was determined by polymerase chain reaction and western blotting analyses, and indirect immunofluorescence was used for identification and immunolocalization of the TRPC5 channel in human sperm. Sperm motility and superoxide dismutase (SOD) activity were also determined with a computer-assisted semen analysis system and assay kit, respectively. Compared with level

Association between expression of TNF-α, P53 and HIF1α with asthenozoospermia.

Reduced sperm motility (asthenozoospermia) accounts for a significant percentage of male infertility and numerous factors have been suggested to explain this phenomenon among which hypoxic and inflammatory markers are the least studied. Therefore, the aim of this study was to assess the main molecular markers involved in hypoxia (P53 and HIF-1α) and inflammation (TNF-α) pathways in infertile men with asthenozoospermia. Expression of these markers were assessed by qRT-PCR, and analysis of data show that mean of hypoxia markers (P53, HIF-1α) and also TNF- α were significantly higher in infertile

Lactylated Proteomic Analysis Reveals Functional Implications of Lysine Lactylation In Asthenozoospermia.

The mechanism underlying asthenozoospermia in male infertility has been a prominent topic in reproductive medicine research. Human sperm function is modified by various protein post-translational modifications (PTMs). Among these, lactylation modification, a relatively novel PTM, has not been previously reported in the context of the male reproductive system. Comparative analyses between asthenozoospermic and normal sperm have revealed a significant down-regulation in the level of lysine lactylation (Kla) in proteins from asthenozoospermic sperm. Based on proteomic studies of protein Kla, 220

Human asthenozoospermia: Update on genetic causes, patient management, and clinical strategies.

BACKGROUND: In mammals, sperm fertilization potential relies on efficient progression within the female genital tract to reach and fertilize the oocyte. This fundamental property is supported by the flagellum, an evolutionarily conserved organelle, which contains dynein motor proteins that provide the mechanical force for sperm propulsion and motility. Primary motility of the sperm cells is acquired during their transit through the epididymis and hyperactivated motility is acquired throughout the journey in the female genital tract by a process called capacitation. These activation processes r

Homozygous variant in DRC3 (LRRC48) gene causes asthenozoospermia and male infertility.

Human infertility affects 10-15% of couples. Asthenozoospermia accounts for 18% of men with infertility and is a common male infertility phenotype. The nexin-dynein regulatory complex (N-DRC) is a large protein complex in the sperm flagellum that connects adjacent doublets of microtubules. Defects in the N-DRC can disrupt cilia/flagellum movement, resulting in primary ciliary dyskinesia and male infertility. Using whole-exome sequencing, we identified a pathological homozygous variant of the dynein regulatory complex subunit 3 (DRC3) gene, which expresses leucine-rich repeat-containing protein

Metabolomics analysis of human spermatozoa reveals impaired metabolic pathways in asthenozoospermia.

BACKGROUND: Infertility is a major health issue, affecting 15% of reproductive-age couples with male factors contributing to 50% of cases. Asthenozoospermia (AS), or low sperm motility, is a common cause of male infertility with complex aetiology, involving genetic and metabolic alterations, inflammation and oxidative stress. However, the molecular mechanisms behind low motility are unclear. In this study, we used a metabolomics approach to identify metabolic biomarkers and pathways involved in sperm motility. METHODS: We compared the metabolome and lipidome of spermatozoa of men with normozo

Association between chromosome 6p21 translocation and asthenozoospermia: A retrospective, observational study.

Asthenozoospermia (AZS) is the commonest cause of male-related infertility. The patients with AZS easily exhibit infertility, with their wives having spontaneous miscarriages or seeking assisted reproductive treatment. Reciprocal chromosomal translocation (RCT) is an important chromosome structural abnormality and has been reported to affect sperm motility. Genetic counseling for male RCT patients with AZS is still a challenge. This study reported 4 RCT carriers, which were 46,XY,t(1;6) (p36.1;p21), 46,XY,t (6;10) (p21;q11.2), 46,XY,t (6;11) (p21;p15), and 46,XY,t (6;17) (p21;q21), respectivel

Research progress on active ingredients of traditional Chinese medicine in the treatment of asthenozoospermia.

Asthenozoospermia, a main cause of male infertility, is characterized by diminished sperm motility and increased morphological abnormalities, presenting a major barrier to natural conception. While current treatments provide limited benefit, their effectiveness is often variable, invasive, and costly. In contrast, Traditional Chinese Medicine (TCM), guided by holistic principles and syndrome differentiation, offers a distinct therapeutic approach. Through multi-component, multi-target, and multi-pathway actions, TCM herbal compounds demonstrate considerable potential to improve sperm quality a

DCDC2C Protein Expression in Human Spermatozoa: A Potential Molecular Marker for Asthenozoospermia.

Background: Infertility is a global public health problem among males, with asthenozoospermia representing the most common cause. Since flagellar activity is vital for proper fertilization, identification of the mechanisms controlling this process is crucial. Microtubules are necessary for sperm functionality; however, their regulatory proteins in humans require further research. DCDC2C belongs to the doublecortin domain-containing family and has a role in microtubule stabilization; therefore, DCDC2C could affect the sperm flagellar structure. Objectives: The goal of the present study was to e

Sperm metabolomic signatures of asthenozoospermia and teratozoospermia in Chinese reproductive-age men.

Asthenozoospermia and teratozoospermia are common causes of male infertility. Despite their prevalence, the underlying metabolic mechanisms remain poorly understood. In this study, we conducted targeted metabolomic profiling of sperm samples from 131 Chinese reproductive-age men (48 normozoospermic controls, 40 asthenozoospermic patients, and 43 teratozoospermic patients) to identify distinct metabolic signatures associated with these conditions. We identified 47 significantly altered metabolites in asthenozoospermia compared to normozoospermia (18 downregulated and 29 upregulated). In teratoz

A novel homozygous mutation in the DNAAF3 gene leads to severe asthenozoospermia and teratospermia.

Primary ciliary dyskinesia (PCD) is an autosomal recessive genetic disorder characterized by ultrastructural defects in the cilia or flagella of cells, causing respiratory abnormalities, sinusitis, visceral transposition, and male infertility. DNAAF3 plays an important role in the assembly and transportation of axonemal dynein complexes in cilia or flagella and has been shown to be associated with PCD. To date, only two cases of PCD with infertility associated with DNAAF3 mutations have been reported, and no mouse models for this gene have been successfully constructed. This study was conducte

Reduction of SLC7A11 and GPX4 Contributing to Ferroptosis in Sperm from Asthenozoospermia Individuals.

Ferroptosis is a newly defined form of regulated cell death, which is involved in various pathophysiological conditions. However, the role of ferroptosis in male infertility remains unclear. In this study, 42 asthenozoospermic and 45 normozoospermic individuals participated. To investigate the ferroptosis level in the two groups, the levels of reactive oxygen species (ROS), malondialdehyde (MDA), and iron were measured, and mitochondrial membrane potential (MMP) was detected as an indicator of mitochondrial injuries. Compared with the normozoospermic group, ROS (p < 0.05), MDA (p < 0.001), and

MiR-4485-3p expression reduced in spermatozoa of men with idiopathic asthenozoospermia.

Asthenozoospermia (AZS), which characterised by reduced forward sperm motility, is a common cause of male infertility. Recently, mitochondrial dysfunction reported in AZS men came to attention for finding the molecular aetiology of AZS. Mitochondria-related microRNAs (miRNAs) are the most important regulators of mitochondrial function through post-transcriptionally modulation of gene expression. Therefore, this study aims to evaluate the expression of four recently reported mitochondrial-related miRNAs (miR-4485-3p/4484/4461 and 4463) in the sperm sample of asthenozoospermic men. RNA was extra

Proteomic analysis of seminal extracellular vesicle proteins involved in asthenozoospermia by iTRAQ.

Asthenozoospermia is a common cause of male infertility, but in most cases its etiology is unknown. The exocytic cell vesicles called seminal extracellular vesicles in the human seminal fluid have been reported to play a pivotal role in promoting the motility of spermatozoa, and functional disorder of seminal extracellular vesicles may cause male infertility. To determine whether abnormal seminal extracellular vesicles are involved in asthenozoospermia, the differential abundance proteins between normozoospermic (NSEV) and asthenozoospermic seminal extracellular vesicles (ASEV) samples were an
